Maleic acid and malonic acid reduced the pathogenicity of Sclerotinia sclerotiorum by inhibiting mycelial growth, sclerotia formation and virulence factors

Fei Yu-chen,Cheng Qin,Zhang Huan,Han Chuang,Wang Xu,Li Yan-feng,Li Shi-qian,Zhao Xiao-hu

Sclerotinia sclerotiorum is a necrotrophic plant pathogenic fungus with broad distribution and host range. Bioactive compounds derived from plant extracts have been proven to be effective in controlling S. sclerotiorum . In this study, the mycelial growth of S. sclerotiorum  was effectively inhibited by maleic acid, malonic acid, and their combination at a concentration of 2 mg/mL, with respective inhibition rates of 32.5%, 9.98%, and 67.6%. The treatment of detached leaves with the two acids resulted in a decrease in lesion diameters. Interestingly, maleic acid and malonic acid decreased the number of sclerotia while simultaneously increasing their weight. The two acids also disrupted the cell structure of sclerotia, leading to sheet-like electron-thin regions. On a molecular level, maleic acid reduced oxalic acid secretion, upregulated the expression of Ss-Odc2 and downregulated CWDE10 , Ss-Bi1 and Ss-Ggt1 . Differently, malonic acid downregulated CWDE2 and Ss-Odc1 . These findings verified that maleic acid and malonic acid could effectively inhibit S. sclerotiorum , providing promising evidence for the development of an environmentally friendly biocontrol agent.

Early identification of Aspergillus spp. in vitro based on HS-GC-IMS and electronic nose

Chen Huajiang,Luo Yanran,Xing Guiqing,Shi Yongqing,Gu Shuang,Wang Xiangyang

Aspergillus spp. are common dominant fungi in the moldy agricultural products. Different Aspergillus species exhibit high morphological and genetic sequence similarity, making identification challenging. The measurement of mycelial growth in these Aspergillus species often requires a relatively long time. Therefore, there is a need to develop a new rapid method for identifying Aspergillus species and monitoring their growth. The volatile compounds released by five Aspergillus species on culture media were detected by gas chromatography-mass spectrometer (GC–MS), electronic nose and headspace-gas chromatography-ion migration spectrometry (HS-GC-IMS), their characteristic volatile signals were captured, the models for identifying Aspergillus species were established, and prediction models for mycelial growth of five Aspergillus species were developed. The results showed that the LDA classification training set model constructed using the fusion of GC-IMS and electronic nose data achieved a 100% accuracy rate for identifying Aspergillus species in the training set and an 83.3% accuracy rate in the test set, significantly outperforming models based on HS-GC-IMS or electronic nose data alone. When the result combined with morphological analysis, a 100% accuracy rate in identifying Aspergillus species could be achieved. The partial least squares regression (PLSR) prediction models for mycelial growth in Aspergillus species, constructed using both fusion electronic nose data and HS-GC-IMS data, demonstrated high accuracy with prediction accuracies greater than 0.972 for all five species. The prediction model based solely on electronic nose data had lower accuracy, while models using HS-GC-IMS data alone or its combined with electronic nose data achieved good results. This fusion approach of HS-GC-IMS and electronic nose might have the potential to serve as a preliminary screening tool for fungi identification.

Inhibitory effects of glutaraldehyde on Geotrichum citri‐aurantii and its possible mechanism

Yang Y.,OuYang Q.,Li L.,Shao X.,Che J.,Tao N.

