In the fields of neuroscience and medicine, the accurate capture and quantitative analysis of dopamine (DA) levels in the blood are of great clinical significance for the early diagnosis of neurological dysfunctions and other pathological conditions. In this study, we developed a laccase (La) sensor based on micromotor technology to accurately detect DA content in blood. Firstly, the magnesium-based micromotor was used to endow the sensor with autonomous motion performance, which significantly increased the contact frequency between La and DA molecules in blood. Secondly, the micromotor-loaded Fe 3 O 4 /PPY/La magnetic nanoparticles provide an efficient electrical signal transmission interface for La, while the presence of polypyrrole (PPY) prevented electrode passivation caused by La. In addition, we used magnetic to collect Fe 3 O 4 /PPY/La after capture and tested it by differential pulse voltammetry. Finally, thanks to the excellent catalytic performance and high selectivity of La, the effective recognition and detection of DA in blood samples were realized, and an efficient electrochemical detection method was developed. This innovative detection scheme has a wide linear detection range and high sensitivity, which is expected to provide a more accurate diagnostic tool for health problems related to DA abnormalities, so as to better cope with and prevent related diseases.

In this work, we developed a novel microfluidic paper-based analytical device to quantify the blood markers of liver function from human fingertips and whole blood samples. The device can quickly acquire information for screening liver injury and supporting clinical decision-making by simultaneously performing quantitative tests for alanine aminotransferase, aspartate aminotransferase, and albumin. We evaluated the detection accuracy and the storage stability of the device using fingertip samples. The yielded results of our device correlated well with those from Mindray BS350s, even under the conditions of 35 °C and 90%RH. Thus, it offers an effective platform for clinical assessment of liver injury particularly in resource-limited areas.

The design, construction and testing for integration of liquid-liquid extraction (EX) and capillary electrophoretic (CE) separation on one glass microchip was reported. In this EX-CE chip, a 1.5 cm-long and 200 μm-wide EX channel was used for extraction based on the two-phase laminar flow , followed by a single-cross CE unit for on-line analysis without any auxiliary devices. One side of the EX channel surface for the organic solvent phase was selectively modified to be hydrophobic while the surface of the other side for the aqueous phase remained hydrophilic, and the extraction product reservoir is also used as the sample reservoir for the subsequent chip separation in the CE channel. With the surface-directed liquid flow behavior and liquid level adjustment in various reservoirs of the EX-CE chip, no disturbance occurred between the extraction (EX) and capillary electrophoretic (CE) units. A small heating block was placed under the chip to accelerate solvent evaporation after liquid-liquid extraction. Sanguinarine (SAN), a plant alkaloid , was used as a model analyte to evaluate the performance of the EX-CE chip. The influences of organic solvent type and liquid flow speed on the extraction efficiency were investigated. Rhodamine 123 (Rh123) was used as an internal standard for quantification of Sanguinarine (SAN) in a physiological buffer (e.g. PBS) or blood samples. A good linearity in the concentration range of 0.05 μg mL −1 to 0.1 mg mL −1 for SAN in PBS was obtained, with the detection limit of 0.5 ng mL −1 . Good repeatibilities for migration times (RSD of SAN is 0.63%, Rh123 is 0.91%, n = 5) and peak area ratio of SAN to Rh123 (RSD is 1.3%, n = 5) were obtained. For blood sample analysis, only 20 μL of sample was needed, and the whole analysis was finished in 17 min. In addition to the advantages in fast analysis speed, minimum sample handling, potential automation, the reported method showed an on-line sample pre-concentration capability.

Water-soluble and biocompatible protein carbon dots (P-CDs) were prepared from egg white simply by the rapid one-step reaction of neutralization heat reaction. Unexpectedly, the so fabricated P-CDs could present the excitation-dependent tunable fluorescence that could be quenched specifically by Fe3+ and Fe2+ ions with obvious color changes. A high-throughput fluorimetric platform was thereby developed by coating the P-CDs onto the capillary array for detection of total iron ions in fish blood samples, with the linear concentration range of 0.25-60.00 μM and the detection limit of 0.085 μM. Moreover, it can allow for the fast evaluation of total iron ions in fish blood samples, showing the recovery efficiencies of 97.25%-99.84%. Such a facile fabrication route for P-CDs from egg white by the rapid neutralization reaction may open a new door for preparing different CDs. More importantly, the developed fluorimetric method may be expected for the wide applications in the fields of diagnosis of iron-relative diseases, monitoring of environmental water, and ensuring of aquatic product safety.

