The oxidative stability of microalgae oil (Schizochytrium aggregatum) and its antioxidant activity after simulated gastrointestinal digestion: Relationship with constituents

Junwei Lv,Xianqing Yang,Haixia Ma,Xiao Hu,Ya Wei,Wanjun Zhou,Laihao Li

The objective of this study was to examine the chemical composition and oxidative stability of microalgae oil, also to explore in vitro bioaccessibility and antioxidant activity of microalgae oil after simulated gastrointestinal digestion. In total, more than 50 fatty acids were identified by GC–MS analysis, with both palmitic acid (38.3%) and DHA (34.5%) being identified as major fatty acids. The contents of total phenolics and flavonoids in the various solvent extracts were measured spectrometrically, and their amounts were 39.33 ± 0.34 µg gallic acid/g and 16.08 ± 4.3 µg rutin/g, respectively. HPLC analysis showed that the contents of β-carotene, α-tocopherols, β- and γ-tocopherols (not separated) and δ-tocopherols were 136 µg/100 g, 164.4 µg/g, 317.3 µg/g, and 43.2 µg/g, respectively. Concerning sterols, cholesterol was the principal sterol at 4210.5 mg/kg and the other six main sterols were campesterol (121.4 mg/kg), 24-methylene cholesterol (192.8 mg/kg), 24-methyl-colest-7-en-3β-ol (144.6 mg/kg), ergosterol (144.8 mg/kg), stigmasterol (260.1 mg/kg) and Δ7,24-stigmastadienol (150.5 mg/kg), respectively. The overall chemical properties of the tested oils indicated that microalgae oil had a great oil quality. A Schaal oven test was used to evaluate the oxidative stability of microalgae oil. Furthermore, in vitro simulated gastrointestinal digestion was performed, and the antioxidant ability of digestion oil was determined by using a 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging assay, a 3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical cation decolourisation activity assay, a reducing power assay, a β-carotene bleaching assay and an oxygen radical absorbance capacity (ORAC) antioxidant assay. The results showed that following simulated gastrointestinal digestion, microalgae oil displayed a good in vitro bioaccessibility and moderate antioxidant capacity. Thus, the antioxidant activity of the microalgae oil was mainly contributed by its abundant antioxidant constituents. Practical applications: Schizochytrium aggregatum oil is a good source of DHA and of effective, bioaccessible antioxidants. The DPPH radical is rather stable, and our study used a relatively fast method for measuring both the hydrophilic and lipophilic substances during in vitro antioxidant activity. The change of violet colour to yellow during the DPPH assay is thought to be due to the attribution of hydrogen atoms by antioxidant substances. The extent of decolourisation is a significant indicator of the sample's antioxidant ability. As shown in figure that the scavenging abilities of all the samples were well correlated with increasing concentrations, and the scavenging activity of BHT was higher than that of microalgae oil. At concentrations ranging from 0.5 to 10 mg/mL, the DPPH radical scavenging ability of digestion oil was determined at 6.8–64.9%. Lower IC 50 values indicated a higher radical scavenging activity. The IC 50 value of microalgae oil in our DPPH radical scavenging study was 5.76 mg/mL, and solutions of BHT had an IC 50 value of 0.56 mg/mL. Although microalgae oil was shown to be weaker than BHT, it demonstrated a fairly good scavenging ability. The DPPH radical scavenging abilities of digestion oil and BHT.

Ru nanoparticles supported on hydrophilic mesoporous carbon catalyzed low-temperature hydrodeoxygenation of microalgae oil to alkanes at aqueous-phase

