Ameliorative potential of ellagic acid via PPARγ against hyperlipidemia: Insights from mice and zebrafish

Junfeng Li,Min Guo,Cuiping Yuan,Tiezhu Li,Jie Zhang,Li Ren

ABSTRACT Peroxisome proliferator-activated receptor γ (PPARγ) was a major transcription factor and performed an essential function in the regulation of lipid metabolism. Ellagic acid (EA) was a natural phytochemical extracted from berries and pomegranates, and research had demonstrated its ameliorative effects on lipid metabolism disorders. We intended to explore the interventional effect and mechanism of action of EA on hyperlipidemia. In vitro experiments revealed that EA could reduce lipid accumulation in HepG2 cells. EA was found to bind stably to PPARγ, suppressing its nuclear translocation as well as its luciferase activity. It could also down-regulate the expression of genes and proteins related to lipid metabolism upstream and downstream of PPARγ. To explore the impact of EA on hyperlipidemia in vivo from different levels and perspectives, we established hyperlipidemia mice and hyperlipidemia zebrafish models. Consistent with the in vitro results, in vivo experiments revealed that EA attenuated lipid accumulation in hyperlipidemia mice and reduced the levels of related inflammatory factors in their serum. EA also inhibited lipid accumulation by decreasing the expression of upstream and downstream lipid-associated genes of pparg in hyperlipidemia zebrafish. In conclusion, EA could effectively improve hyperlipidemia by regulating PPARγ.

Chuling Liu,Li Cheng,Mingtian Yang,Zhengli He,Yanan Jia,Li Xu,Yuansong Zhang

Monascusis a fungus widely used in food fermentation. This study employed microbial technology, combined with microscopic morphological observations and ITS sequence analysis, to isolate, purify, and identify 10 strains of red yeast mold from variousMonascusproducts. After the HPLC detection of metabolic products, the M8 strain containing the toxic substance citrinin was excluded. Using the EWM-TOPSIS model, the remaining nine safeMonascusstrains were evaluated for their inhibitory activities against pancreatic lipase, α-glucosidase, α-amylase, and the angiotensin-converting enzyme. The M2 strain with the highest comprehensive scores for lowering blood sugar, blood lipids, and blood pressure was selected. Its fermentation product at a concentration of 3 mg/mL had inhibition rates of 96.938%, 81.903%, and 72.215%, respectively. The contents of the blood lipid-lowering active substance Monacolin K and the blood sugar and blood pressure-lowering active substance GABA were 18.078 mg/g and 5.137 mg/g, respectively. This strain can be utilized for the biosynthesis of important active substances such as Monacolin K and GABA, as well as for the fermentation production of safe and effective functional foods to address health issues like high blood lipids, high blood sugar, and high blood pressure in people. This study also provides insights into the use of natural fungi to produce healthy foods for combating chronic diseases in humans.

Ultrasonic extraction of polyphenols from roots of Ilex asprella: Structural characterization, antioxidant, antimicrobial, anti-inflammatory, and anti-hyperlipidemic activities

Jiarong Su,Peishan Li,Xinrui Tang,Zihao Chen,Qiang Peng,Huilin Li,Qianhua Zhang,Shengjie Lu,Famin Wen,Huaicheng Qin,Yongguang Bi,Juan Guo,Fansheng Kong,Haijie Jin,Canzhao Chen,Huiyan Xu

Plant polyphenols have gained increasing attention due to their abundant and powerful biological activities. However, polyphenols from roots of Ilex asprella (RIAPs) remain largely unexplored, with limited research on their composition and bioactivity, making it necessary to focus on RIAPs. In the present study, single - factor analysis and response surface optimization were performed for the extraction of RIAPs via an ultrasonic bath extraction method. The findings demonstrated that at 50 % ethanol concentration, 30 min of ultrasonic time, and 300 W of ultrasonic power, the maximum extraction content of RIAPs was 30.14 ± 0.41 mg/g. Compound and structural analysis via high-resolution liquid chromatography-mass spectrometry (LC-MS/MS) and a Fourier transform infrared (FT-IR) spectrometer confirmed that polyphenols were the majority of compounds comprising RIAPs, in addition to small amounts of alkaloids and trace amounts of other compounds such as amino acids. Scanning electron microscope (SEM) images revealed that RIAPs exhibited a densely reticulated and fragmented microstructure. In addition, bioactivity experiments demonstrated significant antioxidant, antibacterial, anti-inflammatory, and anti-hyperlipidemic activities of RIAPs, supporting their potential pharmaceutical applications. This study provides an experimental foundation for further pharmacological research and potential drug development of RIAPs.

