Staphylococcus aureus(S. aureus) is a major foodborne pathogen. Rapid and specific detection is crucial for controlling staphylococcal food poisoning. This study reported aStaphylococcusphage named LSA2302 showing great potential for applications in the rapid detection ofS. aureus. Its biological characteristics were identified, including growth properties and stability under different pH and temperature conditions. The genomic analysis revealed that the phage has no genes associated with pathogenicity or drug resistance. Then, the phage-functionalized magnetic beads (pMB), serving as a biological recognition element, were integrated with ATP bioluminescence assays to establish a biosensing method forS. aureusdetection. The pMB enrichment brought high specificity and a tenfold increase in analytical sensitivity during detection. The whole detection process could be completed within 30 min, with a broad linear range of 1 × 104to 1 × 108CFU/mL and a limit of detection (LOD) of 2.43 × 103CFU/mL. After a 2 h pre-cultivation, this method is capable of detecting bacteria as low as 1 CFU/mL. The recoveries ofS. aureusin spiked skim milk and chicken samples were 81.07% to 99.17% and 86.98% to 104.62%, respectively. Our results indicated that phage-based biosensing can contribute to the detection of target pathogens in foods. Keywords:Staphylococcus aureus;Staphylococcusphage;magnetic beads;ATP bioluminescence assay

In this work, quaternary ammonium carbon quantum dots (QCQD) was successfully synthesized with 2,3-epoxypropyltrimethylammonium chloride and diallyldimethylammonium chloride by a simple green “one-pot” method. QCQD was demonstrated to have satisfactory antibacterial activity against gram-positive bacteria, including Staphylococcus aureus , methicillin-resistant Staphylococcus aureus (MRSA), Staphylococcus epidermidis , Listera monocytogenes and Enterococcus faecalis . QCQD was used to treat MRSA-infected pneumonia in mice, and could promote the regression of pulmonary inflammation in lung of mice. In vivo and in vitro toxicity tests also confirmed that QCQD had acceptable biocompatibility. There is no doubt that all the experimental results confirmed that QCQD is a promising antimicrobial agent for treating infections of gram-positive bacteria. The antibacterial mechanism of QCQD was analyzed and confirmed by the quantitative proteomics based tandem mass tags, revealing that QCQD might interfere with the protein translation, posttranslational modification and protein turnover in bacterial cells by mainly acting on ribosomes and upregulating RNA degradation related proteins. The application of proteomics extended the research on antibacterial mechanism from single target to protein network and realized the research and analysis on multiple targets of bacteria treated by QCQD, providing a new idea and strategy for the development of efficient bactericidal weapon against drug-resistant bacteria.

Antimicrobial resistance is becoming more and more serious and has become a potential hazard to human life and health. The fabrication of some new antibacterial substances against resistant bacteria is demanded. With the wide application and research of carbon nanomaterials, nitrogen-doped carbon quantum dots (NCQDs) were synthesized by a one-step chemical route herein. The particle size of NCQDs in the range of 2–5 nm were characterized by transmission electron microscopy (TEM), atomic force microscopy, and dynamic light scattering. The functional groups and optical properties of NCQDs were investigated by UV–vis absorption spectroscopy, X-ray photoelectron spectroscopy and Fourier transform infrared spectroscopy. Disk-diffusion tests showed that the NCQDs had specific antibacterial activity against Staphylococcus . TEM showed that the NCQDs could destroy the cell structure of Staphylococcus aureus and methicillin-resistant Staphylococcus aureus (MRSA) but could not combat Escherichia coli . The antibacterial mechanism may be that positively charged NCQDs firstly interacted with the negatively charged bacteria, and then specifically anchored on some specific sites on the surface of Staphylococcus . The NCQDs were applied to treat wounds infected with MRSA and showed the same therapeutic effect as vancomycin. Photomicrographs of hematoxylin-eosin-stained histological sections showed that the NCQDs at concentrations effectively killing S. aureus and MRSA caused negligible toxicity to the main rat organs, including heart, liver, spleen, lung, and kidney. Thus, the NCQDs can be developed as a promising antibacterial agent for Staphylococcus . And the NCQDs are likely to treat local infections caused by Staphylococcus clinically, especially S. aureus and MRSA .

