microRNA-14 as an efficient suppressor to switch off ecdysone production after ecdysis in insects

Kang He,Huamei Xiao,Yang Sun,Gongming Situ,Yu Xi,Fei Li

The precise increase and decrease of hormone ecdysone are critical for accurate development in insects. Most previous works focus on transcriptional activation of ecdysone production; however, little is known about the mechanism of switching off ecdysone biosynthesis after ecdysis. Here, we showed that the precursor microRNA-14 (pre-miR-14) encodes two mature miRNAs in silkworm; both of these two mature miRNAs regulate various genes in the ecdysone-signalling pathway. Bmo-miR-14-5p targets on nine genes whereas Bmo-miR-14-3p targets on two genes in the same pathway. These two mature miRNAs increased immediately after the ecdysis, efficiently suppressing the 20-hydroxyecdysone (20E) biosynthesis, the upstream regulation, and the downstream response genes. Knocking down either of two mature miRNAs or both of them delays moult development, impairing development synchrony in antagomir-treated groups. In addition, overexpressing Bmo-miR-14-5p but not Bmo-miR-14-3p significantly affected the 20E titer and increased the moulting time variation, suggesting that Bmo-miR-14-5p, though it is less abundant, has more potent effects in development regulation than Bmo-miR-14-3p. In summary, we present evidence that a pre-miRNA encodes two mature miRNAs targeting on the same pathway, which significantly improves miRNA regulation efficiencies to programmatically switch off ecdysone biosynthesis.

20-Hydroxyecdysone and Receptor Interplay in the Regulation of Hemolymph Glucose Level in Honeybee (Apis mellifera) Larvae

Jing Yu,Hongfang Wang,Wenfeng Chen,Hongyu Song,Ying Wang,Zhenguo Liu,Baohua Xu

The hormone 20-hydroxyecdysone (20E) and the ecdysone receptors (ECR and USP) play critical roles in the growth and metabolism of insects, including honeybees. In this study, we investigated the effect of 20E on the growth and development of honeybee larvae by rearing them in vitro and found reduced food consumption and small-sized pupae with increasing levels of 20E. A liquid chromatography-tandem mass spectrometry (LC-MS/MS)-based analysis of widely targeted metabolomics was used to examine the changes in the metabolites after an exogenous 20E application to honeybee larvae and the underlying mechanisms. A total of 374 different metabolites were detected between the control group and the 20E treatment group, covering 12 subclasses. The most significant changes occurred in 7-day-old larvae, where some monosaccharides, such as D-Glucose and UDP-galactose, were significantly upregulated. In addition, some metabolic pathways, such as glycolysis/gluconeogenesis and galactose metabolism, were affected by the 20E treatment, suggesting that the 20E treatment disrupts the metabolic homeostasis of honeybee larvae hemolymph and that the response of honeybee larvae to the 20E treatment is dynamic and contains many complex pathways. Many genes involved in carbohydrate metabolism, including genes of the glycolysis and glycogen synthesis pathways, were downregulated during molting and pupation after the 20E treatment. In contrast, the expression levels of the genes related to gluconeogenesis and glycogenolysis were significantly increased, which directly or indirectly upregulated glucose levels in the hemolymph, whereas RNA interference with the 20E receptor EcR-USP had an opposite effect to that of the 20E treatment. Taken together, 20E plays a critical role in the changes in carbohydrate metabolism during metamorphosis.Keywords:20-hydroxyecdysone;metabolomics;hemolymph;glucose;ecdysone receptors

Novel regulation pathway of eclosion hormones in Tribolium castaneum by distinct transcription factors through the initiation of 20-Hydroxyecdysone

