Recent studies have indicated that active peptides can induce an improvement in wound repair. Herein, we evaluated egg white peptides (EWPs) as a nutritional supplement to improve mechanical skin damage in BALB/c mice. Two symmetrical circular full-thickness wounds were created with 5 mm biopsy punches in the skin of the mouse dorsal region, and EWPs (200, and 400 mg kg−1) were administrated by gavage for 14 days. We analyzed the EWPs for their in vivo and in vitro antioxidant capability, toxicity, and microscopy of skin wounds, and there was no cytotoxicity or in vivo toxicity. During the period of wound healing, EWPs could promote healthy cell migration, increase serum superoxide dismutase and catalase activities and accelerate the wound healing process in a time- and dose-dependent manner, whereas the levels of malondialdehyde and reactive oxygen species showed the opposite trend. After administration with 400 mg kg−1 EWPs for 10 days, the wound had almost healed. Meanwhile, EWPs significantly enhanced serum amino acids, particularly enhancing the content of Arg, Glu, Pro, Met, and Lys, which could provide sufficient nutrition in the wound healing process. The present study demonstrates that EWPs possess a positive potential to accelerate the wound healing process of mechanical skin damage at the cellular and animal level.

Oral food protein-derived peptides for healthcare have attracted increasing attention. However, most animal proteins (e.g., egg white) are highly in vivo digestible into peptides without additional hydrolysis treatment, which poses evident challenges to consumer choices and applications of peptide-based functional food. Herein, egg white-derived peptides prepared by alcalase hydrolysis (APH) and in vitro simulated digestion (SGDP) were compared to investigate the superiority of non-gastrointestinal hydrolysis in preparing peptides. Results indicated that the small peptide (<1 kDa) ratio, proteolysis degree, and sequence diversity of APH were double that of SGDP owing to broader cleavage sites. APH with C-terminal hydrophobic uncharged amino acid residues and higher positivity exhibited admirable advantages in transepithelial transport, contributing to better bioactivities (e.g., antioxidant and anti-inflammatory) and amino acid supplementation. Moreover, APH preferentially enriched mucus- and short-chain fatty acid-associated probiotics, thereby achieving preferable inflammation relief and microbiota restoration. Overall, this study offers an integrated theoretical basis for the superiority of non-gastrointestinal hydrolysis in preparing egg white-derived peptides for healthcare (e.g., colitis relief). Meanwhile, these findings would be enlightening to the development of enzymatic combination strategies and peptide-based ingredients.

Soft assembly of peptide and curcumin (Cur) molecules enables functional integration by finding dynamic equilibrium states through non-covalent interactions. Herein, we developed two soft assembly systems, curcumin-egg white peptides (Cur-EWP) aggregations (AGs) and Cur-EWP-casein-quaternary chitosan (Cur-EWP-CA-QC) nanoparticles (NPs) to comparatively investigate their therapeutic effects on ulcerative colitis in mice and elucidate their underlying mechanism. Results revealed that Cur-EWP AGs, despite gastrointestinal tract instability, exhibited a propensity for swift accumulation within the colorectal region, enriching mucus-associated and short-chain fatty acid (SCAF)-producing bacteria, restoring the intestinal barrier damage. Whereas, Cur-EWP-CA-QC NPs, benefiting from their remarkable stability and exceptional mucosal adsorption properties, not only enhanced permeability of Cur and EWP in the small intestine to activate the immune response and boost tight junction protein expression but also, in their unabsorbed state, regulated the intestinal flora, exerting potent anti-inflammatory activity. Soft assembly of peptides and hydrophobic nutraceuticals could synergize biological activities to modulate chronic diseases.
