Biodegradation, Biosorption of Phenanthrene and Its Trans-Membrane Transport by Massilia sp. WF1 and Phanerochaete chrysosporium

Gu Haiping,Lou Jun,Wang Haizhen,Yang Yu,Wu Laosheng,Wu Jianjun,Xu Jianming

Reducing phenanthrene (PHE) in the environment is critical to ecosystem and human health. Biodegradation, biosorption, and the trans-membrane transport mechanism of PHE by a novel strain, Massilia sp. WF1, and an extensively researched model fungus, Phanerochaete chrysosporium were investigated in aqueous solutions. Results showed that the PHE residual concentration decreased with incubation time and the data fitted well to a first-order kinetic equation, and the t1/2 of PHE degradation by WF1, spores, and mycelial pellets of P. chrysosporium were about 2 h, 87 days, and 87 days, respectively. The biosorbed PHE was higher in P. Chrysosporium than that in WF1, and it increased after microorganisms were inactivated and inhibited, especially in mycelial pellets. The detected intracellular auto-fluorescence of PHE by two-photon excitation microscopy also proved that PHE indeed entered into the cells. Based on regression, the intracellular (Kdin) and extracellular (Kdout) dissipation rate constants of PHE by WF1 were higher than those by spores and mycelial pellets. In addition, the transport rate constant of PHE from outside solution into cells (KinS/Vout) for WF1 were higher than the efflux rate constant of PHE from cells to outside solution (KoutS/Vin), while the opposite phenomena were observed for spores and mycelial pellets. The amount of PHE that transported from outside solution into cells was attributed to the rapid degradation and active PHE efflux in the cells of WF1 and P. Chrysosporium, respectively. Besides, the results under the inhibition treatments of 4°C, and the presence of sodium azide, colchicine, and cytochalasin B demonstrated that a passive trans-membrane transport mechanism was involved in PHE entering into the cells of WF1 and P. Chrysosporium.

Biodegradation and metabolic pathway of phenanthrene by a newly isolated bacterium Gordonia sp. SCSIO19801

Zhimao Mai,Lin Wang,Qiqi Li,Yingting Sun,Si Zhang

The bacterium Gordonia sp. SCSIO19801, which could effectively utilize phenanthrene as the sole carbon source, was isolated from the seawater of the South China Sea. Its biodegradation characteristics, whole genome sequence, and biodegradation pathway were investigated. The phenanthrene biodegradation process of Gordonia sp. SCSIO19801 was estimated to be a first-order kinetic model with a k value of 0.26/day. Based on the identification of metabolites, utilization of probable intermediates, and genomics analysis of related genes, the degradation of phenanthrene by Gordonia sp. SCSIO19801 was proposed to occur via the salicylate metabolic pathway. This is the first report of a phenanthrene degradation pathway in Gordonia species. In addition, the Gordonia sp. SCSIO19801 could use other aromatic compounds as the sole source of carbon and energy. These characteristics indicate that Gordonia sp. SCSIO19801 can be utilized for developing effective methods for the biodegradation of petroleum hydrocarbons in marine environments.

Biodegradation of Phenanthrene by Mycobacterium sp. TJFP1: Genetic Basis and Environmental Validation

Shuyun Li,Jiazhen Liu,Ping Fang

The development of efficient bioremediation technologies for polycyclic aromatic hydrocarbons contamination is a hot research topic in the environmental field. In this study, we found that theMycobacteriumsp., TJFP1, has the function of degrading low molecular weight PAHs, and further investigated its degradation characteristics using the PAH model compound phenanthrene as a target pollutant. The optimal growth and degradation conditions were determined by single-factor experiments to be 37 °C, pH 9.0, and an initial concentration of 100 mg/L phenanthrene. Under this condition, the degradation efficiency of phenanthrene reached 100% after 106 h of incubation, and the average degradation rate could reach 24.48 mg/L/day. Combined with whole genome sequencing analysis, it was revealed that its genome carries a more complete phenanthrene degradation pathway, including functional gene clusters related to the metabolism of PAHs, such as phd and nid. Meanwhile, intermediates such as phthalic acid were detected; it was determined that TJFP1 metabolizes phenanthrene via the phthalic acid pathway. Simulated contaminated soil experiments were also conducted, and the results showed that the removal rate of phenanthrene from the soil after 20 days of inoculation with the bacterial strain was about 3.7 times higher than that of the control group (natural remediation). At the same time from the soil physical and chemical properties and soil microbial community structure of two levels to explore the changes in different means of remediation, indicating that it can be successfully colonized in the soil, and as a dominant group of bacteria to play the function of remediation, verifying the environmental remediation function of the strains, for the actual inter-soil remediation to provide theoretical evidence. This study provides efficient strain resources for the bioremediation of PAH contamination.

