Possible Insecticidal Mechanism of Cry41-Related Toxin against Myzus persicae by Enhancing Cathepsin B Activity

Xiao-Di Zhao,Bin-Wu Zhang,Li-Jun Fu,Qi-Lin Li,Yi Lin,Xiao-Qiang Yu

Cry toxins produced by Bacillus thuringiensis are well known for their high insecticidal activities against Lepidoptera, Diptera, and Coleoptera; however, their activities against Aphididae are very low. Recently, it has been reported that a Cry41-related toxin exhibited moderate activity against the aphid Myzus persicae, and thus, it is highly desirable to uncover its unique mechanism. In this paper, we report that Cathepsin B, calcium-transporting ATPase, and symbiotic bacterial-associated protein ATP-dependent-6-phosphofructokinase were pulled down from the homogenate of M. persicae as unique proteins that possibly bound to Cry41-related toxin. Cathepsin B has been reported to cleave and inactivate antiapoptotic proteins and plays a role in caspase-initiated apoptotic cascades. In this study, Cathepsin B was expressed in Escherichia coli and purified, and in vitro interaction between recombinant Cathepsin B and Cry41-related toxin was demonstrated. Interestingly, we found that addition of Cry41-related toxin obviously enhanced Cathepsin B activity. We propose a model for the mechanism of Cry41-related toxin as follows: Cry41-related toxin enters the aphid cells and enhances Cathepsin B activity, resulting in acceleration of apoptosis of aphid cells.

Volatiles from essential oils of three Lamiaceae plants repel the winged cotton aphid, disturb its feeding behavior and reduce its fecundity

Ying Zhang,Tao Zhang,Xinhang Wang,Zhipeng Bian,Xiaofang Zhang,Guoqing Yang,Yanhui Lu

BACKGROUND Insects use odor detection to sense their surroundings. Use of volatile compounds, such as essential oils (EOs) of plants, to repel pests and disrupt their olfaction-driven behaviors has great practical potential for use in integrated pest management. Despite the available information on the repellent effects of EOs on herbivorous insects, the olfaction-based mechanisms remain unknown. RESULTS Y-tube olfactometer tests showed that the EOs of three Lamiaceae plants – Mentha arvensis L., Mentha piperita L. and Lavandula angustifolia Mill. – were significantly repellent to winged cotton aphid, Aphis gossypii Glover. Electrical penetration graph (EPG) tests indicated the EOs reduced phloem feeding and increased the level of non-productive probing by the aphids. The EOs also reduced the fecundity of winged Aphis gossypii . Electrophysiological bioassays and gas chromatography–mass spectrometry (GC–MS) identified five physiologically active volatiles, that is menthone, isomenthone, neomenthol and menthol from Mentha piperita ; menthone and menthol from Mentha arvensis ; and linalool from L. angustifolia . Behavioral tests confirmed that all five compounds repelled winged Aphis gossypii . Under field conditions, the growth rate of aphid populations after 7 days was significantly lower in fields treated with these compounds than in the control fields. CONCLUSION Our findings demonstrated that three EOs not only repelled winged Aphis gossypii but also interfered with the aphid's feeding behavior and reduced its fecundity. These EOs and their active constituents have great potential as eco-friendly control products for use against Aphis gossypii . The effects of these EOs also exceed other repellents that only keep pests away from host plants. © 2024 Society of Chemical Industry.

A pH- and enzymatic-responsive nanopesticide to control pea aphids and reduce toxicity for earthworms

Xuqian Zhang,Ying He,Zitong Yuan,Guangmao Shen,Zan Zhang,Jinzhi Niu,Lin He,Jinjun Wang,Kun Qian

