Titanium dioxide nanoparticles (TiO 2 NPs) are widely used in various products and inevitably released with different sizes and forms into aquatic environment. The purpose of this study was to assess the differential immune toxicity of TiO 2 NPs with size difference on mussel hemocytes using flow cytometry (FCM) assays. Hemocyte parameters, including total hemocyte count (THC), hemocyte mortality (HM), phagocytosis activity (PA), lysosomal content (LC), esterase activity (EA), mitochondrial number (MN), mitochondrial membrane potential (MMP) and reactive oxygen species content (ROS) were evaluated in the mussels Mytilus coruscus exposed to two types of TiO 2 NPs (25nm & 100nm: 0.1, 1, 10 mg/L, respectively). In general, size- and concentration-dependent toxicity was pronounced with 25nm-NP and highest concentration (10mg/L) being the most toxic. Alhough a slight recovery from the TiO 2 exposure was observed, significant carry-over effects were still detected. These results highlight the importance of differential size effects of metal oxide NPs on toxicity mechanisms in aquatic animals.

Effectively controlling deep non-compressible bleeding remains a major challenge. In this study, by mimicking nanofiber structure and netting blood cells function of fibrin network in blood clots, we develop a novel bioinspired quaternized chitosan nanofiber sponge with distinct blood cells filtration and blood plasma absorption bifunction for rapid hemostasis. The quaternized chitosan nanofiber sponge possesses a unique gradient pore structure with small pores on the outer surface and large pores in the inner part. The outer small pores effectively capture blood cells with a filtration efficiency as high as 91.7%, while the inner large pores endow with an ultrahigh liquid absorption capacity (93 g/g), surpassing previously reported literature records. The quaternized chitosan nanofiber sponge demonstrates a hemostasis time 2.5 times faster than that of the commercial gelatin® hemostatic sponge when treating the rat liver defect bleeding (noncompressible hemorrhage model). When applied to the rabbit arterial injury bleeding (lethal arterial hemorrhage model), the bioinspired nanofiber sponge achieves bleeding control within only 61.6 s, while both commercial gelatin® hemostatic sponge and commercial collagen® hemostatic sponge fail to stop bleeding even after 240 s. This bioinspired nanofiber sponge derived from the physiological coagulation process may hold great potential for pre-hospital and battlefield first aid.

Background The argus snakehead ( Ophiocephalus argus Cantor) is a highly nutritious, freshwater, cultured bony fish with a high economic value. The health of the fish is closely related to its blood cells, which are critical for oxygen transport, natural defense, and immunity. We investigated the morphometry, microstructure, and cytochemical characteristics of the peripheral blood cells of O. argus . Our results may provide the basic reference values needed to monitor the health of this fish for large-scale cultivation. Methods The number of blood cells in O. argus were counted on a hemocytometer and their size was measured using a micrometer under light microscope. The morphology and classification of the blood cells were studied using Wright’s staining and the cytochemical characteristics were studied using seven chemical stains including peroxidase (POX), Sudan black B (SBB), periodic acid-Schiff (PAS), acid phosphatase (ACP), alkaline phosphatase (ALP), chloroacetic acid AS-D naphthol esterase (AS-D), and α -naphthol acetate esterase ( α -NAE). Results The peripheral blood cells in O. argus can be classified as erythrocytes, leukocytes, and thrombocytes; of which, females had 2.9597 million/mm 3 , 88,400/mm 3 , and 43,600/mm 3 , respectively, and males had 3.0105 million/mm 3 , 105,500/mm 3 , and 34,000/mm 3 , respectively. Leukocytes consisted of neutrophils, monocytes, large lymphocytes, and small lymphocytes. Eosinophils and basophils were not found. Monocytes were the most numerous leukocytes identified, followed by neutrophils and small lymphocytes, while large lymphocytes were the least frequently identified. Cytochemical staining showed that erythrocytes were only positive for PAS staining. Neutrophils were strongly positive for POX, SBB, and ACP, and positive for all the other cytochemical stains. Monocytes were positive for PAS and α -NAE and were weakly positive for ACP and AS-D staining. Large lymphocytes were positive for PAS and were weakly positive for ALP, AS-D, and α -NAE staining. Small lymphocytes were positive for PAS and weakly positive for AS-D and α -NAE staining. Thrombocytes were positive for PAS and were weakly positive for ACP and AS-D, but negative for the remaining cytochemical stains. The morphology of peripheral blood cells in O. argus was generally similar to that of other fish species, while the cytochemical staining patterns showed clear species specificity.

