Selenium (Se) is an essential trace element for fish growth and performs its physiological functions mainly through incorporating into selenoproteins . It is well known that dietary Se regulates fish growth by controlling the synthesis of deiodinase , a kind of selenoproteins. However, recently, as many as 41 selenoproteins have been characterized in teleost fish. We propose a hypothesis if other selenoproteins, besides deiodinase, could also involve in the regulation of fish growth. In the present study, rainbow trout ( Oncorhynchus mykiss ) were fed diets supplemented with or without graded levels of Se (2, 4 or 6 mg/kg, from selenium yeast, Se-yeast) for 10 weeks. At the end of the feeding trial, fish growth and the expressions of a total of 28 selenoprotein genes in tissues have been evaluated. Results showed that dietary Se-yeast supplementation significantly increased fish growth ( P < 0.05) without causing oxidative stress in fish tissues. In addition, dietary Se supplementation caused a general up-regulation of selenoprotein gene expressions either in liver (10 genes) or in muscle (11 genes) ( P < 0.05). Correlation analysis showed that rainbow trout growth was significantly and positively correlated to only 4 selenoprotein genes in liver but all the 11 differentially expressed selenoprotein genes in muscle ( P < 0.05), and it presented the strongest correlation with the mRNA levels of muscle selenoprotein W-like gene ( P < 0.05). These results indicate that dietary Se-yeast supplementation is beneficial for rainbow trout growth, and the enhanced growth performance is closely correlated to the expressions of muscle selenoprotein genes, particularly the selenoprotein W-like gene. This study reveals the importance of muscle selenoprotein gene expressions, and provides a new concept for the regulatory mechanism of dietary Se on fish growth.

Lactic acid bacteriaLigilactobacillus salivariushas been shown to be a substitute for antibiotics in the treatment of bacterial disease in high animals. However, its beneficial mechanism in fish farming is still unclear. This study evaluated the antagonistic effects of theLigilactobacillus salivariusGX118 strain onAeromonas salmonicidaand its regulation of rainbow trout growth in vivo and in vitro. The results found that GX118 produces an antibacterial substance that can directly destroy the cell wall ofA. salmonicida. Whole-genome sequencing of GX118 revealed that Enterolysin A is a type III bacteriocin with antibacterial properties. An in vivo experiment showed that the supplementation of GX118 in diet competitively inhibited the colonization ofA. salmonicidain the intestine. In addition, it was able to improve the growth performance of rainbow trout within a 21-day feeding experiment. The supplementation of GX118 increased the diversity of gut microbiota, in which the abundance ofBacteroidota,Blautia, andRhodobacteraceaeincreased. In addition, the use of GX118 activated the expression ofIFN-γandNF-κBgenes and reduced the expression level ofIL-6andIL-8, thus exhibiting a certain effect on activating the immunity of rainbow trout. This study provides a scientific basis for the development of antibacterial probiotics in the healthy farming of rainbow trout.

Aeromonasspecies are among the main pathogens causing rainbow trout infections. Silver nanoparticles (AgNPs) have a broad spectrum of antimicrobial properties and are usually produced by various green-synthesis methods. However, the application of commercialized AgNPs has not fully been clarified. Thus, the objective of this study was to evaluate the antibacterial activities of commercialized AgNPs (range of sizes 10–12 nm) on two contrastingA.salmonicidastrains (I-1 and I-4), isolated from rainbow trout; the antibacterial mechanism, histopathological alterations and the expression of immune-related genes were investigated. In vitro, the minimal inhibitory concentration (MIC) was 10 µg/mL for I-1, and lowered to 9.5 µg/mL for I-4, respectively. AgNPs were shown to disrupt both the cell wall and membrane of I-1 and I-4, resulting in cell lysis and degradation. In vivo, rainbow trout challenged by immersed or intraperitoneally injected infection, the 10 µg/mL AgNP-treated groups, both showed delayed deaths and lower mortalities compared to the control groups, without any clinical signs and pathological changes. Especially for the virulent I-4, the enhanced expressions of immune-related genes TNF-α, IL-1β, IL-10 and IL-11 were significantly reduced in the AgNP-treated group, indicating a lesser inflammation due to the application of AgNPs. This study would lay theoretical foundation for the wide application of silver nanoparticles in fish diseases.

This study aimed to examine the effects of high-fat diets supplemented with astaxanthin , chlorogenic acid, and bile acid on the growth, lipid accumulation, oxidation resistance, inflammatory response, and liver morphology in Oncorhynchus mykiss cultured in the convection-water cages. Fish were fed five isonitrogenous diets (D1: 20% lipid; D2: 30% lipid; D3: 30% lipid +0.1% BASF astaxanthin; D4: 30% + 0.06% chlorogenic acid; and D5: 30% lipid +0.1% bile acid) for 6 weeks. The results found that fish fed the D2 diet achieved a lower growth, while the optimal growth was identified in D4 diet. The higher crude lipid contents in the whole body were identified in fish fed the D3-D5 diets. Fish fed the high-fat diet showed liver vacuolization and lower superoxide dismutase (SOD) activity and transcriptional levels of NF-E2-related nuclear factor 2 ( Nrf2 ), catalase ( CAT ) , and SOD , as well as higher malondialdehyde content. Supplementing above three additives can reverse above results and enhance the total antioxidant capacity. Besides, the three additives alleviated the inflammatory responses caused by the high-fat diet via inhibiting proinflammatory cytokines. Moreover, high-fat diet reduced the endogenous lipid production and promoted the lipoprotein hydrolysis by increasing the lipoprotein lipase and inhibiting the fatty acid synthesis . The above results showed that three additives could mitigate the negative effects caused by the high-fat diet. Among them, astaxanthin showed the best antioxidant and anti-inflammatory properties and chlorogenic acid could elevate the growth greatest.