Heterogeneous Fenton-like degradation of phenanthrene catalyzed by schwertmannite biosynthesized using Acidithiobacillus ferrooxidans

Xiaoqing Meng,Su Yan,Wenzhu Wu,Guanyu Zheng,Lixiang Zhou

Heterogeneous Fenton-like degradation of phenanthrene in aqueous solution was investigated using schwertmannite biosynthesized by Acidithiobacillus ferrooxidans LX5 as a catalyst. The effects of different reaction parameters including catalyst loading, H2O2 concentration, initial solution pH and inorganic anions on the Fenton-like degradation of phenanthrene were studied. Results showed that the biosynthesized schwertmannite had an effective catalytic ability on phenanthrene degradation. The degradation efficiency of phenanthrene was 99.0% within 3–5 h reaction under conditions of H2O2 200 mg L−1, schwertmannite 1 g L−1, phenanthrene 1 mg L−1 and pH 3.0–4.5. The degradation was mainly via a surface mechanism, in which phenanthrene was readily adsorbed on the surface of schwertmannite and then oxidized by ˙OH produced from H2O2 decomposition. The XPS results of schwertmannite before and after Fenton-like degradation of phenanthrene revealed the change of Fe2+/Fe3+ species on the surface of schwertmannite. Moreover, phthalates, octadecanoic acid and 9,10-phenanthraquinone were identified by GC-MS analyses as the main intermediate compounds during phenanthrene degradation, and all the intermediates were finally mineralized. The repeated use of biosynthesized schwertmannite for phenanthrene degradation illustrated its stability and reusability as a Fenton-like catalyst. Therefore, schwertmannite biosynthesized using A. ferrooxidans is an excellent catalyst for the degradation of phenanthrene in heterogeneous Fenton-like reactions.

High phenanthrene degrading efficiency by different microbial compositions construction

Zhou Guoyan,Qiao Hongtao,Liu Yandong,Yu Xiongsheng,Niu Xiang

Microbial remediation has become the most promising technical means for the remediation of polycyclic aromatic hydrocarbons (PAHs) non-point source contaminated soil due to its low cost of treatment, complete degradation of pollutants, and in-situ remediation. In this study, in order to demonstrate the phenanthrene degrading microbial diversity, phenanthrene was chosen as the representative of PAHs and strains capable of degrading phenanthrene were isolated and screened from the sedimentation sludge and the bottom sludge of oil tank trucks, and high throughput sequencing was used to check the dominant strains with a good degrading effect on phenanthrene. Results showed even more than 50% of phenanthrene was degraded in all samples, the composition of PAH-degrading bacteria was diverse, and different environments constructed different functional microbial groups, which resulted in the microbial adapting to the diversity of the environment. Finally, a series of bacterial species with phenanthrene-degrading functions such as Achromobacter, Pseudomonas, Pseudochelatococcus, Bosea was enriched after nine transferring process. Overall, our study offers value information for the enrichment of functional degrading microbes of phenanthrene or other pollutants that more concern should be paid in not only the degradation rate, but also the diversity variation of microbial community composition.

Phenanthrene Elimination from Soil through the Activation of Peroxymonosulfate by Biogenically Derived Manganese Oxide

Jiang Yuanyuan,Zhang Hongrui,Wu Yanling,Wang Hui,Miran Waheed,Liu Jun,Yang Fei,Long Xizi

