Peripheral nerve injuries represent a prevalent and severe category of damage resulting from traumatic incidents. Predominantly, the deficiency in nerve regeneration can be ascribed to enduring inflammatory reactions, hence imposing substantial clinical implications for patients. Fisetin, a flavonoid derived from plants, is naturally present in an array of vegetables and fruits, including strawberries, apples, onions, and cucumbers. It exhibits immunomodulatory properties through the reduction of inflammation and oxidative stress. In the present research, a nerve defect was addressed for the first time utilizing a scaffold primed for controlled fisetin release. In this regard, fisetin-loaded chitosan hydrogels were incorporated into the lumen of polycaprolactone (PCL) nerve guide conduits (NGCs). The hydrogel maintained a steady release of an appropriate fisetin dosage. The study outcomes indicated that the fisetin/chitosan/polycaprolactone (FIS/CS/PCL) NGCs amplified Schwann cell proliferation and neural expression, curtailed oxidative stress, alleviated inflammation, and improved functions, electrophysiological properties, and morphology. This pioneering scaffold has potential to contribute significantly to the field of neuroengineering. This article is protected by copyright. All rights reserved

DNA methylation plays a crucial role in eukaryotes’ growth and development, which is catalyzed by DNA methyltransferase (MTase). DNA MTase can transfer the methyl group to the C-5 position of cytosine in eukaryote from the methyl group donor of S -adenosylmethionine (SAM). Here, we developed a sensitive electrochemical method for assay of DNA methyltransferase (MTase) activity and screening of DNA MTase inhibitor without labeling processes. This strategy is mainly based on the change of reduction peak current of ferrocenecarboxylic acid (FcA), which is conjugated to the biosensor as electrochemical activity probe. The reduction current of FcA was enhanced with increasing DNA MTase concentration from 0.1 to 70 unit/mL, where the linear range of the concentration was from 0.1 to 1 unit/mL and 1 to 50 unit/mL with the detection limit of 0.045 unit/mL (S/N = 3). The investigation results demonstrated that the activity of DNA MTase depended on MTase concentration and incubation time. Inhibition experiment indicated that fisetin had good inhibition activity on DNA MTase in the presence of catechol- O -methyltransferase. This electrochemical method had the potential application in the screening of DNA MTase inhibitor, provided valuable information for anti-cancer drug research and also for cancer therapy.

DNA methylation is one of important epigenetics events, and responsible to transcription, genomic imprinting and cellular differentiation . Aberrant DNA methylation is always contacted with various diseases. Methyl binding domain (MBD) proteins can specifically bind to the methylated CpG dinucleotides . Conventional assay for DNA methylation normally need bisulfide treatment, methylated nucleotide labeling or PCR amplification. Here, we fabricated a novel electrochemical biosensor for detection of DNA methylation, assay of DNA methyltransferase (MTase) activity and screening of MTase inhibitor based on MBD protein and coomassie brilliant blue G250 (CBB-G250), where the electrochemical signal of CBB-G250 was used to monitor the methylation event. After the hybrids of DNA S1 and DNA S2 were treated with M. Sss I MTase in the presence of S -adenosylmethionine, the MBD proteins were specifically conjugated to the methylation site of CpG dinucleotides, and then, the MBD proteins were stained with CBB-G250. The electrochemical signal of CBB-G250 increased linearly with increasing M. Sss I MTase concentration in the range from 0.1 to 40 unit/mL. Furthermore, the inhibition investigation demonstrates that fisetin and chlorogenic acid can inhibit the M. Sss I MTase activity with the IC 50 value of 153.12 and 137.07 μM, respectively. Therefore, we think that this study may provide a sensitive platform for screening of DNA MTase inhibitors.