Arif Ali,Chen Zhao

The processing of an energy carrier such as microalgae oil into valuable fuels and chemicals is quite promising. Aqueous-phase processing is suitable for this purpose because the separation of intrinsic water from the algae cell is difficult. In this study, we synthesized ruthenium (Ru) nanoparticles supported on highly hydrophilic mesoporous carbon to catalyze the quantitative hydrodeoxygenation (HDO) of microalgae oil to alkanes in a one-pot process at a low temperature (140 °C) in the aqueous phase. The mesoporous carbon was obtained by single-step calcination of starch and zinc chloride in nitrogen. The as-obtained carbon showed high surface areas and pore volumes, allowing high dispersion of Ru nanoparticles. The surface of the carbon material was rich in hydroxyl groups, as evidenced by X-ray photoelectron spectroscopy (XPS), infrared (IR) spectroscopy, and thermogravimetric analysis (TGA) measurements. As a result, the carbon material contacted preferably with the water phase versus the organic phase, improving the accessibility of substrates. On the other hand, the contact angle test results speculated the superior hydrophilic nature of mesoporous Ru/C (ZnCl 2 , starch) than commercial Ru/C. Both kinetics modeling and in situ IR monitoring in water revealed the superior performance of the hydrophilic mesoporous and hydrophilic Ru/C compared to a commercial Ru/C for the tandem hydrogenation of stearic acid and decarbonylation of stearyl alcohol. The herein designed hydrothermal carbon material was highly active, environmentally benign, sustainable, and recyclable material, and could be potentially used for other hydrogenation reactions in the aqueous phase.

Insight into low methoxyl pectin enhancing thermal stability and intestinal delivery efficiency of algal oil nanoemulsions

Yufeng Chen,Jipeng Gu,Yi Sun,Yicheng Ding,Xuan Yang,Siqi Lan,Jiayue Ding,Yuting Ding

BACKGROUND Algae oil has garnered widespread acclaim due as a result of its high purity of docosahexaenoic acid (DHA) and excellent safety profile. The present study aimed to develop stable nanoemulsions (NEs) systems containing DHA from algae oil through thermal sterilization by combining modified whey protein concentrate (WPC) with low methoxyl pectin (LMP), as well as to investigate the impact of LMP concentration on the thermal stability and the gastrointestinal delivery efficiency of DHA NEs. RESULTS The addition of LMP enhanced the stability of the emulsion after sterilization, at the same time as improving the protective and sustained release effects of DHA in the gastrointestinal tract. Optimal effect was achieved at a LMP concentration of 1% (10 g kg −1 sample), the stability of the emulsion after centrifugation increased by 17.21 ± 5.65% compared to the group without LMP, and the loss of DHA after sterilization decreased by only 0.92 ± 0.09%. Furthermore, the addition of 1% LMP resulted in a substantial reduction in the release of fatty acids from the NEs after gastrointestinal digestion simulation, achieving the desired sustained-release effect. However, excessive addition of 2% (20 g kg −1 sample) LMP negatively impacted all aspects of the NEs system, primarily because of the occurrence of depletion effects. CONCLUSION The construction of the LMP/WPC-NEs system is conducive to the protection of DHA in algae oil and its sustained-release in the gastrointestinal tract. The results of the present study can provide reference guidance for the application of algae oil NEs in the food field. © 2024 Society of Chemical Industry.

Extraction of microalgae oil and further production for second-generation biodiesel

He Taotao,Wang Fei,Feng Junfeng,Liu Yancong,Chen Yuwei,Guo Feihong,Liu Peng,Jiang Xiaoxiang,Jiang Jianchun

To efficiently convert the microalgae to valuable and renewable fuels, the microalgae oil was first extracted and then transformed into the second-generation biodiesel. Among all solvents that were used for the microalgae oil extraction, the polar single solvent system methanol achieved the optimal value (47.8% extraction yield) because the polar cell membrane was easily dissolved by the polar solvent. Hexadecanoic acid (38.54%) and docosanoic acid (54.39%) dominated in fatty acid of microalgae oil. In the catalytic deoxygenation of microalgae oil, the Co-Al nanosheet displayed a higher performance than Co/Al 2 O 3 because of its high Co content and lamellar structure, exposing massive Co sites with high dispersion and small particle size. Moreover, when the reaction temperature was raised to 240 °C, the 100% conversion and 100% alkane yield were obtained, and the further temperature rise in temperature led to the cracking reaction. Preparation of astaxanthin by lipase-catalyzed hydrolysis from its esters in a slug-flow microchannel reactor

