In order to improve the ability of starch to absorb menthone, corn starch was modified by enzymatic treatment (amyloglucosidase and α-amylase) combined with octenyl succinic anhydride (OSA) esterification. The oil absorption rate of starch modified by enzymatic treatment followed by OSA (P-OSA) reached 101.33%, whereas that of samples with reverse action sequences (OSA-P) was only 59.67%. The degree of substitution of OSA-P was also generally lower than that of P-OSA. At high OSA addition, OSA-P had a smaller specific surface area with fewer pores because octenyl succinic (OS) groups impeded the enzymatic treatment. Compared with OSA-P, the lamellar structure of P-OSA is sparser and less ordered. Owing to its pores, P-OSA was beneficial for the reaction to occur inside the granules, which was observed by Raman spectroscopy and laser confocal microscopy. At high OSA addition, the loading of P-OSA to menthone could reach 64.34 mg/g.

Objective The characteristic constituents of essential oils from aromatic plants have been widely applied as antimicrobial agents in the last decades. However, their mechanisms of action remain obscure, especially from the metabolic perspective. The aim of the study was to explore the antimicrobial effect and mechanism of menthone, a main component of peppermint oil, against methicillin resistant Staphylococcus aureus (MRSA).Methods An integrated approach including the microbiology and the high-coverage lipidomics was applied. The changes of membrane properties were studies by the fluorescence and electron microscopical observations. The lipid profile was analyzed by ultra-high performance liquid chromatography coupled with quadruple Exactive mass spectrometry (UHPLC-QE-MS). The lipid-related key targets which were associated with the inhibitory effect of menthone against MRSA, were studied by network analysis and molecular docking. Results Menthone exhibited antibacterial activities against MRSA, with minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) of 3,540 and 7,080 μg/mL, respectively. The membrane potential and membrane integrity upon menthone treatment were observed to change strikingly. Further, lipids fingerprinting identified 136 significantly differential lipid species in MRSA cells exposed to menthone at subinhibitory level of 0.1× MIC. These metabolites span 30 important lipid classes belonging to glycerophospholipids, glycolipids, and sphingolipids. Lastly, the correlations of these altered lipids, as well as the potential metabolic pathways and targets associated with menthone treatment were deciphered preliminarily. Conclusion Menthone had potent antibacterial effect on MRSA, and the mechanism of action involved the alteration of membrane structural components and corresponding properties. The interactions of identified key lipid species and their biological functions need to be further determined and verified, for the development of novel antimicrobial strategies against MRSA.

This study evaluated the effects of three extraction methods, hydrodistillation (HD), steam distillation (SD), and lipophilic solvent extraction (LSE), on the yield, chemical composition, and in vitro / in silico biological activities of Schizonepeta tenuifolia (Benth.) Briq essential oil (STEO). The highest yield of STEO was obtained by HD (0.750±0.040%), followed by SD (0.440±0.020%) and LSE (0.350±0.030%). Although their contents varied, menthone and pulegone predominated in HDEO, SDEO, and LSEO. HDEO displayed the strongest antioxidant ability, with a 2,2-diphenyl-1-picrylhydrazyl (DPPH) IC50 value of 14.164±0.090 mg/mL and 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulfonate) (ABTS) value of 0.326±0.023 mg/mL. SDEO demonstrated the highest antimicrobial activity against Staphylococcus aureus and Escherichia coli and HDEO the highest potent anti-inflammatory activity. The molecular docking of menthone and pulegone demonstrated strong binding to inflammatory targets, including prostaglandin-endoperoxide synthase 1 (PTGS1) and 2 (PTGS2) and tumor necrosis factor alpha (TNF-α). In short, the extraction method significantly affected the yield, composition, and biological activity of STEO.

