Dynamics of oppositely charged emulsion droplets

Liu Zhou,Wyss Hans M.,Fernandez-Nieves Alberto,Shum Ho Cheung

We study the dynamics of two pinned droplets under the influence of an applied electric stress. We find that at a sufficiently strong field, this stress is sufficient to induce contact of the droplets. Interestingly, upon such contact, the dynamic behavior sensitively depends on the separation distance between the droplets. Besides the classical “coalescence” regime, we identify two other dynamic regimes: “fuse-and-split” and “periodic non-coalescence.” In the “fuse-and-split” regime, the droplets first fuse to form a jet, which subsequently breaks up into two droplets. In the “periodic non-coalescence” regime, the droplets contact and bounce away periodically without coalescence. Further analysis indicates that while the electric stress stretches the droplets into shapes that depend on the initial droplet separation, the surface tension stress dominates over the electric stress as soon as the droplets touch. We show that the shapes of the contacting droplets determine their subsequent dynamics. Our work provides a rationale for understanding the interplay between surface tension and electric stresses that govern the behavior of charged droplets and could inspire new methods for characterizing emulsion stability and surfactant performance.

High Strength and Shape Memory Spinal Fusion Device for Minimally Invasive Interbody Fusions

Min Liu,Bo Liu,Ziyang Liu,Zhen Yang,Thomas J Webster,Huan Zhou,Lei Yang

Introduction Lumbar interbody fusion is widely employed for both acute and chronic spinal diseases interventions. However, large incision created during interbody cage implantation may adversely impair spinal tissue and influence postoperative recovery. The aim of this study was to design a shape memory interbody fusion device suitable for small incision implantation. Methods In this study, we designed and fabricated an intervertebral fusion cage that utilizes near-infrared (NIR) light-responsive shape memory characteristics. This cage was composed of bisphenol A diglycidyl ether, polyether amine D-230, decylamine and iron oxide nanoparticles. A self-hardening calcium phosphate-starch cement (CSC) was injected internally through the injection channel of the cage for healing outcome improvement. Results The size of the interbody cage is reduced from 22 mm to 8.8 mm to minimize the incision size. Subsequent NIR light irradiation prompted a swift recovery of the cage shape within 5 min at the lesion site. The biocompatibility of the shape memory composite was validated through in vitro MC3T3-E1 cell (osteoblast-like cells) adhesion and proliferation assays and subcutaneous implantation experiments in rats. CSC was injected into the cage, and the relevant results revealed that CSC is uniformly dispersed within the internal space, along with the cage compressive strength increasing from 12 to 20 MPa.Conclusion The results from this study thus demonstrated that this integrated approach of using a minimally invasive NIR shape memory spinal fusion cage with CSC has potential for lumbar interbody fusion.

Shielding Electric Fields to Prevent Coalescence of Emulsions in Microfluidic Channels Using a 3D Metallic Coil

Jingmei Li,Zhou Liu,Haibo Huang,Ho Cheung Shum

In microfluidics, electric fields are widely used to assist the generation and the manipulation of droplets or jets. However, uncontrolled electric field can disrupt the operation of an integrated microfluidic system, for instance, through undesired coalescence of droplets, undesired changes in the wettability of the channel wall or unexpected death of cells. Therefore, an approach to control the distribution of electric fields inside microfluidic channels is needed. Inspired by the electro-magnetic shielding effect in electrical and radiation systems, we demonstrate the shielding of electric fields by incorporating 3D metallic coils in microfluidic devices. Using the degree of coalescence of emulsion drops as an indicator, we have shown that electric fields decrease dramatically in micro-channels surrounded by these conductive metallic coils both experimentally and numerically. Our work illustrates an approach to distribute electric fields in integrated microfluidic networks by selectively installing metallic coils or electrodes, and represents a significant step towards large-scale electro-microfluidic systems. Keywords:droplet coalescence;electric field;microfluidics;coils;shielding