Background Timely diagnosis and prevention of diseases require rapid and sensitive detection of biomarkers from blood samples without external interference. Abnormal electrolyte ion levels in the blood are closely linked to various physiological disorders, including hypertension. Therefore, accurate, interference-free, and precise measurement of electrolyte ion concentrations in the blood is particularly important. Results In this work, a colorimetric sensor based on a biphasic microdroplet extraction is proposed for the detection of electrolyte ions in the blood. This sensor employs mini-pillar arrays to facilitate contact between adjacent blood microdroplets and organic microdroplets serving as sensing phases, with any color changes being monitored through a smartphone's colorimetric software. The sensor is highly resistant to interference and does not require pre-treatment of the blood samples. Remarkably, the sensor exhibits exceptional reliability and stability, allowing for rapid enrichment and detection of K + , Na + , and Cl − in the blood within 10 s (Cl − ), 15 s (K + ) and 40 s (Na + ) respectively. Significance The colorimetric sensor based on biphasic microdroplet extraction offers portability due to its compact size and ease of operation without the need for large instruments. Additionally, it is location-independent, making it a promising tool for real-time biomarker detection in body fluids such as blood.

The accurate quantification of lysine is crucial for animal health and nutrition. In this work, we report the development of a novel electrochemical sensor for rapid, highly selective, and pretreatment-free quantification of lysine in whole blood. The sensor was fabricated by electrodepositing Prussian blue on screen-printed carbon electrodes, followed by electropolymerization of a lysine-imprinted polymer layer. We implemented a computational design strategy, integrating molecular docking and density functional theory calculations, to screen and identify the optimal functional monomer with high lysine binding affinity. The developed sensor exhibited a linear response to lysine concentration in simulated serum, with a limit of detection of 28.1 μM. It demonstrated high selectivity against structural analogs and common blood interferents. Validation using swine serum samples showed excellent agreement with results obtained from an amino acid analyzer. Furthermore, the sensor successfully quantified lysine in whole blood samples without any pretreatment, providing a simple, reliable, and efficient platform for on-site lysine detection in resource-limited settings.

Two novel fluorescent probes were designed to detect the biothiol in foods using the highly efficient Michael addition reaction between maleimide-derived probes and the biothiol. First, maleimide functionalized GQDs (M-GQDs) were synthesized and used for biothiol identification according to the Michael addition principle. The biothiol can be detected in the range of 5 × 10 −9 to 4 × 10 −7 mol/L and the detection limit was 1.69 × 10 −9 mol/L. Then, a fluorescence resonance energy transfer (FRET) system between M-GQDs and tetrakis (4-aminophenyl) porphyrin (TAPP) for biothiol detection was developed. However, the process of FRET was switched off in the presence of biothiols due to the switch of M-GQDs fluorescence emission to the“ON” mode following the Michael addition mechanism. The system could quickly and accurately detect the biothiol with a detection range of 6.7 × 10 −10 to 2 × 10 −7 mol/L and a detection limit of 2.34 × 10 −10 mol/L. Compared to the single detection system, the FRET system had a wider detection range and lower detection limit, and the related biomolecules did not interfere with the quantitative identification of the biothiol. The proposed method was successfully applied for the determination of the biothiol in foods and human blood samples.

Quinine, chloroquine, and hydroxychloroquine are effective antimalarial drugs that have shown antiviral activity against SARS-CoV-2 in initial in vitro studies. However, their excessive use can lead to adverse effects in humans. Therefore, rapid and accurate analysis of these inhibitors in whole blood samples is crucial for public health, although this presents significant challenges. In this study, perfluorooctyl-functionalized halloysite nanotubes were synthesized and used as sorbents for dispersive solid-phase extraction. The developed sorbents interacted with inhibitors through various mechanisms, including hydrophobic interactions, electrostatic interactions, F–π interactions, and hydrogen bonding. Notably, their adsorption capacity was twice that of unmodified halloysite nanotubes. By combining the perfluorooctyl-functionalized halloysite nanotubes-based dispersive solid-phase extraction method with liquid chromatography-tandem mass spectrometry, an analytical method was developed to determine concentrations of the three inhibitors in whole blood samples. Under optimized conditions, the limits of detection ranged from 1.2 to 2.1 ng/mL. The recoveries achieved 91.9%–101.5% for intra-day and 92.8%–101.2% for inter-day. The inter- and intra-day relative standard deviations were in the range of 0.6%–3.8% and 2.3%–8.1%, respectively. This study introduces a novel approach for developing sorbents to target drugs and presents an innovative analytical method for therapeutic monitoring. These advancements will facilitate the evaluation of the pharmacokinetics of these inhibitors in patients and support ongoing clinical trials.

