Simultaneous determination of umbelliferone and scopoletin in Tibetan medicine Saussurea laniceps and traditional Chinese medicine Radix angelicae pubescentis using excitation-emission matrix fluorescence coupled with second-order calibration method

Li Wang,Hai-Long Wu,Xiao-Li Yin,Yong Hu,Hui-Wen Gu,Ru-Qin Yu

A chemometrics-assisted excitation-emission matrix (EEM) fluorescence method is presented for simultaneous determination of umbelliferone and scopoletin in Tibetan medicine Saussurea laniceps (SL) and traditional Chinese medicine Radix angelicae pubescentis (RAP). Using the strategy of combining EEM fluorescence data with second-order calibration method based on the alternating trilinear decomposition (ATLD) algorithm, the simultaneous quantification of umbelliferone and scopoletin in the two different complex systems was achieved successfully, even in the presence of potential interferents. The pretreatment is simple due to the “second-order advantage” and the use of “mathematical separation” instead of awkward “physical or chemical separation”. Satisfactory results have been achieved with the limits of detection (LODs) of umbelliferone and scopoletin being 0.06   ng   mL −   1 and 0.16   ng   mL −   1 , respectively. The average spike recoveries of umbelliferone and scopoletin are 98.8   ±   4.3% and 102.5   ±   3.3%, respectively. Besides, HPLC-DAD method was used to further validate the presented strategy, and t- test indicates that prediction results of the two methods have no significant differences. Satisfactory experimental results imply that our method is fast, low-cost and sensitive when compared with HPLC-DAD method.

Phytochemical Analysis, Antioxidant and Analgesic Activities of Incarvillea compacta Maxim from the Tibetan Plateau

Jiajia Guo,Dan Zhang,Chao Yu,Ling Yao,Zhuo Chen,Yanduo Tao,Weiguo Cao

Incarvillea compactaMaxim is a traditional Tibetan plant widely used to treat rheumatic pain and bruises. We conducted qualitative analyses by liquid chromatography-mass spectrometry and quantitative analyses of the total phenols, flavonoids, and alkaloids content of different extracts ofI. compactaMaxim. Antioxidant and analgesic activity were analyzed. The results showed that the methanol extract had the highest content of the various ingredients. A total of 25 constituents were identified, of which compounds1–23were found for the first time in this plant. The water extract had the highest capacity to clear free radicals in the antioxidant test. The water extract had dose-dependent analgesic effects in the first and second phase in a formalin test. The latency of pain from a hot-plate test was augmented by the water extract when the dose was greater than or equal to 30 g/kg. The water extract significantly decreased the amount of writhing in a dose-dependent manner compared with the control group in the acetic acid-induced writhing test. These results showed thatI. compactaMaxim is a new antioxidant and analgesic agent, and this study provides information on its ingredients for further study.Keywords:LC-MS;anti-oxidation;analgesia;formalin test;Tibetan plant

Chemical Composition and Anti-arthritic Potential of Himalayan Marmot Oil Against Type II Collagen-induced Rat Rheumatoid Arthritis

