Awareness of toxicokinetics at the subcellular level is crucial to deciphering the underlying intoxication processes of metal(loid)s, although this information is often lacking. Here, the toxicokinetics of two non-essential metal(loid)s (Cd and As) and one essential metal (Zn) in both the whole body and subcellular fractions of earthworm ( Eisenia fetida ) were assessed. Earthworms were exposed to natural soils originating from a gradient of metal(loid) pollution for 14 days followed by a 14-day elimination phase in clean soil. Clearly distinct toxicokinetic patterns were found in the earthworms according to the metal(loid) considered. An obvious concentration-dependent increase was observed in earthworms or subcellular compartments where no equilibrium was reached (with slow or no elimination) for Cd and As throughout the experiment. As for Zn, the earthworms were able to retain a steady-state concentration of Zn in its body or each fraction without a clear intake behavior via the dynamic trade-off between uptake and elimination at different pollution levels. These differences in toxicokinetics at the subcellular level supported the observed differences in bioaccumulation patterns and were indicative of the strategy by which non-essential and essential elements are handled by earthworms. Notably, the concentration of Cd and As in subcellular compartments showed the same pattern as for Zn in the order of cellular cytosol > cellular debris > metal-rich granules, which might be associated with the binding of non-essential/essential elements with metallothionein enriched in the cytosol. Our findings enhance the understanding of the underlying mechanisms for metal(loid) accumulation kinetics in earthworms from the perspective of subcellular partitioning, and will be beneficial for accurate risk assessment of Cd, As, and Zn.

Purpose Decabromodiphenyl ethane (DBDPE) and lead (Pb), two prevalent contaminants in e-waste dismantling areas, pose significant threats to soil ecosystem health. However, the ecotoxicological effects of their co-exposure on soil invertebrates remain poorly characterized. Methods Avoidance behavior, survival rate, and growth-inhibition assays were performed to assess the combined toxic effects of DBDPE and Pb on earthworm ( Eisenia fetida ) in an artificial soil system over exposure periods of 7–21 d. Responses from multiple biochemical biomarkers, including superoxide dismutase (SOD), catalase (CAT), malondialdehyde (MDA), glutathione S-transferase (GST), and acid phosphatase (ACP) were also measured, and the data were integrated by the biomarker response index (BRI). Interaction between Pb and DBDPE on BRI was further analyzed by calculation of effect addition index (EAI). Results Avoidance behavior emerged as the most sensitive endpoint and exhibited concentration-dependent escalation under co-exposure conditions. While individual exposure to DBDPE (0–0.75 mg/kg) induced negligible impacts on growth and mortality, its combination with Pb (1000 mg/kg) resulted in significant synergistic effects. Similarly, biochemical biomarkers—including SOD, CAT, MDA, GST, and ACP—showed markedly greater alterations in co-exposure groups compared to single DBDPE treatments. The BRI revealed only slight alterations in biological health across most treatments. The EAI values indicated that antagonistic effects dominated the interaction between DBDPE and Pb. Conclusion Our findings demonstrated significant variation in biomarker sensitivity to pollutant exposure. Notably, short-term sub-lethal co-exposures may elicit synergistic effects on avoidance, growth, and mortality, revealing a time-concentration toxicity dependency. These results provide novel insights into the interaction between heavy metals and organo-pollutants and highlight the ecological hazards posed by emerging brominated flame retardants (e.g., DBDPE) in heavy metal-contaminated soil systems.

Thiacloprid is a new chlorinated nicotinoid insecticide against stinging-oral pests, such as aphids. It is less toxic to bees but more toxic to earthworms. In this study, a pH- and amylase-responsive MOF (ZIF-8) was constructed for site-specific delivery of thiacloprid to control pea aphids and more safety for earthworms. Thiacloprid from α-cyclodextrin@Thiacloprid@ZIF-8 (α-CD@T@ZIF-8) could be released quickly in pea aphids, which was ascribed to disintegration of ZIF-8 at low pH values in pea aphid intestines and degradation of α-CD under the action of α-amylase. The release results showed a significant pH dependence of α-CD@T@ZIF-8, with an approximately 65 % release amount at pH = 7 and a 95 % release amount at pH = 5 for 7 d. The results of the pot experiment and biosafety showed that for α-CD@T@ZIF-8, 88 % pea aphids could be killed compared with 32 % aphids for commercially available formulation on the 7th day after application. Meanwhile the LC 50 of thiacloprid OD was 0.034 μg/cm 2 and the LC 50 of α-CD@T@ZIF-8 was 0.564 μg/cm 2 on earthworms, and it was more safety for pea and lower acute toxicity and enrichment for the earthworms. α-CD@T@ZIF-8 could be used for intelligently controlled release of other insecticides against aphids.

