20-Hydroxyecdysone and Receptor Interplay in the Regulation of Hemolymph Glucose Level in Honeybee (Apis mellifera) Larvae

Jing Yu,Hongfang Wang,Wenfeng Chen,Hongyu Song,Ying Wang,Zhenguo Liu,Baohua Xu

The hormone 20-hydroxyecdysone (20E) and the ecdysone receptors (ECR and USP) play critical roles in the growth and metabolism of insects, including honeybees. In this study, we investigated the effect of 20E on the growth and development of honeybee larvae by rearing them in vitro and found reduced food consumption and small-sized pupae with increasing levels of 20E. A liquid chromatography-tandem mass spectrometry (LC-MS/MS)-based analysis of widely targeted metabolomics was used to examine the changes in the metabolites after an exogenous 20E application to honeybee larvae and the underlying mechanisms. A total of 374 different metabolites were detected between the control group and the 20E treatment group, covering 12 subclasses. The most significant changes occurred in 7-day-old larvae, where some monosaccharides, such as D-Glucose and UDP-galactose, were significantly upregulated. In addition, some metabolic pathways, such as glycolysis/gluconeogenesis and galactose metabolism, were affected by the 20E treatment, suggesting that the 20E treatment disrupts the metabolic homeostasis of honeybee larvae hemolymph and that the response of honeybee larvae to the 20E treatment is dynamic and contains many complex pathways. Many genes involved in carbohydrate metabolism, including genes of the glycolysis and glycogen synthesis pathways, were downregulated during molting and pupation after the 20E treatment. In contrast, the expression levels of the genes related to gluconeogenesis and glycogenolysis were significantly increased, which directly or indirectly upregulated glucose levels in the hemolymph, whereas RNA interference with the 20E receptor EcR-USP had an opposite effect to that of the 20E treatment. Taken together, 20E plays a critical role in the changes in carbohydrate metabolism during metamorphosis.Keywords:20-hydroxyecdysone;metabolomics;hemolymph;glucose;ecdysone receptors

iTRAQ-based quantitative proteomic analysis identified Eno1 as a cadmium stress response gene in Propsilocerus akamusi (Tokunaga) hemolymph

Xianyun Zheng,Ye Gao,Wanghong Li,Shu Wang

Propsilocerus akamusi (Tokunaga) is a common species of midge in Siberia , Japan, and China and an important prey species for fish and aquatic birds. Furthermore, this species has been shown to have an extraordinary capacity to resist cadmium (Cd) toxicity. In this study, isobaric tags for relative and absolute quantitation (iTRAQ) coupled liquid chromatography tandem mass spectrometry (LC-MS/MS) was used to analyze relative changes in the P. akamusi hemolymph proteome following exposure to a sublethal concentration of Cd 2+ . The results showed that Cd 2+ stress affects energy metabolism in P. akamusi . After examining the differentially expressed proteins (DEPs), only one up-regulated protein associated with metabolism, α-enolase (Eno1) was identified and further isolated and characterized. Sequence alignments showed that the deduced P. akamusi Eno1 amino acid sequence is highly conserved, with similarities of 77–95% noted when compared to other Dipteran Eno1 sequences. Furthermore, prolonged Cd 2+ exposure impacted Eno1 transcription, protein expression and enzyme activity levels. These results suggest that Eno1 may play a role in the response to Cd 2+ stress in P. akamusi .

Developing a low fishmeal diet for juvenile Pacific white shrimp, Litopenaeus vannamei, using the nutritional value of FM as the reference profile

S. Xie,J. Niu,W. Zhou,Y. Liu,L. Tian

An 8-week feeding trial was conducted to evaluate the effect of fish-meal replacement on growth performance, antioxidative ability, immune capacity and haemolymph metabolites of Litopenaeus vannamei . A 250 g/kg fish-meal diet was used as control (Diet 1). Other two diets contained 100 g/kg fishmeal, Diet 2 was supplemented with three indispensable amino acids, and Diet 3 was supplemented with microbalance components and phytase. Weight gain was lower in shrimp fed the Diet 2 than shrimp fed the Diet 1. Feed efficiency, protein efficiency ratio and survival were higher in shrimp fed the Diet 1 than other groups. Glutathione and nitric oxide contents in haemolymph were decreased in shrimp fed the Diet 2, and nitric oxide synthetase, phenoloxidase, acid phosphatase and myeloperoxidase activities showed the similar tendency. In hepatopancreas, antioxidative ability and immune capacity also lower in shrimp fed the Diet 2. In haemolymph, 21 differential metabolites were identified, and 13 metabolism pathways were affected by different diets. Generally, the fishmeal inclusion level could be reduced from 250 g/kg to 100 g/kg without affecting the growth performance of L. vannamei , and meanwhile, the protein utilization efficiency and antioxidative ability of shrimp fed the low fishmeal diets should be more focused on in future studies.

