Study on the components changes of polysaccharides and saponins during nine steaming and drying of Polygonatum sibiricum

Qihong Zhang,Xinyu Lin,Weike Su

BACKGROUND In this study, the content and structure of Polygonatum sibiricum polysaccharides and saponins during different processing stages were determined. RESULTS After processing of Polygonatum , the content of polysaccharide and glucose decreased, and the content of galactose, glucuronic acid and sugar substitution gradually increased. The content of total saponins increased significantly. Only 18 compounds were found in raw Polygonatum and 17 new compounds were presented in processed Polygonatum . During the processing of Polygonatum , the polysaccharide was partially degraded into oligosaccharides, the molecular weight gradually decreased, and the neutral sugar was converted into uronic acid, resulting in a decrease in polysaccharide content. The saponins were partially degraded into sapogenins or modified. CONCLUSION This study clarifies the changes in the content and structure of polysaccharides and saponins in processed Polygonatum , which will pave the way for elucidating the processing mechanism. © 2024 Society of Chemical Industry.

Anti-prostate cancer components of black ginseng based on the "nine steaming and nine sun-drying" process

Ye Huang,Xiaoku Ran,Deqiang Dou

This study was to identify the main active components of black ginseng responsible for prostate cancer treatment. HPLC fingerprints of 10 batches of processed ginseng from the "nine steaming and nine sun-drying" procedure were established. MTT test was performed to evaluate the inhibitory activity of black ginseng on human prostate cancer (DU145) cells. The spectrum-effect relationship analysis was assessed by correlation coefficient and grey relational analysis, and the active ginsenosides were further validated by bioactivity assay. Results indicated the anti-prostate cancer activity of black ginseng increased until the 7th cycle times of steaming and sun-drying, and the primary anti-prostate cancer ingredients of black ginseng were disclosed to be ginsenosides Rg5, F4, Rh4, Rk1, R-Rg3, and S-Rg3, of which Rk3, Rg5, and F4 exerted the highest effects. It is for the first time that a spectrum-effect relationship based on the ingredient changes of the "Nine Steaming and Nine Sun-drying" process was used to identify the active anticancer constituents of ginseng systemically and could further the stronger anticancer activity of black ginseng as well as provide a theoretical basis for the quality evaluation of black ginseng.

Study on the varied content of Polygonatum cyrtonema polysaccharides in the processing of steaming and shining for nine times based on HPLC-MS/MS and chemometrics

Baolei Fan,Guoliang Wei,Xiaofeng Gan,Tingting Li,Zengyi Qu,Sheng Xu,Chao Liu,Chunqi Qian

In this study, we reported the various contents of Polygonatum cyrtonema by extracting polysaccharides (PCPs) using steaming and shining procedures, thus providing scientific basis for the processing of Polygonatum cyrtonema and content analysis of polysaccharides. We established the characteristic fingerprint to identify and analyze polysaccharide component changes in distillated and processed Polygonatum cyrtonema by PMP precolumn derivatization and HPLC-MS/MS technology. We also classified two kinds of solutions by comparing the composition difference of 10 polysaccharide components in the samples with principal component and cluster analysis. There was a huge difference between them and seven monosaccharides. Processed PCPs mainly consisted of galactose along with mannose, followed by glucose. Meanwhile, we found that the results of cluster analysis and principal component analysis were the same. The polysaccharide components extracted from processed Polygonatum cyrtonema and distillate were mainly galactose and mannose. The results of cluster analysis and principal component analysis were the same. The distilled liquid can be divided into 5 categories, and the processed dry extract of Polygonatum cyrtonema can be divided into 4 categories. The changing trend of polysaccharide components of Polygonatum cyrtonema and distilled liquid was revealed during the multiple cycles of steaming and sun drying. The similarity and difference of polysaccharide components in the extracted liquid and the distilled liquid of each batch were shown.

Dynamic profiling of metabolite changes and health-promoting functions in ‘yuling paste’ during nine steaming and nine sun-drying processes

Yue E,Weimiao Li,Hongbin Guo,Xianman Zhang,Qinggele Caiyin,Yi Yuan

Yuling paste, a traditional Chinese health food derived from longan pulp and American ginseng, undergoes a unique processing method involving nine cycles of steaming and sun-drying. Ultra-high-performance liquid chromatography tandem mass spectrometry combined with widely targeted metabolomics has been used to examine the dynamic change in metabolite profiles through the processing. A total of 758 metabolites were identified. Processing significantly affects metabolite changes, and network pharmacology is subsequently used to explore potential pharmacological ingredients. After processing, the contents of active ingredients such as ginsenoside rh2, oleanolic acid, choline, d -glucose, and D-galacturonic acid were found to increase significantly. These increases can be correlated to the enhancement of five distinct pathways, and the contents of naringenin-7-O-glucoside, adenosine, pantothenic acid, and D-sucrose decreased after the processing, correlating with decreases in two different pathways. This study provides a comprehensive reference and scientific basis for understanding the health benefits associated with this traditional health food.

