Today India is the main producer of peppers especially the hot peppers , albeit mostly for domestic use. The thrips and mites, and the virus diseases transmitted by them along with fungal diseases like fruit rot, powdery mildew , bacterial wilt and leaf spots are the limiting factors in pepper productivity. The solution for managing these pests on a sustained basis exists in adopting eco-friendly approaches like using resistant cultivars. Fortunately huge natural genetic diversity exists in pepper and therefore, essential research efforts in finding out resistant sources and their utilization have been by and large dynamic and successful. Despite continuous scientific efforts there is a dire need for new cultivars with resistant traits for various pests suitable to varied climatic conditions, consumption and quality preferences all over the world. Efforts need to be intensified to find out useful genetic material and to introduce genes of resistance against insects, fungal and virus diseases into commercial cultivars. Genetic resources that have been used intensively in pepper breeding are for developing sweet peppers, hot peppers, bell peppers in various shapes, sizes and colors. Germplasm repositories at the World Vegetable Research Center (AVRDC) and United States Department of Agriculture (USDA) do possess number of genotypes resistant to insect pests , nematode, fungi, bacteria and virus diseases. At AVRDC resistant genotypes originating from several pepper growing countries to most virus diseases like Tobacco mosaic virus , Cucumber mosaic virus , Pepper veinal mottle virus , Chilli veinal mottle virus , Peanut bud necrosis virus etc., were identified from its rich pepper diversity. Exploitation of rich genetic diversity resulted in the development of new cultivars encompassing resistance to various pests and good agronomical traits. This has eventually resulted in meeting the demanding situations of diverse domestic as well as global production requirements. The chile database ( Chile Database, 2010 ) highlights the exemplary utilization of germplasm and development of large number of cultivars and hybrids resistant to most diseases and nematodes around the world. This paper also depicts information on Indian sources of resistance to thrips, mites, leaf curl complex, nematodes and diseases, while presenting the scope for exploitation of diversity available in the Indian National Gene Bank and other repositories all over the world. An attempt is also made to identify the gaps in the genetic diversity and cultivars against the biotic constraints and the augmentation efforts required to be initiated toward enrichment of the gene pool for domestic as well as global use.

Tribolium castaneum ( T. castaneum ) is a worldwide pest of stored grain that mainly harms flour, and not only causes serious loss of flour quality but also leads to deterioration of flour quality. Chemical detection plays a key role in insect behavior, and the role of odorant-binding proteins (OBPs) in insect chemical detection has been widely studied. However, the mechanism of OBPs in insect defense against exogenous toxic substances is still unclear. In this study, biochemical analysis showed that eugenol , the active component of A. vulgaris essential oil , significantly induced the expression of the OBP gene OBPC12 from T. castaneum ( TcOBPC12 ). The mortality of late larvae treated with eugenol was higher than that of the control group after RNA interference (RNAi) against TcOBPC12 , which indicates that the OBP gene is involved in the eugenol defense mechanism and leads to a decrease in sensitivity to eugenol. Tissue expression profiling showed that the expression of TcOBPC12 in the epidermis, hemolymph, and intestine was higher than in other larval tissues, and TcOBPC12 was expressed mainly in the epidermis, head, and fat body of adults. The developmental expression profile showed that the expression of TcOBPC12 in late eggs, early and late larval stages, and late adult stages was higher than in other developmental stages. These data suggest that TcOBPC12 may be involved in the absorption of exogenous toxic substances by the larvae from T. castaneum . Our results provide a theoretical basis for the metabolism and degradation mechanism of exogenous toxic substances and help explore more potential target genes of insect pests .

This study elucidates the molecular mechanism by which flavin-containing monooxygenase (FMO) mediates metabolic resistance to nereistoxin insecticides in lepidopteran pests. A field population of Spodoptera exigua exhibited 50-fold resistance with upregulated SeFMO expression. Using FMO-specific inhibitors, recombinant protein expression, and mass spectrometry, we confirmed that FMO catalyzes N-oxidation of nereistoxin insecticide at the tertiary amine nitrogen. Molecular docking revealed that insect FMO’s catalytic mechanism resembles that of human FMO. Transgenic Drosophila models demonstrated that the FMO-mediated N-oxidation enhances insecticide resistance, indicating evolutionary conservation. This highlights FMO’s role in insecticide detoxification and its conserved function across species, providing new insights into pest resistance mechanisms.