Damage to the behavior and physiological functions of Apis mellifera (Hymenoptera: Apidae) by monocrotaline via the modulation of tryptophan metabolism and the corazonin receptor

Qi Hua,Xuepeng Chi,Wei Zhang,Hongyu Song,Ying Wang,Zhenguo Liu,Hongfang Wang,Baohua Xu

Monocrotaline (MCT) is a toxic pyrrolizidine alkaloid found in plants of the Crotalaria genus. As primary pollinators of Crotalaria plants, honeybees come into contact with this harmful substance. However, limited research has been conducted on the effects of MCT on Apis mellifera , particularly the risks of long-term exposure to sublethal concentrations. Through evaluating the proboscis extension reflex (PER) ability, analyzing the honeybee brain transcriptome, and analyzing the honeybee hemolymph metabolome, we discovered that sublethal concentrations of MCT impair the olfactory and memory capabilities of honeybees by affecting tryptophan (Trp) metabolism. Furthermore, MCT upregulates the expression of the corazonin receptor ( CrzR ) gene in the honeybee brain, which elevates reactive oxygen species (ROS) levels in the brain while reducing glucose levels in the hemolymph, consequently shortening the honeybees' lifespan. Our findings regarding the multifaceted impact of MCT on honeybees lay the foundation for exploring its toxicological pathways and management in honeybee populations.

Nano titanium dioxide alleviates the toxic effects of tris (2-chloropropyl) phosphate on the digestive gland and hemolymph of thick-shell mussel Mytilus coruscus

Jiani Liu,Yueyong Shang,Fujing Deng,Zhihua Feng,Menghong Hu,Youji Wang

In the marine environment, nanoparticles play a role in adsorbing and catalytically degrading organic pollutants, thereby mitigating their toxic effects on aquatic organisms. This study aimed to investigate the impact of nano titanium dioxide (nTiO 2 ) and tris (2-chloropropyl) phosphate (TCPP) on the hemolymph and digestive function of the thick-shell mussel Mytilus coruscus . Mussels were divided into a control group, a group exposed to TCPP alone, a group exposed to a combination of TCPP and 0.5 mg/L nTiO 2 , and a group exposed to a combination of TCPP and 1 mg/L nTiO 2 . After 14 days of exposure, oxidative stress responses, including superoxide dismutase (SOD) activity and malondialdehyde (MDA) content, immune defense responses, including acid phosphatase (ACP) and alkaline phosphatase (AKP) activities, and gene expression, including HSP70 expression, were measured in the hemolymph and digestive glands of the mussels. Compared to the control group, mussels solely exposed to 100 μg/L TCPP exhibited a significant reduction in SOD activity in the hemolymph. When TCPP was co-exposed with 0.5 mg/L nTiO 2 , there were significant increases in MDA content and AKP activity in both the digestive gland and hemolymph compared to the control group. Upon co-exposure of TCPP with 1 mg/L nTiO 2 , MDA content and AKP activity in the digestive gland significantly decreased, while SOD, ACP, and AKP activity in the hemolymph significantly increased and MDA content significantly decreased, returning to the control group levels. Furthermore, in the combined exposure, HSP70 gene expression significantly decreased as the nTiO 2 concentration increased from 0.5 mg/L to 1 mg/L. In summary, TCPP impacted the hemolymph and digestive function of mussels, whereas a concentration of 1 mg/L nTiO 2 effectively alleviated the toxic effects of TCPP. This study is crucial for assessing the ecological risks of nanoparticles and emerging organic pollutants in marine environments, and provides new insights into the interaction between nTiO 2 and TCPP, as well as the influence of nTiO 2 concentration on mitigating TCPP toxicity.

Nutritional geometry framework of sleep

Mei-Ling Lai,An-Qi Li,Alistair M. Senior,G. Gregory Neely,Stephen J. Simpson,Qiao-Ping Wang

Aims Sleep is a fundamental physiological function and is essential for all animals. Sleep is affected by diet compositions including protein (P) and carbohydrates (C), but there has not been a systematic investigation on the effect of dietary macronutrient balance on sleep. Main methods We used the nutritional geometry framework (NGF) to explore the interactive effects on sleep of protein (P) and carbohydrates (C) in the model organism Drosophila . Both female and male flies were fed various diets containing seven ratios of protein-to-carbohydrates at different energetic levels for 5 days and sleep was monitored by the Drosophila Activity Monitor (DAM) system. Key findings Our results showed that the combination of low protein and high carbohydrates (LPHC) prolonged sleep time and sleep quality, with fewer sleep episodes and longer sleep duration. We further found that the effects of macronutrients on sleep mirrored levels of hemolymph glucose and whole-body glycogen. Moreover, transcriptomic analyses revealed that a high-protein, low-carbohydrate (HPLC) diet significantly elevated the gene expression of metabolic pathways when compared to the LPHC diet, with the glycine, serine, and threonine metabolism pathway being most strongly elevated. Further studies confirmed that the contents of glycine, serine, and threonine affected sleep. Significance Our results demonstrate that sleep is affected by the dietary balance of protein and carbohydrates possibly mediated by the change in glucose, glycogen, glycine, serine, and threonine.

