Background Ralstonia solanacearum, one of the most devastating bacterial plant pathogens, is the causal agent of bacterial wilt. Recently, several studies on resistance to bacterial wilt have been conducted using the Arabidopsis-R. solanacearum system. However, the progress of R. solanacearum infection in Arabidopsis is still unclear. Results We generated a bioluminescent R. solanacearum by expressing plasmid-based luxCDABE. Expression of luxCDABE did not alter the bacterial growth and pathogenicity. The light intensity of bioluminescent R. solanacearum was linearly related to bacterial concentrations from 104 to 108 CFU·mL−1. After root inoculation with bioluminescent R. solanacearum strain, light signals in tomato and Arabidopsis were found to be transported from roots to stems via the vasculature. Quantification of light intensity from the bioluminescent strain accurately reported the difference in disease resistance between Arabidopsis wild type and resistant mutants. Conclusion sBioluminescent R. solanacearum strain spatially and quantitatively measured bacterial growth in tomato and Arabidopsis, and offered a tool for the high-throughput study of R. solanacearum-Arabidopsis interaction in the future.

BACKGROUND Research on selenium nanoparticles (SeNPs) in chemical defense and chemotherapy of plants has developed rapidly owing to their high microbial toxicity, environmental safety, and degradability. Pine wilt disease (PWD) threatens pine forests worldwide; however, it is difficult to kill the nematodes ( Bursaphelenchus xylophilus ) inside the tree that cause PWD using traditional pesticide formulations. SeNPs could be the key to controlling PWD. RESULTS In this study, approximately 50 nm SeNPs were prepared using a simple and green method, and chitosan was used to increase their biocompatibility and stability. The preparation and characterization results showed that the prepared SeNPs coated with chitosan (SeNPs@CS) were spherical and evenly dispersed. The bioassay results showed that SeNPs@CS had an LC 50 of 15.627 mg L −1 against B. xylophilus . In addition, the killing mechanism of SeNPs@CS against B. xylophilus was studied. Confocal microscopy and transmission electron microscopy demonstrated that B. xylophilus were killed by reactive oxygen species, and the penetration of nano-form materials to B. xylophilus was higher than that of non-nano-form materials. To verify the effective penetration of SeNPs in pine tissues, Cy5-labeled SeNPs@CS was observed inside pine needles and branches using frozen sections and confocal microscopy. In addition, the cytotoxicity of SeO 2 and SeNPs@CS was tested, and the results showed that the cytotoxicity of SeNPs@CS to MC3T3-E1 cells was reduced. CONCLUSION These results show that SeNPs are expected to be used as a new strategy for the control of PWD with oxidative damage and high penetration to B. xylophilus and effective target penetration and biosafety. © 2022 Society of Chemical Industry.

Bursaphelenchus xylophilus(PWN) causes pine wilt disease (PWD), which is one of the most devastating pine diseases worldwide. Cytochrome P450 (CYP) catalyzes the biosynthetic metabolism of terpenoids and plays an important role in the modification of secondary metabolites in all living organisms. We investigated the molecular characteristics and biological functions ofBx-cyp29A3inB. xylophilus. The bioinformatics analysis results indicated thatBx-cyp29A3has a transmembrane domain and could dock with L(−)-carvone. The gene expression pattern indicated thatBx-cyp29A3was expressed in 0.2, 0.4, 0.6, 0.8, and 1.0 mg/mL L(−)-carvone solutions. TheBx-cyp29A3expression increased in a dose-dependent manner and peaked at 24 h of exposure when the L(−)-carvone solution concentration was 0.8 mg/mL. However, the gene expression peaked at 0.6 mg/mL after 36 h. Furthermore, RNA interference (RNAi) indicated thatBx-cyp29A3played an essential role in the response to L(−)-carvone. The mortality rates of theBx-cyp29A3knockdown groups were higher than those of the control groups in the 0.4, 0.6, 0.8, and 1.0 mg/mL carvone solutions after 24 h of exposure or 36 h of exposure. In summary, bioinformatics provided the structural characteristics and conserved sequence properties ofBx-cyp29A3and its encoded protein, which provided a target gene for the study of the P450 family ofB. xylophilus. Gene silencing experiments clarified the function ofBx-cyp29A3in the immune defense ofB. xylophilus. This study provides a basis for the screening of new molecular targets for the prevention and management ofB. xylophilus. Keywords:cytochrome P450;L(−)-carvone;Bursaphelenchus xylophilus;RNAi