Jie Zhou,Weilong Li,Xina Yu,Ye Wu,Pei Luo,Qingquan Kong,Zhifeng Zhang

BackgroundAnimal-derived remedies are prominent in folk medicine worldwide, and discovering potent anti-inflammatory agents is attractive for rheumatoid arthritis (RA) treatment. Himalayan marmot oil (HMO), a traditional Tibetan medicine topically or orally used for burns and arthritis, has been common in Tibetan areas for centuries. However, the composition and systematic pharmacological evaluation of HMO are still unclear.PurposeThis study focused on the chemical composition and anti-inflammatory effects of HMO in vitro and in vivo to expand the topical value of the application of HMO in RA.Materials and MethodsFirst, the chemical composition of HMO was explored by gas chromatography–mass spectrometry (GC–MS). Then, we established a bioassay system for L929 cells and RAW 264.7 cells to evaluate the effect of HMO on cytotoxicity and inflammatory responses. The anti-arthritic evaluation involved testing for paw swelling, swelling inhibition, and histological analysis in collagen-induced arthritis (CIA) rats. Enzyme-linked immunosorbent assay, Western blot analysis, and quantitative real-time polymerase chain reaction were performed to determine the effect of HMO on the production and secretion of inflammatory cytokines.ResultsGC–MS results showed that seven categories of chemical substances, including 261 compounds in HMO, and the main components of HMO were saturated fatty acids and unsaturated fatty acids. HMO showed low toxicity in cells according to the CCK-8 tests. In CIA model rats, HMO obviously inhibited hind paw swelling and reduced the arthritis index. Pathological staining results and micro-CT imaging indicated that HMO attenuated synovial hyperplasia and inflammatory cell infiltration in joint tissues. Meanwhile, in vivo and in vitro, HMO significantly inhibited interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) levels, while the expression of interleukin-10 (IL-10) was increased.ConclusionThe current work identified an excellent anti-inflammatory animal-derived medicinal agent, HMO, in alleviating synovial inflammation and joint destruction by regulating IL-6, IL-10, and TNF-α expression with low toxicity, which provides a protective effect against RA. Our study expands the topical value of the application of HMO to inhibit the progression of inflammation, and it could be a safe, novel RA complementary therapy.

An immobilization enzyme for screening lipase inhibitors from Tibetan medicines

Jia Liu,Run-Tian Ma,Yan-Ping Shi

With the increasing demand for lipase inhibitors and new drugs used in the clinical treatment of obesity, it is of great significance to screen lipase inhibitors from traditional Chinese medicines (TCMs) via capillary electrophoresis. In this work, Fe 3 O 4 @TiO 2 nanoparticles was fabricated by solvothermal method and employed as an improved magnetic support to immobilize lipase through electrostatic interaction. By the method of transmission electron microscopy, fourier transform infrared spectroscopy and X-ray diffraction, the magnetic nanoparticles were characterized. The immobilized enzyme possessed advantages of a wider range for pH and temperature endurance, better storage stability and reusability. The kinetics performances of the immobilized lipase were studied. When p-Nitrophenyl palmitate (pNPP) was used as enzyme substrate, the Michaelis-Menten constant was calculated to be 2.51 mM and its inhibition constant for Orlistat was ascertained to be 13.41 μM. Ultimately, the established method was applied to lipase inhibitors screening from 6 Tibetan medicines with lipase inhibitory activity and Oxytropis falcate Bunge was screened out for its supreme lipase inhibitory activity. 11 compounds in the Oxytropis falcate Bunge were further screened, five compounds exhibited similar inhibitory activity to Orlistat, and one compound (kaempferol) presented better inhibitory activity than Orlistat, which is the most commonly used drugs to treat obesity in clinic. This work not only developed a method for new anti-obesity drugs discovery, but also provided inspiration for exploring new medicinal value of the TCMs.

3,5-Dimethyl-8-methoxy-3,4-dihydro-1H-isochromen-6-ol Attenuates Ulcerative Colitis via Targeting NLRP3 in Mice

Mengru Zhou,Xiaoyao Ma,Jiaqi Zhang,Xuan Lu

3,5-Dimethyl-8-methoxy-3,4-dihydro-1H-isochromen-6-ol (DMD) is a polyketide compound obtained from the endophytic fungus Penicillium sp. HJT-A-10 of Rhodiola tibetica. R. tibetica is a nourishing food and also used in traditional Chinese medicine and Xizang medicine. In dextran sulfate sodium (DSS)-induced ulcerative colitis (UC) mice, DMD significantly alleviated the pathological symptom of UC. Network pharmacology studies have shown that nucleotide-binding domain-like receptor family pyrin domain containing (NLRP) 3 is the primary target protein of DMD associated with anti-UC. In molecular biology studies, DMD suppressed the activation of NLRP3 and decreased the expression of downstream inflammatory proteins and pro-inflammatory cytokines in UC. The finding was further verified in knockout mice. DMD lost the effect of attenuating DSS-induced UC in NLRP3–/– mice. In conclusion, this study demonstrates that DMD reduces inflammatory response and balances the barrier integrity to attenuate UC via targeting NLRP3, and DMD is a potential natural agent or dietary supplement for attenuating UC.