Thiacloprid is a new chlorinated nicotinoid insecticide against stinging-oral pests, such as aphids. It is less toxic to bees but more toxic to earthworms. In this study, a pH- and amylase-responsive MOF (ZIF-8) was constructed for site-specific delivery of thiacloprid to control pea aphids and more safety for earthworms. Thiacloprid from α-cyclodextrin@Thiacloprid@ZIF-8 (α-CD@T@ZIF-8) could be released quickly in pea aphids, which was ascribed to disintegration of ZIF-8 at low pH values in pea aphid intestines and degradation of α-CD under the action of α-amylase. The release results showed a significant pH dependence of α-CD@T@ZIF-8, with an approximately 65 % release amount at pH = 7 and a 95 % release amount at pH = 5 for 7 d. The results of the pot experiment and biosafety showed that for α-CD@T@ZIF-8, 88 % pea aphids could be killed compared with 32 % aphids for commercially available formulation on the 7th day after application. Meanwhile the LC 50 of thiacloprid OD was 0.034 μg/cm 2 and the LC 50 of α-CD@T@ZIF-8 was 0.564 μg/cm 2 on earthworms, and it was more safety for pea and lower acute toxicity and enrichment for the earthworms. α-CD@T@ZIF-8 could be used for intelligently controlled release of other insecticides against aphids.

Effects of Endosymbiont Disruption on the Nutritional Dynamics of the Pea Aphid Acyrthosiphon pisum

Ning Lv,Lei Wang,Wen Sang,Chang-Zhong Liu,Bao-Li Qiu

Pea aphid (Acyrthosiphon pisum) is a worldwide pest that feeds exclusively on the phloem sap of numerous host plants. It harbours a well-known primary endosymbiontBuchneraaphidicolathat helps to overcome the nutritional deficiency of a plant-based diet. However, how theBuchneracontributes to the nutritional and energy metabolism of its aphid host is unclear to date. In the current study, the function ofBuchnerain relation to nutritional synthesis of pea aphid was investigated by disrupting the primary endosymbiont with an antibiotic rifampicin. Our findings revealed that the disruption ofBuchneraled to infertility and higher loss in body mass of aphid hosts. Body length and width were also decreased significantly compared to healthy aphids. The detection of nutrition indicated that the quantity of proteins, soluble sugars, and glycogen in aposymbiotic pea aphids increased slowly with the growth of the aphid host. In comparison, the quantities of all the nutritional factors were significantly lower than those of symbiotic pea aphids, while the quantity of total lipid and neutral fat in aposymbiotic pea aphids were distinctly higher than those of symbiotic ones. Thus, we concluded that the significant reduction of the total amount of proteins, soluble sugars, and glycogen and the significant increase of neutral fats in aposymbiotic pea aphids were due to the disruption ofBuchnera, which confirmed that the function ofBuchnerais irreplaceable in the pea aphid. Keywords:Buchnera;pea aphid;endosymbiont;antibiotics;nutrition

The low-lethal concentrations of rotenone and pyrethrins suppress the population growth of Rhopalosiphum padi

Xu Li,Wu Zhenghua,Li Jingkun,Xu Yihao,Zhou Feng,Zhang Fulong,Li Dongzhi,Zhou Lin,Liu Runqiang

As an important pest on winter wheat, Rhopalosiphum padi (L.) causes damage to the wheat yield by sucking plant nutrients, transmitting plant viruses and producing mildew. R. padi has been reported to develop resistance to pyrethroids and neonicotinoids. To explore potential alternative approaches for R. padi control, the activity of 10 botanical insecticides was evaluated. Results suggested that the toxicity of rotenone and pyrethrins to R. padi were the highest and near to the commonly used chemical insecticides. When exposed to the low-lethal concentrations (LC 10 , LC 30 ) of rotenone or pyrethrins for 24 h, the lifespan and fecundity of adults in F0 generation decreased significantly compared to control. The negative effect could also be observed in the F1 generation, including the decreased average offspring, longevity of adult, and prolonged nymph period. The population parameters in F1 generation of R. padi were also inhibited by exposing to the low-lethal concentrations of rotenone or pyrethrins, including the decreased net reproductive rate, intrinsic rate of natural increase, finite rate of population increase, and gross reproduction rate. Co-toxocity factor results showed that mixtures of rotenone and thiamethoxam, pyrethrins and thiamethoxam showed synergistic effect. Our work suggested that rotenone and pyrethrins showed negative effect on the population growth under low-lethal concentrations. They are suitable for R. padi control as foliar spraying without causing population resurgence.

