A self-fused peptide-loaded hydrogel with injectability and tissue-adhesiveness for preventing postoperative peritoneal adhesions

Zequn Zhang,Chao Yin,Xianwen Song,Xi Liu,Chonglei Zhong,Jun Zheng,Yaqiong Ni,Rujuan Shen,Yihang Guo,Xiaorong Li,Changwei Lin,Yi Zhang,Gui Hu

Peritoneal adhesions commonly occur following abdominal or pelvic surgery and can cause serious complications. Currently, physical barriers are the primary approach used in clinical practice to prevent adhesion, although their effectiveness is frequently inadequate. In this study, we developed an injectable peptide-loaded hydrogel with multiple functions, including self-fusion, tissue-adhesiveness, anti-inflammation, anti-cell adhesion and anti-angiogenesis. To assess the effectiveness of these hydrogels, which are stabilized by dynamic imine bonds and acetal connections, in preventing postoperative abdominal adhesions, we utilized both a rat abdominal adhesion model and a rat model simulating repeated-injury adhesions. In comparison to the commercially available HA hydrogel, as-prepared hydrogels exhibited significant reductions in inflammation, fibrosis, and angiogenesis, leading to an obvious decrease in peritoneal adhesions. Moreover, this peptide-loaded hydrogel demonstrated an ideal degradation time, maintaining an in vivo viability for about 10 days. We believe this peptide-loaded hydrogel presents a promising solution for the challenging clinical issue of postoperative abdominal adhesions.

Fusion Peptide-Engineered Polyetheretherketone Implants with Photo-Assisted Anti-Pathogen and Enhanced Angiogenesis for in vivo Osseointegrative Fixation

Shuai He,Chunyan Duan,Song Wang,Yue Yu,Yau Kei Chan,Xiuyuan Shi,Jiahu Huang,Shouteng Wang,Shaojun Peng,Yi Deng

Owing to its superior mechanical and physicochemical characteristics, polyetheretherketone (PEEK) is considered a potent orthopedic material. Yet, poor bactericidal, osseointegrative and angiogenic properties of bioinert PEEK collectively hamper its clinical adoption. Herein, we present a strategy for developing multifunctional hierarchical coatings bearing a synthetic fusion peptide and polydopamine/silver nanoparticles (pDA/AgNPs) bio-heterojunctions (bio-HJs), in an attempt to empower PEEK with triple-modal abilities, which prevents bacterial infection, enhances ossification and promotes re-vascularization for physiological osseointegrative fixation. By harnessing the adhesion of bioinspired pDA, AgNPs are facilely immobilized onto peripheral surface and incorporated into interior cavity of micro/nano-porous sulfonated PEEK (SP) to form hierarchical pDA/AgNPs heterostructures. The bio-HJs coatings exhibit robust and repeatable bacteria-eradicating ability, assisted by near-infrared illumination, due to the synergistic effects of the metal ion/photothermal therapy. The fusion peptide-engineered PEEK exhibits osteogenesis activity in terms of promoting osteoblast proliferation, spreading, boosting the synthesis and secretion of osteogenesis-related genes. Besides, the fusion peptide-engineered PEEK also has the ability to promote HUVECs growth, reproduction, and tube formation, facilitating the synthesis and secretion of angiogenesis-associated genes. Moreover, the results of the in vivo experiments further corroborate the in vivo osteointegration and angiogenic abilities of materials. Expectantly, the multifunctional coatings with the presented triple-therapeutic effects represent a prospective clinical solution to address the current clinical challenges associated with the use of PEEK implants.

Fusion peptide engineered “statically-versatile” titanium implant simultaneously enhancing anti-infection, vascularization and osseointegration

Junjian Chen,Guansong Hu,Tianjie Li,Yunhua Chen,Meng Gao,Qingtao Li,Lijing Hao,Yongguang Jia,Lin Wang,Yingjun Wang

Although antimicrobial titanium implants can prevent biomaterial-associated infection (BAI) in orthopedics, they display cytotoxicity and delayed osseointegration . Therefore, versatile implants are desirable for simultaneously inhibiting BAI and promoting osseointegration, especially “statically-versatile” ones with nonessential external stimulations for facilitating applications. Herein, we develop a “statically-versatile” titanium implant by immobilizing an innovative fusion peptide (FP) containing HHC36 antimicrobial sequence and QK angiogenic sequence via sodium borohydride reduction promoted Cu(I)-catalyzed azide-alkyne cycloaddition (CuAAC-SB), which shows higher immobilization efficiency than traditional CuAAC with sodium ascorbate reduction (CuAAC-SA). The FP-engineered implant exhibits over 96.8% antimicrobial activity against four types of clinical bacteria ( S. aureus , E. coli , P. aeruginosa and methicillin-resistant S. aureus ), being stronger than that modified with mixed peptides. This can be mechanistically attributed to the larger bacterial accessible surface area of HHC36 sequence. Notably, the implant can simultaneously enhance cellular proliferation , up-regulate expressions of angiogenesis-related genes/proteins (VEGF and VEGFR-2) of HUVECs and osteogenesis-related genes/proteins (ALP, COL-1, RUNX-2, OPN and OCN) of hBMSCs . In vivo assay with infection and non-infection bone-defect model reveals that the FP-engineered implant can kill 99.63% of S. aureus , and simultaneously promote vascularization and osseointegration. It is believed that this study presents an excellent strategy for developing “statically-versatile” orthopedic implants .