In this study we sought to elucidate the therapeutic effects of fenchone on constipation-predominant irritable bowel syndrome (IBS-C) and the underlying mechanisms. An IBS-C model was established in rats by administration of ice water by gavage for 14 days. Fenchone increased the reduced body weight, number of fecal pellets, fecal moisture, and intestinal transit rate, and decreased the enhanced visceral hypersensitivity in the rat model of IBS-C. In addition, fenchone increased the serum content of excitatory neurotransmitters and decreased the serum content of inhibitory neurotransmitters in the IBS-C rat model. Meanwhile, western blot and immunofluorescence experiments indicated that fenchone increased the expressions of SCF and c-Kit. Furthermore, compared with the IBS-C model group, fenchone increased the relative abundance of Lactobacillus , Blautia , Allobaculum , Subdoligranulum , and Ruminococcaceae_UCG-008 , and reduced the relative abundance of Bacteroides , Enterococcus , Alistipes , and Escherichia-Shigella on the genus level. Overall, fenchone ameliorates IBS-C via modulation of the SCF/c-Kit pathway and gut microbiota, and could therefore serve as a novel drug candidate against IBS-C.

s Background Exploring effective and eco-friendly corrosion inhibitors is critical for Metal protection against an aggressive environment. This work investigated the performance of a newly synthesized menthone (5-methyl-2-(1-methyl-ethyl) cyclohexanone) and chitosan Schiff base for Q235 carbon steel in 1.0 M HCl. Methods The newly synthesized structure of MCS is confirmed using the FT-IR and 1H NMR spectrum. The inhibitor performance was evaluated using the weight loss. technique across a 298 – 328 K temperature range, electrochemical Impedance Spectroscopy (EIS) and Potentiodynamic Polarization (PDP). SEM, AFM images, and X-ray photoelectron spectroscopy is employed to analyze the Q235 carbon steel Surface. Additionally, the adsorption behavior of MCS was studied using the Langmuir model isotherm and computational studies, including DFT, Fukui functions, and molecular dynamics simulations. Significant findings The results demonstrated the effectiveness of the MCS inhibitor. Achieving the highest inhibition efficiency of 95.9 % at an optimized concentration of 600 mg. L -1 . The electrochemical investigations suggest a mixed mechanism for the inhibitor. Surface analysis indicates that the adsorption of MCS molecules on the metal surface reduces corrosion. The adsorption mechanism of the inhibitor on the surface of the Fe metal occurs through physio-chemisorption, as inferred from the Gibbs free energy (ΔG ads ) value of -34 kJ. mol -1. Molecular dynamics simulations confirmed the stable adsorption of the inhibitor on the Fe (110) surface, with an adsorption energy of –941.564 kcal/mol for the neutral form.

Rapid and sensitive detection of volatile compounds in complex matrices remains challenging due to limited ionization efficiency. Herein, we developed a novel hybrid ionization source by coaxially integrating low-temperature plasma with desorption electrospray ionization (LTP-DESI). The LTP-DESI hybrid source was equipped with a triple quadrupole mass spectrometer to analyze volatile aroma compounds. Under the LTP-DESI mode, volatile aroma compounds exhibited significant signal enhancements, with intensities increased by up to 2 orders of magnitude compared with DESI or LTP. This enhancement arises from a three-stage synergistic mechanism: (1) droplet and plasma-driven desorption, (2) primary ionization by both charged droplets and plasma species, and (3) synergistic ionization enhancement via plasma–droplet interaction. The spatial and temporal overlap of the charged droplet and plasma plume significantly promotes [M + H]+ generation through Penning ionization, proton transfer and charge exchange under atmospheric pressure. The quantitative and qualitative capability of the LTP-DESI-MS platform was validated through analyses of commercial perfumes and banana tissues, achieving recoveries of 75.3–108.5% with relative standard deviations of 2.7–17.1%. These results highlight the potential of the LTP-DESI system as a robust, practical technique for rapid ambient analysis of aromas in diverse complex samples.

