Metabolic profiling unravels the effects of enhanced output and harvesting time on royal jelly quality

Chuan Ma,Licui Zhang,Mao Feng,Yu Fang,Han Hu,Bin Han,Lifeng Meng,Jianke Li

Royal jelly (RJ), a natural secretion of nurse bees, is a functional food with health-promoting properties. To investigate RJ quality changes with enhanced output and harvesting time, we compared metabolic profiles of RJ harvested at 24 h, 48 h, and 72 h after larval grafting into queen cells from Italian bees (ITBs) and high RJ-producing bees (RJBs) selected from ITBs. A total of 77 high-abundance compounds were identified with reverse-phase liquid chromatography– and hydrophilic interaction liquid chromatography–high-resolution mass spectrometry. Metabolite abundance, water content, and antibacterial activity were found largely unchanged between both bee strains, indicating that a similar RJ quality was achieved. Notably, the observed 10-hydroxy-2-decenoic acid content in RJBs (1.98%) and ITBs (2.41%) meet the standard for premium RJ in China. RJ harvested at 48 h and 72 h had similar metabolic profiles, indicating little influence of the two harvesting times on RJ quality. The harvesting at 24 h could lead to a higher RJ quality with higher abundance levels of many bioactive compounds and lower water content. Our data based mainly on RJ metabolic composition constitute evidence for mass production of high-quality RJ.

Gas chromatography-mass spectrometry analysis reveals the differences in volatile components of royal jelly from different honeybee stocks

Dandan Qi,Chengying Ma,Wenwen Wang,Licui Zhang,Jinghong Hao,Jianke Li

The aim of this study is to reveal differences of volatile components of royal jelly (RJ) produced by different honeybee stocks. Volatile components of three RJ samples from high and low RJ production honeybee stocks were extracted by headspace-solid phase micro-extraction and analyzed by gas chromatography-mass spectrometry. Principal component analysis, hierarchical clustering analysis of the volatile components are suggestive of the fact that honeybee stock selected for increasing RJ yields (royal jelly bees) has shaped distinct volatile component profile compared with the unselected Italian bees. In total, 37 components were identified that could distinguish three RJ samples, including 5 aldehydes, 5 esters, 5 alkanes, 14 alcohols and phenols, 4 ketones and 4 other components. Our study provides the first characterization of volatile components in RJs and uncovered the differences among royal jelly secreted by different bee stocks.

Identification of freshness and metabolite changes of royal jelly during storage using Nano-ESI-MS and UPLC-Q/TOF-MS

Shuai Zhuang,Huihui Ming,Wenjie Yu,Liping Luo

Royal jelly (RJ) is a highly perishable food during storage. However, the quality of RJ is hard to evaluate due to its complex and variable compositions. Nanoliter electrospray ionization mass spectrometry (nano-ESI-MS) is an emerging direct mass spectrometry method suitable for rapid analysis of complex food components and food quality. In this study, we investigated the freshness and metabolite changes of RJ stored at 4 °C and 25 °C using nano-ESI-MS and verified the results with UPLC-Q/TOF-MS. The nano-ESI-MS identified 20 compounds and quantified 9 metabolites with high stability and reproducibility. Multivariate analysis of the nano-ESI-MS data distinguished RJ of different freshness and suggested 45–60 and 7 days as the freshness threshold of RJ stored at 4 °C and 25 °C, respectively. The multivariate analysis of UPLC-Q/TOF-MS data output similar results to that of nano-ESI-MS. The relative abundance changes of metabolites identified by UPLC-Q/TOF-MS were approximately consistent with the quantification results of nano-ESI-MS, indicating the reliability of nano-ESI-MS in monitoring RJ freshness. This study proved the potential of nano-ESI-MS as a rapid method for monitoring RJ freshness and thus promoted the quality control of RJ during production.

Royal Jelly and 10-Hydroxy-2-Decenoic acid activate autophagy through mTOR/ULK1 pathway to improve cognitive function in diabetic mice

Lu Xiu,Rongguang Ge,Siyu Lu,Linwei Li,Wenqi Huang,Guojun Du,Zengli Zhang,Jie Zhang,Zhongxiao Wan,Jie Chang

Diabetes, a ruthless chronic metabolic disorder, is increasingly considered to jeopardize global well-being over the decades. Royal jelly (RJ) is a complex natural product secreted by worker bees containing a unique fatty acid known as 10-hydroxy-2-decenoic acid (10-HDA). This study aimed to verify the protective effect on cognitive function of RJ and 10-HDA in diabetic mice and explore the underlying mechanism. Briefly, our animal experiment results suggested that 10-HDA had a significant hypoglycemic effect and improved cognitive performance. Through western blotting, RJ and 10-HDA intervention notably increased Atg7, Atg16L1, Beclin-1, Atg5, Atg12, Atg3, and LC3-II expression and decreased p62 expression. Meanwhile, decreased mTOR expression and increased ULK1 expression were also observed in the 10-HDA intervention group. In conclusion, 10-HDA can regulate mTOR/ULK1 pathway to activate autophagy to alleviate hippocampal damage, thereby improving cognitive function in mice with diabetes, which may provide a new option for real-world cognitive impairment management.