Peroxymonosulfate (PMS) advanced oxidation is gaining recognition as a promising method for tackling persistent soil pollutants. However, developing an efficient PMS activator remains a formidable task. This study harnessed Shewanella oneidensis MR-1, a model dissimilatory metal-reducing bacterium (DMRB), to synthesize Mn 2 O 3 nanoparticles by oxidizing Mn(II). These nanoparticles were employed to activate PMS for phenanthrene degradation in soil. Remarkably, biogenic Mn 2 O 3 outperformed chemically synthesized Mn 2 O 3 , removing 77.4% of phenanthrene compared to 55.7%. This superior performance is attributed to biogenic Mn 2 O 3 's faster electron transfer rate and higher Mn(III) ratio, facilitating electron donation to PMS. Additionally, we assessed the feasibility of PMS advanced oxidation for soil remediation by examining microbial community diversity. Given manganese's prevalence in natural soil and groundwater, in-situ biogenic Mn 2 O 3 synthesis emerges as an innovative soil remediation strategy. Graphical Abstract

Lab-scale transport and activation of peroxydisulfate for phenanthrene degradation in soil: A comprehensive assessment of the remediation process, soil environment and microbial diversity

Xiupeng Yue,Yaping Zhang,Yongping Shan,Kai Shen,Wentao Jiao

Electrokinetic transport followed by electrical resistance heating activation of peroxydisulfate is a novel in situ soil remediation method. However, the strategy of electrokinetic transport coupled with electrical resistance heating and the comprehensive evaluation of restored soil need to be further explored. In this study, a lab-scale simulation device for in situ electrokinetic transport coupled with electrical resistance heating activation of peroxydisulfate was constructed to monitor the transport and transfer of peroxydisulfate, target pollutants, and process parameters, and the physicochemical properties and bacterial community of treated soil were evaluated. The results showed that adding 10 wt% peroxydisulfate to both the anode and cathode resulted in the optimized transfer rate and cumulative concentration of peroxydisulfate under electrokinetics. After 8 h, the cumulative concentration of peroxydisulfate reached 66.15– 166.29 mmol L −1 , which was attributed to the migration of a large amount of S 2 O 8 2− from the cathode to the soil under electromigration . Additionally, the anodic interfacial electric potential was improved, which was more conducive to electroosmotic transport of peroxydisulfate from the anode chamber. By alternating electrokinetic transport and electrical resistance heating activation of peroxydisulfate for two cycles, the phenanthrene degradation efficiency in four evenly distributed wells between electrodes reached 75.4 %, 87.6 %, 92.3 %, and 94.4 %. With slight variations in soil morphology and structure, the electrokinetic transport coupled with electrical resistance heating activation of peroxydisulfate elevated the soil fertility index. The abundance and diversity of bacterial communities in treated soil recovered to above the original soil level after 15 days. Our findings may support the application of electrokinetic transport coupled with electrical resistance heating activation of peroxydisulfate as a promising green ecological technology for the in situ remediation of organic-contaminated soil.

Synergistic division of labor in a bacterial consortium for enhanced phenanthrene mineralization under cadmium stress: mechanisms of degradation-detoxification coordination

Tianzuo Cheng,Pengyu Zhou,Mingle Zhang,Tianyin Huang,Bingdang Wu,Jinlong Zhuang,Bin Wang,Xiaoyi Xu

The remediation of co-contamination by polycyclic aromatic hydrocarbons (PAHs) and heavy metals poses a significant challenge. Although microbial consortia present a promising approach, their synergistic mechanisms under stress conditions are not fully understood. To address this gap, we constructed a functionally specialized bacterial consortium (KZ) by assembling Klebsiella sp. CW-D3T and Arthrobacter sp. SZ-3, which synergistically enhanced phenanthrene (PHE) degradation and mineralization under cadmium stress (25 mg/L Cd 2+ ), outperforming monocultures by 1.2–1.9-fold. Through biomass-normalized enzyme activity assays, we uncovered a structured division of labor: SZ-3 exhibited superior upstream catalytic activity (50 % higher 2H1N conversion), while CW-D3T dominated downstream mineralization (>80 % contribution). Mechanistic investigations via metagenomics revealed that CW-D3T utilized high-expression efflux pumps (ZntA/zinT) and antioxidant genes ( yhcN ) to mitigate cadmium toxicity, whereas SZ-3 employed the frnE -mediated oxidative stress response and limited Cd 2+ uptake via mntH . This study elucidates a synergistic mechanism for concurrent PAH degradation and heavy metal detoxification, offering a novel bioresource for remediating co-contaminated environments.