ObjectiveThis study aims to investigate the effect of Gallic Acid (GA) on the alleviation of chemotherapy-induced bone marrow suppression, with a comparison to Diyu sheng bai tablets (DYSB) and RhG-CSF.MethodsA mouse model of bone marrow suppression was established in BALB/c mice using intraperitoneal injections of cyclophosphamide (CTX). All procedures were performed after obtaining ethical clearance from the institutional animal ethics committee. Mice were treated with low (100 mg/kg/d), medium (200 mg/kg/d), and high (400 mg/kg/d) doses of Gallic Acid (GA) to mitigate CTX-induced bone marrow suppression. In parallel, mice in the positive control group were also treated with DYSB and RhG-CSF at their respective standard doses (DYSB: 100 mg/kg/day, RhG-CSF: 125 mg/kg/day). The efficacy of GA in alleviating chemotherapy-induced bone marrow suppression was evaluated through blood cell counts, immune organ (thymus and spleen) indices, bone marrow nucleated cell (BMNC) counts, cell cycle analysis, apoptosis, histopathology of bone marrow and spleen, and analysis of splenic hematopoietic factors.ResultsCTX induced a decrease in peripheral blood cells and BMNC counts, reduced spleen and thymus indices, and diminished abnormal pathology of bone marrow and spleen, as well as decreasing disturbances in hematopoietic factors. GA was able to alleviate these abnormalities in the bone marrow. It modulated cell proliferation and apoptosis, adjusted the proportion of cells in the G0/G1 phase, and reduced apoptosis in femoral bone marrow.ConclusionGallic Acid (GA) alleviates chemotherapy-induced bone marrow suppression by improving immune organ function, promoting bone marrow cell recovery, and inhibiting apoptosis. These findings support GA as a potential adjunct therapy for chemotherapy, with promising clinical applications.

Metastasis is one of the most important factors related to malignant tumor development and therapeutic failure. Phytic acid (PA), a poly-phosphorylated inositol derivative in plant seeds and grains, has been shown to have antitumoral activity. However, the effect of PA on tumor metastasis and the underlying mechanisms remain uncharacterized. In the current study, we aimed to identify the effect of PA on tumor metastasis by employing a Drosophila model of malignant tumor Raf GOF Scrib −/− . We observed that PA supplementation in food inhibited the invasion and migration of tumors. Further studies indicated that PA drives Yki phosphorylation and inactivation through Lats kinase, which subsequently results in decreased expression of the JAK/STAT pathway ligands unpaired cytokines (Upds). Moreover, the reduced JAK/STAT pathway inhibits the recruitment and proliferation of hemocytes. To summarize, these findings provide valuable pathological and mechanistic insights into the potential application of PA for functional food or natural medicine development.

Nano-TiO 2 is inevitably released into aquatic environment with increasing of nanotechnology industries. Study pointed that different individuality showed divergent behavioral and physiological response when facing environmental stress. However, the effects of nano-TiO 2 on tolerance of bivalves with different individualities remain unknown. In the study, clams were divided into two types of individuality - proactive and reactive by post-stress recovery method. It turned out that proactive individuals had quicker shell opening level, stronger burrowing behavior, faster feeding recovery, higher standard metabolic rate and more rapid ammonia excretion ability than reactive individuals after exposed to air. Then, the survival rate, hemocytes response and oxidase activity of classified clams were evaluated after nano-TiO 2 exposure. Results showed that after 30 d exposure, proactive individuals accelerated burrowing behavior with higher survival rate. Moreover, proactive clams had better adaptability and less hemocytes response and oxidative damage than reactive clams. The study highlights the individualities of marine shell fish determine individual capacity to adapt to environmental changes, play important roles in aquaculture and coastal ecosystem health.

Diagnosing hematological disorders based on the separation and detection of cells in the patient's blood is a significant challenge. We have developed a novel barcode particle-based suspension array that can simultaneously capture and detect multiple types of blood cells. The barcode particles are polyacrylamide (PAAm) hydrogel inverse opal microcarriers with characteristic reflection peak codes that remain stable during cell capture on their surfaces. The hydrophilic PAAm hydrogel scaffolds of the barcode particles can entrap various plasma proteins to capture different cells in the blood, with little damage to captured cells.

