Background The vast majority of systemic lupus erythematosus patients develop lupus nephritis (LN) with severe renal manifestations, such as inflammatory responses, oxidative stress, and fibrosis. The purpose of this research was to investigate Cordyceps cicadae as a potential therapeutic target for treating inflammatory responses, oxidative stress, and fibrosis in LN. Methods The effects of C. cicadae on lupus symptoms in mice with LN were determined. MRL/lpr mice were treated with C. cicadae (4 g/kg/day, i.e., CC group, n = 8) or an equal volume of saline (model group, n = 8), and MRL/MP mice were treated with an equal volume of saline (control group, n = 8). Renal function indices, renal pathology, inflammatory markers, oxidative stress markers, and renal interstitial fibrosis levels were evaluated after C. cicadae treatment. Western blot analysis was performed to investigate the effect of C. cicadae on the expression of fibrosis biomarkers and the phosphatidylinositol 3-kinase (PI3K)/mammalian target of rapamycin (mTOR)-mediated autophagy pathway in the renal tissues of mice. Results C. cicadae ameliorated renal lesions, the inflammatory response, and oxidative stress damage in MRL/lpr mice. C. cicadae treatment inhibited renal fibrosis (16.31 ± 4.16 vs. 31.25 ± 5.61) and downregulated the expression of the fibrosis biomarkers alpha-smooth muscle actin, fibronectin, and collagen I (COL I) in the kidneys of MRL/lpr mice. In addition, further research showed that the PI3K/mTOR-mediated autophagy pathway was involved in C. cicadae -mediated effects on renal fibrosis in MRL/lpr mice. Furthermore, the therapeutic effect of C. cicadae on repairing renal fibrosis and damage in MRL/lpr mice was abolished by the PI3K agonist 740 Y-P. Conclusions The findings of the present research showed that C. cicadae could alleviate inflammatory responses, oxidative stress, and fibrosis in the renal tissues of mice with LN by targeting the PI3K/mTOR-mediated autophagy pathway.

In this study, the in vitro digestion and fermentation of two intra-polysaccharide fractions (IPS1 and IPS2) from Paecilomyces cicadae TJJ1213 were investigated. The constituent monosaccharides of IPS1 and IPS2 were not changed after simulated saliva, gastric and small intestinal digestion. However, they can be hydrolyzed and utilized by gut microbiota, and short-chain fatty acids (SCFAs) level were increased after IPS1and IPS2 treatments. Furthermore, 16S rRNA sequencing analysis of fermentation samples were performed. Alpha-diversity, beta-diversity and taxonomic composition differences analysis revealed that IPS1 and IPS2 promoted the proliferation of beneficial bacteria and modulated the overall structure of gut microbiota. Taxonomic comparison analysis found that IPS1 increased the relative abundances of beneficial bacteria including Megamonas , Bifidobacterium and Lactobacillus , while IPS2 could increase the abundance of Bacteroides , Parabacteroides and Phascolarctobacterium. In addition, they can also decrease the levels of pathogenic bacteria containing Escherichia-Shigella , Klebsiella and Fusobacterium . These results indicated that IPS from P. cicadae TJJ1213 could be used as potential candidates for new functional foods.

Two intracellular polysaccharide fractions (IPS1 and IPS2) were obtained from the mycelium of Paecilomyces cicadae TJJ1213, and the structures were conducted. Results showed that they were homogenous with the average molecular weight of 2.40 × 10 6 Da and 6.79 × 10 5 Da. Two fractions were composed of mannose, glucose and galactose with molar ratios of 1.35: 6.93: 1.0 and 2.04: 1.0: 1.87, respectively. The backbone of IPS1 was → 4)-α-D-Glc p (1 → and → 3,4)-α-D-Man p (1 → residues with a side chain consisted of T-α-D-Gal p . IPS2 was consisted of → 4)-α-D-Glc p -(1→, →3,4)-α-D-Man p -(1 → and → 2,6)-α-D-Man p -(1 → residues and the branches were also consisted of T-α-D-Gal p . In addition, the scanning electron microscope and atomic force microscope images presented different features of IPS1 and IPS2, respectively. Furthermore, two fractions exhibited better immunomodulatory effects. They could markedly promote the proliferation of RAW264.7 cells and enhance phagocytosis, nitric oxide release and cytokines production. These results indicated that IPS1 and IPS2 had potential to enhance immune responses.

This study aimed to explore the in vitro simulated digestion and fecal fermentation behaviors of two purified exopolysaccharide fractions (EPS1 and EPS2) from Paecilomyces cicadae TJJ1213 and its effects on human gut microbiota composition. Results showed that EPS1 and EPS2 could not be digested by saliva-gastrointestinal. After fecal fermentation, however, the molecular weight of EPS1 and EPS2 significantly decreased, and the molar ratios of the monosaccharide composition remarkably changed, indicating that EPS1 and EPS2 could be degraded and consumed by human gut microbiota. Notably, EPS1 and EPS2 could significantly modulate the composition, via increasing the relative abundances of Bacteroides and Phascolarctobacterium and decreasing the pathogenic bacteria Escherichia-Shigella, Klebsiella and Fusobacterium . In addition, EPS1 and EPS2 also could promote the production of short-chain fatty acids during fermentation for 24 h. These results suggested that EPS from Paecilomyces cicadae TJJ1213 can be used as a functional food to improve health and prevent diseases by promoting gut health.