Lycopene detection in cherry tomatoes with feature enhancement and data fusion

Yuanhao Zheng,Xuan Luo,Yuan Gao,Zhizhong Sun,Kang Huang,Weilu Gao,Huirong Xu,Lijuan Xie

Lycopene, a biologically active phytochemical with health benefits, is a key quality indicator for cherry tomatoes. While ultraviolet/visible/near-infrared (UV/Vis/NIR) spectroscopy holds promise for large-scale online lycopene detection, capturing its characteristic signals is challenging due to the low lycopene concentration in cherry tomatoes. This study improved the prediction accuracy of lycopene by supplementing spectral data with image information through spectral feature enhancement and spectra-image fusion. The feasibility of using UV/Vis/NIR spectra and image features to predict lycopene content was validated. By enhancing spectral bands corresponding to colors correlated with lycopene, the performance of the spectral model was improved. Additionally, direct spectra-image fusion further enhanced the prediction accuracy, achieving R P 2 , RMSEP, and RPD as 0.95, 8.96 mg/kg, and 4.25, respectively. Overall, this research offers valuable insights into supplementing spectral data with image information to improve the accuracy of non-destructive lycopene detection, providing practical implications for online fruit quality prediction.

Photo-controlled Hierarchical Assembly and Fusion of Coumarin-containing Polydiacetylene Vesicles

Jingguo Li,Hao Jiang,Wenlong Hu,Hongyan Xia,Gang Zou,Qijin Zhang

Herein, we synthesize a coumarin-substituted diacetylene monomer (CODA) and report the novel photo-controlled reversible assembly and disassembly behavior of the polymerized CODA (PCODA) vesicles. The photo-triggered dimerization and cleavage reactions of the coumarin groups within the surface of the adjacent PCODA vesicles can be utilized as the driving force to induce assembly and disassembly of PCODA vesicles. Moreover, the boundary of PCODA vesicles in the aggregates becomes more obscure when the irradiation time exceeds 30 min. Fusion occurs upon close docking of target membranes, driven by sufficient dimerization of the coumarin groups within the surface of PCODA vesicles.

Light-Cured Junction Formation and Broad-Band Imaging Application in Thermally Mismatched van der Waals Heterointerface

Liyuan Cheng,Qinglin Quan,Liang Hu

Van der Waals (vdW) heterostructures are mainly fabricated by a classic dry transfer procedure, but the interface quality is often subject to the vdW gap, residual strains, and defect species. The realization of interface fusion and repair holds significant implications for the modulation of multiple photoelectric conversion processes. In this work, we propose a thermally mismatched strategy to trigger broad-band and high-speed photodetection performance based on a type-I heterostructure composed of black phosphorus (BP) and FePS3(FPS) nanoflakes. The BP acts as photothermal source to promote interface fusion when large optical power is adopted. The regulation of optical power enables the device from pyroelectric (PE) and/or alternating current photovoltaic (AC–PV) mode to a mixed photovoltaic (PV)/photothermoelectric (PTE)/PE mode. The fused heterostructure device presents an extended detection range (405~980 nm) for the FPS. The maximum responsivity and detectivity are 329.86 mA/W and 6.95 × 1010Jones, respectively, and the corresponding external quantum efficiency (EQE) approaches ~100%. Thanks to these thermally-related photoelectric conversion mechanism, the response and decay time constants of device are as fast as 290 μs and 265 μs, respectively, superior to current all FPS-based photodetectors. The robust environmental durability also renders itself as a high-speed and broad-band imaging sensor.