AimsThis study investigated the inhibitory effect of glutaraldehyde (GA) on sour rot in citrus fruit and the underlying antifungal mechanism on mycelial growth of the causative pathogen Geotrichum citri‐aurantii. Methods and ResultsGlutaraldehyde exhibited a strong inhibitory effect on G. citri‐aurantii, with a minimum inhibitory and fungicidal concentration (MFC) of 1·00 µl ml−1. In addition, in vivo application of GA (1 × MFC and 5 × MFC) reduced the disease incidence of sour rot in citrus fruit by 60 and 80% respectively. Scanning electron microscopy results revealed that the morphology of G. citri‐aurantii mycelia was greatly altered by GA treatment. Propidium iodide and Calcofluor White Staining revealed that the membrane permeability, rather than the cell wall integrity, of G. citri‐aurantii mycelia was severely disrupted after the addition of GA. Massive accumulation of malonaldehyde and reactive oxygen species as well as an increase in lipoxygenase activity were observed.ConclusionThese results indicate that GA may inhibit the mycelia growth of G. citri‐aurantii through a membrane damage mechanism induced by membrane peroxidation.Significance and Impact of the StudyGlutaraldehyde is expected to be a novel fungicide for controlling sour rot in citrus fruit.

Dipicolinic acid reduces Epicoccum sorghinum symptoms on maize and inhibits tenuazonic acid biosynthesis

Xin-Yu Ji,Bing-Yi Wang,Yi-Feng Zhang,Yu-Jing Zhang,Ya-Jie Lai,Yang Yang,Xiang-Chen Wang,Su-Yan Wang,Pedro Laborda,Xin-Chi Shi

BACKGROUND Epicoccum sorghinum is a pathogenic fungus that causes leaf spot in a wide range of plants, including maize, and synthesizes the mycotoxin tenuazonic acid (TEA), which is carcinogenic. Despite the relevant economic and yield losses caused by E. sorghinum worldwide, methods for the control of this pathogen are lacking. RESULTS In this work, the efficacy of Bacillus -produced dipicolinic acid (DPA) for control of E. sorghinum was evaluated using in vitro and in vivo assays, and compared with the efficacy of three commercial fungicides, including carbendazim, prochloraz, and thiram. DPA inhibited E. sorghinum mycelial growth, and conidia germination, and produced important alterations in E. sorghinum hyphae. Interestingly, 10 mM DPA reduced TEA biosynthesis by 86.6%. Although DPA rapidly degraded on maize leaves, 10 mM DPA showed higher preventive (67.4% lesion length inhibition) and inhibitory (89.5% lesion length inhibition) efficacies for the control of E. sorghinum on maize leaves compared to the commercial fungicides. CONCLUSIONS Collectively, this study presents the first method for the control of E. sorghinum on maize and demonstrates that DPA application is a suitable approach to inhibit E. sorghinum symptoms in plants and TEA biosynthesis. © 2024 Society of Chemical Industry.

Comparative physiological and transcriptome analysis provide insights into the inhibitory effect of osthole on Penicillium choerospondiatis

Duo Lai,Delin Wang,Xuehua Shao,Jian Qin,Qingli Zhuang,Hanhong Xu,Weiqiang Xiao

Blue mold induced by Penicillium choerospondiatis is a primary cause of growth and postharvest losses in the fruit of Phyllanthus emblica . There is an urgent need to explore novel and safe fungicides to control this disease. Here, we demonstrated osthole, a natural coumarin compound isolated from Cnidium monnieri , exhibited a strong inhibitory effect on mycelia growth, conidial germination rate and germ tube length of P. choerospondiatis , and effectively suppressed the blue mold development in postharvest fruit of P. emblica . The median effective concentration of osthole was 9.86 mg/L. Osthole treatment resulted in cellular structural disruption, reactive oxygen species (ROS) accumulation, and induced autophagic vacuoles containing cytoplasmic components in fungal cells. Transcriptome analysis revealed that osthole treatment led to the differentially expressed genes mainly enriched in the cell wall synthesis, TCA cycle, glycolysis/ gluconeogenesis , oxidative phosphorylation . Moreover, osthole treatment led to increase genes expression involved in peroxisome , autophagy and endocytosis . Particularly, the autophagy pathway related genes ( PcATG1 , PcATG3 , PcATG15 , PcATG27 , PcYPT7 and PcSEC18 ) were prominently up-regulated by osthole. Summarily, these results revealed the potential antifungal mechanism of osthole against P. choerospondiatis . Osthole has potentials to develop as a natural antifungal agent for controlling blue mold disease in postharvest fruits.

