Absorption and utilization of polysaccharides from Cordyceps militaris fruiting bodies and their protective effects against CCl4-induced acute liver injury in mice

Lin Lan,Jingxin Wang,Hao Luo,Min Cao,Nianhong Guo,Guiling Fan,Shengyu Wang,Zaixing Sun,QiaoLing He,Yufeng Li

Cordyceps militaris , a medicinal and edible fungus, has garnered significant interest for its polysaccharides, known for their diverse physiological activities and health benefits. The purpose of this study was to extract polysaccharides (CM-1) from Cordyceps militaris fruiting bodies using hot water extraction and column chromatography, and evaluate its structural characteristics, antioxidant capacity, and in vivo absorption effects. Additionally, the study aimed to assess its protective effects against carbon tetrachloride (CCl 4 ) induced liver injury in mice. The results showed that CM-1 significantly decreased the expression levels of serum ALT, AST, TNF-α, IL-6, and IL-1β. Pre-treatment with CM-1 also increased the activities of superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GSH-Px). Furthermore, histopathological examination of liver tissues confirmed the protective effects of CM-1 against acute liver injury induced by CCl 4 . These results indicate CM-1 promising potential as a dietary supplement for liver injury prevention and treatment.

Bifidogenic effects of Cordyceps sinensis fungal exopolysaccharide and konjac glucomannan after ultrasound and acid degradation

Ang-Xin Song,Yu-Heng Mao,Ka-Chai Siu,Jian-Yong Wu

The bifidogenic effects of exopolysaccharide (EPS) of a medicinal fungus ( Cordyceps sinensis ) and a well-known food polysaccharide konjac glucomannan (KGM) with different molecular weight (MW) ranges were evaluated through in vitro experiments in liquid cultures of Bifidobacteria . Native EPS and KGM were partially degraded with power ultrasound (US) to improve the water solubility, and further hydrolysed with trifluoroacetic acid to much lower MW. The acid-hydrolysed fractions (EPS-AH and KGM-AH) supported the growth of all five tested bifidobacterial species, while the US-degraded high MW fractions, EPS-US and KGM-US, could only slightly support the growth of some species. All EPS fractions increased the acetic acid production of most bifidobacterial species. Most remarkably, the high MW EPS-US, EPS-AH and KGM-US fractions significantly enhanced the cell viability with much higher colony forming unit (CFU) counts, suggesting a protective effect of these high MW polysaccharides for the bacterial survival . The results have shown that MW was a significant factor on the bifidogenic properties of partially degraded EPS and KGM.

Omics data reveal the unusual asexual-fruiting nature and secondary metabolic potentials of the medicinal fungus Cordyceps cicadae

Lu Yuzhen,Luo Feifei,Cen Kai,Xiao Guohua,Yin Ying,Li Chunru,Li Zengzhi,Zhan Shuai,Zhang Huizhan,Wang Chengshu

Background Ascomycete Cordyceps species have been using as valued traditional Chinese medicines. Particularly, the fruiting bodies of Cordyceps cicadae (syn. Isaria cicadae ) have long been utilized for the treatment of chronic kidney disease. However, the genetics and bioactive chemicals in this fungus have been largely unexplored. Results In this study, we performed comprehensive omics analyses of C. cicadae , and found that, in contrast to other Cordyceps fungi, C. cicadae produces asexual fruiting bodies with the production of conidial spores instead of the meiotic ascospores. Genome sequencing and comparative genomic analysis indicate that the protein families encoded by C. cicadae are typical of entomopathogenic fungi, including the expansion of proteases and chitinases for targeting insect hosts. Interestingly, we found that the MAT1-2 mating-type locus of the sequenced strain contains an abnormally truncated MAT1-1-1 gene. Gene deletions revealed that asexual fruiting of C. cicadae is independent of the MAT locus control. RNA-seq transcriptome data also indicate that, compared to growth in a liquid culture, the putative genes involved in mating and meiosis processes were not up-regulated during fungal fruiting, further supporting asexual reproduction in this fungus. The genome of C. cicadae encodes an array of conservative and divergent gene clusters for secondary metabolisms. Based on our analysis, the production of known carcinogenic metabolites by this fungus could be potentially precluded. However, the confirmed production of oosporein raises health concerns about the frequent consumption of fungal fruiting bodies. Conclusions The results of this study expand our knowledge of fungal genetics that asexual fruiting can occur independent of the MAT locus control. The obtained genomic and metabolomic data will benefit future investigations of this fungus for medicinal uses.

Surface-carboxylated cobaltous oxide with oxidase-like activity for conveniently assessing total antioxidant capacity of Cordyceps militaris at neutral pH

Xuechao Xu,Juanli Yang,Qiao Guo,Lu Gao,Bo Wang,Suhui Tan,Kun Wang,Yun-xia Li,Shengyan Zhao,Yi Li,Zhen-quan Yang