Pesticides and heavy metals commonly coexist in soil. In this study, the influence of Cd and Cu on the toxicity of rac -dinotefuran and the enantioselective behavior of dinotefuran enantiomers in soil-earthworm microcosms were investigated. The acute toxic tests showed that S -dinotefuran has higher toxic than that of R -dinotefuran. The rac -dinotefuran and Cd has an antagonistic effect on earthworms, and the Cu and rac -dinotefuran has a synergistic effect. Earthworms maybe promoted the enantioselective behavior of dinotefuran in soil. Co-exposure to Cd or Cu inhibited the dissipation of dinotefuran enantiomers ( S -dinotefuran and R -enantiomers), and slightly reduced the enantioselectivity in soil. The earthworms were found to be preferentially enriched with S -dinotefuran. However, Cd or Cu attenuated the accumulation of dinotefuran enantiomers in earthworms and decreased the enantioselectivity. The effect of Cd and Cu on the environmental behaviors of dinotefuran enantiomers were correlated positively with the dose of Cd/Cu. These results showed that Cd and Cu alter the environmental behaviors and the toxicity of dinotefuran enantiomers in soil-earthworm microcosms. Thus, the influence of coexistent heavy metals on the ecological risk assessment of chiral pesticides should be considered.

Soil microbial extracellular polymeric substances (EPS) are important components of soil organic carbon (SOC), which are thought to promote the formation of soil structures and induce the stabilization of SOC. Earthworms may not only stimulate the production of EPS through the promotion of microbial activity and microbial biomass, but might also accelerate the decomposition of EPS via ingestion, resulting in SOC sequestration or loss. However, the exact impacts of earthworms on EPS in soils have never been evaluated. Here, a pioneering experiment involving two species of endogeic earthworms ( Drawida gisti and Metaphire guillelmi ) with different body sizes was conducted to investigate their impacts on soil EPS. It was found that the D. gisti and M. guillelmi earthworms reduced the soil EPS-protein concentrations by 12.6 % and 17.6 %, respectively; however, they did not affect the EPS-polysaccharide concentration. Moreover, the presence of these endogeic earthworms increased soil microbial biomass, and elevated microbial respiration, microbial metabolic quotient, dissolved organic carbon, and soil pH. The results of this study provide the first evidence for the decoupling effects of earthworms on extracellular residual components. This suggests that earthworms may alter the molecular composition of SOM, thereby influencing its persistence and soil-atmosphere carbon exchange.

Purpose In this study, the adsorption behavior of diuron in a typical black soil and its bioavailability to crops and earthworms were investigated by adsorption and incubation experiments, combined with biochar amendment and aging time. Methods In this study, the adsorption of diuron, and its uptake by corn and earthworms were investigated in black soil with 0.1–5% (w/w) bamboo charcoal. Results Uptake of diuron by corns was reduced by 45% in black soil aged for 0 day in 0.1–5% biochar-amended soil, the concentration of diuron in both roots and stems of corn plants tended to decrease with the increase of aging time, and the enrichment coefficient of diuron in corn also tended to decrease in 0.1–5% biochar-amended soil. When aging time was 0 day and the biochar amendment in black soil was 0.1–5%, the accumulation concentration of diuron in earthworms decreased by 63%; the bioconcentration coefficient of diuron in earthworms in black soil with different levels of biochar decreased continuously with the increase of aging time. Conclusion Biochar is an efficient adsorbent for diuron and could significantly reduce the uptake of diuron by plants and earthworms from contaminated soil. Significant correlations indicated that regression analyses could be constructed between the bioconcentration factor and sorption coefficients to predict the bioavailability of diuron residues in soil to corns and earthworms. Biochar amendment could be used as an option to immobilize diuron in soil and protect non-target organisms from diuron contamination.