Jianfeng Mei,Xiaoyun Zhao,Yu Yi,Yanlu Zhang,Xudong Wang,Guoqing Ying

Natural astaxanthin is widely used as a food and cosmetics additive because of its multiple biological activities. However, astaxanthin produced by Haematococcus pluvialis is generally esterified, and its activity is far less than that of free astaxanthin. Hydrolysis of astaxanthin esters to free astaxanthin by enzymes can overcome the drawbacks of chemical saponification methods. In this paper, a slug-flow microchannel reactor was constructed and tested in enzymatic hydrolysis of astaxanthin esters. The reactor consists of a “T” slug-flow generator, a stainless-steel microchannel, two constant-flow pumps, and a temperature controller. The reactor has the advantages of simple configuration and easy scale-up, and is suitable for two-phase biochemical reactions. Using the microchannel reactor, astaxanthin esters in H. pluvialis oil were efficiently hydrolyzed to free astaxanthin by lipase from Aspergillus niger . After hydrolysis, the content of free astaxanthin in H. pluvialis oil was 18.8 mg/L, 7.83-times higher than that before hydrolysis (2.13 mg/L). The hydrolysis rate reached 75.4 %. These results indicate that the microchannel reactor can be useful for the production of free astaxanthin from its esters.

Enhancing the oxidative stability of algal oil powders stabilized by egg yolk granules/lecithin composites

Junhua Li,Yimei Shen,Jiali Zhai,Yujie Su,Luping Gu,Cuihua Chang,Yanjun Yang

This study reported a powder formulation containing omega-3-rich algal oil emulsions stabilized by egg yolk granules (EYGs)/lecithin composites. The improved physical stability of the algal oil samples due to increasing pH and lecithin addition was beneficial to the oxidative stability through analysis of free radical scavenging activities, metal ion chelating activities, and the release of primary and secondary oxidation products during accelerated storage (12 days, 60 °C). In addition, the effect of three antioxidants, i.e. ascorbic acid (VC), ascorbyl palmitate (AP), and α-tocopherol (VE), on lipid oxidation was investigated. Results showed that antioxidant partitioning at different regions of the emulsion system influenced its ability to prevent oxidation with the effectiveness of AP (at the O/W interface) > VE (in the oil phase) > VC (in the aqueous phase). This study developed a new powder-based emulsion formulation for algal oils with superior oxidative stability as an alternative source of omega-3.

Microfluidic design of bio-nanocapsules for algal oil delivery: Particle stability, oxidative stability, and bioaccessibility

Yan Chen,Qinlin Zhang,Anqi Yang,Lexiang Zhang,Xing Chen,Liqiang Zou,Chemgmei Liu,Wei Liu

Algal oil is a sustainable source of docosahexaenoic acid (DHA), but its high susceptibility to oxidation and hydrophobic nature limits its applications. To address these challenges, biopolymer nanocapsules (40.87–169.27 nm) were fabricated from shellac and phospholipids using microfluidic-mediated co-precipitation for algal oil delivery. Nanocapsule size increased from 78.51 nm to 152.96 nm with higher poor solvent ratios and decreased from 169.27 nm to 61.65 nm as total fluid velocity increased. Additionally, particle size was influenced by the phospholipid fraction, ranging from 40.87 nm to 61.65 nm, with the smallest size achieved at a shellac-to-phospholipid ratio of 2:3. Nanocapsule composition further affected particle stability, oxidative stability, and DHA bioaccessibility. A higher phospholipid fraction enhanced stability against pH fluctuations and thermal stress, while all nanocapsules exhibited stability during centrifugation and static storage, retaining over 93.01% DHA after 28 days at 25 °C. During accelerated storage (37 °C, 20 days), nanocapsules reduced hydroperoxide levels by 5.44- to 3.30-fold and secondary oxidation products by 2.62- to 1.79-fold compared to pure algal oil, with higher shellac content providing superior oxidative protection. Furthermore, the nanocapsules enhanced DHA bioaccessibility by a factor of 5.43–8.23 compared to pure algal oil, achieving a maximum DHA bioaccessibility of 54.31% at the highest phospholipid level. These findings provide valuable insights into designing advanced nano-colloids for enhancing the stability and bioavailability of bioactive lipids.