A highly sensitive and selective near-infrared (NIR) fluorescent probe, (E)-2-(2-(6-((2-carboxy-8-oxo-7-(2-phenylacetamido)-5-thia-1-azabicyclo[4.2.0]oct-2-en-3-yl)methoxy)-2,3-dihydro-1H-xanthen-4-yl)vinyl)-3,3-dimethyl-1-propyl-3H-indol-1-ium (1), is developed for the determination of β-lactamase. The probe is designed and synthesized by incorporating the specific substrate (cephalosporin) of β-lactamase into a stable hemicyanine skeleton. The fluorescence of 1 itself is very weak due to the alkylation of the hydroxyl group of the hemicyanine fluorophore; however, β-lactamase can selectively react with its substrate (β-lactam ring) in the probe, thereby causing a spontaneous fragmentation. This action leads to the release of the fluorophore and a large fluorescence enhancement at 707 nm (λex = 680 nm). Under the optimized conditions, the fluorescence response of probe 1 is directly proportional to the concentration of β-lactamase in the range of 0.05–2 nM, with a detection limit of 0.02 nM. The validity of the probe has been confirmed by determining β-lactamase in human urine samples in comparison with that determined by iodimetry. Moreover, by taking advantage of its high sensitivity and NIR emission feature, the probe has also been utilized to image β-lactamase in three types of Staphylococcus aureus, including methicillin-resistant S. aureus ATCC BAA44, penicillin-resistant strain ATCC 11632, and penicillin-susceptible strain ATCC 29213, which clearly reveals the significantly different expression levels of β-lactamase in these S. aureus.

Recently, continuous emergence of resistant bacteria has appeared as one of the most serious threats to human health. Therefore, systematic exploration of new antibacterial materials is of guiding significance. In this study, a series of photosensitizer-polypeptide conjugate (PPa-cP) is readily synthesized through simple ring-opening reactions to realize the synergistic antibacterial effects on Staphylococcus aureus ( S. aureus ) and methicillin-resistant S. aureus (MRSA) under light irradiation. Compared with free PPa, the cationic PPa-cP shows enhanced binding ability with the negative surface of S. aureus through electrostatic interaction, exhibiting effective antibacterial activity against both S. aureus and MRSA in vitro under light irradiation. Among the synthesized PPa-cP, PPa-cP5 with the degree of polymerization of 37 and modified with a 1-methylimidazole side group exhibits the best antibacterial activity with a minimum inhibitory concentration value of 2 µ m without light irradiation and 0.25 µ m with light irradiation. Moreover, PPa-cP5 shows good hemocompatibility. The above-mentioned results elucidate that the positively charged PPa-cP5 can significantly increase the efficiency of photodynamic therapy and effectively eradicate S. aureus biofilm due to its potent penetration ability into S. aureus biofilms. Overall, the present study establishes an efficient strategy for the treatment of S. aureus and S. aureus biofilm infections.

Staphylococcus aureus ( S. aureus ) infection, especially its drug-resistant bacterial infection, is a great challenge often faced by clinicians and patients, and it is also one of the most important threats to public health. Finding a safe and effective antibacterial agent is of great significance for the prevention and treatment of S. aureus infection. Lysozyme is known to have antibacterial effects against Gram-positive bacteria including S. aureus . Here, high-quality lysozyme with a purity of more than 99% and an activity of more than 60, 000 U/mg was prepared from egg white, which showed excellent antibacterial activity against three strains of S. aureus , especially against MRSA . Furthermore, an antibacterial cream loaded with lysozyme was prepared and tested in scald wound healing. The lysozyme-loaded cream exhibited the effect of preventing wound infection and promoting wound healing on scalds, and no toxicity was found in animal organs. Overall, lysozyme showed great application potential in the prevention and treatment of infections caused by S. aureus and scalded wound healing. The most remarkable discovery in this work is the unexpectedly powerful inhibitory effect of lysozyme on the drug-resistant bacterial, especially MRSA , which is usually very difficult to deal with using normal antibacterial drugs.