High-throughput design and performance validation of superior latent heat eutectic salt materials

Fengyi Yang,Yimin Xuan,Xianglei Liu

Eutectic salts are widely utilized as latent heat storage materials in various thermal energy storage and management applications. However, developing efficient and accurate methods to predict the thermophysical properties of molten salts and guide their design remains a significant challenge. This study focuses on thermal properties, constructing an element gene feature database, and introducing an integrated machine-learning strategy for the design, screening, and evaluation of eutectic salts. Marginal contribution analysis reveals that the melting point is primarily influenced by features related to structural stability, while the fusion enthalpy is strongly associated with features affecting atomic bonding strength. Based on these insights, five novel eutectic salts are designed and experimentally validated. Among them, NaCl-NaF (66.0–34.0 mol%) exhibits a melting point of 683.7 °C and a fusion enthalpy of 588.8 J/g. High-temperature testing demonstrates that the prediction accuracies for the melting point and fusion enthalpy of the five salts are 94.49 % and 95.01 %, respectively. This study presents an effective and efficient approach for the precise targeted design of novel eutectic salts, offering expanded possibilities and options for the efficient utilization of renewable energy.

Non-coalescence and chain formation of droplets under an alternating current electric field

Xin Huang,Limin He,Xiaoming Luo,Ke Xu,Yuling Lü,Donghai Yang

The dynamic behaviors of two droplets and droplet cluster under an alternating current (AC) electric field are investigated. Two droplets generally undergo transformation from complete coalescence to partial coalescence and finally to non-coalescence as the electric capillary number Ca p increases. The critical electric capillary number Ca pc for complete coalescence in the AC electric field remains unchanged and is twice as large as that in the direct current (DC) electric field when the frequency f ≥ 250 Hz. Charge transfer and reversal of electric field result in the reversal of the direction of electric force, which is the fundamental mechanism of non-coalescence of two droplets and chain formation in droplet cluster. The number of rebounds dramatically increases as f increases, promoting the stability of droplet chain. The droplet chains in the high-frequency AC electric field are longer and more stable than those in the low-frequency AC electric field.

ATG2A acts as a tether to regulate autophagosome-lysosome fusion in neural cells

Ze Zheng,Cuicui Ji,Hongyu Zhao,Yan G. Zhao

The macroautophagy/autophagy proteins ATG2A and ATG2B transfer lipids for phagophore membrane growth. They also form stable complexes with WDR45 and WDR45B. Our previous study demonstrated that WDR45 and WDR45B mediate autophagosome-lysosome fusion in neural cells. Given the defective autophagosome formation in cells lacking both ATG2s, their role in later autophagy stages is hard to explore. Here, we report that in neuroblastoma-derived Neuro-2a (N2a) cells, knocking down (KD) Atg2a, but not Atg2b, results in significant accumulation of SQSTM1/p62 and MAP1LC3-II/LC3-II, indicating impaired autophagy. Atg2a deficiency does not affect autophagosome formation, but reduces colocalization of autophagosomal LC3 with late endosomal/lysosomal RFP-RAB7, suggesting impaired autophagosome-lysosome fusion. ATG2A interacts with the SNARE proteins STX17, SNAP29, and VAMP8, facilitating their assembly. Overexpression of ATG2A partially rescues the autophagosome-lysosome fusion defects in Wdr45- and Wdr45b-deficient cells. ATG2 and another tether protein, EPG5, function partially redundantly in mediating autophagosome-lysosome fusion. Thus, ATG2A plays a key role in neural autophagy by tethering autophagosomes with lysosomes for fusion.Abbreviations: AAV: adeno-associated virus; ATG2Ar: RNAi-resistant ATG2A; Baf: bafilomycin A1; co-IP: co-immunoprecipitation; CQ: chloroquine; DKD: double knockdown; DKO: double knockout; ER: endoplasmic reticulum; KD: knockdown; KO: knockout; MIL: membrane-impermeable Halo ligand; MPL: membrane-permeable Halo ligand; N2a: Neuro-2a; NC negative control; PG: phagophore; PtdIns3K: phosphatidylinositol 3-kinase; PtdIns3P: phosphatidylinositol-3-phosphate; TEM: Transmission electron microscopy; TM: transmembrane domain; WT: wild-type.