The aim of this study was to identify peptides fromLumbricus terrestriswith neuroprotective effects. Two peptides (GYSFTTTAER and AVFPSIVGR) isolated from earthworms improved cell viability of the SH-SY5Y human neuroblastoma cell line treated with 1-methyl-4-phenyl-1,2,3-tetrahydropyridinehydrochloride (MPP+), a commonly used model of Parkinson’s disease (PD). Both peptides increased the mitochondrial membrane potential and upregulated the mRNAs of mitophagy regulators PINK1 and Parkin in the MPP+-damaged cells. The in vitro assay and molecular docking indicated that both peptides exhibited moderate PINK1 agonistic activity. Furthermore, GYSFTTTAER and AVFPSIVGR extended the lifespan, improved locomotor behavior, and raised the ATP and dopamine levels at all ages inPINK1B9mutant flies, a PD model characterized by loss-of-function of PINK1. These findings suggest that earthworm-derived peptides possess anti-neurodegenerative properties and hold potential for the development of health products and therapeutic agents for PD.

The toxicity of nanoparticles has attracted much attention, but the toxicity of nanoparticle types to soil fauna remains unclear. In this study, the earthworm ( Eisenia fetida ) was selected as a representative soil fauna to evaluate the toxicity of nano-hydroxyapatite (n-HAP), -zeolite (n-zeolite), and -iron tetroxide (n-Fe 3 O 4 ) to soil fauna. The results showed that the skin ultrastructure, mortality, growth, antioxidant systems, and genetic material of the earthworms were affected to varying degrees by different nanoparticles. A filter paper test indicated that dermal exposure to nanoparticles primarily caused lesions in the cuticle, microvilli, and nucleus of the epithelial cells of earthworm skin. Soil incubation experiments showed that all types of nanoparticles could cause weight loss in earthworms, but only n-Fe 3 O 4 resulted in their death. The reactive oxygen species and glutathione contents in earthworms exposed to n-Fe 3 O 4 were much higher than those in earthworms exposed to n-HAP and n-zeolite. All nanoparticles stimulated the catalase and lactate dehydrogenase activity and damaged the DNA of earthworms. Integrated biomarker responses showed that the toxicity to earthworms increased in the following order: n-HAP < n-zeolite < n-Fe 3 O 4 . The characterization results indicated that the differences in surface hydroxyl groups and surface charge may be the cause of the differences in toxicity between n-HAP and n-zeolite with similar properties. The highest toxicity of n-Fe 3 O 4 could be due to the size, spherical morphology, positive surface charge and the presence of Fe(II).

Earthworms are keystone regulators of carbon exchange between terrestrial ecosystems and the atmosphere. However, exactly how earthworms regulate the composition of microbial and plant-derived carbon in soil organic matter remains poorly understood. Here we conducted a microcosm experiment with two species of endogeic earthworms ( Drawida gisti and Metaphire guillelmi ) to investigate their effects on cellular and extracellular-microbial residues versus fast and slow-decaying plant materials. We found that both species of earthworms reduced microbial residues (amino sugars or the protein content of extracellular polymeric substances (EPS)) and facilitated the decomposition of microbial residues rather than their formation. Neither earthworm species affected slow-decaying plant residues (lignin phenols). However, their effects on the fast-decaying fraction of plant residues (particulate organic matter (POM)) depended on the earthworm species. Principal component analysis (PCA) revealed that earthworms mediated two gradients between microbial and plant residues. The first gradient was between the nitrogenous fraction of microbial residues (e.g., amino sugars and EPS-protein) versus slow-decaying plant lignin, while the second gradient was between the fast-decaying POM versus EPS-polysaccharide. Our results suggest that earthworms play vital roles in mediating plant and microbial residue fractions in soil through their multifaceted mechanisms in regulating the chemical composition of organic carbon, and in understanding biological control of the global soil carbon cycle.