The emergence of antibiotic resistance represents a significant and growing threat to global public health. The development of novel and antibiotic-free antibacterial strategies is therefore highly importance. In this context, chemodynamic therapy (CDT), which is based on Fenton or Fenton-like chemistry, is a promising strategy. CDT utilizes Fe 2+ to convert H 2 O 2 into highly reactive hydroxyl radicals (•OH), which can oxidize biomolecules to achieve antibacterial effects. The antibacterial efficacy of CDT is contingent upon both the catalyst and substrate. We developed a straightforward, convenient and economical approach to fabricate Fe-caffeic acid metal–polyphenol networks (Fe-CA MPNs) by mixing CA and Fe 3+ . The Fe-CA MPNs are acid-sensitive and self-enhanced CDT agents with remarkable antibacterial efficacy against both Staphylococcus aureus ( S. aureus ) and methicillin-resistant S. aureus (MRSA). Antibacterial efficacy was evaluated via a variety of methods, including the zone of inhibition, spread plate, turbidimetric, and live/dead bacterial staining tests. Specifically, the inhibition rates of Fe-CA MPNs for S. aureus and MRSA were 96.33 ± 0.33 % and 96.33 ± 0.15 %, respectively. CaO 2 @Fe-CA nanocomposites with H 2 O 2 self-supply properties were synthesized by depositing Fe-CA MPNs on the surface of CaO 2 nanoparticles which function as a sustained-release source of H 2 O 2 . The antibacterial effect of the CaO 2 @Fe-CA nanocomposites was comparable to that of Fe-CA MPNs combined with H 2 O 2 . Both Fe-CA MPNs and CaO 2 @Fe-CA possess favorable in vitro biosafety profiles. This work will facilitate the development of innovative antibiotic-free CDT antibacterial strategies based on •OH.

The aim of this study was to evaluate the inhibitory activity of shikimic acid (SA) against biofilm formation of Staphylococcus aureus and to explore its potential molecular mechanism. Crystal violet staining assay showed that SA clearly reduced the biomass of the biofilm at its sub-MICs. Light microscopic images and scanning electron microscope (SEM) observations suggested that SA inhibited the biofilm adhesion on glass slides and induced apparent collapse of biofilm architecture. In addition, XTT reduction assay and confocal laser scanning microscopic images illustrated that the metabolic activity and the viability of the SA-treated biofilm cells significantly decreased. Cell mobility of the SA-treated S. aureus was also inhibited. These observations suggested that SA inhibited the initial biofilm formation by interfering the metabolic activity, the cell viability, and the cell mobility of S. aureus . However, SA failed to disrupt the one-day-old mature biofilm of S. aureus even at the concentrations higher than MIC, suggesting that SA disturbed the early steps in the biofilm formation of S. aureus . qRT-PCR analyses showed that SA down-regulated the transcription of sarA , while up-regulated the transcription of agrA , which resulted in the prevention of biofilm formation. So, SA could be a potential antibiofilm agent to inhibit biofilm formation of S. aureus .

Background Acne has become one of the most prevalent skin disorders , affecting mostly young people's physical and mental health globally. Cryptotanshinone (CPT) is a potential drug for acne, but its mechanism of acne treatment has not been thoroughly studied on the microbiota . Till date, only a few studies are directed to the impact of acne therapy on skin microbiota and lipid metabolites. Purpose The action mechanism of CPT treatment of acne was investigated by the strategy of microbiome integration with lipidomics . Methods The 16Sr DNA sequencing was used to detect skin microbiota composition, and absolute quantitative lipidomics was utilized to identify lipid metabolites profiles levels. Four key proteins of the glycolysis pathway were detected with the immunochemistry method. Antibacterial analysis was used to evaluate CPT treatment of acne. Results CPT significantly inhibited Staphylococcus epidermidis and Staphylococcus aureus . Combination of the skin microbiome and lipidomics analysis, 29 types of differentially expressed flora (DEFs) and 782 differentially expressed lipid metabolites (DELMs) were significantly altered, especially Staphylococcus, Corynebacterium , Ralstonia , Enhydrobacter, Burkholderia , and Streptococcus . Cer was mainly regulated by Staphylococcus and Corynebacterium , whereas TG and DG were mainly regulated by Ralstonia, Enhydrobacter, Burkholderia , and Streptococcus . The glycolysis pathway was significantly regulated by Staphylococcus on CPT treatment of acne. The energy metabolism, lipid metabolism , immune system, glycan biosynthesis , and metabolism could be reversed by CPT. Conclusion CPT might help acne rats rebuild their skin microbiota and alter lipid metabolism signatures. Furthermore, since skin microbes and skin lipid metabolites have a close correlation and are both regulated by CPT, the findings potentially provide a research foundation for the discovery of biomarkers of skin microbiome imbalance and targeted treatment of acne development mechanisms.