BACKGROUND Alternaria alternata is a globally distributed plant pathogen infecting >400 plant species. Trans -2-decenal is a notable plant-derived secondary metabolite with strong volatility and antifungal properties, although its specific activity and mode-of-action against A. alternata remain unclear. RESULTS Toxicity assays showed that trans -2-decenal inhibited A. alternata with a median inhibitory concentration (IC 50 ) of 12.23 mg L −1 . Treated with trans -2-decenal resulted in hyphal abnormalities, reduced spore production and suppressed spore germination. Mechanistically, it disrupted fungal cell membrane integrity, as confirmed by propidium iodide staining and increased leakage of DNA and soluble proteins in conductivity assays. Further investigations revealed that trans -2-decenal disrupted redox homeostasis by inducing reactive oxygen species (ROS) accumulation. DCFH-DA fluorescence staining showed elevated ROS levels, whereas antioxidant enzyme assays revealed dose-dependent changes, including decreased superoxide anion (O 2 . − ) and catalase (CAT) activity, and significantly increased hydrogen peroxide (H 2 O 2 ), superoxide dismutase (SOD) and malondialdehyde (MDA) levels, collectively indicating oxidative stress and damage. Transcriptome analysis identified 2319 differentially expressed genes (DEGs), with gene ontology (GO) enrichment highlighting inhibition of pathways associated with oxidative stress response, transmembrane transport and ribosome biogenesis. Molecular docking further suggested interactions between trans -2-decenal and key antioxidant enzyme and membrane transporter of A. alternata . In vivo assays showed that 32 mg L −1 trans -2-decenal fumigation achieved protective and curative efficacies of 97.14% and 96.54%, respectively, against yam leaf spot disease. CONCLUSION Trans -2-decenal inhibited A. alternata through a multitarget synergistic mechanism, providing theoretical support for its development as a plant-derived biofumigant and a promising tool for the green management of A. alternata -induced diseases. © 2025 Society of Chemical Industry.

BACKGROUND Insects use odor detection to sense their surroundings. Use of volatile compounds, such as essential oils (EOs) of plants, to repel pests and disrupt their olfaction-driven behaviors has great practical potential for use in integrated pest management. Despite the available information on the repellent effects of EOs on herbivorous insects, the olfaction-based mechanisms remain unknown. RESULTS Y-tube olfactometer tests showed that the EOs of three Lamiaceae plants – Mentha arvensis L., Mentha piperita L. and Lavandula angustifolia Mill. – were significantly repellent to winged cotton aphid, Aphis gossypii Glover. Electrical penetration graph (EPG) tests indicated the EOs reduced phloem feeding and increased the level of non-productive probing by the aphids. The EOs also reduced the fecundity of winged Aphis gossypii . Electrophysiological bioassays and gas chromatography–mass spectrometry (GC–MS) identified five physiologically active volatiles, that is menthone, isomenthone, neomenthol and menthol from Mentha piperita ; menthone and menthol from Mentha arvensis ; and linalool from L. angustifolia . Behavioral tests confirmed that all five compounds repelled winged Aphis gossypii . Under field conditions, the growth rate of aphid populations after 7 days was significantly lower in fields treated with these compounds than in the control fields. CONCLUSION Our findings demonstrated that three EOs not only repelled winged Aphis gossypii but also interfered with the aphid's feeding behavior and reduced its fecundity. These EOs and their active constituents have great potential as eco-friendly control products for use against Aphis gossypii . The effects of these EOs also exceed other repellents that only keep pests away from host plants. © 2024 Society of Chemical Industry.

The formation and stabilization mechanisms of inclusion complexes (ICs) between β-cyclodextrin (β-CD) and five C10 aroma molecules were simultaneously investigated using experimental methods and molecular dynamics (MD) simulation. XRD, thermodynamic, FTIR and NMR analyses confirmed the formation of inclusion complexes, which had a new crystal structure and better thermal stability. MD simulation based on the molecular level including molecular structure (RMSD and Rg), solubility (RDF and SASA), interaction energy (Coul, vdW and H-bonds), and solvation and binding free energy , further clarified the properties of the inclusion complexes. The aroma molecules cause conformational changes in the inclusion complexes. The solubility of the inclusion complexes was lower than that of β-CD. The main driving force for the spontaneous entrance of aroma molecules into the cavity is van der Waals interactions. H-bonds are closely related to conformational changes, interactions and solubility.