Hypolipidemic effects and mechanisms of Val-Phe-Val-Arg-Asn in C57BL/6J mice and 3T3-L1 cell models

Xing Zhang,Wen Shi,Hui He,Ruge Cao,Tao Hou

Val-Phe-Val-Arg-Asn (VFVRN) has been identified and screened from lipid-lowering chickpea peptides (ChPs) previously. This work explored its lipid-lowering mechanisms by using a high-fat diet C57BL/6J mice model and 3T3-L1 preadipocyte cell model. Administration of VFVRN to hyperlipidemic mice significantly decreased the levels of total cholesterol (TC) and total triglyceride (TG) in liver and serum (P < 0.05), and increased the fecal triglyceride excretions. The expressions of sterol regulatory element-binding protein (SREBP)-2, liver X receptor (LXR)α, peroxisome proliferator-activated receptors (PPAR)α and their related target genes were regulated via AMP-activated protein kinase (p-AMPK) pathway in liver. Compared with model group, the levels of PPARα and PPARγ in 10 mg/kg VFVRN group were up-regulated by 36.96% and 36.49% in adipose tissues, while LXRα and ATP binding cassette G5/8 transporters (ABGC5/8) were lowered by 87.26% and 145% in small intestine respectively. Moreover, VFVRN promoted 3T3-L1 preadipocyte apoptosis by regulating the expressions of BaX, Caspase-3 and Bcl-2.

Comparison of the antioxidant activities of nonfumigated and sulphur-fumigated Chrysanthemum morifolium cv. Hang-ju induced by oxidative stress

Hongyan Ma,Shanshan Liu,Wenshan Qu,Qi Huang,Linyuan Li,Fujiang Chu,Yuyun Zhu,Xinlin Lv,Zhimin Wang,Jingjing Zhu

Context The traditional drying method, sun drying, for Chrysanthemum morifolium Ramat. cv. Hang-ju (Compositae) (HJ) is widely replaced by sulphur fumigation (SF), which has an unknown effect on its efficacy. Objective To investigate protective effects of nonfumigated HJ (NHJ) and sulphur-fumigated HJ (SHJ) water extracts against oxidative stress and lipid peroxidation.Materials and methods Sprague-Dawley rats were administered high-fat diet to induce hyperlipidaemia and randomly divided into eight groups (n = 6): control, fenofibrate, NHJ and SHJ extracts (1, 2 or 4 g crude drugs/kg/d; intragastric administration for 8 weeks). Serum total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), superoxide dismutase (SOD) and malondialdehyde (MDA) levels were detected. Human umbilical vein endothelial cells (HUVECs) were treated with NHJ and SHJ extracts (50, 100 or 200 μg/mL) for 24 h, followed by oxidized low-density lipoprotein (ox-LDL, 20 μg/mL) for 2 h in vitro. Cellular reactive oxygen species (ROS), SOD and MDA levels and apoptosis were evaluated. Results NHJ was more effective than SHJ in decreasing serum TG, TC, LDL-C, LDL/HDL and MDA while increasing serum HDL-C and SOD levels at high doses. SHJ (IC50=19.9 mg/mL) suppressed HUVEC growth stronger than NHJ (IC50=186.7 mg/mL). At 200 μg/mL, NHJ was more effective than SHJ in downregulating ROS and MDA levels, reducing HUVECs apoptosis rate and elevating SOD activity in ox-LDL-treated HUVECs.Conclusions SF causes oxidative damage and attenuates antioxidative activity in ox-LDL-treated HUVECs, which promotes lipid peroxidation. SF is not recommended for processing HJ.

Chemometrics strategy coupled with high resolution mass spectrometry for analyzing and interpreting comprehensive metabolomic characterization of hyperlipemia

Qiqi Zhao,Aihua Zhang,Wenjing Zong,Na An,Huamin Zhang,Yihan Luan,Hongxin Cao,Hui Sun,Xijun Wang

Hyperlipidemia (HLP) is a metabolic disorder which is characterized by a disturbance in lipid metabolism and is a primary risk factor for cardiovascular disease and atherosclerosis. Patients who were diagnosed with hyperlipidemia are usually with a serious dyslipidemia, which can be evaluated by current clinical markers such as triglyceride, total cholesterol, low density lipoprotein cholesterol, and high-density lipoprotein. Metabolic profiling has great potential to help the diagnosis and prognosis of hyperlipidemia patients in an earlier stage. To explore the metabolic profiling, we used ultraperformance liquid chromatography mass spectrometry, monitored the metabolites' alterations and impaired pathways in HLP patients. Of note, a total of 37 serum metabolites from HLP patients were tentatively identified. Furthermore, they were involved in 11 pathways such as linoleic acid metabolism, glycerophospholipid metabolism, terpenoid backbone biosynthesis, sphingolipid metabolism, glycosylphosphatidylinositol-anchor biosynthesis, steroid hormone biosynthesis, arachidonic acid metabolism, pentose and glucuronate interconversions, starch and sucrose metabolism, glycerolipid metabolism and purine metabolism. These results provide potential biomarkers for early risk assessment of HLP and offer further insights into the complex metabolic pathway changes in HLP. They also suggest the distinction of utilizing UPLC-Q-TOF-MS for exploring metabolic characterization of HLP patients.

