Development and Verification of a Precolumn Derivatization LC-MS/MS Method for the Pharmacokinetic Study of Houttuynine of Houttuynia Essential Oil

Yuanyuan Liu,Yanfang Yang,Bangyuan Wang,Renyun Wang,Jianmei Pang,Yu Jiang,Yuling Liu

Houttuynia essential oil (HEO) has excellent antiviral, anti-inflammatory, and other pharmacological effects, but the lack of effective analytical methods to quantify HEO in plasma has hindered its better clinical monitoring. Houttuynine (Hou) is one of the main active ingredients and quality control substances of HEO, so the pharmacokinetic study of HEO could be conducted by determining Hou blood concentration. Hou is active and not stable in plasma, which makes its blood concentration difficult to measure. In this work, a novel liquid chromatography tandem mass spectrometry (LC-MS/MS) method for Hou determination in rat blood was established that involves Hou being derivatized with 2, 4-dinitrophenylhydrazine to form a stable compound to prevent degradation. Herein,p-Tolualdehyde-2,4-dinitrophenylphenylhydrazone was selected as an internal standard substance and the LC-MS/MS method was evaluated for selectivity, precision, accuracy, calibration limit, matrix effect, recovery, and stability. Good linearity (r2= 0.998) was reached in the range of 2–2000 ng/mL, and the lower limit of quantification of Hou was determined to be 2 ng/mL. The mean intra-assay accuracy ranged from 77.7% to 115.6%, whereas the intra-assay precision (relative standard deviation, RSD) was below 11.42%. The matrix effect value for Hou in rat plasma was greater than 75%, and for the internal standard (IS) it was 104.56% ± 3.62%. The extraction recovery of Hou were no less than 90%, and for the IS it was 96.50% ± 4.68%. Our method is sensitive and reliable and has been successfully applied to the pharmacokinetic analysis of Hou in rats given HEO via gavage and injection. Keywords:Houttuynia cordataThunb.;houttuynine;houttuynia essential oil;LC-MS/MS;pharmacokinetic study

Deep Eutectic Solvent-Based Ultrasound-Assisted Extraction of Flavonoids from Houttuynia cordata

Xinxin Wu,Ling Yan,Jingda Li,Zhijian Tan

Recently, deep eutectic solvents (DESs) have attracted much attention in the extraction and separation field because of their green characteristics, and they are widely used to extract various bioactive substances from plants. In this study, ten choline chloride (ChCl)-based mixtures were selected for extracting flavonoids from Houttuynia cordata. Then, the optimal conditions for the DES-based extraction of Houttuynia cordata flavonoids (HCFs) were determined through one-way experiments and response surface optimization (RSM). The optimal conditions were a 60 min extraction time, an extraction temperature of 40 °C, a water content of 30%, a solid−liquid ratio of 1:22 g·mL −1 , a molar ratio of 1:2, and an ultrasound power of 420 W. The antioxidant capacities of HCFs in terms of ABTS radical, DPPH radical, and hydroxyl radical scavenging capacity and nitrite inhibition capacity were determined. DESs can be efficiently recycled after extraction. This study developed an effective and environmentally friendly method for HCF extraction, thereby also supporting the development and utilization of natural products.

Study of the Contents of Analogues of Aristolochic Acid in Houttuynia cordata by Ultra-High Performance Liquid Chromatography Tandem Mass Spectrometry