In this article, we presented an electrochemical method for detection of DNA methylation and assay of DNA methyltransferase (MTase) activity using methylene blue (MB) as electrochemical indicator. After the DNA hybrid was methylated by T. aq I MTase, it could not be cleaved by Hinc II endonuclease. On the contrary, the double DNA without methylation could be cleaved, which would decrease the amount of intercalated MB. Thus, the DNA methylation status and MTase activity could be determinated based on the voltammetric signal change of MB. The methylation site could be found based on this method. The electrochemical signal of MB increased linearly with increasing T. aq I MTase concentration from 0.1 to 100 U/mL. Moreover, the inhibition investigation demonstrated that fisetin could inhibit the T. aq I MTase activity with the IC 50 value of 280 μM. Therefore, the screening of the inhibitors of MTase could be accomplished using the novel method.

Human skin fibroblast collagenase also known as Matrix Metalloproteinase-1 (MMP-1) is a key enzyme in remodeling and degradation of extracellular matrix, and the inhibitors of human MMP-1 are effective drug candidates for the treatment of cancer. In this study, we report an improved method for high-level expression of soluble human MMP-1 catalytic domain (cd-MMP-1) in E.coli. The enzymatic activity is found maximum at pH 7.5 and temperature 40°C with a Km value of 13.02 µM. Effects of 17 structure-related flavonoids on MMP-1 activity are evaluated using a fluorescent assay, 6 inhibitors are identified with IC50 < 10 µM. Fisetin is the most active agent with an IC50 value of 1.35 µM and is identified as a mixed type inhibitor. Our improved soluble cd-MMP-1 expression method provides a basis for inhibitors identification and may be beneficial to discover novel anti-cancer agent targeting human MMP-1.

Objective Parkinson's disease (PD) is characterized by the loss of dopaminergic neurons in substantia nigra (SN). Our previous study demonstrated kukoamine A (KuA) to exhibit strong neuroprotective effects through antioxidative stress, and autophagy in MPTP/MPP + -induced PD models in vivo and in vitro . It is necessary to evaluate the efficacy of the anti-PD effects under various models. Methods In the present study, total chemical synthesis was used to obtain KuA, which performed low content in Lycii Cortex . Then, 6-OHDA-induced PD model of PC12 cells was used to investigate the effects of KuA on PD. Results Our results demonstrated that KuA ameliorated cell loss and mitochondrial membrane potential (MMP) loss, and inhibited Bax/Bcl-2 ratio increase that were induced by 6-OHDA. Iron accumulation in SN is thought to participate in neuronal death in PD, which subsequently resulted in oxidative stress and overexpression of α-synuclein caused by iron metabolism protein disorder. In our study, KuA could chelate cellular iron content and decrease iron influx. Moreover, KuA could upregulate the expression of ferroportin1 and Hephaestin, downregulate the expression of DMT1, TfR, and Ferritin to maintain cellular iron homeostasis avoiding neuronal death from cellular iron deposition. Moreover, KuA could decrease the expression of a-synuclein in cells. All the results indicated that KuA protected against neurotoxin-induced PD due to the apoptosis inhibition and iron homeostasis maintaining. Conclusion KuA treatment might represent a neuroprotective treatment for PD.

A simple and efficient matrix solid-phase dispersion via high-performance liquid chromatography coupled with a photodiode array detector was developed to analyze the following flavonoids of Rhus verniciflua Stokes: fisetin, fustin, butein, sulfuretin, garbanzol, and quercetin. The optimum conditions for the procedure was the use of Zeolite Socony Mobil-twenty-two molecular sieves as the adsorbent, sample:adsorbent ratio of 2:5, grinding for 3 min, and use of 8 mL of 70% methanol:water as the elution solvent. The method was validated for linearity, precision, reproducibility, limit of detection, and limit of quantification. The method exhibited excellent linearity for all six flavonoids. The intra- and interday precisions over a range of concentrations were below 3.0% and limits of quantification for the six flavonoids were 0.16 and 0.50 μg/mL. Compared with other published methods, the proposed method was more effective, rapid, and required less reagents. Therefore, the combination of matrix solid-phase dispersion and high-performance liquid chromatography coupled with photodiode array detector showed excellent reproducibility and simplicity and could be suitable for the extraction and quantification of multiple flavonoids in R. verniciflua Stokes samples.