Evaluation of floral-derived volatile blend for attracting aphid parasitoids and lady beetles in the tobacco fields

Shaolong Wu,Fengli Liu,Weiai Zeng,Zhipeng Xiao,Jiaying Li,Kai Teng,Qianshaung Guo,Jinhao Zhao,Yongjun Du

In tobacco planting in China, the artificial release of aphid parasitoids Aphidius gifuensis has been applied to control aphids for a decade. However, due to seasonal effects and crop rotation, the sustainability of releasing parasitoids and the aphid control efficacy have not been properly evaluated. In this study, effective semiochemical-based attractants were further screened and optimized in the tobacco fields. Film lures were developed to release the synthetic attractant blends. In tobacco planting fields, the synthetic mixture of β-pinene, methyl salicylate , linalool , and limonene(6:6:2:20 mg) significantly increased the population of natural enemies, including A. gifuensis and ladybeetles ( Coccinella septempunctata , Harmonia axyridis and Hippodamia variegate ) monitored using using malaise traps. The increase in the natural enemy population was related to the season and the lure placement density in the tobacco fields, as well as the distribution of surrounding habitats and flowering plants. A density of 135 lures per hectare significantly increased the number of parasitoids, reduced the population of aphids, elevated the rate of parasitized aphids by 42%, and effectively suppressed the aphid population in the tobacco fields. Combined with the protection of flowering plants around tobacco fields, synthetic lures may provide a technical measure for conservative biological control.

Eucalyptol-loaded microcapsules combined with Cynanchum komarovii extracts provide long-term and low-risk management of Chinese wolfberry (Lycium barbarum L.)

Rui Wang,Jian Zhang,Zhengyi Sun,Xuewen Jian,Yue Xu,Xuan Zhou,Xiaojie Liang,Jin Lin,Beixing Li,Wei Mu,Yuekun Li

Realizing eco-friendly, long-term, and low-risk aphid control on Lycium barbarum (medicinal cash crop) using a Cynanchum komarovii extracts and eucalyptus oil-loaded microcapsules (EOMCs) formulation compositions is viable. In this study, the aim is to optimize the composition of Cynanchum komarovii extracts and EOMCs formulation for effective control of aphids, the release of EOMCs was controlled by changing the cross-linking degree of the shell to match the aphid control characteristics of Cynanchum komarovii extracts. Four types of polyamines were used as cross-linking agents for the preparation of EOMCs by interfacial polymerization. The bioactivity, wettability, and field application efficacy of Cynanchum komarovii extracts and different EOMCs formulation compositions were evaluated. These EOMCs exhibited an encapsulation efficiency exceeding 85 %. The control efficiency of the formulation compositions of microcapsules with a moderate release rate and Cynanchum komarovii extracts on aphids remained at 62.86 %, while the control efficiency of the combination of microcapsules with the fastest and slowest rates with Cynanchum komarovii extracts was only 48.62 % and 57.11 %, respectively. The formulation compositions of Cynanchum komarovii extracts with all four types of EOMCs were found to be safe for Chinese wolfberry plants. Overall, by selecting appropriate polyamines during fabrication, the release rate can be effectively controlled to achieve sustainable and low-risk aphid control in Lycium barbarum through compounding with selected microcapsules.

An efficient mutagenesis system to improve the propamocarb tolerance in Lecanicillium lecanii (Zimmermann) Zare & Gams

