Preparation of mint oil microcapsules by microfluidics with high efficiency and controllability in release properties

Du Yuhan,Mo Liangji,Wang Xiaoda,Wang Hongxing,Ge Xue-hui,Qiu Ting

Mint oil is a complex mixture of comparatively volatile and labile components, making it challenging to be efficiently encapsulated and retained in microcapsules using traditional approaches. Microencapsulation, by encapsulating the substance with certain shells, can effectively reduce the diffusion, evaporation and the reactions of the inside actives with the external environment. The microencapsulation of mint oil and other aromatic actives have been a prevailing method to improve the stability, prolong the storage time, and enhance the controllability of the actives. Monodisperse core–shell microcapsules based on double emulsions prepared by microfluidic technology with controllable structure, pH-responsive ability, and adjustable release properties are successfully developed. Different flow rates, glutaraldehyde concentration, solidification time and pH have been used to adjust the core sizes, the shell thickness, the crosslinking density and the shrinkage degree of the shell. By monitoring their mint oil sustained release ability through an ultraviolet spectrophotometer, appropriate conditions to form the mint oil microcapsules with high encapsulation efficiency and controllability in release properties are achieved, building the foundation for better mint oil storage, transportation, and their applications.

The preparation of peppermint oil/2-hydroxypropyl-β-cyclodextrin/chitosan composite microcapsule and their prolonged retaining ability

Ge Xue-hui,Du Yu-han,Huang Xiaolian,Mo Liangji,Wang Hongxing,Qiu Ting

In this paper, the peppermint oil/HP-β-CD (2-hydroxypropyl-β-cyclodextrin)/chitosan composite microcapsules were prepared. The release ability of these microcapsules was improved for peppermint oil through the combination of molecular encapsulation by HP-β-CD and physical encapsulation by chitosan shell. First, stable peppermint oil/HP-β-CD pre-emulsion was prepared by stirring, based on which the optimal conditions were determined for producing the pre-emulsions with the highest encapsulation efficiency. Then, the pre-emulsion was moved into the flow-focusing microfluidic device as the inner phase and was cut into droplets by chitosan aqueous solution as the middle phase and the octanol as the outer phase to obtain templated emulsions. They were collected using a Petri dish with n-octane coagulation bath containing glutaraldehyde and 2 wt% span80 to be cured into microcapsules. The crosslinking time and the ratio of HP-β-CD in the templated emulsions were adjusted to investigate their impact on the microcapsule structures and correspondingly the capability of the microcapsules to retain peppermint. It was found out that the extended crosslinking time and the addition of HP-β-CD can improve the capability of the composite microcapsules to retain peppermint oil. It provides a straightforward method to produce composite microcapsules. Besides, the microcapsules with improved retaining ability were obtained by combing molecular and physical encapsulation, thus laying a foundation for the exquisite, efficient and polylaminate protection of the volatile perfume or other physically or chemically active components.

Fabrication of PGFE/CN-stabilized β-carotene-loaded peppermint oil nanoemulsions: Storage stability, rheological behavior and intelligent sensory analyses

Hujun Xie,Yutong Zhang,Mengna Cao,Chengzhi Liu,Yuezhong Mao,Gerui Ren,Zunyi Wu,Sheng Fang,Shiyi Tian,Di Wu

High stability nanoemulsions emulsified by combination of tripolyglycerol monostearate (PGFE) and casein (CN) were fabricated for loading functional components of peppermint oil and β-carotene, and the physicochemical properties, storage stability and sensory characteristics were explored. The β-carotene-loaded peppermint oil nanoemulsions exhibited fine physical stability when the mass ratio of PGFE/CN was less than 0.5. The β-carotene nanoemulsions were pseudoplastic fluid since the flow behavior index was lower than 1, based on the Herschel-Bulkley model. Intelligent sensory analyses clarified that electronic sensory evaluation methods, including colorimeter, electronic nose and electronic tongue, are convenient and effective to distinguish various nanoemulsions with different ratios of mixing emulsifiers. The formation mechanism of nanoemulsions was further explored to reveal the important role of hydrogen bonding and electrostatic interactions to yield nanoemulsions.

Preparation of zein-lecithin-EGCG complex nanoparticles stabilized peppermint oil emulsions: Physicochemical properties, stability and intelligent sensory analysis

Hujun Xie,Fangfang Ni,Jian Gao,Chengzhi Liu,Jieyu Shi,Gerui Ren,Shiyi Tian,Qunfang Lei,Wenjun Fang

Peppermint oil emulsions were prepared by using zein-lecithin-EGCG (Z-L/E) complex nanoparticles as emulsifiers. The preparation conditions of emulsions were optimized via measuring the particle size, surface tension and stability of emulsions, and peppermint oil of 3% (particle size = 375 nm, polydispersity index (PDI) = 0.45), the zein:lecithin ratio of 4:1 ( w/w ) (particle size = 396 nm), and the zein:EGCG ratio of 10:1 ( w/w ) (surface tension = 47.32 N/m) was the optimal condition. The rapid stability analysis showed that the instability mechanism of emulsions was ascribed to creaming and stratification, and the stability mechanism of emulsions was explored, indicating that the complex nanoparticles adsorbed on the surface of oil droplets to give Pickering emulsions. Electronic tongue experiments showed that the Z-E/L 4:1 stabilized emulsion was distinguished from the other three samples due to its good stability. The electronic nose experiment could distinguish the emulsions with different droplet sizes.