Huiling Zhou,Gaoke Lei,Yusi Li,Peng Chen,Zhiping Liu,Chengjun Li,Bin Li

Eclosion hormone (EH) is not only a key trigger of insect ecdysis, but is also involved in the regulation of important physiological processes such as development, diapause, metamorphosis and reproduction. EH is an ideal target for RNAi treatment and prevention of the Tribolium castaneum . However, two EH genes in Tribolium castaneum demonstrate distinct replication and functional conversion relationships, and the mechanisms of transcriptional regulation of eclosion hormone remain largely unexplored and poorly understood. In this study, the activity of highly active promoter fragments and potential transcription factors of TcEH and TcEHL were identified using the Dual-Luciferase reporter system and TANSFAC. TcSlbo and TcCAD were revealed to be important transcription factors for TcEH and TcEHL , respectively. Knockdown of TcSlbo failed to slough off the old epidermis of Tribolium castaneum and prevented them from developing into adults. Furthermore, we demonstrated for the first time that 20-hydroxyecdysone (20E) affects the expression of TcEH and TcEHL by regulating the transcriptional activities of TcSlbo and TcCAD. This study provides new insights into the transcription regulation of TcEH and TcEHL , their roles in insect growth and development, and the involvement of 20E in eclosion regulation, offering potential molecular targets for future pest management strategies.

Herbicide prometryn adversely affects the development and reproduction of Tigriopus japonicus by disturbing the ecdysone signal pathway and chitin metabolic pathway

Dong Wang,Guangxin Yang,Shaoguo Ru,Zhenzhong Zhang,Yuejiao Li,Jun Wang

Prometryn, a widely used triazine herbicide in agriculture and aquaculture, has been commonly detected in marine environments, but its effects on the marine copepod are unknown. In this study, marine copepod Tigriopus japonicus was chronically exposed to environmentally relevant concentrations of prometryn to investigate its impacts and potential mechanism of action. The results showed that 0.5, 5, and 50 μg/L prometryn delayed the first spawning time and hatching time, reduced the fecundity, and inhibited the population growth rate. Moreover, exposure to 0.5, 5 and 50 μg/L prometryn decreased food ingestion, the content of C and N elements, nutrient accumulation and body size, but increased the content of 20-hydroxyecdysone (20E). Transcriptome analysis showed that 50 μg/L prometryn down-regulated 1431 genes, which were mainly enriched in lysosome pathway and chitin binding and cuticle construction process. The results of qRT-PCR showed that the expression of key genes involved in juvenile hormone synthesis and chitin metabolic pathways were also inhibited after prometryn exposure. Molecular docking revealed that prometryn could bind to ecdysone receptor (EcR) and UDP-N-acetylglucosamine pyrophosphorylase (UAP), components of the ecdysteroid nuclear receptor complex. Therefore, environmental relevant prometryn delayed the molting and development of T. japonicus by disrupting the ecdysone signal pathway and chitin metabolic pathway through binding to EcR and UAP. This study provides new insights into toxic effects and molecular mechanisms of prometryn on marine copepods.

The loss of Halloween gene function seriously affects the development and reproduction of Diaphorina citri (Hemiptera: Liviidae) and increases its susceptibility to pesticides

Cong Zhang,Bin Wan,Meng-Ru Jin,Jing Wang,Tian-Rong Xin,Zhi-Wen Zou,Bin Xia

The citrus industry has suffered severe losses as a result of Huanglongbing spread by Diaphorina citri . Controlling the population of D. citri is the key to preventing and controlling the spread of Huanglongbing. Ecdysteroids are key hormones that regulate insect development and reproduction. Therefore, the Halloween gene family involved in the ecdysone synthesis of D. citri is an ideal target for controlling the population growth of this insect. In this study, we successfully cloned four Halloween genes expressed during D. citri development. Silencing of one of the four genes resulted in a significant decrease in 20E titers in nymphs and significant decreases in the developmental, survival and emergence rates. Inhibiting Halloween gene expression in adults impeded the growth of the female ovary , diminished yolk formation , lowered vitellogenin transcription levels, and hence impaired female fecundity. This showed that Halloween genes were required for D. citri development and reproduction. DcCYP315A1 and DcCYP314A1 were highly expressed when D. citri was exposed to thiamethoxam and cypermethrin, and silencing these two genes made D. citri more sensitive to these two pesticides . Inhibition of DcCYP315A1 and DcCYP314A1 expression not only significantly delayed the development and reproduction of D. citri but also increased its susceptibility to pesticides. Therefore, these two genes are more suitable as potential target genes for controlling D. citri .