Bacterial wilt is a significant soil-borne disease that poses a threat to mulberry production yield and quality of agricultural production worldwide. However, the disease resistance mechanisms dependent on root exudates are not well understood. In this present study, we investigated the antibacterial mechanisms of the main active substances (erucamide, oleamide, and camphor bromide) present in mulberry root exudates (MRE) againstRalstonia pseudosolanacearum(Rp), the causal agent of bacterial wilt. Our findings revealed that these three active substances inhibited the growth activity ofRpby affecting the cell morphology and extracellular polysaccharide content, as well as triggering a burst of reactive oxygen species. The active substances induced oxidative stress, leading to a decrease inRpgrowth. Additionally, the expression levels of key genes in the hrp gene cluster (hrpB,hrpX, andhrpF) and other virulence-related genes (such asripAW,ripAE,Rs5-4819,Rs5-4374,ace,egl3, andpehB) were significantly reduced upon treatment with the active substances. Further pathogenicity experiments demonstrated that root exudates (at a concentration of 1.5 mg·mL−1) delayed or slowed down the occurrence of bacterial wilt in mulberry. These findings provide valuable insight into the antimicrobial mechanisms of MRE againstRpand lay a theoretical foundation for the development and application of biocontrol agents to control mulberry bacterial wilt.

Integrating pesticides and mineral elements into a multi-functional stimuli-responsive nanocarrier can have a synergistic effect on protecting plants from pesticides and the supply of nutrients. Herein, a pH dual-responsive multifunctional nanosystem regulated by coordination bonding using bimodal mesoporous silica (BMMs) as a carrier and coordination complexes of ferric ion and polymethacrylic acid (PMAA/Fe3+) as the gatekeeper was constructed to deliver prochloraz (Pro) for the smart treatment of wilt disease (Pro@BMMs−PMAA/Fe3+). The loading capacity of Pro@BMMs−PMAA/Fe3+nanoparticles (Nps) was 24.0% and the “PMMA/Fe3+” complexes deposited on the BMMs surface could effectively protect Pro against photodegradation. The nanoparticles possessed an excellent pH dual-responsive release behavior and better inhibition efficacy againstRhizoctonia solani. Fluorescence tracking experiments showed that Nps could be taken up and transported in fungi and plants, implying that non-systemic pesticides could be successfully delivered into target organisms. Furthermore, BMMS−PMAA/Fe3+nanocarriers could effectively promote the growth of crop seedlings and had no obvious toxicological influence on the cell viability and the growth of bacteria. This study provides a novel strategy for enhancing plant protection against diseases and reducing the risk to the environment. Keywords:prochloraz;mesoporous silica nanoparticles;pH-responsive release;micronutrient;biosafety

The present study focuses on the inhibitory effect of volatile metabolites released byBacillus velezensisCT32 onVerticillium dahliaeandFusarium oxysporum, the causal agents of strawberry vascular wilt. The CT32 strain was isolated from maize straw compost tea and identified asB. velezensisbased on 16S rRNA gene sequence analysis. Bioassays conducted in sealed plates revealed that the volatile organic compounds (VOCs) produced by the strain CT32 possessed broad-spectrum antifungal activity against eight phytopathogenic fungi. The volatile profile of strain CT32 was obtained by headspace solid-phase microextraction (HS-SPME) coupled with gas chromatography-mass spectrometry (GC-MS). A total of 30 volatile compounds were identified, six of which have not previously been detected in bacteria or fungi: (Z)-5-undecene, decyl formate, 2,4-dimethyl-6-tert-butylphenol, dodecanenitrile, 2-methylpentadecane and 2,2’,5,5’-tetramethyl-1,1’-biphenyl. Pure compounds were tested in vitro for their inhibitory effect on the mycelial growth ofV. dahliaeandF. oxysporum. Decanal, benzothiazole, 3-undecanone, 2-undecanone, 2-undecanol, undecanal and 2,4-dimethyl-6-tert-butylphenol showed high antifungal activity, with benzothiazole and 2,4-dimethyl-6-tert-butylphenol being the most potent compounds. These results indicate that the VOCs produced byB. velezensisCT32 have the potential to be used as a biofumigant for management of vascular wilt pathogens. Keywords:biocontrol;Bacillus velezensis;volatile organic compounds;vascular wilt pathogens