Untargeted components and in vivo metabolites analyses of Polygonatum under different processing times

Yue Sun,Lin Zhou,Xiao Shan,Tiantian Zhao,Meirong Cui,Wenqian Hao,Binbin Wei

Polygonatum is a medicinal and edible homologous food, for which nine steaming and nine drying (SAD-9) is the most used processing method. The processed polygonatum not only changes its components, but also causes the fluctuation of metabolites in vivo . UPLC-QTOF/MS (Ultraperformance liquid chromatography-Quadrupole time of flight mass spectrometry) was employed to analyze the changes in components and the plasma untargeted metabolomics, thus, the differences in vitro of three groups of polygonatum (Raw Polygonatum, RP; two steaming and two drying, SAD-2; nine steaming and nine drying, SAD-9) and their effects on metabolic fluctuations in vivo were revealed. Results showed that the components, such as protein, fat, polysaccharide, flavonoids, saponins and isoflavones changed after processing. Moreover, 4 types of Lysophosphatidylcholines, the major differential metabolites, were highly expressed in rats’ plasma of SAD-9 group. Furthermore, through network pharmacological analysis, it was found that liquiritigenin and 4′,5-dihydroxyflavone activated PLA2 type enzymes to promote the production of lysophosphatidylcholine, and the processing method of SAD-9 was conducive to the role of these two components. These findings indicated the dissimilarity of components and metabolic profiles of polygonatum under different processing times, and revealed the advantages of SAD-9 processing methods.

A comprehensive investigation on the chemical changes of traditional Chinese medicine with classic processing technology: Polygonum multiflorum under nine cycles of steaming and sunning as a case study

Fan Xinyu,Zhou Lin,Xing Yanchao,Wang Liming,Choi Shin Sik,Zhang Zixin,Zhang Xu,Liu Caixiang,Zhu Yu,Fu Zhifei,Han Lifeng

The processing of traditional Chinese medicine (TCM) plays an important role in the clinical application, which usually has the function of "increasing efficiency and reducing toxicity". Polygonum multiflorum (PM) has been reported to induce hepatotoxicity, while it is believed that the toxicity is reduced after processing. Studies have shown that the hepatotoxicity of PM is closely related to the changes in chemical components before and after processing. However, there is no comprehensive investigation on the chemical changes of PM during the processing progress. In this research, we established a comprehensive method to profile both small molecule compounds and polysaccharides from raw and different processed PM samples. In detail, an online two-dimensional liquid chromatography coupled with quadrupole-orbitrap mass spectrometry (2D-LC/Q-Orbitrap MS) was utilized to investigate the small molecules, and a total of 150 compounds were characterized successfully. After multivariate statistical analysis, 49 differential compounds between raw and processed products were screened out. Furthermore, an accurate and comprehensive method for quantification of differential compounds in PM samples was established based on ultra-high performance liquid chromatography/Q-Orbitrap-MS (UHPLC/Q-Orbitrap-MS) within 16 min. In addition, the changes of polysaccharides in different PM samples were analyzed, and it was found that the addition of black beans and steaming times would affect the content and composition of polysaccharides in PM significantly. Our work provided a reference basis for revealing the scientific connotation of the processing technology and increasing the quality control and safety of PM. Graphical Abstract

Reverse tracing anti-thrombotic active ingredients from dried Rehmannia Radix based on multidimensional spectrum-effect relationship analysis of steaming and drying for nine cycles

Pu-Yang Gong,Yu-Jie Guo,Yu-Shan Tian,Li-Fei Gu,Jin Qi,Bo-Yang Yu

Ethnopharmacological relevance In traditional Chinese medicine (TCM) and modern pharmacodynamics , dried Rehmannia Radix (DRR) possesses prominent anti-thrombotic activity that decreases after processing by nine steaming and drying cycles to develop processed Rehmannia Radix (PRR). Due to the complexity of the DRR components, the chemical mechanism leading to efficacy changes of DRR caused by processing is still unclear. Aim of study This study aimed to trace the anti-thrombotic active compounds of DRR and different degrees of processed RR (PRR) and to evaluate the synergistic effects among different active components. Materials and methods The anti-thrombotic active chemical fraction of DRR extracts was evaluated. Targeted fractions of the processed products of RR were prepared at different processing stages. The changes in monosaccharides , oligosaccharides and secondary metabolites during processing were characterized by multidimensional high-performance liquid chromatography (HPLC). The anti-thrombotic effects of targeted fractions of different RR samples were evaluated by analyzing the length of tail thrombus (LT) and serum biochemical indicators in carrageenan-induced tail-thrombus mice. The spectrum-effect relationships were investigated by partial least squares regression (PLSR) analysis and gray correlation analysis (GRA). Finally, the active compounds were screened by spectrum-effect relationship analysis and validated in vivo , and their synergistic effects were determined by Webb's fraction multiplication method. Results Six ingredients highly associated with anti-thrombotic activities were screened out by the spectrum-effect relationship analysis, of which oligosaccharides (stachyose, sucrose and raffinose) and iridoid glycosides (catalpol, leonuride and melitoside) possessed a synergistic effect on tumor necrosis factors (TNF-α), interleukin 1β (IL-1β) and plasminogen activator inhibitor 1 (PAI-1)/tissue-type plasminogen activator (t-PA) ratio in vivo with synergistic coefficient (SC) > 1. Conclusion The main material basis of the anti-thrombotic activities of DRR is oligosaccharide components of stachyose, raffinose and sucrose, iridoid glycosides components of catalpol , leonuride and melittoside. The two kinds of components exert synergistic anti-thrombotic effects by inhibiting the expression of inflammatory factors and regulating the balance of the fibrinolysis system.