Vitro UPLC analysis and mass method identification, and in vivo or cellular immune anti-inflammatory function of Sanhuang Xiexin Decoction (SHXD)

Yang Gao,Shi-Qiao Liu,Jia-Long Wang,Han-Ming Cui,Qiu-Yan Zhang,Lei Wang,Yi-Xin Zhang,Jian Li,Yu Dong,Yuan-Hui Hu

Ethnopharmacological relevance Sanhuang Xiexin Decoction (SHXD), consisting of Coptis chinensis Franch., Scutellaria baicalensis Georgi and Rheum palmatum L. , is t raditionally used for relieving fever, purging fire for removing toxins, eliminating phlegm and haemostasis, eliminating the wetness-evil from the upper warmer, clearing away the heat-evil and expelling superficial evils. Each of the three herbs contained in SHXD has been indicated to have anti-inflammatory effects in vivo , but its effects on rat NK-cell phenotypes remain unexplored, and the comprehensive mechanism of this compound SHXD in curing the inflammation induced by lipopolysaccharides (LPS) remains to be revealed. Aim of the study The study aim was to assess the effect of SHXD on LPS-induced fever and inflammation in a rat model, reduce NLRP3 activation in NK cells expressing specific cell phenotype antibodies and determine the therapeutic value of this approach in vivo . Materials and methods SHXD extract was prepared and analysed by the developed ultra-performance liquid chromatography (UPLC) method for the simultaneous detection of 14 compounds. The main peaks were firstly identified on an Orbitrap via high resolution tandem mass spectrometry (MS). Then, the extract was used in the rat model of LPS-induced inflammation and fever for pharmacologically study the effects of drug treatment. Peripheral blood lymphocyte cells were isolated from the animals, including those subjected to the SHXD extract treatment, and the cell phenotype was determined prior to cell culture and after treating the cell cultures with the extract. The phenotypes of cells harvested using CD3, CD4, CD8a, CD81, CD161 and CD86 antibodies were used to verify the enhanced memory of the peripheral blood lymphocytes cells (PBMC) that were induced into nature killer (NK) cells. Results The SHXD extract was prepared, analysed and identified via quality control equipment and was observed to have pharmacological effects that reduced NLRP3 activation and fever in rats. The production of NK cells and peripheral blood lymphocytes was induced by the SHXD extract, which manifested as increased levels of CD4 + , CD8a+, CD81 + , CD161+ and CD86 + cells. The levels of CD3 + cells were significantly different between the model group and the drug-treated or control groups (p < 0.01) with dose independence, while the levels of CD4 + cells were not significantly different between the drug-treated and control groups, with a trend towards lower levels in the model group with dose independence. The levels of CD4 + cells was significantly different between the drug-treated group and the model groups with dose independence (p < 0.05). The levels of CD86 + cells were not significantly different between the drug-treated group and the model and control groups. The levels of CD8a + cells was significantly different between the model group and the drug and control groups (p < 0.05, dose 2.0 μg/ml), with higher levels in the drug-treated group. The levels of CD3 + , CD4 + , CD8a + cells in the drug treated group have dose dependence with SHXD. Conclusions This experiment revealed that SHXD reduced NLRP3 activation in the blood of LPS-treated rats, which occurred through the activation of NK cells that expressed CD3, CD8a and CD161. SHXD may possess anti-inflammatory effect via activacting the one of major pharmacology effcet of NK cells that expressed CD3, CD8a and CD161 phenotypes expression. This result demonstrates that SHXD may possess ability to enhance the memory of peripheral blood lymphocytes and natural killer cells.

Microplastics and Nanoplastics Alter the Physicochemical Properties of Willow Trees and Lead to Mortality in Leaf Beetle Larvae

Peipei Zhu,Yanping Zhang,Mengqi Deng,Yuxin Zhang,Jing Luo,Runhua Han,Letian Xu

Polystyrene micro- and nanoplastics (MNPs) are increasingly found in terrestrial environments, posing risks across the food web. However, the potential impacts of MNPs transfer on plant-insect interactions remains largely unknown. In this study, consumption of willow plants ( Salix maizhokunggarensis ) exposed to 10.0 mg/L MNPs for 21 days inhibited survival and reduced body weight in Plagiodera versicolora larvae unlike those exposed to lower concentrations or shorter durations (0.1, 1.0 and 10.0 mg/L MNPs for 7 or 14 days). MNPs exposure increased lignin content and leaf thickness in willows, leading to decreased leaf consumption and increased mouthpart wear in P. versicolora larvae. Transcriptome and gut microbiota analyses revealed significant downregulation of genes related to digestion, intestinal homoeostasis, immunity, and growth/development along with profound alterations in gut microbiota composition. Notably, the abundance of the pathogenic bacterium Pseudomonas increased significantly. The gut barrier was disrupted, allowing gut bacteria to translocate into the haemolymph, accelerating larval mortality. Overall, MNPs altered plant physiology, making willow plants unsuitable for herbivore consumption and indirectly influenced herbivore survival by modulating gut bacteria. These findings offer novel insights into the cascading ecological effects and risks of MNPs, highlighting potential impacts on plant-herbivore interactions, biodiversity, and ecosystem health in terrestrial ecosystems.