As the number of diabetic patients increases year by year, the demand for a self-monitoring blood glucose test also increases. Additionally, more in-depth studies of blood glucose biosensors have led to increasing accuracy requirements. However, at present, most enzyme glucose sensors have difficulty solving issues related to hematocrit (HCT) interference. Most studies on anti-HCT interference in blood glucose sensing have mainly focused on correction factors, such as glucose correction, and electrical impedance or resistivity characteristics to correct the impact of HCT. In this work, we developed an anti-HCT-interference blood glucose sensor based on a fiber paper-based screen-printed carbon electrode with a mediator (potassium ferricyanide) and glucose oxidase (GOD), which were fixed through inkjet printing. When testing whole blood samples with HCT levels of 20%–70% and a fluid dropping method, the results of a consensus error grid analysis showed that 91% of the data points lied within zones A and B. The glucose sensor was exclusively designed with a 16 μL chamber in the working area of the sensor. When the testing fluid was taken in by capillary sucking, the effect of HCT was further reduced, and 95% of the data points lied within zones A and B in the consensus error grid.

PANoptosis is an emerging form of regulated cell death (RCD) characterized by simultaneous activation of pyroptotic, apoptotic, and necroptotic signaling that not only participates in pathologies of inflammatory diseases but also has a critical role against pathogenic infections. Targeting PANoptosis represents a promising therapeutic strategy for related inflammatory diseases, but identification of inhibitors for PANoptosis remains an unmet demand. Baicalin (黄芩苷) is an active flavonoid isolated from Scutellaria baicalensis Georgi (Huangqin), a traditional Chinese medicinal herb used for heat-clearing and detoxifying. Numerous studies suggest that baicalin possesses inhibitory activities on various forms of RCD including apoptosis/secondary necrosis, pyroptosis, and necroptosis, thereby mitigating inflammatory responses. In this study we investigated the effects of baicalin on PANoptosis in macrophage cellular models. Primary macrophages (BMDMs) or J774A.1 macrophage cells were treated with 5Z-7-oxozeaenol (OXO, an inhibitor for TAK1) in combination with TNF-α or LPS. We showed that OXO plus TNF-α or LPS induced robust lytic cell death, which was dose-dependently inhibited by baicalin (50−200 μM). We demonstrated that PANoptosis induction was accompanied by overt mitochondrial injury, mitochondrial DNA (mtDNA) release and Z-DNA formation. Z-DNA was formed from cytosolic oxidized mtDNA. Both oxidized mtDNA and mitochondrial Z-DNA puncta were co-localized with the PANoptosome (including ZBP1, RIPK3, ASC, and caspase-8), a platform for mediating PANoptosis. Intriguingly, baicalin not only prevented mitochondrial injury but also blocked mtDNA release, Z-DNA formation and PANoptosome assembly. Knockdown of ZBP1 markedly decreased PANoptotic cell death. In a mouse model of hemophagocytic lymphohistiocytosis (HLH), administration of baicalin (200 mg/kg, i.g., for 4 times) significantly mitigated lung and liver injury and reduced levels of serum TNF-α and IFN-γ, concomitant with decreased levels of PANoptosis hallmarks in these organs. Baicalin also abrogated the hallmarks of PANoptosis in liver-resident macrophages (Kupffer cells) in HLH mice. Collectively, our results demonstrate that baicalin inhibits PANoptosis in macrophages by blocking mitochondrial Z-DNA formation and ZBP1-PANoptosome assembly, thus conferring protection against inflammatory diseases. PANoptosis is a form of regulated cell death displaying simultaneous activation of pyroptotic, apoptotic, and necroptotic signaling. This study shows that induction of PANoptosis is linked to mitochondrial dysfunction and mitochondrial Z-DNA formation. Baicalin inhibits PANoptosis in macrophages in vitro via blocking mitochondrial dysfunction and the mitochondrial Z-DNA formation and thereby impeding the assembly of ZBP1-associated PANoptosome. In a mouse model of hemophagocytic lymphohistiocytosis (HLH), baicalin inhibits the activation of PANoptotic signaling in liver-resident macrophages (Kupffer cells) in vivo, thus mitigating systemic inflammation and multiple organ injury in mice.