Fimbrin associated with Pmk1 to regulate the actin assembly during Magnaporthe oryzae hyphal growth and infection

Li Yuan-Bao,Shen Ningning,Deng Xianya,Liu Zixuan,Zhu Shuai,Liu Chengyu,Tang Dingzhong,Han Li-Bo

The dynamic assembly of the actin cytoskeleton is vital for Magnaporthe oryzae development and host infection. The actin-related protein MoFim1 is a key factor for organizing the M. oryza e actin cytoskeleton. Currently, how MoFim1 is regulated in M. oryzae to precisely rearrange the actin cytoskeleton is unclear. In this study, we found that MoFim1 associates with the M. oryzae mitogen-activated protein (MAP) kinase Pmk1 to regulate actin assembly. MoFim1 directly interacted with Pmk1, and the phosphorylation level of MoFim1 was decreased in Δ pmk1 , which led to a change in the subcellular distribution of MoFim1 in the hyphae of Δ pmk1. Moreover, the actin cytoskeleton was aberrantly organized at the hyphal tip in the Δ pmk1 , which was similar to what was observed in the Δ mofim1 during hyphal growth . Furthermore, phosphorylation analysis revealed that Pmk1 could phosphorylate MoFim1 at serine 94. Loss of phosphorylation of MoFim1 at serine 94 decreased actin bundling activity. Additionally, the expression of the site mutant of MoFim1 S94D (in which serine 94 was replaced with aspartate to mimic phosphorylation) in Δ pmk1 could reverse the defects in actin organization and hyphal growth in Δ pmk1 . It also partially rescues the formation of appressorium failure in Δ pmk1. Taken together, these findings suggest a regulatory mechanism in which Pmk1 phosphorylates MoFim1 to regulate the assembly of the actin cytoskeleton during hyphal development and pathogenesis.

Nobiletin enhances the antifungal activity of eugenol nanoemulsion against Penicillium italicum in both in vitro and in vivo settings

Yanchi Liu,Lintao Zhao,Hongyang Chen,Zimao Ye,Long Guo,Zhiqin Zhou

The study prepared and used eugenol nanoemulsion loaded with nobiletin as fungistat to study its antifungal activity and potential mechanism of Penicillium italicum (P. italicum) . The results showed that the minimum inhibitory concentration (MIC) of eugenol nanoemulsion loaded with nobiletin (EGN) was lower than that of pure eugenol nanoemulsion (EG), which were 160 μg/mL and 320 μg/mL, respectively. At the same time, the mycelial growth inhibition rate of EGN nanoemulsion (54.68 %) was also higher than that of EG nanoemulsion (9.92 %). This indicates that EGN nanoemulsion is more effective than EG nanoemulsion. Compared with EG nanoemulsion, the treatment of EGN nanoemulsion caused more serious damage to the cell structure of P. italicum . At the same time, in vitro inoculation experiments found that EGN nanoemulsion has better control and delay the growth and reproduction of P. italicum in citrus fruits. And the results reflected that EGN nanoemulsion may be considered as potential resouces of natural antiseptic to inhibit blue mold disease of citrus fruits, because it has good antifungal activity.

Biofortification of Pleurotus geesteranus via exogenous selenium species: Selenium distribution, speciation and its effect on mineral elements accumulation

Lingyang Yao,Huan Tian,Zhengyu Bao,Shuyun Xie,Tao Feng,Min Sun,Hui Ma,Lingyun Yao,Qiangjun Lang