Antimicrobial peptides are important components of the host innate immune system, forming the first line of defense against infectious microorganisms. Among them, liver-expressed antimicrobial peptides (LEAPs) are a family of antimicrobial peptides that widely exist in vertebrates. LEAPs include two types, named LEAP-1 and LEAP-2, and many teleost fish have two or more LEAP-2s. In this study, LEAP-2C from rainbow trout and grass carp were discovered, both of which are composed of 3 exons and 2 introns. The antibacterial functions of the multiple LEAPs were systematically compared in rainbow trout and grass carp. The gene expression pattern revealed that rainbow trout and grass carp LEAP-1, LEAP-2A, LEAP-2B and/or LEAP-2C were differentially expressed in various tissues/organs, mainly in liver. After bacterial infection, the expression levels of LEAP-1, LEAP-2A, LEAP-2B and/or LEAP-2C in the liver and gut of rainbow trout and grass carp increased to varying degrees. Moreover, the antibacterial assay and bacterial membrane permeability assay showed that rainbow trout and grass carp LEAP-1, LEAP-2A, LEAP-2B and LEAP-2C all have antibacterial activities against a variety of Gram-positive and Gram-negative bacteria with varying levels through membrane rupture. Furthermore, cell transfection assay showed that only rainbow trout LEAP-1, but not LEAP-2, can lead to the internalization of ferroportin, the only iron exporter on cell surface, indicating that only LEAP-1 possess iron metabolism regulation activity in teleost fish. Taken together, this study systematically compared the antibacterial function of LEAPs in teleost fish and the results suggest that multiple LEAPs can enhance the immunity of teleost fish through different expression patterns and different antibacterial activities to various bacteria.

Salmo salar is one of the most popular salmon species due to its meaty texture and quality protein. Oncorhynchus mykiss , which has a muscle texture similar to that of Salmo salar and is less expensive, is often used as a substitute for Salmo salar . As Salmo salar and Oncorhynchus mykiss belong to the same subfamily of Salmonidae, traditional methods are ineffective in the specific detection of the two. In this study, we combined hue-change with CRISPR/Cas12a lateral flow assay to detect the Salmo salar adulteration. This method detected S. salar genomic DNA at a vLOD of 5 copies, and was able to accurately identify adulterated samples containing 5 % w /w Salmo salar within one hour. In addition, the detection of Salmo salar in processed food products was achieved with the naked-eye at a concentration range of 0 % ∼ 70 % w/w, and the detection accuracy is between 93.3 % ∼ 100 %.

Clostridium autoethanogenum protein (CAP) is a promising protein source for aquaculture. Previous studies proved that high levels of dietary CAP retarded the muscle development in turbot through mammalian target of rapamycin (mTOR)/amino acid response (AAR)/adenosine monophosphate-activated protein kinase (AMPK) pathways. However, how CAP regulating these pathways was still unclear and there was no information on the comprehensive analysis for the mechanisms of CAP on fish muscle. In the present study, the integration of metabolomic and proteomic was applied on the muscle of turbot (initial weight of 150 ± 3 g) fed with fishmeal-based diet or diet with 60 % of fishmeal protein replaced by CAP for 70 days. The present work confirmed that CAP decreased the growth, fillet yield and induced the muscle atrophy. Integrated analysis of metabolomics and proteomics showed that CAP negatively influenced the energy production by inhibiting the key enzymes of glycolysis and mitochondrial respiratory chain. Second, CAP inhibited mitogen-activated protein kinase (MAPK) and phosphatidylinositol-3-kinase (PI3K) pathways to down-regulate mTOR pathways and protein synthesis. Third, the amino acid contents in turbot muscle were imbalanced by CAP, which would be perceived by general control nonderepressible 1 in AAR signal pathways. Finally, the up-regulated AAR induced amino acids catabolism through purine-based combined deamination. In conclusion, the present work provided the universal knowledge on the working principle of CAP and laid the foundation for CAP improvement in the future.

Plant essential oils lose their activity due to unstable chemical properties and volatility, and the coating can improve their stability by encapsulating. The three-layer coatings were prepared by tape casting method with flaxseed gum (FG) and chitosan (CS) as film-forming materials, eugenol (EG) and laurel essential oil (LEO) as preservatives. The composite coatings were characterized, and their physicochemical properties, release properties, antibacterial and antioxidant properties were determined. Meanwhile, the protective effect of the composite coatings on rainbow trout fillets myofibril protein was studied. The mechanical properties of the FG/CS/FG coatings are better than FG coating. The release of EG and LEO from the coatings are followed simple diffusion mechanism. After added essential oils, the antibacterial and antioxidant properties of the composite coatings are significantly enhanced. In the preservation process of the rainbow trout fillets, the composite coatings can reduce the carbonyl content, increase the sulfhydryl content and Ca 2+ -ATPase activity. The β-sheet content is 6.09%–15.63% higher than that of control, indicating the coatings are helpful to maintain the order of myofibril protein. The composite coatings slowed down the decrease of antioxidant enzyme activity, thus delay the protein oxidation. Because of long-term antibacterial and antioxidant properties, the composite coatings have potential value in food preservation or food packaging materials.