Long-term in vitro cultured crayfish cells can serve as a valuable tool for investigating various aspects of crayfish biology, encompassing cell physiology and viral mechanisms. Regrettably, they are difficult to maintain a highly active state and survive during long-term and continuous culture in vitro . In order to generate highly viable crayfish cell cultures, it is crucial to optimize dependable culture conditions and utilize a suitable cell medium. In this study, we have successfully developed reliable in vitro long-term culture systems for hematopoietic (HPT) cells of crayfish Cherax quadricarinatus by formulating an optimal crayfish cell growth medium (OCCM), including under two-dimensional (2D) and three-dimensional (3D) conditions. The novel medium was optimized by a mix of exogenous nutrients, crayfish plasma (CP) and shrimp serum-derived amino acids and saccharides within the Leibovitz's L-15 (L-15) medium. Our newly developed crayfish cell growth medium (CCM-28) enabled the culturing of HPT cells for over 142 days under 2D and over 60 days under 3D conditions, with consistent active cell proliferation observed over a 30-day incubation period. This proliferation was comparable to in vivo conditions, with a cell division rate exceeding 20%, suggesting that a high mitotic rate has been successfully achieved during long-term in vitro culture. Flow cytometry analysis further elucidated the mitosis-arrest of HPT cells cultured in vitro was predominantly observed in the G 0 /G 1 phase. Furthermore, the cell subculture techniques have been established using mechanical dissociation method (in 2D) and substrate exchange method (in 3D). Additionally, the susceptibility of these cell cultures to covert mortality nodavirus (CMNV) and white spot syndrome virus (WSSV) was confirmed by capturing cytopathic effects (CPE) and analyzing viral copy numbers. Species identification using the cytochrome oxidase subunit I ( COI ) gene established the origin of the cell cultures from C. quadricarinatus . In conclusion, the successful development of highly viable crayfish cells in long-term in vitro culture systems, as demonstrated in this study, will significantly advance the research on immortalization in crayfish cells and the study of mechanisms related to viruses.

Crab cell line, especially continuous crab cell line, can provide us a useful tool for studies on the virology, immunology, and molecular biology of crabs. However, no continuous crab cell line has been available due to the lacking of suitable medium and the occurrence of mitosis-arrest. In this study, long-term in vitro culture conditions for both two- (2D) and three-dimensions (3D) were successfully developed for the circulating hemocytes of swimming crab Portunus trituberculatus , designated as PTH cells. In 2D culture, a novel crab basic medium in osmolarity of 990–1 100 mOsm/kg was optimized for the first time, which is different from Leibovitz’s L-15 medium in mainly the components of amino acids, containing double strengths of the contents of free amino acid mixture in the crab serum. Then an optimal crab growth medium was developed by supplementing 5% fetal bovine serum, 50-g/L yeast extract powder, 20-µg/L basic fibroblast growth factor and epidermal growth factor into the optimal crab basic medium, and found that it could support a long-term survival of PTH cells in a healthy monolayer up to 347 days and partially break through the mitosis-arrest of crab cells evidenced by the obvious increase of proliferating potential detected in the 10-d primarily cultured PTH cells. These 2D cultured PTH cells could be successfully sub-cultured for 11 times by physical flushing method and well cryopreserved in liquid nitrogen. In 3D culture, using the same crab growth medium, the PTH cell aggregates could be easily formed and healthily maintained on the surface of solidified Matrigel or in the ultra-low-attachment plate with a survival rate of 50%–60% on Day 103. This work largely improved the primary culture and subculture of crab cells and will facilitate the establishment of continuous crab cell line.

This investigation is aimed to develop a novel iron supplement, iron oxide nanoparticles modified with Angelica sinensis polysaccharide (IONPs-ASP), for the treatment of iron deficiency anemia (IDA). IONPs-ASP was synthesized using the co-precipitation method and characterized for their structural features, physicochemical properties, and biocompatibility by TEM, SEM, FTIR, etc. The therapeutic effects of IONPs-ASP were assessed in a rat IDA model. IONPs-ASP was administered intravenously, and blood parameters were monitored. In addition, the preliminary safety of IONPs-ASP was evaluated by organ coefficients and histopathological staining. IONPs-ASP was successfully constructed, with a small particle size, core–shell structure, hydrophilicity, and good biocompatibility. Compared to unmodified IONPs, IONPs-ASP showed better stability and dispersion. In the IDA rat model, IONPs-ASP exhibited superior therapeutic efficacy than that of ASP or IONPs monotherapy. Furthermore, no abnormalities were observed in the organ coefficients and tissue section. IONPs-ASP not only has a superior therapeutic effect on IDA but also has the effect of the ASP on supplementing blood. Hence, it may be used as a new iron supplementing agent with double therapeutic efficacy on blood supplementation for the treatment of IDA. Graphical Abstract In this study, we constructed Angelica sinensis polysaccharide (ASP)–modified iron oxide nanoparticles (IONPs) and demonstrated the therapeutic effects of IONPs-ASP on iron deficiency anemia (IDA), which was associated with IONPs supplementation and APS-stimulated hematopoietic cell generation.