It is important to study the response of plant pathogens to the antibiosis traits of biocontrol microbes to design the efficient biocontrol strategies. In this study, we evaluated the role of volatile organic compounds (VOCs) produced by a biocontrol strain Bacillus amyloliquefaciens SQR-9 on the growth and virulence traits of tomato wilt pathogen Ralstonia solanacearum (RS). The VOCs of SQR-9 significantly inhibited the growth of RS on agar medium and in soil. In addition, the VOCs significantly inhibited the motility traits, production of antioxidant enzymes and exopolysaccharides, biofilm formation and tomato root colonization by RS. The strain SQR-9 produced 22 VOCs, but only nine VOCs showed 1–11% antibacterial activity against RS in their corresponding amounts; however, the consortium of all VOCs showed 70% growth inhibition of RS. The proteomics analysis showed that the VOCs of SQR-9 downregulated RS proteins related to the antioxidant activity, virulence, carbohydrate and amino acid metabolism, protein folding and translation, while the proteins involved in the ABC transporter system, amino acid synthesis, detoxification of aldehydes and ketones, methylation, protein translation and folding, and energy transfer were upregulated. This study describes the significance and effectiveness of VOCs produced by a biocontrol strain against tomato wilt pathogen.

To further develop new antiviral agents, several novel cyclized derivatives of ferulic acid were designed and synthesized. Their antiviral activities were evaluated against the cucumber mosaic virus (CMV), pepper mild mottle virus (PMMoV), and tomato spotted wilt virus (TSWV). The results showed that some ferulic acid derivatives exhibited desirable antiviral activities. Particularly, compound 5e exhibited excellent protective activities against CMV, PMMoV, and TSWV, with EC50 values of 167.2, 102.5, and 145.8 μg mL–1, respectively, which were superior to those obtained for trans-ferulic acid (581.7, 611.2, and 615.4 μg mL–1), dufulin (312.6, 302.5, and 298.2 μg mL–1), and ningnanmycin (264.3, 282.5, and 276.5 μg mL–1). Thereafter, the protective mechanisms of 5e were evaluated through photosynthesis evaluation, transcriptome profiling, and proteomic analysis. The results indicated that 5e significantly activated the expression levels of photosynthesis-related regulatory genes and proteins in tobacco plants and promoted the accumulation of defense molecules to resist viral infection. Thus, the findings of this study indicated that novel cyclized ferulic acid derivatives are potential antiviral agents that act via regulating photosynthesis in the host.

Root exudate-mediated microbial community assembly is critical for host health and growth. However, disease-induced variations in plant-microbe interactions remain ambiguous. Here, we explored the intrinsic distinctions and interactions between the secretion patterns and microbial community composition of diseased and healthy tomatoes. Our results showed that rhizosphere microbial communities in healthy and diseased tomatoes were dominated by potentially beneficial genera ( Bacillus , Rhodanobacter , Pseudolabrys , Gemmatimonas , Dongia , and Bradyrhizobium ) and putative pathogens ( Ralstonia and Neocosmospora ), respectively, which were correlated with differential metabolites. Further metabolite addition experiments demonstrated the differential regulation mechanism of specific metabolites on host health. Drawing inspiration from synthetic communities, we displayed a way to construct the synthetic metabolites (SynMets), and we found that the SynMets (cortisol, quercetin, pyridoxal, and levodopa), which were enriched in healthy tomatoes could resist diseases by inhibiting pathogen growth and constructing beneficial microbial communities. Conversely, the SynMets (pyridoxine, N-benzylformamide, isoquinoline, and xanthine) enriched in diseased tomatoes could result in microbial imbalances by facilitating pathogen growth, thereby causing disease occurrence and growth limitation. In total, our research indicated the importance of SynMets-mediated pathogen reproduction and microbial community assembly for plant health and lays a foundation for targeted regulation of rhizosphere microecology through synthetic metabolites.

Introduction: Tomato bacterial wilt (TBW) is a destructive soil-borne bacterial infection caused by Ralstonia solanacearum. Various nanoparticles have been employed as antibacterial agents to manage TBW via soil application. However, research on the effects of nanoparticles on plant endophytes remains limited. Methods: Here, an analysis of the endophytic bacterial community was performed on healthy and infected tomatoes that were treated with Cu-Ag nanoparticles and thiodiazole-copper via high-throughput 16S rRNA gene amplicon sequencing. Results: The relative abundance levels of beneficial bacteria, including Acidobacteriota, Firmicutes, Actinobacteriota, and Myxococcota, were higher in infected tomato roots treated with Cu-Ag nanoparticles compared with thiodiazole-copper. Functional predictions show that Cu-Ag nanoparticles may affect pyruvate metabolism, oxidative phosphorylation, purine metabolism, carbon metabolism, secondary metabolite production, and the metabolic pathways associated with microbial communities.Discussion: These results could reveal the mechanism by which nanoparticles influence the endophytic microbiomes of plant roots and direct the rational application of nanoparticlesin sustainable agriculture.