Phytosterol-mediated glycerosomes combined with peppermint oil enhance transdermal delivery of lappaconitine by modulating the lipid composition of the stratum corneum

Ruan Hang,Shen Lina,Hou Xiaolin,Li Jiaqi,Guo Teng,Zhu Chunyun,Feng Nianping,Zhang Yongtai

Although the introduction of glycerosomes has enriched strategies for efficient transdermal drug delivery, the inclusion of cholesterol as a membrane stabilizer has limited their clinical application. The current study describes the development and optimization of a new type of glycerosome (S-glycerosome) that is formed in glycerol solution with β-sitosterol as the stabilizer. Moreover, the transdermal permeation properties of lappaconitine (LA)-loaded S-glycerosomes and peppermint oil (PO)-mediated S-glycerosomes (PO-S-glycerosomes) are evaluated, and the lipid alterations in the stratum corneum are analyzed via lipidomics. The LA-loaded S-glycerosomes prepared by the preferred formulation from the uniform design have a mean size of 145.3 ± 7.81 nm and an encapsulation efficiency of 73.14 ± 0.35%. Moreover, the addition of PO positively impacts transdermal flux, peaking at 0.4% (w/v) PO. Tracing of the fluorescent probe P4 further revealed that PO-S-glycerosomes penetrate deeper into the skin than S-glycerosomes and conventional liposomes. Additionally, treatment with PO-S-glycerosomes alters the isoform type, number, and composition of sphingolipids, glycerophospholipids, glycerolipids, and fatty acids in the stratum corneum, with the most notable effect observed for ceramides, the main component of sphingolipids. Furthermore, the transdermal administration of LA-loaded PO-S-glycerosomes improved the treatment efficacy of xylene-induced inflammation in mice without skin irritation. Collectively, these findings demonstrate the feasibility of β-sitosterol as a stabilizer in glycerosomes. Additionally, the inclusion of PO improves the transdermal permeation of S-glycerosomes, potentially by altering the stratum corneum lipids. Graphical Abstract

Electrospun PVA/gelatin based nanofiber membranes with synergistic antibacterial performance

Sirui He,Lei Jiang,Jia Liu,Jie Zhang,Wei Shao

Synergistic antimicrobial action as a new strategy against various bacteria is attractive in the development of novel antibacterial materials. Based on this, the combination of peppermint oil (PO) and amoxicillin (AM) was constructed in order to achieve a synergistic antimicrobial effect with small amount of antibiotic addition. Then the synergistic antimicrobial PO and AM were introduced into polyvinyl alcohol/gelatin matrix to fabricate excellent antibacterial nanofiber membranes by electrospinning technique. Their morphologies and compositions of the prepared nanofiber membranes were investigated. The antibacterial ability and the antibacterial mechanism of the fabricated nanofiber membranes were determined. The co-incorporation of PO and AM into the nanofiber membranes showed strong antimicrobial activity with good cytocompatibility on intestinal epithelioid cell line 6. Therefore, this combination of PO and AM loaded nanofiber membrane as antibacterial material with synergistic antibacterial mechanism is very promising in the various antibacterial and bacterial inhibition applications.

Controllable prepared PDMS/SiO2/PVDF membrane for the separation of gaseous peppermint aromatic water

Qin Liu,Yan Wang,Zhi Guo,Siyuan Wu,Wancheng Li,Chuanrun Li,Bo Wu

One of the main challenges in oil–water separation of traditional Chinese medicines (TCM) is to obtain essential oils from the aromatic water of TCM. In this study, silicon dioxide/polyvinylidene fluoride (SiO 2 /PVDF) membranes were prepared using nonsolvent induce phase separation. Then polydimethylsiloxane (PDMS) was coated to obtain PDMS/SiO 2 /PVDF membranes. Separated essential oils and water from aromatic water in the gaseous state by vapor permeation membrane separation technology. The relationship between membrane structure and membrane separation effect was investigated. Response surface methodology was used to develop a quadratic model for the separation factor, membrane permeation separation index and membrane preparation process. The optimal process parameters for the membrane separation were 12.31% (mass) concentration of PVDF solution, 9.6% (mass) of N,N -dimethylacetamide in the solidification bath, and 0.2 g hydrophobic nano-SiO 2 incorporation, with a separation factor of 14.45, and a membrane flux of 1203.04 g·m –2 ·h –1 . Compared with the PDMS/PVDF membranes, the separation factor and membrane flux were increased by 68.59% and 3.46%, respectively. Compared with the SiO 2 /PVDF membranes, the separation factor and membrane flux were increased by 478% and 79.33%, respectively. Effectively mitigated the limitations of traditional polymer membrane material performance affected by the “trade-off” effect. Attenuated total internal reflection-Fourier transform infrared spectroscopy, contact angle, scanning electron microscopy and energy dispersive spectroscopy were used to characterize the PDMS/SiO 2 /PVDF membranes, and gas chromatography was used to characterize the permeate. In addition, the contents of L -menthol, L -menthone, menthyl acetate and limonene in the permeate, conformed to the European Pharmacopoeia standards . This study provided an effective preparation strategy of a feasible hydrophobic powder polymer membrane for the separation of essential oils from gaseous peppermint aromatic water.