Bioengineered nanovesicles for efficient siRNA delivery through ligand-receptor-mediated and enzyme-controlled membrane fusion

Cui Lele,Cui Yongsheng,Liu Jing,Li Wei,Wu Mengdan,Wei Xiawei,Lai Ying,Mi Peng

A major obstacle in knocking down oncogenes for tumor therapy is the efficient delivery of siRNA into the cytosolic spaces of cancer cells. Here, we genetically bioengineer biomimetic nanovesicles with tumor-recognition and enzyme-controlled membrane fusion functions for efficiently delivering small interfering RNA into cancer cells towards gene silencing tumor therapy. The siRNA@eS-BNVs are formulated by encapsulating siRNA inside the core and coating with genetically engineered HEK293T ACE2- cell membranes encoded with functional S protein, which can recognize cancer cells and initiate membrane fusion when triggered by the enzyme. The siRNA@eS-BNVs demonstrate better efficacy for cytosolic siRNA delivery and RNA interference than conventional formulations. By intravenous injection, siRNA@eS-BNVs are highly accumulated in tumors and potently inhibited tumor and lung metastasis by simultaneously silencing the epidermal growth factor receptor gene in vivo. The cancer cell-targeting and enzyme-activatable nanovesicles provide a valuable strategy for effective and precise drug delivery.

Mitochondrial fusion reduces T cell susceptibility to HIV infection through citrate modulation

Song Zichen,Wang Jiangrong,Zheng Zhihang,He Zhixiang,Xun Jingna,Gu Ling,Shen Yinzhong,Chen Jun

Inhibiting the metabolic activity of CD4+ T cells can effectively reduce HIV infection. Mitochondria, as critical organelles in eukaryotic metabolism, play a significant role in the progression of many diseases. The change of mitochondrial dynamics is an important process of mitochondrial regulation of cell metabolic activity. However, it remains uncertain whether regulating mitochondrial dynamics is a viable approach to reducing HIV infection. In this study, we demonstrated that promoting mitochondrial fusion in Jurkat cells through treatment with the mitochondrial fusion promoter M1 and the dynamin-related protein 1 (Drp1) inhibitor Mdivi1 conferred resistance to single-round VSVG-HIVNL4-3-GFP viral infection. Targeted metabolomics analysis revealed and subsequently confirmed the potential involvement of citrate in reducing HIV infection, which has been subsequently verified. And we found that plasma citrate level was negatively associated with HIV disease progression. Multi-omics results showed that citric acid leads to a decrease in the level of nucleotide metabolism in Jurkat cells. In conclusion, increased citrate levels resulting from mitochondrial fusion significantly impair the ability of HIV to infect cells, which may due to regulate nucleotide metabolism

Bisphenol A inhibits autophagosome-lysosome fusion and lipid droplet degradation

Dan Song,Yu Chen,Binran Wang,Diana Li,Chao Xu,Hong Huang,Saifei Huang,Rong Liu

Bisphenol A (BPA) is an artificial xenoestrogen widely used in consumer products containing polycarbonate plastics and epoxy resins . Exposure to BPA occurs through various channels, including ingestion of contaminated food and water. Autophagy is an important catabolic pathway that plays an important role in liver lipid metabolism . Evidence suggests that BPA exposure causes abnormal lipid droplet accumulation in liver, but the mechanism remains unknown. Here, we investigate the function of BPA in lipid metabolism and autophagy. BPA exposure increases lipid droplet and ROS accumulation which is accompanied by a defect in the fusion of the autophagosome to the lysosome . BPA exposure decreases the translocation of Stx17 to lysosome resulting in the autophagogome-lysosome fusion defect. There is no defect in the formation of the autophagosome indicated by increased LC3-II, p62 level, GFP/mRFP-LC3 ratios and decreased colocalization between LAMP2 with LC3. Mechanistically, BPA exposure reduces autophagy SNARE complex formation. Promoting autophagy by autophagy inducer (Torin2) partially reverses lipid droplet accumulation caused by BPA exposure. In summary, our results demonstrate BPA exposure inhibits autophagy resulting in decreased lipid droplet degradation and increased ROS levels. These results also provide a novel implication between autophagosome-lysosome fusion.