RNA interference targeting β-N-acetylhexosaminidase genes impairs molting and development of Tetranychus urticae

Ming Liu,Qianwen Wang,Bin Lai,Yan Chen,Rongchumu Ge,Shuo Yan,Chunya Bu

β- N -acetylhexosaminidases (HEXs) are key chitin-degrading enzymes in insects. Here, we identified TuHex1 and TuHex2 using insect orthologous genes by searching Tetranychus urticae genome and transcriptome database to investigate their roles in mite molting. TuHex1 and TuHex2 expression was induced by 20-hydroxyecdysone (20E), and inhibition of TuHex1 and TuHex2 expression by RNAi resulted in wrinkled cuticle or an inability to shed the old cuticle in nymphs, which may be due to a reduction in particle deposition in the exocuticle and lamellar structure in the endocuticle as revealed by scanning electron microscopy and transmission electron microscopy. The results suggest that the TuHex1 and TuHex2 genes play an essential role in the molting and developmental process of the mite. TuHex2 , with a mortality rate of 67.41 % in the leaf disc assay, was a potential RNAi target by oral feeding. Spraying of nanocarrier-delivered bacteria expressing ds TuHex2 at 500 ng/μL kept spider mites at a consistently low level throughout the 14 days and showed good mite control comparable to that of matrine. In addition, nanocarrier-delivered ds TuHex2 is safe for Neoseiulus californicus in our experiments, providing its potential for green mite management.

Transcriptome Analysis Reveals the Endocrine Regulation on the Expression of IAG in Litopenaeus vannamei

Kangxuan Chen,Shihao Li,Jianhai Xiang,Amir Sagi,Fuhua Li

The insulin-like androgenic gland hormone (IAG) plays a key role in male sexual differentiation and spermatogenesis in crustaceans. The expression of IAG is usually negatively regulated by neuropeptide hormones through the “eyestalk-AG” endocrine axis. However, the underlying mechanism is still largely unknown. In the present study, we performed a comparative transcriptome analysis on the androgenic gland (AG), the main secretory organ of IAG, ofL. vannameibefore and after unilateral eyestalk ablation. A total of 67 differentially expressed genes (DEGs) were identified, including some putative genes involved in sexual development. Interestingly, several genes related to molting and endocrine processes were found differentially expressed between the two treatments. Further investigation on the expression profiles of these genes andLv-IAGshowed that their expression patterns were closely correlated throughout the molting cycle. In addition, injection of 20-hydroxyecdysone (20E) could inhibit the expression ofLv-IAGat different molting stages in a dosage-dependent manner. The data indicated that ecdysteroids played important roles in regulating theLv-IAGexpression and might bridge the endocrine axis between eyestalk and AG. The present study provided new insights into understanding the regulation of sexual development in male crustacean.Keywords:insulin-like androgenic gland hormone;sexual development;endocrine axis;ecdysteroids;crustacean

Functional analysis of ecdysteroid biosynthetic enzymes of the rice planthopper, Nilaparvata lugens

Xiang Zhou,Yi-Zhou Ye,Mari H. Ogihara,Mika Takeshima,Daiki Fujinaga,Cheng-Wen Liu,Zhen Zhu,Hiroshi Kataoka,Yan-Yuan Bao