Although bacteriophage-based biosensors are promising tools for rapid, convenient, and sensitive detection of Staphylococcus aureus in food products, the effect of biosensors using temperate phages as biorecognition elements to detect viable S. aureus isolates remains unclear. In this study, three temperate S. aureus phages were isolated and their biological features (one-step growth, host range, pH stability, temperature stability, and adsorption rate) were evaluated as the biological element. The selected phage SapYZUs8 was immobilized on the nanozyme Cu-MOF via electrostatic interactions to generate SapYZUs8@Cu-MOF, and its detection performance in real food (skim milk and pork) was then evaluated. Compared with phages SapYZUm7 and SapYZUs16, phage SapYZUs8 exhibited a broader host range, greater pH stability (3–12), and a better absorption rate (92 %, 8 min) suitable for S. aureus detection, which is likely the result of the DNA replication (DNA helicase) and phage tail protein genes in the SapYZUs8 genome. Therefore, phage SapYZUs8 was fixed on Cu-MOF to generate SapYZUs8@Cu-MOF, which exhibited good sensitivity and specificity for rapid colourimetric detection of viable S. aureus. The method took <0.5 h, and the detection limit was 1.09 × 10 2 CFU/mL. In addition, SapYZUs8@Cu-MOF was successfully employed for the colourimetric detection of S. aureus in food samples without interference from different food additives, NaCl concentrations, or pH values. With these benefits, it allows rapid visual assessment of S. aureus levels.

Staphylococcus aureus ( S . aureus ) is the leading cause of a broad range of human infections and associated with food poisoning worldwide. Nisin can be considered as a natural preservative and is widely used by the food industry. However, resistance of nisin has been reported in several species of bacteria. Identification of antibacterial agents that are synergistic with nisin is a method of minimising resistance to nisin. Cinnamaldehyde (CA), a well-studied essential oil isolated from cinnamon bark, has been reported to exhibit a broad spectrum of antimicrobial activity across a wide range of microorganisms. Previous studies have demonstrated that CA could be used in the food industry due to its active antibacterial activity and its safety. In this study, the synergistic antibacterial activity in vitro of nisin and CA against 13 food-borne isolates of S. aureus and S. aureus ATCC 29213 were evaluated by a checkerboard assay. The FICI values of nisin combined with CA ranged from 0.1875 to 0.375, which demonstrated a synergistic effect. The positive interactions were verified using the challenge tests in pasteurized milk and agar diffusion assays. The LIVE/DEAD BacLight experiment and scanning electron microscope (SEM) assay were also performed to distinguish the intact and damaged both of cell walls and membranes with the compounds, the combination group displayed stronger damage both of cell wall and cell membrane than when nisin or CA was used alone. These findings indicated that the combination of nisin and CA could not only be used as a promising naturally sourced food preservative but also to reduce the problem of bacterial resistance.

Conventional methods for Staphylococcus aureus ( S. aureus ) detection using mammalian immunoglobulins could yield false positive results, owing to the presence of the protein G-producing Streptococcus . In this study, because canine IgM can bind to protein A but not to protein G, we firstly designed a colorimetric platform based on canine IgM to identify S. aureus . Magnetic beads (MBs) functionalized with canine IgM acted as captors to enrich S. aureus , relying on the binding interaction between the Fc region of canine IgM and protein A in the bacterial cell wall, while horseradish peroxidase (HRP)-labeled rabbit anti- S. aureus IgG served as a tracer in the sandwich-type immunoassay, catalyzing the colorimetric reaction. Under optimal conditions, the proposed strategy achieved a satisfactory analytical performance with a broad linear range from 1.6 × 10 3 CFU/mL to 1.0 × 10 5 CFU/mL, a colorimetric minimum resolution of as low as 1.4 × 10 2 CFU/mL in 100 μL phosphate-buffered saline (PBS), and good selectivity in differentiating target from protein G-producing Streptococcus . Moreover, the proposed approach successfully identified S. aureus in various application scenarios (except for samples containing IgG), can be completed in less than 90 min, and was not dependent on any specialized equipment.