Biosurfactant rhamnolipid and “soil ecosystem engineers” earthworms are often used to remediate contaminated soils. However, the effect of rhamnolipid on earthworm intestinal flora and microbial community in soil with earthworms is not understood. In our study, microcosm experiments were carried out to reveal the rhamnolipid on microbial abundance, composition, metabolism and contaminant degradation capacity. The results showed that both rhamnolipids and earthworms could have positive effects on soil bacteria. Bacteria in soil with rhamnosis have higher abundance and metabolic activity than in earthworm-only soils, while the improvement of rhamnolipid in bacterial composition and their contaminant degradation capacity were lower than earthworms. Notably, these changes were further amplified when rhamnolipids and earthworms coexisted. Specifically, the (log(copies/g) value increased from 9.50 (CK) to 10.25 (E), 10.50 (R) and 11.00 (RE). Comparing the soil with only earthworms (E), the relative abundance of dominant phyla Proteobacteria increased from 41.66% to 51.67% in the soil with both rhamnolipid and earthworms (RE), and more pollutant-degrading bacteria were also enriched in soil RE. Therefore, the increase in bacterial abundance and the enrichment of degrading bacteria lead that the removal rate of soil dioxins was earthworms + rhamnolipid (RE, 77.28%) > earthworms (E, 59.83%) > rhamnolipid (R, 24.65%) > control treatment (4.71%). Moreover, the addition of rhamnolipid enhanced the abundance of bacterial functional genes involved in “Metabolism” and “Environmental information processing”. In addition, the composition and diversity of bacteria in earthworms’ gut was conspicuously affected by rhamnolipid too, and the relative abundance of Microbacterium and Shewanella increased significantly (p<0.05). In conclusion, this study reveals that rhamnolipid will prominently influence the abundance, composition and metabolism of the microbial community in earthworms’ gut and soil, further promoting the degradation rate of dioxins. This study provides theoretical support for optimizing the combined application of rhamnolipid and earthworms in bioremediation engineering and the assessment of the ecological impacts of rhamnolipids.

Perfluorooctane sulfonamide (PFOSA) is a precursor of perfluorooctane sulfonic acid (PFOS) and can be broken down to PFOS in environment and biota. In the present work, PFOSA was spiked in soil and its biodegradation in soil, uptake and metabolism in wheat ( Triticum aestivum L.) and earthworms ( Eisenia fetida ) were investigated. The results indicated that PFOSA could be biodegraded to highly stable PFOS, which has the same perfluorinated carbon chain length as PFOSA, by microbes in soil. PFOSA could be taken up by wheat root and earthworm from soil with higher bioaccumulation ability than PFOS. In both wheat and earthworms, PFOSA also degraded to PFOS. However, other shorter-chain perfluoroalkane sulfonates (PFSAs), including perfluorohexane sulfonate (PFHxS) and perfluorobutane sulfonate (PFBS), were observed in wheat, but not in soil and earthworms, suggesting that wheat displayed distinctly different degradation mechanisms to PFOSA from soil microbes and earthworms.

Agricultural soil contamination with plastic film has become a critical global environmental problem, requiring greater research on the possible occurrence and biological risk of microplastics (MPs) and their additives in soil ecosystems. The presence of MPs and tris (2-chloroethyl) phosphate (Tcep) in agricultural soil was investigated at nine sites in the present study. Polyethylene MPs (PE-MPs) and Tcep were found at all nine sites. To study co-exposure effects on soil microbiota and earthworms, and to mimic a realistic exposure scenario, 0.05 % (w/w) PE-MPs with three particle size ranges were combined with Tcep (1.0 mg/kg). After 28 days of exposure, there was no indication that added PE-MPs and/or Tcep significantly affected the soil microbial community structure. In earthworms, size-selective intake, digestion and egestion of PE-MPs may occur, with Tcep co-exposure affecting the residual Tcep concentration in earthworm intestines (3.52–9.31 μg/g dw). Long-term earthworm PE-MPs intake caused intestinal damage, and Tcep co-exposure increased oxidative stress, thereby influencing their feeding behavior and growth, resulting in weight loss (3.42 %–14.96 %), especially for the most common PE-MPs sizes (0–300 μm). High performance liquid chromatography-mass spectrometry (LC-MS) was used for metabolomic analysis, revealing the significant up-regulation of citrate ( p < 0.001) and down-regulation of l -glutamate ( p < 0.05) in co-exposure groups. Co-exposure resulted in the alteration of most metabolic pathways, thereby impairing nervous, digestive and excretory systems in the earthworm, with an associated decrease in amino acid metabolism and changes in tricarboxylic acid (TCA) cycle intermediates. Gut microbiota, such as Proteobacteria ( Verminephrobacter and Bradyrhizobium ) and Firmicutes ( Bacillus ), are critically important in maintaining earthworm metabolic homeostasis, particularly for the TCA cycle and amino acid metabolism. Overall, MPs and Tcep co-exposure in agricultural soil enhanced their toxicity to earthworms and may potentially endanger the development of agricultural sustainability.