Polyphenol-Enriched Protein Oleogels as Potential Delivery Systems of Omega-3 Fatty Acids

Gao-Shang Wang,Hong-Yu Chen,Guang-Xin Feng,Yang Yuan,Zhi-Li Wan,Jian Guo,Jin-Mei Wang,Xiao-Quan Yang

Omega-3 polyunsaturated fatty acids (n-3 FAs) are essential nutrients and are considered effective in improving human health. Recent studies highlight the importance of the combination of n-3 FAs and polyphenols for limiting the oxidation of n-3 FAs and exhibiting synergistic beneficial effects. Herein, we developed a novel formulation technology to prepare oleogels that could be used for the codelivery of n-3 FAs and polyphenols with high loading efficacy and oxidative stability. These oleogels are made from algal oil with polyphenol-enriched whey protein microgel (WPM) particles as gelling agents via simple and scalable ball milling technology. The oxidative status, fatty acid composition, and volatiles of protein oleogels during accelerated storage were systematically assessed by stoichiometry and gas chromatography–mass spectrometry. These results showed that protein oleogels could overcome several challenges associated with the formulation of n-3 oils, including long-term oxidative stability and improved sensory and textural properties. The protein oleogel system could provide an excellent convenience for formulating multiple nutrients and nutraceuticals with integrating health effects, which are expected to be used in the care of highly vulnerable populations, including children, the elderly, and patients.

Two Birds with One Stone: Enzymatic Degumming and Deacidification for Arachidonic Acid-Enriched Algae Oil Using PLA1@MCM-41-C8

Zhonghui Liu,Yi Zhang,Tieliang Liu,Qi Zhou,Yandaizi Zhou,Kheng-Lim Goh,Siew-Young Quek,Fenghong Huang,Mingming Zheng

Algae oil contains polyunsaturated fatty acids (PUFAs) that are beneficial to human health and development. Enzymatic refining is a sustainable approach that can improve the quality and nutritional value of algae oil. However, this approach involves the use of specific enzymes to remove unwanted impurities such as phospholipids (PLs) and free fatty acids (FFAs). This study proposes a novel immobilized phospholipase, immobilized phospholipase A1 (PLA1@MCM-41-C 8 ), which serves a dual purpose: it is effective for both degumming arachidonic acid (ARA)-enriched algal oil and its subsequent deacidification. The use of PLA1@MCM-41-C 8 achieved a degumming rate of 95.9% for crude algal oil, with an ARA retention rate of up to 97.3%. The acidity of the degummed oil decreased from 14.5 to 1.1 mg KOH∙g −1 , while simultaneously producing 29.2% high-value diacylglycerol (DAG). By comparing enzymatically degummed and deacidified ARA-enriched algae oil (EDDO) with crude ARA-enriched algae oil (CO), commercialized refining algae oil (CRO), it was found that enzymatic refining had minimal impact on antioxidant stability, fatty acid composition, and oil content. In particular, the flavor characteristics of algal oil before and after enzymatic refining with PLA1@MCM-41-C 8 remained unchanged and were dominated by the main volatile compounds: alcohols and aldehydes. Compared with free phospholipase A1 (PLA1), PLA1@MCM-41-C 8 exhibited significantly improved thermostability, pH tolerance, solvent tolerance, and long-term reusability. These characteristics make it a promising candidate for complex oil refining in industrial applications.

Microencapsulation of algae oil by complex coacervation of chitosan and modified starch: Characterization and oxidative stability

Hongyan Mu,Zhaoxia Song,Xin Wang,Deda Wang,Xiaoqing Zheng,Xiaodan Li

The formation of complex coacervation using chitosan and octenyl succinic anhydride modified starch (OSA starch) and microencapsulation of algae oil were investigated in this study. The zeta-potential, turbidity and coacervate yield were evaluated as a function of pH and the chitosan- OSA starch mass ratio. The highest coacervate yield was achieved at pH 6.0 with a chitosan to OSA starch ratio of 1:3 ( w/w ). Isothermal titration calorimetry (ITC) indicated favorable affinity (Ka = 1.51 × 10 5  M −1 ) between chitosan and OSA starch. The microcapsules yielded an encapsulation efficiency (EE) in the range of 42.8 ± 0.8%- 93.1 ± 1.2%, the loading capacity ranged between 30.4 ± 2.7% and 58.3 ± 1.3%. Fourier transform infrared spectroscopy (FT-IR) spectra and scanning electron microscopy (SEM) further confirmed the microencapsulation. In comparison with the bulk oil, the microencapsulated algae oil exhibited improved oxidative stability during storage.