Royal jelly acid alleviates diet-induced hyperlipidemia through regulation of oxidative stress and tryptophan metabolism

Dan-dan Zhi,Xi-ying He,Lin-fu Yang,Yun-fei Xue,Yi-qiu Liu,Dan Yue,Yi-nan Feng,Kun Dong,Ya-kai Tian

Royal jelly acid is a unique unsaturated fatty acid isolated from royal jelly. Recently, royal jelly acid was proposed to have potential therapeutic effects on hyperlipidemia. However, its effect on hyperlipidemia and the underlying molecular mechanism remains unclear. Therefore, in this study, we analyzed the mechanism of anti-hyperlipidemia of royal jelly acid through animal experiments and plasma metabolomics in conjunction with human network pharmacology and molecular docking analyses. We found that royal jelly acid could significantly decrease the serum lipid levels, ameliorate hepatic pathological injury, and reduce the level of oxidative stress in the experimental rats. A total of 41 key metabolites and 10 hub targets played key roles in the exertion of anti-hyperlipidemic effects, including tumor necrosis factor (TNF), insulin (INS) and epidermal growth factor receptor (EGFR). A total of 24 pathways, including tryptophan, citrate cycle, and arachidonic acid metabolisms, were identified as the key pathways involved in royal jelly acid-alleviated hyperlipidemia. The present findings provide new insights into the pathogenesis, diagnosis, and treatment targets of hyperlipidemia as well as contribute to the development and utilization of royal jelly acid related products.

Characterization of polyvinyl alcohol/chitosan nanofibers loaded with royal jelly by blending electrospinning for potential wound dressings

Hongying Yu,Di Chen,Wenjing Lu,Cen Zhang,Haiyan Wang,Zhengju Peng,Han Jiang,Chaogeng Xiao

This study aimed to fabricate a polyvinyl alcohol/chitosan (PVA/CS) nanofiber loaded with royal jelly (RJ) using blending electrospinning for potential wound dressings. The different PVA/CS ratios in electrospun nanofibers resulted in continuous nanofibers with an average diameter ranging from 219 to 299 nm. The FTIR spectra indicated that RJ was successfully incorporated into the nanofibers through hydrogen bonding with PVA/CS, which was further confirmed by the subsequent TGA experiments. Meanwhile, the RJ/PVA/CS nanofibers exhibited excellent water vapor permeability and hydrophilic properties. The encapsulation efficiency of RJ reached the maximum value of 89.00 %, while the cumulative release rate was up to 84.87 %. Furthermore, the RJ/PVA/CS nanofibers could inhibit the growth of Gram-positive ( Staphylococcus aureus ) and Gram-negative bacteria ( Escherichia coli ). The optimal PVA/CS ratio was determined to be 7:3, achieving inhibition rates of 97.83 % for S. aureus and 72.08 % for E. coli , demonstrating an excellent antibacterial performance. Therefore, this study successfully fabricated a wound dressing nanofiber with potential antibacterial efficacy.

Sugaring-Out Assisted Liquid-Liquid Extraction Combined with High-Performance Liquid Chromatography-Fluorescence Detection for the Determination of Bisphenol A and Bisphenol B in Royal Jelly

Tu Xijuan,Wu Siyuan,Liu Weiyi,Gao Zhaosheng,Huang Shaokang,Chen Wenbin

An analytical method based on sugaring-out assisted liquid-liquid extraction (SULLE) and HPLC-fluorescence detection has been developed for the determination of bisphenol A (BPA) and bisphenol B (BPB) in royal jelly. Experimental parameters of SULLE including the extraction solvent, amount of phase separation agent, initial concentration of acetonitrile (ACN), and sample size were investigated. The optimal SULLE procedure involves 0.3 g sample extracted by 2 mL ACN-water mixture (50%, v / v ), followed by phase separation with the addition of 0.4 g glucose. The obtained extract was injected into the reversed-phase HPLC system without clean-up step. The chromatography separation of BPA and BPB from royal jelly matrix was achieved in the mobile phase composed of 40% ACN and 60% water with isocratic elution at 1.0 mL/min. Limits of quantification for BPA and BPB were 40 μg/kg and 45 μg/kg, respectively, and limits of detection for BPA and BPB were 16 μg/kg and 18 μg/kg, respectively. Average recoveries of BPA and BPB at three spiked levels were in the range of 88.32~93.59% and 95.14~97.48%, respectively. Precision expressed as relative standard deviations (RSDs) in the interday and intraday analysis were all lower than 5%. The developed method was applied to the analysis of ten commercial and eight raw royal jelly samples. Results indicated that none of BPA and BPB were detected in these 18 samples. The proposed method is simple, fast, and sensitive, and could be used for the monitoring of BPA and BPB contamination in royal jelly.