In this study, four Se species including selenite [Se (VI)], selenate [Se (VI)], potassium,2-selenocyanatoacetate (PSeCA), and selenium ore powder (SeOP) were used for biofortification of Pleurotus geesteranus to obtain Se-enriched mushroom. Based on their effects on mycelium growth of P. geesteranus Jinxiu, [Se (VI)] in the concentration range of 100–1000 μg/kg and other three Se species in the range of 100–2000 μg/kg were adopted in spawn running and fruiting body growth stage, with their effects on production yield and elements accumulation evaluated. The results revealed that PSeCA treatment (200 μg/kg) had the highest fruiting body production (290.61 g/bag), followed by 2000 μg/kg of SeOP treatment (286.24 g/bag). SeMet revealed to be dominant species detected in mushroom, with [Se (VI)] exhibited the highest biological efficiency among the four Se species, and 2000 μg/kg of PSeCA treatment exhibited the highest Se content (10.25 mg/kg) in terminal phase of fruiting body. The results suggested that high concentration of PSeCA would be more favored for the Se biofortification. Moreover, Se treatment could greatly alter the mineral elements content of fruiting bodies, both synergistic and antagonistic relationships on accumulation of mineral elements were observed for each tested Se species.

Non-targeted metabonomics and transcriptomics revealed the mechanism of mulberry branch extracts promoting the growth of Sanghuangporus vaninii mycelium

Huo Jinxi,Sun Yuqing,Pan Meiliang,Ma Huanyan,Lin Tianbao,Lv Zhiqiang,Li Yougui,Zhong Shi

Sanghuangprous vaninii is a wood-inhabiting fungus, and its mycelium and fruiting body show excellent medicinal values. Mulberry is one of the major hosts of S. vaninii, however, the mechanism of mulberry affecting the growth of S. vaninii has not been reported. In the present study, a mulberry-inhabiting strain of S. vaninii was selected to explore the effects of mulberry branch extracts (MBE) on the growth of the strain. Results showed that MBE could significantly promote the growth of S. vaninii mycelium at the concentration of 0.2 g/l. After 16 days of liquid culture, the dry weight of mycelium in 0.2 g/l MBE medium was higher by three times compared with that in the control. The non-targeted metabonomic analysis of the culture medium at different culture times and concentrations was conducted to find the key components in MBE that promoted the growth of S. vaninii mycelium. Under the different concentrations of MBE culture for 10 and 16 days, 22 shared differential metabolites were identified. Next, in accordance with the peak value trend of these metabolites, HPLC–MS and liquid culture validation, four components derived from MBE (i.e., scopoletin, kynurenic acid, 3,5-dihydroxybenzoic acid and 2,4-dihydroxybenzoic acid) could significantly increase the growth rate of mycelium at the concentration of 2 mg/l. Transcriptomic and qRT-PCR analyzes showed that MBE could upregulate hydrolase-related genes, such as serine–glycine–asparaginate–histidine (SGNH) hydrolase, alpha-amylase, poly-beta-hydroxybutyrate (PHB) depolymerase, glycosyl hydrolase family 61, cerato-platanin protein and Fet3, which might enhance the nutrient absorption ability of S. vaninii. Importantly, MBE could significantly increase the content of harmine, androstenedione and vesamicol, which have been reported to possess various medicinal effects. Results suggested that MBE could be an excellent additive for liquid culture of S. vaninii mycelium, and these hydrolase-related genes also provided candidate genes for improving the nutrient absorption capacity of S. vaninii.

Combination of sodium dehydroacetate and sodium silicate reduces sour rot of citrus fruit

Lu Li,Xu Tang,Qiuli Ouyang,Nengguo Tao

Sodium dehydroacetate (SD) and sodium silicate (Na 2 SiO 3 ) are food preservatives with antifungal activities against many pathogens. In this paper, the antifungal activity of the combination treatment of SD and Na 2 SiO 3 against Geotrichum citri-aurantii , the postharvest pathogen of sour rot in citrus fruit, was tested. SD and Na 2 SiO 3 combination (1:4, w/w) showed a synergistic inhibitory effect compared to single compound on the mycelial growth of G. citri-aurantii , with the minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) being 1.8 and 3.6 g L −1 , respectively. The spores’ germination and the plasma membrane integrity of the pathogen could also be visibly influenced by the 1× MFC SD + Na 2 SiO 3 treatment. The application of various SD + Na 2 SiO 3 concentrations (1×, 5×, and 10× MFC) to citrus fruit inoculated with G. citri-aurantii significantly ( P < 0.05) decreased the incidence of sour rot during the entire storage period. After 8 d of storage, the disease incidences in SD + Na 2 SiO 3 (10×, 5× or 1× MFC)-treated fruit were only 20%, 30%, 65%, respectively, in contrast to 100% of the control fruit. Meanwhile, SD + Na 2 SiO 3 treatments maintained the fruit quality of citrus fruit. Results indicate that the combination of SD and Na 2 SiO 3 is a promising approach in controlling the postharvest decay of citrus fruit.