Yanjun Zhang,Xiao Zhang,Weiliang Qiu

Lecanicillium lecanii (Zimmermann) Zare & Gams is used as an effective biopesticide for the control of sap-sucking insect pests on agricultural crops. However, low fungicide tolerance limits its large-scale field application. To improve the propamocarb tolerance in L. lecanii , a composite mutagenesis system was established by using UV-light (U), N-Methyl-N′-nitro-N-nitrosoguanidine (NTG) (N) and N + ion-beam (I). The permutation type of three agents was a consecutive mutagenesis treatment (I/N/U) after an intermittent treatment (U + N + I). The “U” mutagenesis was performed at 254 nm for 60 s and at a distance of 45 cm under a 20 W germicidal lamp, the “N” mutagenesis was performed at a concentration of 1.0 mg/mL NTG for 60 min, and the “I” mutagenesis was performed by low energy N + ion-beam using a dose of 10 × 10 13 ions/cm 2 at 30 keV. This composite mutagenesis system was recorded as the “U + N + I + I/N/U,” and then the mutagenesis efficiency in improving propamocarb tolerance was assessed by analyzing changes of mutants in the propamocarb sensitivity, mitotic stability, mycelial growth speed on plates or in liquid, sporulation on plates or aphids, conidial germination, 50% lethal concentration (LC 50 ) and 50% lethal time (LT 50 ) to aphids, lipid constituent and cell membrane permeability and control against aphids in the presence or absence of propamocarb. Compared to the wild-type isolate with a 50% effective concentration (EC 50 ) value of 503.6 μg/mL propamocarb, the Ll-IC-UNI produced by the “U + N + I + I/N/U” had the highest EC 50 value of 3576.4 μg/mL and a tolerance ratio of 7.1. The mutant was mitotically stable in 20-passage cultivation and did not show any unfavorable changes in growth and virulence indicators. The mutant showed the highest ability to resist or avoid the damaging effects of propamocarb as reflected by the alternations of lipid constituents and membrane permeability. The interval time for applying fungal agent was significantly shortened in this mutant after spraying a field recommended dose of 550 μg/mL propamocarb. In conclude, the “U + N + I + I/N/U” composite mutagenesis mode was efficient and useful to improve the propamocarb-tolerance of L. lecanii and the obtained Ll-IC-UNI could have commercial potential for field application.

Combination of functional plants conserves predators, repels pests, and enhances biological control of Aphis spiraecola in apple orchards

Zhihui Liu,Fan Wang,Yunhe Zhang,Eresbek Temir,Xiaotong Zhou,Yilei Shangguan,Donghai Zhang,Zhiping Cai

Functional plants possess the ability to attract natural enemies or repel pests, and they play an important role in ecological regulation. While most studies have focused on the impacts of individual plants or plant combinations with similar functions on pests, there is a scarcity of research on the effects of combining plants with different functions, particularly in the case of orchard pests. In this study, we combined plants ( Cnidium monnieri (L.) or Brassica napus L.) that attract predators with plants ( Ocimum basilicum L. or Mentha haplocalyx Briq.) that repel pests to determine if these combinations can enhance biocontrol of Aphis spiraecola Patch on apple trees. All four combinations reduced the A. spiraecola population on apple trees. B. napus & M. haplocalyx achieved the highest reduction rate (88.58%), followed by C. monnieri & O. basilicum (88.06%), B. napus & O. basilicum (86.11%), and C. monnieri & M. haplocalyx (79.41%). Cage exclusion tests on 29 June and 4 July 2023 showed that biocontrol services index of all combinations was significantly higher than that of the control. Rubidium marking test showed that 61.90%–71.43% of Chrysoperla sinica (Tjeder) individuals in trees had transferred from functional plants. Behavioral responses and field trapping indicated that compounds such as cis -citral, nepetalactone, caryophyllene, beta-pinene, and eugenol, which are present in repellent plants, had significant repellency against A. spiraecola . The use of functional plant combinations in apple orchards can enhance biocontrol of A. spiraecola by promoting predators and by repelling aphid. This supports the potential of functional plant combinations in agroforestry ecosystems for pest biocontrol.

Virus-induced changes in host plant phenotype cue behavioral changes in Aphis glycines that enhance acquisition and transmission of soybean mosaic virus

Fang Haibo,Gao Le,Michaud J. P.,Chen Haowei,Liu Xiaoxia,Zhang Songdou,Li Zhen

Vector-borne viruses can modify the phenotype of their host plants to manipulate the behavior of their vectors and maximize their dissemination. The soybean aphid, Aphis glycines Matsumura (Hemiptera: Aphididae), is a primary vector of soybean mosaic virus (SMV), a non-persistent potyvirus that is a serious disease of soybean. We investigated mechanisms by which SMV changes A. glycines feeding and dispersal behavior via virus-induced modifications of plant phenotype. These changes affect gustatory, visual and olfactory cues that alter the behavior of both viruliferous and virus-free aphids to enhance virus acquisition and transmission. SMV-infected soybean seedlings exhibited up-regulation of the amino acids threonine and tyrosine which rendered viruliferous aphids more restless and prone to dispersal. Whereas dispersing viruliferous A. glycines preferred to settle and feed on virus-free soybean seedlings, virus-free aphids preferred SMV-infected seedlings. Volatile cues emanating from SMV-infected seedlings, primarily up-regulated methyl salicylate and down-regulated ( Z )-3-hexen-1-ol, were implicated in repulsion of viruliferous aphids, whereas the mottled yellow coloration of SMV-infected seedlings was a visual attractant for aphids regardless of virus status. Our results reveal how a non-persistent virus uses multimodal 'push and pull' mechanisms to manipulate vector behavior for its own benefit, and suggests new avenues for exploring the mechanisms of altered behavior in viruliferous aphids.