Preventive Effect of Royal Jelly and 10-HDA on Skin Damage in Diabetic Mice through Regulating Keratinocyte Wnt/β-Catenin and Pyroptosis Pathway

Xinqi Huang,Lu Xiu,Yumei An,Yuan Gong,Sunao Li,Xueshi Chen,Chao Liu,Jianghuiwen Lu,Haiyan Shan,Jie Chang,Mingyang Zhang

The objective of this study is to elucidate how Royal jelly (RJ) and 10-hydroxy-2-decanoic acid (10-HDA) prevents diabetic skin dysfunction by modulating the pyroptosis pathway. Type 2 diabetes models are induced by fat diet consumption and low dose of streptozotocin (STZ) in C57BL/6J mice and treated with RJ (100 mg kg –1 day –1 ) and 10-HDA, the major lipid component of royal jelly (100 mg kg −1 day −1 ) for 28 weeks. The results show that serum concentrations of glucose and triglyceride are significantly lower in the RJ group or 10-HDA than diabetes mellitus (DM) group. Compared to the control group, pyroptosis proteins, GSDMD, ASC, Caspase-1, and IL-1β are increased in the skin of the diabetic model, accompanied by the activation of the Wnt/β-catenin signal pathway. Further evaluations by RJ exhibit superior improvement of skin damage, repress activation of the Wnt/β-catenin pathway, and attenuate keratinocyte pyroptosis, but 10-HDA cannot completely suppress the activation of Wnt/β-catenin pathway and pyroptosis, which shows a relatively weak protective effect on skin damage which shows that RJ is a better effect on skin injury after DM.

The effect of halogenation on the antimicrobial activity, antibiofilm activity, cytotoxicity and proteolytic stability of the antimicrobial peptide Jelleine-I

Fengjing Jia,Yi Zhang,Jiayi Wang,Jinxiu Peng,Ping Zhao,Lishi Zhang,Haiyan Yao,Jingman Ni,Kairong Wang

Antimicrobial peptides (AMPs) are believed to be a promising class of antimicrobial agents against bacteria and fungi. To promote the clinical use of AMPs, their antimicrobial activity and susceptibility to protease degradation should be further improved. The antimicrobial peptide Jelleine-I was originally isolated from the royal jelly of honeybees ( Apis mellifera ) with a short sequence of PFKLSLHL-NH 2 (953.24 Da). Here, a series of halogenated derivatives of the antimicrobial peptide Jelleine-I were designed and synthesized. The results showed that the in vitro antimicrobial activity, antibiofilm activity and in vivo antimicrobial efficacy were enhanced 1-8-fold after halogenation. Additionally, the proteolytic stability of Jelleine-I was improved 10-100-fold by halogenation. Meanwhile, the halogenated derivatives retained negligible hemolytic activity and cytotoxicity. Among these derivatives, the antimicrobial activity and antibiofilm activity of chlorine-Jelleine-I (Cl-J-I), bromine-Jelleine-I (Br-J-I), and iodine-Jelleine-I (I-J-I) were better than those of fluorine-Jelleine-I (F-J-I). The stabilities of Br-J-I and I-J-I against the degradation of enzymes and the serum were better than those of F-J-I and Cl-J-I. In conclusion, this study may offer a useful strategy to enhance antimicrobial efficacy and proteolytic stability by halogenation. The halogenated derivatives Cl-J-I, Br-J-I and I-J-I may be considered as potential antimicrobial agents against microbial infection.