Magnetic Microdimer as Mobile Meter for Measuring Plasma Glucose and Lipids

Yu Shimin,Sun Zhongqi,Zhang Zhanxiang,Sun Haoran,Liu Lina,Wang Wuyi,Li Mu,Zhao Qingsong,Li Tianlong

With the development of designed materials and structures, a wide array of micro/nanomachines with versatile functionalities are employed for specific sensing applications. Here, we demonstrated a magnetic propelled microdimer-based point-of-care testing system, which can be used to provide the real-time data of plasma glucose and lipids relying on the motion feedback of mechanical properties. On-demand and programmable speed and direction of the microdimers can be achieved with the judicious adjustment of the external magnetic field, while their velocity and instantaneous postures provide estimation of glucose, cholesterol, and triglycerides concentrations with high temporal accuracy. Numerical simulations reveal the relationship between motility performance and surrounding liquid properties. Such technology presents a point-of-care testing (POCT) approach to adapt to biofluid measurement, which advances the development of microrobotic system in biomedical fields.

Molecular hybridization method for obtaining paeonol-based fibrate derivatives with potent lipid-lowering and hepatoprotective activity

Quan Lina,Guo Ying,Wang Siyao,Sun Mengfei,Pang Yan,Cui Chunli,Wang Jinrui,Wei Jinlian,Wei Peifeng,Xie Yundong

Molecular hybridization method was applied to design and synthesize a series of target compounds paeonol-based fibrate derivatives. The target compound was screened using a Triton WR-1339 induced hyperlipidemia mouse model, and compound T9 was found to have good lipid-lowering activity. The dose-dependent study of its lipid-lowering activity was also conducted. To further evaluate the lipid-lowering activity of compound T9, a hyperlipidemic mouse model induced by high fat emulsion can be used. The findings of the research illustrate that T9 is capable of significantly decreasing blood lipid levels, including TG, TC, LDL-C, and increasing HDL-C. The results of liver tissue oil red O staining and HE staining demonstrated that T9 improved the hepatic lipid accumulation, thus decreasing AST and ALT levels and protecting against hyperlipidemic liver injury. Studies into the lipid-lowering effect of T9 have indicated that it can upregulate PPAR-α protein expression in liver tissue, while simultaneously decreasing the expression of HMG-CoA protein. T9 was further demonstrated to possess antioxidant properties, as evidenced by an increase in SOD and a decrease in MDA, as well as anti-inflammatory effects, demonstrated by a decrease in TNF-α, IL-1β, and IL-6, thus confirming its potential as a hypolipidemia and hepatoprotective agent. Glucomannans Alleviated the Progression of Diabetic Kidney Disease by Improving Kidney Metabolic Disturbance

Haihong Chen,Qixing Nie,Jielun Hu,Xiaojun Huang,Ke Zhang,Shaoping Nie

Scope This study is aimed to investigate the kidney protective effects of glucomannans from Dendrobium officinale stem, konjac, and Aloe vera leaves on type 2 diabetic rats and explore its potential mechanisms. Methods and results Pathological and metabolomics analysis methods are adopted in this study. Compared with the model group, lower levels of fasting blood glucose, serum insulin, and glycated serum protein are observed in glucomannan-treated groups. Concentrations of serum low-density lipoprotein cholesterol, total cholesterol, triacylglycerols, and nonesterified fatty acid are significantly decreased by glucomannan treatment. Furthermore, glucomannan treatment significantly decreased the levels of uric acid, creatinine, urea in serum, and glycosuria, ketone body, and protein in the urine. Histopathological analysis showed that glucomannan treatment normalized the architecture of glomerulus. Metabolomic analysis indicated glucomannan treatment could improve urea cycle, metabolism of lipid, glucose, and amino acids on diabetes. In particular, the konjac glucomannan treatment is more effective in lipid and glucose regulation. Glucomannan from D. officinale is more effective in balancing the urea cycle and amino acid metabolism. Conclusions The disturbance of lipid, glucose, and amino acid metabolism is closely associated with the advancement of diabetic kidney disease, and glucomannan treatment could be efficient in the management of diabetic kidney disease.