Ecdysteroids , insect steroid hormones , play key roles in regulating insect development and reproduction. Hemipteran insects require ecdysteroids for egg production; however, ecdysteroid synthesis (ecdysteroidogenesis) details have not been elucidated. We identified all known genes encoding ecdysteroidogenic enzymes in Nilaparvata lugens and clarified their necessity during nymphal and ovarian development . We confirmed that N. lugens utilized 20-hydroxyecdysone as an active hormone. Assays using heterologous expression of enzymes in Drosophila S2 cells showed conserved functions of enzymes Neverland, CYP306A2, CYP314A1 and CYP315A1, but not CYP302A1. RNA interference and rescue analysis using 20-hydroxyecdysone demonstrated that most of the genes were necessary for nymphal development. The identified N. lugens enzymes showed conserved functions and pathways for ecdysteroidogenesis . Knockdown of ecdysteroidogenic enzyme genes in newly molted females caused failure of egg production: less vitellogenic and mature eggs in ovaries , fewer laid eggs and embryonic development deficiency of laid eggs. Considering the high expressions of ecdysteroidogenic enzyme genes in adults and ovaries, ecdysteroidogenesis in ovaries was critical for N. lugens ovarian development. Our study presents initial evidence that hemipteran insects require ecdysteroidogenesis for ovarian development.

Knockdown of the β-N-acetylhexosaminidase genes by RNA interference inhibited the molting and increased the mortality of the white-backed planthopper, Sogatella furcifera

Pan-Pan Guo,Xi-Bin Yang,Hong Yang,Cao Zhou,Gui-Yun Long,Dao-Chao Jin

β -N-Acetylglucosaminidases and/or β - N -acetylhexosaminidases (NAGs / Hexes) are crucial exonucleases, playing a crucial role in the insect molting process. SfHex3 and SfHex4 contain conserved catalytic domains of GH20 and GH20b, clustered into NAG2 and NAG1 group, respectively. SfHex3 and SfHex4 were mainly highly expressed in the 4th–5th instar nymphs, as well as in the integument and ovary. The expression level of SfHex3 gradually decreased in male and female adults, and SfHex4 on the first day of female was significantly higher than that on the first day of male. In addition, RNA interference (RNAi) results demonstrated that the downregulation of SfHex3 and SfHex4 expression in 5th-instar nymphs resulted in failed molting, and a high mortality. Furthermore, after RNAi with SfHex3 and SfHex4 , the transcript levels on key genes of the chitin metabolism pathway ( SfCHS1 , SfCHS1a , SfCHS1b , SfTRE1 , SfTRE2 , SfCht5 , and SfCht7 ) were significantly decreased compared to the control group. Meanwhile the expression levels of SfHex3 and SfHex4 were up-regulated after 6 h and 12 h of 20E treatment. And the transcription levels of SfHex3 and SfHex4 were significantly inhibited at nitenpyram LC 20 , LC 50 , and LC 90 after 96 h of treatment, in 3rd nymphs of Sogatella furcifera . In conclusion, SfHex3 and SfHex4 play important roles in the nymphal development of S. furcifera , contributing to the molting process from nymph to adult. This study not only enhances our understanding of the nitenpyram in pest control, but also provides a foundation for the development of new control strategies using RNAi to targeting SfHex3 and SfHex4 .

Acute and Chronic Toxicity of Carbamazepine on the Release of Chitobiase, Molting, and Reproduction in Daphnia similis