Reassessment of total antioxidant capacity (TAC) in Cordyceps militaris is imperative for providing dietary guidance and facilitating health monitoring. Herein, surface-carboxylated cobaltous oxide (COOH-Co 3 O 4 ) nanozyme was achieved via a dual-stage procedure encompassing reduction preparation and surface modification. COOH-Co 3 O 4 exhibited exceptional oxidase-like activity under neutral pH conditions, facilitating conversion of dissolved oxygen into superoxide anion free radicals and subsequently catalyzing oxidation of colorless 3, 3′, 5, 5′-tetramethylbenzidine (TMB) to yield blue oxidized TMB (TMBox) at neutral pH. Additionally, antioxidants can eliminate O 2 · − produced in the above reaction system. Moreover, the absorbance at 652 nm of the COOH-Co 3 O 4  + TMB reaction system gradually diminishes with an increase in ascorbic acid concentration. Based on it, a colorimetric evaluation system (using ascorbic acid as a reference) for TAC of C. militaris was established. This innovative approach offers an impressively low limitation of detection (LOD) calculated as 0.78 μM. Furthermore, it was employed to evaluate TAC of real C. militaris samples, yielding a satisfactory recovery (96.1 %–102.3 %), thereby demonstrating its exceptional accuracy. The TAC of C. militaris extracted using different solvents was determined, indicating that the highest TAC is observed in C. militaris extracted by deionized water. This study not only offers a rapid, cost-effective, and convenient analytical tool for evaluating TAC of C. militaris but also expands the potential applications of nanozymes.

Protective Effect of Purified Polysaccharides from Cordyceps militaris Substrate on Acute Ethanolic Hepatotoxicity in Male Mice

Wu Yulong,Zhou Feng,Jiang Haitao,Wang Zhengjiong,Hua Chun,Wang Renlei

Two novel polysaccharides fractions were isolated from Cordyceps militaris substrate polysaccharides (CMSP) by DEAE-52 cellulose and Sephadex G-100 column chromatography, and named as CMSP-1 and CMSP-2. The CMSP-1 was a heteropolysaccharide mainly composed of sorbin, D-mannose, D-glucose, galactose, arabinose, with the molar ratios of 1.00:6.32:10.23:2.86:1.02, and the molecular weight of CMSP-1 was 10715.2 Da. The CMSP-1 was used to investigate the protective effect on acute ethanolic hepatotoxicity (AAH) in male mice. The results indicated that oral administration of CMSP-1 could significantly decrease the liver index, and increase body weight in mice with AAH. CMSP-1 could also significantly increase the levels of SOD, GSH and GSH-PX, and decrease the levels of ALT, AST, TG, VLDL and MDA in AAH model mice. Moreover, Histopathological examination of liver showed that CMSP-1 could significantly alleviate the symptoms of liver injury caused by ethanol, such as swelling of hepatocytes, loose cytoplasm and lipid changes. Therefore, CMSP-1 has obvious protective effect on AAH model mice, and has a good prospect in functional foods or natural medicine.

Impact of cell disruption technologies on the structure, functionality, and digestibility of Cordyceps militaris protein isolates

Houle Xiang,Yuqing Zhang,Shuo Zhang,Jingwei Kou,Hui Wang,Fengxia Zou,Wei Zhuang,Pengpeng Yang,Jinglan Wu,Hanjie Ying

Cell disruption is essential for extracting intracellular proteins from fungal biomass. However, its effects on the structural and functional properties of proteins remain insufficiently understood. This study used Cordyceps militaris as a model to evaluate three cell disruption techniques—high-pressure homogenization (HPH), ultrasonication (US), and cellulase hydrolysis—for extracting C. militaris protein isolates (CMPI). All methods significantly improved protein extraction compared to alkaline extraction alone, with HPH yielding the highest rate (70.75 %, p  < 0.05). Structural analysis revealed a decrease in α-helix and β-sheet content, along with increased β-turns, random coils, surface hydrophobicity, and modifications in disulfide–sulfhydryl balance. CMPI-HPH and CMPI-US showed superior water-holding capacities (6.23 and 5.95 g/g) and oil-holding capacities (9.97 and 7.70 g/g), as well as enhanced foaming (69.57 % and 65.22 %) and emulsifying activities (32.07 and 31.13 m 2 /g). CMPI-HPH also exhibited the highest foaming stability (87.50 %). In vitro digestibility was significantly greater in CMPI-US (80.05 %) and CMPI-HPH (71.71 %) than in the control ( p  < 0.05). These results demonstrate that cell disruption, especially HPH, effectively improves protein extraction efficiency, structural flexibility, and techno-functional properties, offering new potential for the sustainable application of fungal proteins in food systems.

Differential Analysis of Pomelo Peel Fermentation by Cordyceps militaris Based on Untargeted Metabolomics

Yannan Xiang,Siyi Tian,Xinyu Luo,Chenggang Cai,Yaowen Du,Hailong Yang,Haiyan Gao

The content of differentially abundant metabolites in the fermentation broth of grapefruit peels fermented byCordyceps militarisat different fermentation times was analyzed via LC–MS/MS. Small molecule metabolites and differential metabolic pathways were analyzed via multivariate analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment. A total of 423 metabolites were identified at 0, 2, 6, and 10 days after fermentation. Among them, 169 metabolites showed differential abundance, with significant differences observed between the fermentation liquids of every two experimental groups, and the metabolite composition in the fermentation liquid changed over the fermentation time. In summary, the upregulation and downregulation of metabolites in cancer metabolic pathways collectively promote the remodeling of cancer cell metabolism, facilitating increased glycolysis, alterations in TCA cycle flux, and enhanced biosynthesis of the macromolecules required for rapid proliferation and survival. This study provides new perspectives on the development of high-value-added agricultural and forestry byproducts and the development and research of functional foods.