Xuan Yu,Yuan Gao,Ying Xu,Xian Guo,Lan Guo,Ting Tan,Fan Liu,Yiqun Wan

In this study, a new and simple method was established for the simultaneous determination of analogues of aristolochic acids (aristolochic acid I, aristolochic acid II, aristolactam I and aristolactam AII) inHouttuynia cordataby ultra-high performance liquid chromatography–quadrupole/time-of-flight mass spectrometry (UHPLC–Q/TOF-MS). The samples were ultrasonically extracted with methanol, purified with HC-C18, and then separated on a C18 column (75 × 2.1 mm, 2.0 μm) at 35 °C. Under the optimized conditions, aristolochic acid I (AA-I), aristolochic acid II (AA-II), aristolactam I (AL-I) and aristolactam AII (AL-AII) all showed good linear regression (not less than 0.9987). The average recoveries of the four analytes were within the range of 72.3–105.5%, with the relative standard deviations (RSDs) being ≤7.6%. The proposed method was then applied to the determination ofHouttuynia cordatasamples collected from different regions in China. The results showed that none of the three carcinogenic substances (aristolochic acid I, aristolochic acid II and aristolactam I) were detected in any of the 22 samples collected from 22 different regions of China, while aristolactam AII, which has not been reported to have genotoxicity, was detected in all samples. This study provides a valuable reference for the further safety assessment ofHouttuynia cordata. Keywords:Houttuynia cordata;aristolochic acid analogues;detection;UHPLC-Q/TOF-MS

An analytical method for the determination of flavonoids in Houttuynia cordata Thunb. by UPLC-TQ-MS and analysis of its antioxidant activity

Zhang Zhiqi,Li Lin,Wang Xinyang,Liu Liyan

Houttuynia cordata Thunb. (H.cordata) , as a traditional Chinese medicinal herb and food, has been used to treat various diseases because of rich content in flavonoids. Here, an ultra-high performance liquid chromatography with triple quadrupole tandem mass spectrometry (UPLC-TQ-MS/MS) method was proposed for the determination of flavonoids profiling in H.cordata . Single factor experiment and response surface methodology were used to optimize the pretreatment process of H.cordata samples. The optimal conditions were as follows: ultrasonic temperature: room temperature (25 °C); ultrasonic time: 5 min; solid-liquid ratio: 1:40 and concentration of ethanol: 60%. The method showed the good linearity ( r  ≥ 0.9855), the satisfactory intra-day (RSD%≤10.94%) and inter-day precision (RSD% ≤ 11.2%) and low limit of quantification (< 0.01 µg/mL). The content of flavonoids in flowers and leaves was much higher than in roots and stems. The flavonoids in the leaves of H.cordata from Chongqing was the highest (9.22 mg/g), and that in the flowers of H.cordata from Hubei province was the highest (6.69 mg/g). In addition, the antioxidant activity of the extracts of H.cordata was evaluated using the scavenging effect of the extracts on five free radicals and the iron ion reducing ability. The best antioxidant activity was observed in scavenging DPPH free radicals with EC 50 value of 295.16 µg/mL. The scavenging of ABTS•+ free radicals and PTIO• free radicals was also effective with EC 50 values of 818.82 µg/mL and 685.43 685.43 µg/mL, respectively. Thus, this study will be helpful to the development and utilization of flavonoids from H.cordata in food and pharmaceutical industries.

Transport, pollution, and health risk of heavy metals in “soil-medicinal and edible plant-human” system: A case study of farmland around the Beiya mining area in Yunnan, China

Jiayi Hu,Xiaofang Yang,Huajian Chi,Xin Liu,Ning Lu,Ya Liu,Shengchun Yang,Xiaodong Wen

Heavy metals (HMs) pose a significant threat to soil quality, plant growth, and human health. However, the impacts (transport, pollution, and health risk) of HMs in medicinal and edible plants with rich value growing in high background soil for a long time remain unclear. So the effects of HMs on the “soil-medicinal and edible plant-human” system were comprehensively studied. Houttuynia cordata Thunb. (HCT) and growing soil were collected from the farmland around Beiya mining area located in Three Parallel Rivers of Yunnan Protected Areas, China. The transport, pollution, and health risks of HMs in the system were investigated by determining the content of active ingredients, HMs, and physicochemical properties in HCT and soil. The results of comprehensive analysis using various measurement and analysis methods showed that Zn, Cu, Mn, and Co in soil could affect the contents of available nutrients (available P (AP), available K (AK), and organic matter (OM)), while AP, OM, pH, Co, and Fe in soil were closely related to the formation of HCT quality. Cr and Fe had the strongest transport capacity in stems and leaves of HCT, respectively. Furthermore, it is worth noting that Cr, Pb, and Cu had caused serious pollution to HCT in the area. Pb and Cu are tolerable levels for human health and may pose a carcinogenic risk. Our study demonstrates that HCT is expected to be a marker of heavy metal pollution in the area and provides a scientific basis for making effective regulations on HMs control and scientific planting.