Construction of pyrethrin nanopesticides based on zeolite Imidazole-8 framework via one-pot method for effectively wheat aphid control

Jiangtao Dong,Hexin Li,Guoquan Wang,Haoming Li,Yanli Zhao,Xinan Li,Li Xu,Feng Zhou,Runqiang Liu

Pesticides play an important role in controlling plant diseases and insect pests to increase food production. Pyrethrin (Py), as a plant-derived insecticide, can effectively control wheat aphids owing to its fast knockdown, low concentration, and broad-spectrum insecticidal activity. However, Py is sensitive to UV irradiation and decomposes easily under sunlight, which leads to the loss of insecticidal activity. In this paper, we prepared one kind of metal organic framework nanopesticides based on zeolite imidazole-8 framework (ZIF-8) loaded with Py (Py@ZIF-8) via one-pot method for wheat aphid control. The prepared Py@ZIF-8 nanopesticides appeared acidic/basic pH and PO 4 3- stimuli-responsive properties for the controlled release of packaged Py by the disassembly of MOF skeleton and enhanced leaf wettability. The control efficacies of Py@ZIF-8 nanopesticides against wheat aphids reached ca. 86.67 ± 4.56 % under normal conditions, ca. 82.56 ± 3.88 % after water washing, and ca. 78.92 ± 4.34 % under exposure to UV irradiation, which appeared remarkably improvement compared with technical Py under UV irradiation and rain erosion condition, respectively. Even through the nanopesticides inevitably enter into soil or water, they appear good biosafety on wheat and cucumber seedlings by seed dressing, as well as zebrafish ( D. rerio ). Importantly, the eco-friendly nanocarriers will be ultimately degraded against bioaccumulation in crops based on the stimuli-responsive mechanisms. These results indicated that the prepared nanopesticides have an efficient wheat aphid control owing to the intelligent control release performance, as well as good UV and washing resistance, and good biosafety on non-target species, which have potential applications in agriculture against wheat aphids.

Preparation and insecticidal activities of betulin-cinnamic acid-related hybrid compounds and insights into the stress response of Plutella xylostella L.

Xiaobo Huang,Nan Hao,Lili Shu,Zhenyang Wei,Jiawei Shi,Yuee Tian,Genqiang Chen,Xinling Yang,Zhiping Che

BACKGROUND The emergence and increasing intensification of insect pest resistance greatly shortens the service life of commercial insecticides. Hence, the continuous development of novel insecticides is extremely necessary. Discovering new insecticide molecules based on natural products with insecticidal activity has been a popular technique and has made outstanding contributions in this field. RESULTS A series of betulin-cinnamic acid-related hybrid compounds was prepared by molecular hybridization through the bridge of acyloxy groups and their chemical structures were unambiguously confirmed. Against A. pisum , compounds 2e , 3a-3e , 3g , 3i , 3j , and 3m showed complete antifeedant activity at 2 mg/mL; compound 5b exhibited 2.7 times the aphicidal activity of the precursor betulin. Against P. xylostella , compound 2l displayed 3.9 times the insecticidal activity of betulin. Meanwhile, some interesting structure–activity relationships were obtained. Furthermore, compound 2l could destroy the ultrastructure of midgut cells and significantly inhibit the activity of α-amylase in P. xylostella larvae. Molecular docking indicated that the lupane skeleton and p -nitrocinnamoyloxy groups played a key role in the binding of compound 2l to α-amylase. CONCLUSION Compounds 5b and 2l could be further optimized structurally as the secondary lead compounds for the discovery of novel insecticide molecules. © 2025 Society of Chemical Industry.