Royal jelly attenuates LPS-induced immune dysfunction of dendritic cells via Nrf2/HO-1 axis

Jinyuan Wang,Yinyan Yin,Nuo Xu,Bangyue Zhou,Tao Qin

In the bacterial infection of humans, sepsis is of great concern based on its high fatality rate. Correction of immune dysfunction by targeting dendritic cells (DCs) is a promising strategy for sepsis therapy. Royal jelly, a natural health product, is produced by worker bees for feeding queens. In the study, we evaluated whether royal jelly could attenuate LPS-induced immune dysfunction of DCs. We found that royal jelly reduced the expression levels of mature phenotype (MHCII, CD40, CD80, and CD86), activation marker (CD69), and inflammatory cytokines (TNF-α, IL-6, and IL-10) in LPS-induced DCs. In addition, royal jelly enhanced the endocytosis levels and abrogated allogeneic CD4 + T cell proliferation in LPS-induced DCs. Finally, we found that the Nrf2/HO-1 axis played a vital role in the inhibition of LPS-induced immune dysfunction of DCs by royal jelly. These results indicated that royal jelly had a potential application in sepsis therapy by regulating DC function.

Royal jelly acid inhibits NF-κB signaling by regulating H3 histone lactylation to alleviate IgE-mediated mast cell activation and allergic inflammation

Xue-Ting Xu,Jun-Kai Fan,Xi Chen,Si-Yi Que,Run-Xin Zhang,Yu-Yan Xie,Tian-Ci Zhou,Kunmei Ji,Zhen-Fu Zhao,Jia-Jie Chen

Background Mast cells (MCs) mediate high-affinity IgE Fc receptor (FcεRI)-mediated allergic reactions. Royal jelly acid (RJA), the major lipid constituent of royal jelly, has anti-inflammatory, anti-tumor, antibacterial, and immunomodulatory properties. Purpose This study aimed to investigate how RJA affects IgE-mediated MC activation and to elucidate mechanisms underlying anti-allergic effects. Methods Mouse bone marrow-derived mast cells (BMMCs) and rat basophilic leukemia cells-2H3 (RBLs) were stimulated with IgE-antigen (Ag). β-hexosaminidase (β-hex) and histamine (HA) release, inflammatory cytokines expression, and cellular morphology changes were examined in stimulated cells. Transcriptome changes associated with RJA-treated activated-MCs were analyzed. Network pharmacology, gene expression, and western blot analyses were employed to explore the relationship between H3 lactylation and anti-allergic effects of RJA on MC. In vivo studies were conducted in IgE-mediated passive cutaneous anaphylaxis (PCA), ovalbumin (OVA)-induced active systemic anaphylaxis (ASA), and HA-induced passive systemic anaphylaxis (PSA) mouse models. Results RJA inhibited IgE-dependent MC activation as evidenced by reduced β-hex and HA release. RJA inhibited MAPK and NF-κB signaling, down-regulated RELA and NFκB1 transcripts, and decreased NF-κB reporter activity. RJA inhibited cellular H3 lactylation and H3K9 lactylation (H3K9la), leading to reduced histone lactyllysine enrichment in RELA or NFκB1 promoter loci. RJA alleviated allergic symptoms in PCA and ASA mice by intraperitoneal injection or oral administration, but did not affect HA-induced hypothermia. Conclusion RJA inhibits IgE-dependent MC activation by reducing cellular H3K9la and suppressing NF-κB signaling via reduced histone lactyllysine enrichment in RELA or NFκB1 promoter sites. RJA may support MC inhibition in the prevention and treatment of allergic diseases.

In situ formed scaffold with royal jelly-derived extracellular vesicles for wound healing

Dehong Tan,Wenxiang Zhu,Lujie Liu,Yuanwei Pan,Yangtao Xu,Qinqin Huang,Lingling Li,Lang Rao

Background: Safe and effective wound healing can be a major clinical challenge. Inflammation and vascular impairment are two main causes of inadequate wound healing. Methods: Here, we developed a versatile hydrogel wound dressing, comprising a straightforward physical mixture of royal jelly-derived extracellular vesicles (RJ-EVs) and methacrylic anhydride modified sericin (SerMA), to accelerate wound healing by inhibiting inflammation and promoting vascular reparation. Results: The RJ-EVs showed satisfactory anti-inflammatory and antioxidant effects, and significantly promoted L929 cell proliferation and migration in vitro . Meanwhile, the photocrosslinked SerMA hydrogel with its porous interior structure and high fluidity made it a good candidate for wound dressing. The RJ-EVs can be gradually released from the SerMA hydrogel at the wound site, ensuring the restorative effect of RJ-EVs. In a full-thickness skin defect model, the SerMA/RJ-EVs hydrogel dressing accelerated wound healing with a healing rate of 96.8% by improving cell proliferation and angiogenesis. The RNA sequencing results further revealed that the SerMA/RJ-EVs hydrogel dressing was involved in inflammatory damage repair-related pathways including recombinational repair, epidermis development, and Wnt signaling. Conclusion: This SerMA/RJ-EVs hydrogel dressing offers a simple, safe and robust strategy for modulating inflammation and vascular impairment for accelerated wound healing.