An S-scheme TiO2/g-C3N4 nanocomposite effectively degrades phenanthrene under visible light

Ting-Ting Li,Meng-Meng Xie,Hong-Yang Wang,Jun-Sheng Wang,Jia-Horng Lin,Ching-Wen Lou

It is a challenge to effectively degrade phenanthrene (PHE) pollutants, which are widely present in aquatic environments, in order to reduce harm to humans and ecosystems. In this study, an S-scheme TiO 2 /g-C 3 N 4 photocatalytic system is constructed using 0D TiO 2 nanospheres and 2D g-C 3 N 4 nanosheets for the removal of PHE from water under sunlight irradiation. The effects of irradiation time, quality ratio of TiO 2 to g-C 3 N 4 , and cycle time on the performance of the TiO 2 /CN photocatalyst are investigated. The experimental results show that the ratio of TiO 2 to g-C 3 N 4 significantly affects the photocatalytic activity of the photocatalyst. Under the optimal ratio of TiO 2 to g-C 3 N 4 (50 % TiO 2 /CN), the apparent reaction rate constant for phenanthrene reached 0.00796 min −1 , which is 11.5 times higher than that of pure TiO 2 (0.00069 min −1 ). The tests of optical performance and photoelectrochemical properties further confirmed that the construction of the TiO 2 /g-C 3 N 4 S-type photocatalyst successfully enhanced the spatial separation efficiency of photogenerated carriers and ensured a continuous supply of energy during the redox reaction process. Converting highly toxic phenanthrene into a non-toxic green degradation product provides an practical strategy for the safe treatment of PHE in aqueous environments through the use of visible-light-driven heterojunction photocatalysts. Additionally, the data collected on phenanthrene degradation in this study will provide valuable references for developing degradation methods for other PAHs, such as naphthalene, anthracene, and pyrene.

Enhanced Catalytic Efficiency of CeO2/BC@Fe3O4 Magnetic Catalyst in Activating Peroxymonosulfate for Effective Degradation of Polycyclic Aromatic Hydrocarbons in Water

Runnan Zhang,Yanyan Liu,Wanqi Ou,Yuying Ye,Kai Xu

Polycyclic Aromatic Hydrocarbons (PAHs) are persistent organic pollutants that are widely present in aquatic environments and pose a significant threat to the ecological environment and human health. This study introduces a novel magnetic biochar-supported metal oxide catalyst (CeO 2 /BC@Fe 3 O 4 , CBF) as a peroxymonosulfate (PMS) activator in phenanthrene (PHE) degradation in aqueous environments. The degradation performance of the CeO 2 /BC@Fe 3 O 4 composite material was investigated through degradation experiments. The results showed that under the optimal reaction conditions, the degradation efficiency of 20 mg/L PHE solution within 60 minutes could reach 99.55% and maintained stability within a wide range of pH values. Radical quenching and EPR studies confirmed the involvement of SO 4 ·- , ·OH and 1 O 2 in the removal of PHE during the degradation process. The possible degradation pathways were proposed based on the identification of intermediates. The catalyst demonstrated excellent reusability and was effective in real water samples and for other organic pollutants, highlighting its potential for practical environmental applications.

Autochthonous bioaugmentation accelerates phenanthrene degradation in acclimated soil

Haiping Gu,Jie Yan,Yuhao Liu,Xuewei Yu,Yan Feng,Xuanyi Yang,Su Shiung Lam,Mu. Naushad,Cheng Li,Christian Sonne

Bioaugmentation helps to obtain a microbiome capable of remediating polycyclic aromatic hydrocarbons (PAHs). In this study, acclimation of microorganisms to soil supplemented with phenanthrene (PHE) led to enrichment with PAH-degraders, including those in Actinobacteriota and in the genera Streptomyces , Rhodococcus , Nocardioides , Sphingomonas , and Mycobacterium . Aqueous (28 °C, pH 6.5) and soil cultures inoculated with PHE-acclimated soil showed a high PHE (ca. 50 mg L −1 ) degradation efficiency. The PHE degradation kinetics in aqueous and soil incubations fitted to the Gompertz equation and the first-order kinetic equation, respectively. Indigenous microorganisms adapted to PHE in their environment, and this increased their capacity to degrade PHE. The effect of co-contaminants and pathway intermediates on PHE degradation showed that the degradation of PHE improved in the presence of diesel while being hindered by lubricant oil, catechol, salicylic and phthalic acid. Our findings provide theoretical and practical support for bioremediationof PAHs in the environment.