Huihui Chen,Xiaohong Gu,Qingfei Zeng,Zhigang Mao

As one of the most frequently detected pharmaceutical compounds in aquatic environments, carbamazepine (CBZ) has recently been shown to cause acute and chronic toxicity in a variety of non-target aquatic organisms. However, little is known about the ecotoxicological effects it has on the molting and reproduction of crustaceans. The aim of the present work was to evaluate the acute and chronic toxic responses to CBZ in the crustaceanDaphnia similis. After acute exposure (4 days), CBZ did not cause lethal toxicity at the tested concentrations. However, CBZ did inhibit the molting and release of chitobiase at concentrations higher than 6.25 μg/L, with 96 h EC50(median effective concentration) values of 864.38 and 306.17 μg/L, respectively. The results of chronic exposure showed that the mean number of molts, size of the first brood, mean number of offspring per brood, mean number of broods per female, and total offspring per female decreased significantly with increasing CBZ concentrations. Significant effects of CBZ on the molting or fecundity inD. similiswere observed even at concentrations as low as 0.03 μg/L. In conclusion, CBZ can cause inhibition of molting, delayed reproduction, and reduced fecundity inD. similis. CBZ toxicity toD. similisdepends on the timing and duration of the exposure. Moreover, our results indicated that CBZ would act as an endocrine disrupter inD. similis, as with vertebrates (e.g., fish). Keywords:carbamazepine;molting;Daphnia similis;chitobiase;chronic toxicity;reproduction

Effects of Juvenile Hormone Analogue and Ecdysteroid on Adult Eclosion of the Fruit Fly Bactrocera minax (Diptera: Tephritidae)

Wang Jia,Zhou Hong-Yan,Zhao Zhi-Mo,Liu Ying-Hong

The univoltine Bactrocera minax (Enderlein) is a devastating pest of citrus in Nepal, India, Bhutan, and China. To better understand the overwintering pupal diapause, we investigated the juvenile hormone III (JH III) and ecdysteroid 20-hydroxyecdysone (20E) titers across the developmental stages, and the effects of juvenile hormone analog (JHA) and 20E treatments on adult emergence patterns. The results showed that both JH III and 20E levels fluctuated from the late larval stage to the late pupal stage. JHA and 20E treatments at the late larval stage had marginal effect on the adult emergence patterns, with slightly faster adult eclosion and higher eclosion rate. Similarly, JHA treatment at the early pupal stage did not affect the adult emergence. However, 20E treatment at the early pupal stage remarkably shortened the duration of diapause, but lowered the eclosion rate, especially with relatively high dose. These findings demonstrated that 20E treatment at the early pupal stage is a suitable method to hasten the diapause termination of B. minax, which will contribute to the rearing of B. minax in the laboratory as needed for routine experiments.

Silencing of LIM homeodomain transcription factor 1 alpha (Lmx1a) gene caused larval molting failure and adult reproductive deficiency in Henosepilachna vigintioctopunctata

Junna Liu,Zhaoyang Li,Zexin Zhong,Jianxing Li,Qi Chen,Ran Gao,Xuguo Zhou,Youjun Zhang,Chunxiao Yang,Huipeng Pan

LIM-HD (homeodomain) protein is essential for the specific differentiation of various cells and tissues due to its conserved HD. Within the LIM subclass, the LIM-HD transcription factor 1 alpha ( Lmx1a ) remains poorly characterized in insects. This study investigates the expression patterns of HvLmx1a in the 28-spotted lady beetle, Henosepilachna vigintioctopunctata , at both developmental stage and tissue levels. Furthermore, RNA interference (RNAi)-mediated knockdown of HvLmx1a resulted in increased mortality during the early larval stage. Injection of ds HvLmx1a at the beginning of the 4th instar caused a reduction in 20E titer, disrupted normal molting and pupation processes, and led to the emergence of larval-pupal abnormalities. Notably, HvLmx1a expression was significantly down-regulated on d 2 and 4 post-injection, which coincided with a marked inhibition of HvCHS1 and genes associated with 20E and bursicon signaling pathways. Additionally, silencing HvLmx1a induced qualitative changes in male testes and female ovaries, resulting in infertility and increased mortality. Ultimately, these findings suggest that HvLmx1a influences ovarian morphology and development through lipid metabolism. In conclusion, this study provides the first evidence of the diverse physiological roles of Lmx1a in H. vigintioctopunctata and highlight its potential as a target for RNAi-based biological control strategies.