Simple SummaryPlant bugs’ locating of mates mainly relies on peculiar chemical signals known as sex pheromones, which can be utilized to develop environmentally friendly pest control methods. This study investigated the sex pheromones ofPolymerus pekinensis, a phytophagous insect of alfalfa crops in East Asia. By analyzing chemical extracts from both male and female insects, we identified two antenna-active compounds: octyl acetate (OA) and decyl acetate (DA). Females produced higher amounts of OA, while males produced greater quantities of DA. Field experiments demonstrated that lures loaded with solely OA strongly attracted maleP. pekinensis, making it a valuable tool for monitoring this pest. However, the addition of DA in larger amounts significantly decreased the attractivity of lures. Our findings offer new strategies for monitoring and managing these insects in a manner that does not harm non-target insects.Insect sex pheromones have been widely used in integrated pest control due to their efficiency, non-toxicity, specificity, and environmental sustainability. They are considered a key component of green pest management techniques.Polymerus pekinensisis a phytophagous plant bug on alfalfa (Medicago sativaL.) in East Asia. This study used gas chromatography–electroantennogram detection (GC–EAD) and gas chromatography–mass spectrometry (GC–MS) to analyze the whole-body extracts from male and femaleP. pekinensis. Octyl acetate (OA) and decyl acetate (DA) elicited the antennal response of males and were identified as the predominant components of female and male extracts, respectively. Subsequent field trials demonstrated that OA (>8 mg per lure) showed the strongest attraction to conspecific males. However, when DA was added in a lure (≥2 mg), a significant decline in captures occurred. These findings provide new insights into the understanding of sex pheromones in Miridae and benefit the development of sustainable management ofP. pekinensis.

Platycladus orientalis plants in China are experiencing a serious problem with wood-boring beetles, but conventional physical detection methods cannot perform well due to hidden larvae and less obvious symptoms in the early infestation. In this study, the volatile organic compounds (VOCs) of P. orientalis were investigated using gas chromatography-ion mobility spectrometry (GC-IMS) to identify its internal infestation. An image-based evaluation method of the degree of internal wood damage was proposed, the principal component analysis (PCA) based on specific markers was applied for natural clustering of samples, and unsupervised learning approaches based on specific markers and automatically located peaks were used to create predictive models for sample classification. The results showed that the herbivore-induced plant volatiles (HIPVs) of P. orientalis samples had two main characteristics compared with undamaged plants. One was the emergence of several compounds such as C 3 -C 6 ketones, aldehydes, and alcohols, while the other was the obvious content variation of several monoterpenes and their derivatives. The classification models based on specific markers and automatically located peaks could correctly classify at least 89.5% and 87.9% of the samples, respectively. In a word, these results proved that GC-IMS based approaches combined with different feature extraction methods have the potential to identify the internal infestation of wood-boring beetles in P. orientalis .

Phthalate esters (PAEs) were ubiquitous in agricultural soils and could be metabolized after being absorbed by crops, posing significant implications for crop yield and quality. We hypothesize that monophthalates (mPAEs), the hydrolyzed products of PAEs, might mimic phytohormone jasmonic acid (JA) to activate the JA signaling pathway, therefore enhancing the defense towards pests and inhibiting the rice plant growth. Taking dibutyl phthalate (DBP) as a representative PAE, our study discovered that DBP exposure significantly induced JA-related outcomes including decreased larval weight (9.58–18.8%), and rice biomass (11.7–34.2%). Under the conditions where the JA content remained unchanged, monobutyl phthalate (MBP), the hydrolyzed product of DBP, triggered the JA signaling pathway, evidenced by significantly upregulated genes encoding coronatine insensitive 1 (COI1) (1.56–1.73 fold), jasmonate ZIM-domain (JAZ) (4.33–7.71 fold), MYC2 transcription factor (2.07–2.87 fold), and promoted phytoalexins production in downstream signaling. MBP conjugated with isoleucine, and the conjugate subsequently mimicked a JA bioactivator (JA-isoleucine conjugate) to occupy the binding site of COI1-JAZ co-receptor protein, thereby initiating the JA signaling pathway. These JA-related outcomes and mechanism were consistently evidenced in rice exposed to other four typical PAEs, and the aliphatic chain length of selected PAEs indicated a negative contribution to these observations. In this study, we discovered a unconventional mechanism through which the transformation metabolites of PAEs elicit pest defense while simultaneously inhibiting rice growth, providing insights into the risk assessment of PAEs on crop yields and quality.

Mechanical stimuli can affect plant growth, development, and defenses. The role of water spray stimulation, as a prevalent mechanical stimulus in the environment, in crop growth and defense cannot be overlooked. In this study, the effects of water spray on tomato plant growth and defense against the chewing herbivore Helicoverpa armigera and necrotrophic fungus Botrytis cinerea were investigated. Suprathreshold water spray stimulus (LS) was found to enhance tomato plant defenses against pests and pathogens while concurrently modifying plant architecture. The results of the phytohormone and chemical metabolite analysis revealed that LS improved the plant defense response via jasmonic acid (JA) signaling. LS significantly elevated the level of a pivotal defensive metabolite, chlorogenic acid, and reduced the emissions of volatile organic compounds (VOCs) from tomato plants, thereby defending against pest and pathogen attacks. The most obvious finding to emerge from this study is that LS enhances tomato plant defenses against biotic stresses, which will pave the way for further work on the application of mechanical stimuli for pest management.