Widely targeted metabolomic deciphers the vertical spatial distribution of flavor substances in Houttuynia cordata Thunb

Shuai Zhang,Huixia Zhang,Shiqi Chen,Liu Yang,Xin Chen,Haiyang Jiang

The volatile components in Houttuynia cordata Thunb. have a distinctive odor, which is the source of their unique flavor as food . Although volatile compounds are widely present in Houttuynia cordata Thunb., little is known about their accumulation in vertical distribution in different organs. In this study, gas chromatography-mass spectrometry (GC-MS) and chemometrics were used to determine volatile substance’s types and relative contents, especially 2-undecanone in different parts of Houttuynia cordata Thunb. The results showed that 933 species and 16 classes of volatile compounds were identified in the leaves, aerial stems, subterranean stems, and roots. Among them, the accumulation of 2-undecanone, d -limonene, and α-pinene were significantly higher in subterranean stems than in other organs. The related KEGG pathway-limonene and pinene degradation, was identified. The study's results can provide a reference for the industrial extraction and utilization of volatile compounds from different parts of Houttuynia cordata Thunb.

A dispersive solid-phase extraction method for the determination of Aristolochic acids in Houttuynia cordata based on MIL-101(Fe): An analytes-oriented adsorbent selection design

Rui Cheng,Xuejin Mao,Jiaying Yu,Fan Liu,Lan Guo,Dongmei Luo,Yiqun Wan

In view of the molecular structure of Aristolochic acid I (AA-I) and Aristolochic acid II (AA-II), MIL-101(Fe) was selected as the sorbent to develop a dispersive solid-phase extraction (d-SPE) method for capturing the two analytes from Houttuynia cordata. The interactions between the sorbent and analytes were investigated by FT-IR, XPS and UV–Vis DRS spectra. The optimized method demonstrated good linearity with R 2  > 0.9999. The limit of detections (LODs) were 0.007 mg/L and 0.014 mg/L for AA-I and AA-II, respectively, lower than the limit stipulated by Chinese Pharmacopoeia (0.001 %, w/w). The recoveries for AA-I and AA-II were within the range of 73.3–106.4 %. The precisions of intra-day and inter-day were 0.9–5.8 % and 2.1–5.8 %, respectively. Thus, the established method demonstrated to be efficient and reliable to determine AA-I and AA-II in Houttuynia cordata.

Anti-inflammatory mechanism of Houttuynia cordata polysaccharides against ulcerative colitis based on multi-omics conjoint analysis

Yueqi Hu,Li Zhou,Jian Yang,Ruibin Bai,Eric Marchioni,Minjie Zhao,Li Zhou

The Houttuynia cordata polysaccharide (HCP) was extracted from the traditional Chinese medicine, Houttuynia cordata , known for its anti-inflammatory properties. It has an acidic heteropolysaccharide with a molecular weight of approximately 13.38 kDa, consisting of 7 monosaccharides such as galactose, galacturonic acid, and glucose. Mouse ulcerative colitis (UC) model experiments demonstrated its effective anti-inflammatory activity at concentrations of 100 mg/kg and 300 mg/kg respectively. The objective of this study was to investigate the mechanism of action underlying the therapeutic effects of HCP in UC through omics analysis method. A total of 724 different metabolites and 246 differential lipids were identified. Through metabolomic analysis, six metabolic pathways including the linoleic acid metabolic pathway, caffeine metabolic pathway, mannose and fructose metabolic pathways, methyl histidine metabolic pathway and fatty acid biosynthesis, which were significantly associated with colon-related diseases. Subsequently, lipidomics analysis revealed that the metabolic pathways of α-linolenic and linoleic acid, fatty acid biosynthesis, and glycerolipid metabolism exhibited significant associations with serum lipid metabolism. These findings suggested that HCP had potential therapeutic effects in treating UC.