Antifungal activity of volatile compounds from Bacillus tequilensis XK29 against Botrytis cinerea causing gray mold on cherry tomatoes

Jianheng Guo,Yue Xu,Sunyan Liang,Zhengkun Zhou,Chunmei Zhang,Kui Li,Xue Peng,Sheng Qin,Ke Xing

Botrytis cinerea is a necrotrophic fungal pathogen that causes gray mold disease in more than 1400 plant species. The biocontrol effect of microbial volatile compounds on postharvest diseases has become a research hotspot. This study revealed the biocontrol effects of volatiles released by Bacillus tequilensis XK29 on B. cinerea in vitro and in vivo . The results showed that the strain XK29 volatiles inhibited the mycelial growth of B. cinerea . The inhibition effect of strain XK29 volatiles against B. cinerea was increased by optimizing the wheat seeds weight, suspension concentration and inoculation volume of strain XK29, and the pre-culture time. The volatiles of strain XK29 affected the mycelial development, the formation of conidia , and the metabolic activity of B. cinerea . Selected individual monomers and artificial mixtures showed strong antifungal activity against B. cinerea. The volatiles of strain XK29 reduced the lesion diameter and disease severity of cherry tomatoes after inoculation with B. cinerea. These results indicate that the fumigation with B. tequilensis XK29 volatiles is a promising approach to protect cherry tomatoes from gray mold disease. This is also the first study to prove the antifungal effect of B. tequilensis on B. cinerea and its biocontrol effect on postharvest cherry tomatoes.

Fungicidal activity of curcumol against Phytophthora capsici via inhibiting phosphatidylcholine biosynthesis and its systemic translocation in plants

Jingjing Yang,Hai Zhu,Wenhao Liang,Xingzeng Zhao,Min Yin,Xu Feng,Bi Wang

Phytophthora capsici is an infamously soil-borne pathogen that poses a serious threat to agricultural production. Curcumol is a natural plant-derived sesquiterpene lactone, whose antimicrobial effect against plant pathogens remains unclear. In this study, curcumol exhibited pronounced antifungal activity against a diverse range of plant pathogens, particularly against plant pathogenic oomycetes, which including P. capsici , Phytophthora infestans , Phytophthora parasitica , and Phytophthora sojae . The median effective concentration values of curcumol against P. capsici for spore germination and mycelial growth were 4.75 and 2.11 μg mL −1 , respectively. After treatment with curcumol, mycelia of P. capsici exhibited morphological and ultrastructual defects, which included swelling, hyperbranching, dissolution of plasma membrane, and loss of organelles. In addition, curcumol effectively inhibited the synthesis of phosphatidylcholine (PC), a primary component of cell membrane, by downregulating the expression levels of genes participated in PC synthesis such as Phospholipid N-methyltransferase and Cholinephosphotransferase . This inhibition decreased the accumulation of PC and phospholipids within the cell, thereby increasing the cell membrane permeability and damaging its integrity. In the in vivo antifungal tests, curcumol reduced the disease incidence of P. capsici on tomato leaves as well as pepper seedlings. The systemicity tests further validated the strong phloem and xylem mobility of curcumol in both upward and downward directions. Taken together, these results indicated that curcumol could effectively combat diseases caused by P. capsici and had the potential for development into a novel fungicide for P. capsici management.