3D-printable docosahexaenoic acid-rich algal oil emulsion gels functionalized by multi-shaped starch nanorods

Yuanliang Zhong,Minxuan Chen,Siyu Yao,Qingqing Zhu,Xiangli Kong,Huang Zhang,Donghong Liu,Enbo Xu

The poor stability and low bioavailability of docosahexaenoic acid (DHA) from algal oil restrict its application in functional foods, particularly in three-dimensional (3D) printing systems where both printing performance and lipid protection are required. Here, starch nanorods (SNRs) with prismatic, ellipsoidal, and spindle-shaped morphologies were prepared and employed to stabilize DHA-rich algal oil emulsion gels. The influences of SNR shape and concentration on emulsion stability, gel structure, rheology, and printability were evaluated. At 4 % ( w / v ), spindle-shaped SNRs from normal corn starch (labeled as N20) achieved nearly complete oil encapsulation, significantly suppressed water mobility and reinforced gel elasticity, resulting in pronounced shear-thinning and excellent self-supporting capacity. The N20-stabilized gel exhibited superior printing fidelity, forming stable 3D constructs without oil leakage. When incorporated into plant-based tuna analogues, the emulsion gel yielded products closely resembling real tuna in texture and color, while enriching DHA content in fatty acids to 43.54 %, far exceeding that of commercial tuna. These findings highlight the critical role of particle shape in modulating interfacial stabilization and multiscale gel properties, providing a practical route for designing functional inks in food 3D printing for developing plant-based seafood with both structural and nutritional enhancements.

Microalgal Oil from Schizochytrium sp. Prevents HFD-Induced Abdominal Fat Accumulation in Mice

Jinhui Yu,Yong Ma,Jie Sun,Liyuan Ran,Youwei Li,Ning Wang,Tao Yu,Wenting Gao,Wenbin Jia,Rujiao Jiang,Meihua Guo,Yuping Bi,Yingjie Wu

Objective: Dietary n-3 polyunsaturated fatty acids (PUFAs), especially eicosapentaenoic acids (EPA) and docosahexaenoic acid (DHA), are proved to be effective in obesity reduction. Microalgal oil (MO) is an important alternative source of n-3 PUFAs that effectively alleviates obesity. The aim of the present study was to explore the anti-obesity effects of microalgal oil from Schizochytrium sp. (SMO) and to compare the effects of 2 SMOs (SMO1 and SMO2) with different levels of purity of n-3 PUFAs on high fat diet (HFD)-induced obesity in male C57BL/6J mice. Methods: Mice were randomly divided into 5 groups: (1) regular chow (RC); (2) HFD; (3) HFD + fish oil (FO); (4) HFD + SMO1; and (5) HFD + SMO2. Body weight and food intake were weekly monitored. After 16 weeks of treatment, a glucose tolerance test (GTT) and an insulin tolerance test (ITT) were performed. Serum lipid profile, morphological changes in the liver and epididymal white adipose tissue (eWAT), and the mRNA expression of lipid metabolism–related genes were also examined. Results: SMO treatment significantly decreased HFD-induced abdominal fat accumulation, lowered the levels of triglycerides, cholesterol, and low-density lipoprotein, as did the positive control treated with FO. Morphological examination revealed a remarkable reduction in lipid droplet formation in the liver tissue and the particle size of eWAT. An alleviation of inflammation infiltration in eWAT caused by a high-fat diet was also observed. Real-time reverse transcription–polymerase chain reaction analysis examination confirmed that microalgal oil inhibited the gene expression of fatty acid synthase, sterol responsive element-binding protein-1c, and acetyl-CoA carboxylase but promoted that of hormone-sensitive lipase and lipoprotein lipase, carnitine palmitoyltransferase-1, and uncoupling proteins in the liver and eWAT. Moreover, similar anti-obesity effects were obtained with the same dosage but different purity of n-3 PUFAs.Conclusions: As an alternative n-3 PUFAs resource, dietary intake of SMO might be beneficial to prevent HFD-induced abdominal fat accumulation.