The whitefly Bemisia tabaci ( B. tabaci ) stands as the exclusive identified vector for tomato yellow leaf curl virus (TYLCV), the causative agent of tomato yellow leaf curl disease (TYLCD). Current disease management strategies face significant challenges due to the absence of effective antiviral chemicals, necessitating heavy reliance on whitefly population control. However, the occurrence of insecticide resistance and the invasiveness of B. tabaci have severely compromised conventional control methods, highlighting an urgent demand for innovative disease management approaches. This study presents a kind of LDH superpolymer as a nano-insecticide to control B. tabaci . It was generated through self-assembly of LDH SLNSs gel with supplement of matrine (MT) to form nanohybrids. We found that the 150 nm LDH nanosheets displayed a high efficiency to permeate tomato leaf stomata, enabling accumulation in intercellular cavities and mesophyll cell cytoplasm. The loading efficiency of MT-LDH nanohybrids achieved to 16 %. Compared to conventional MT formulations, foliar application of 500 mg/L MT-LDH increased 50.39–75.17 % phyllospheric MT retention in tomato leaves. The nanomaterial exhibited efficient showing 2.83-fold bioactivity against B. tabaci adults than that of aqueous MT solutions. Moreover, we revealed that the biocontrol activity of the MT-LDH nanohybrids conferred dual protective benefits: (1) activation of plant defenses to inhibit mitigation of TYLCV; and (2) enhanced systemic acquired resistance against viral infection. These findings establish a novel antiviral paradigm that both killed vector insect and activate host defence. The MT-LDH nanohybrids offered a potential solution to control TYLCV epidemics with reduced pesticide reliance.

Fungal diseases caused by Botrytis cinerea and insect infestations by aphids often occur simultaneously during plant growth. A co-delivery nanosystem for dual-pesticide may enhance pesticide efficiency and reduce the required dosage of pesticides through synergistic effects. Therefore, in this study, we developed a dual pH-responsive nanocomposite AZOX/Thi@BMMs@MIL-88B (ATBM) based on bimodal mesoporous silica (BMMs) and metal organic framework MIL-88B, which was loaded with fungicide azoxystrobin (AZOX) and insecticide thiamethoxam (Thi) to control grey mould and aphid pests. The loading capacities of the two pesticides in the nanocomposite were 18.56 % (AZOX) and 37.38 % (Thi). The nanoparticles (NPs) were spindle-shaped with a hydrated particle size of 771.60 ± 12.55 nm and possessed excellent pH-responsive controlled release behavior. The NPs also demonstrated good wettability and stability due to the entrapment of nanocarriers. Fluorescence tracking experiments verified that the NPs could be effectively absorbed and translocated by pathogenic fungal mycelium and pea seedlings. The bioactivities of ATBM against B. cinerea and A. pisum increased by 9.58 % (AZOX) and 39.48 % (Thi), respectively, compared to the commercialized technical control (TC). This nanocomplex promoted the growth of pea seedlings through the introduction of iron ions and by activating the oxidative stress system, and the excellent biosafety of NPs could minimize environmental risk. In summary, this hybrid bi-carrier dual-pesticide nanopesticide-fertilizer showed significant promise for application in sustainable agriculture.

The development of novel, safe, and efficient pest and disease control technologies for agricultural crops remains a pivotal area of research. In this study, by combining ZIF-8 and ZIF-90, a water-stable, pH-responsive bilayer MOF nanoparticle (NP) named Z8@Z90 was created, and tebuconazole (TEB) was added to form T@Z8@Z90, used for controlling peanut southern blight. The loading efficiency of TEB within the T@Z8@Z90 reached 26.15%, enabling rapid release in acidic environments triggered by oxalic acid (OA) secreted by Sclerotium rolfsii. In vitro experiments showed that T@Z8@Z90 can regulate the oxalic acid secretion of S. rolfsii and destroy its cell membrane structure. Additional experiments revealed that T@Z8@Z90 reduced sclerotial formation, decreased the total protein content of sclerotia, and influenced their sensitivity to pesticides, thereby mitigating the risk of reinfection by S. rolfsii. Notably, T@Z8@Z90 exhibited efficient translocation within peanut seedlings, being absorbed through the roots and transported to the leaves. At a concentration of 200 mg/L, T@Z8@Z90 exhibited high safety profiles for peanut seedling growth compared to the TEB suspension. Moreover, T@Z8@Z90 is safer for earthworms than TEB SC. Overall, this study offers valuable insights for the management of soil-borne diseases in agriculture and contributes to the advancement of sustainable agricultural practices.