Effects of brown seaweed oligosaccharides on obesity and constipation managements

Jingyu Gao,Fan Wu,Mingzhu Yan,Xuejiang Wang,Yan Chi,Yu Zhang,Yu Peng,Mo Li,Yuanying Ni,Xin Wen

Brown seaweed oligosaccharides (BSO) have demonstrated potential as nutraceuticals with cholesterol-lowering, anti-obesity, and anti-constipation properties. In this study, we initially examined how BSO impact body weight, blood lipid levels, and adipose tissue in a rat model of obesity induced by a high-fat diet. Our findings revealed that BSO administration significantly attenuated body weight gain, ameliorated dyslipidemia, and reduced visceral adiposity. Additionally, we evaluated the prophylactic efficacy of BSO in a loperamide hydrochloride-induced constipation mouse model by assessing defecation parameters and gastrointestinal motility. Our results indicated that BSO intervention significantly increased fecal pallet output and fecal water content, as well as improved intestinal transit. In conclusion, this study lays the foundation for a deeper understanding of how BSO may influence obesity and constipation, offering novel insights into its potential safe and efficacious clinical applications.

PPP2CB aggravates atherosclerosis-related dyslipidemia via LOX-1/MAPK/ERK signaling pathway

An He,Cheng Dong-liang,Xia Xian-ru,Li Xian-dong,Ruan Zhi-hua,Peng Chun-yan

Background Dyslipidemia has been extensively documented as a key driver of cardiovascular pathology. Regulating lipid homeostasis holds promise for treating atherosclerosis (AS). Although the protein phosphatase 2 catalytic subunit beta (PPP2CB) is involved in post-transcriptional gene regulation, its role in AS-associated dyslipidemia is not well understood. Methods The study included both human participants and animal models. The following techniques were employed: cell culture, extraction of exosomes, preparation of pooled hyperlipidemic serum (HS), transfection, western blotting, immunofluorescence staining, quantitative reverse transcription polymerase chain reaction (qRT-PCR), co-immunoprecipitation, low-density lipoprotein cholesterol (LDL-C) uptake assay, biochemical assays, assessment of aortic atherosclerotic lesions, as well as statistical analysis. Results This study identified a marked upregulation of PPP2CB expression in peripheral blood leukocytes of AS patients, artery plaque of ApoE −/− mice given a high-fat diet, and hepatic cells exposed to hyperlipidemic stimuli. Overexpression of PPP2CB in hepatic cells exacerbated lipid accumulation and low-density lipoprotein uptake, whereas silencing PPP2CB mitigated this effect. Immunofluorescence co-localization and co-immunoprecipitation analysis confirmed a direct interaction between PPP2CB and lectin-like oxidized LDL receptor-1 (LOX-1). Notably, PPP2CB manipulation disrupted hyperlipidemia-induced LOX-1 expression. Additionally, PPP2CB-mediated lipid dysregulation was linked to the activation of the LOX-1/ mitogen-activated protein kinase (MAPK)/ extracellular signal-regulated kinase (ERK) signaling cascade. Conclusions These results unveil PPP2CB as a novel lipid regulator in the progression of pathological AS and highlight its involvement in signaling regulation during abnormal lipid metabolism. PPP2CB could be considered a promising candidate for biomarker development and therapeutic intervention in AS.

An Optimized Aerogel-Based Apheresis Device for Targeted Lipid Clearance in Elderly Hyperlipidemia Patients

Hao Zheng,Jiangbangrui Chu,Shikang Li,Xun Tang,Shicheng Bi,Kerong Chen,Anwei Zhou,Youwen Yang,Xinghai Ning,Yurui Xu

Elderly patients with hyperlipidemia often exhibit resistance to conventional hypolipidemic treatments, underscoring the need for more effective strategies to address lipid imbalances in this high-risk group. This study introduces LipClean, an aerogel-based apheresis device specifically designed to remove harmful plasma lipids. LipClean is constructed using hydrophilic cellulose fibers, which serve as a supramolecular platform for synthesizing hydrophobic conjugated polymers through a Sonogashira-Hagihara reaction. These conjugated polymers are then cross-linked with the cellulose fibers via phosphorylation, generating an aerogel monolith with an interpenetrating network of hydrophilic fibers and hydrophobic polymers. Unlike bilayer aerogels that separate hydrophilic and hydrophobic layers, LipClean's interpenetrating structure is precisely engineered through polymer design and gradient cross-linking. This optimization enhances both bodily fluid flow and lipid adsorption while minimizing the removal of essential plasma components and ensuring unobstructed cell passage. In preclinical testing, LipClean significantly reduced triglyceride and cholesterol levels in an elderly rat model of hyperlipidemia and normalized lipid levels in blood samples from hypertensive patients. Importantly, purified blood maintained normal levels of blood cells and physiological and biochemical indicators after apheresis, highlighting LipClean's potential for managing hyperlipidemia-related disorders. This study, therefore, underscores the importance of interdisciplinary collaboration in driving medical device innovation.