Fibrinolytic activity assay is particularly important for the detection, diagnosis, and treatment of cardiovascular disease and the development of fibrinolytic drugs. A novel efficacious strategy for real-time and label-free dynamic detection of fibrinolytic activity based on ordered porous layer interferometry (OPLI) was developed. Fibrin or a mixture of fibrin and plasminogen (Plg) was loaded into the highly ordered silica colloidal crystal (SCC) film scaffold to construct a fibrinolytic response interference layer to measure fibrinolytic activity with different mechanisms of action. Fibrinolytic enzyme-triggered fibrinolysis led to the migration of interference fringes in the interferogram, which could be represented by optical thickness changes (ΔOT) tracked in real time by the OPLI system. The morphology and optical property of the fibrinolytic response interference layer were characterized, and the Plg content in the fibrinolytic response interference layer and experimental parameters of the system were optimized. The method showed adequate sensitivity for the fibrinolytic activity of lumbrokinase and streptokinase, with wide linear ranges of 12–6000 and 10–2000 U/mL, respectively. Compared with the traditional fibrin plate method, it has a lower detection limit and higher linearity. The whole kinetic process of fibrinolysis by these two fibrinolytic drug models was recorded in real time, and the Michaelis constant and apparent kinetic parameters were calculated. Importantly, some other blood proteins were less interfering with this system, and it showed reliability in fibrin activity detection in real whole blood samples. This study established a better and more targeted research method of in vitro fibrinolysis and provided dynamic monitoring data for the analysis of fibrinolytic activity of whole blood.

The effective capture, release and reanalysis of circulating tumor cells (CTCs) are of great significance to acquire tumor information and promote the progress of tumor therapy. Particularly, the selective release of multiple types of CTCs is critical to further study; however, it is still a great challenge. To meet this challenge, we designed a smart DNAzyme probe-based platform. By combining multiple targeting aptamers and multiple metal ion responsive DNAzymes, efficient capture and selective release of multiple types CTCs were realized. Sgc8c aptamer integrated Cu2+-dependent DNAzyme and TD05 aptamer integrated Mg2+-dependent DNAzyme can capture CCRF-CEM cells and Ramos cells respectively on the substrate. With the addition of Cu2+ or Mg2+, CCRF-CEM cells or Ramos cells will be released from the substrate with specific selectivity. Furthermore, our platform has been successfully demonstrated in the whole blood sample. Therefore, our capture/release platform will benefit research on the molecular analysis of CTCs after release and has great potential for cancer diagnosis and individualized treatment.

The authors describe a photometric method for the determination of free cholesterol based on the oxidation of cholesterol by the catalytic action of the enzyme cholesterol oxidase. The hydrogen peroxide formed is used to oxidize the chromogenic compound 3,3′,5,5′-tetramethylbenzidine (TMB) by exploiting the peroxidase-like activity of carbon nanotube-supported Prussian blue (PB). This enzyme mimic was synthesized from a mixture of ferric chloride and hexacyanoferrate(III) in the presence of intrinsically reducing multi-walled carbon nanotubes. The composite can trigger the formation of a blue-green dye from TMB in the presence of H 2 O 2 . The findings were exploited to design a photometric (652 nm) assay that has a linear response in the 4 to 100 μM cholesterol concentration range and a 3 μM detection limit. The practicability of the method was verified by the successful analysis of free cholesterol in human blood samples.
Schematic of a carbon nanotube-supported Prussian blue (PB/MWCNT) hybrid used as a promising peroxidase mimic. It has the advantages of low cost, simple synthesis, high activity, and good stability. PB/MWCNTs along with cholesterol oxidase (ChOx) are used in a colorimetric assay for the determination of free cholesterol.

Sulfur quantum dots (S-dots) show great potential for applications in various field, due to their favorable biocompatibility, high stability, and antibacterial properties. However, the use of S-dots in chemical sensing is limited by the lack of functional groups on the surface. In this work, a fluorescence glutathione (GSH) assay is developed based on the GSH modulated quenching effect of Cu 2 O nanoparticles (NP) on S-dots. The fluorescence of S-dots is effectively quenched after forming complex with Cu 2 O NP through a static quenching effect (SQE). Introducing of GSH can trigger the decomposition of Cu 2 O NP into GSH-Cu(I) complex, which leads to the weaken of SQE and the partial recover of the fluorescence. The intensity of recovered fluorescence shows a positive correlation with the concentration of GSH in the concentration range of 20 to 500 μM. The fluorescence GSH assay shows excellent selectivity and robustness towards various interferences and high concentration salt, which endow the successful detection of GSH in human blood sample. The presented results provide a new door for the design of fluorescence assays, which also provides a platform for the applications in nanomedicine and environmental science.
