Rehmanniae Radix Praeparata in Blood Deficiency Syndrome: UPLC-Q-TOF-MS Profiling, Network Pharmacology, and PI3K-AKT Activation

Ruxi Gao,Fanyi Wang,Xiang Liu,Chu Yuan,Guoshun Shan

This study investigated the therapeutic mechanism of Rehmanniae Radix Praeparata (RRP) in treating blood deficiency syndrome (BDS) through integrated chemical analysis and pharmacological validation. UPLC-Q-TOF-MS identified chemical components of Rehmanniae Radix (RR) and RRP, with network pharmacology analysis suggesting AKT1 and NOS3 in the PI3K-AKT pathway as potential therapeutic targets. Pharmacodynamic evaluations using ELISA, hematological analysis, histopathology, and immunohistochemistry demonstrated RRP’s efficacy in improving hematological parameters, energy metabolism, and organ pathology in BDS mice. Experimental validation via RT-qPCR and Western blot confirmed significant upregulation of AKT1 and NOS3 mRNA and protein expression following RRP treatment. The findings indicate that RRP alleviates BDS by activating the PI3K-Akt signaling pathway to modulate AKT1 and NOS3 expression, providing mechanistic insights into its therapeutic actions.

Characterization of effective constituents in Acanthopanax senticosus fruit for blood deficiency syndrome based on the chinmedomics strategy

Chunlei Wan,Xijun Wang,Hongda Liu,Qingyu Zhang,Guangli Yan,Zhineng Li,Heng Fang,Hui Sun

The fruit of Acanthopanax senticosus (Rupr. and Maxim.) has been newly developed for the treatment of blood deficiency syndrome clinically, but the effective constituents are still unclear, restricting its quality control and the new medicinal development based on it. This study elucidated the efficacy of A. senticosus fruit (ASF) for treating blood deficiency syndrome and accurately characterize the constituents. Chinmedomics strategy was used to identify the metabolic biomarkers of the model and the overall effect of ASF was evaluated based on the biomarker when it showed intervention effects for blood deficiency syndrome. ultrahigh performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) was used to analyze the components in the blood absorbed from A. senticosus fruit, and the components highly relevant to the biomarker are regarded as potential effective constituents for blood deficiency syndrome. Twenty-two of the 28 urine metabolites of blood deficiency syndrome were significantly regulated by A. senticosus fruit, 97 compounds included 20 prototype components, and 77 metabolites were found in vivo under the acting condition. The highly relevant constituents were isofraxidin, eleutheroside B, eleutheroside B1, eleutheroside E, and caffeic acid, which might be the effective constituents of A. senticosus fruit. It is a promising new medicinal resource that can be used for treating blood deficiency syndrome.

Metabolomics and molecular docking to compare of the efficacies of wild-simulated and transplanted Astragali Radix based on its compatibility with Angelicae Sinensis Radix against blood deficiency

YueTao Liu,ZiYing Wang,XueMei Qin

Astragali Radix (AR) has been widely used as bulk medicines in China. Currently, there are mainly two types in the market, wild-simulated AR (WAR) and transplanted AR (TAR). The present work was designed to compare their effects in treatment of blood deficiency with its compatibility with Angelicae Sinensis Radix, a classical formula Danggui Buxue decoction (DBD). UHPLC-Q-Exactive-MS-based serum metabolomics was firstly applied to compare their metabolic regulations in blood deficiency mice. Metabolic results showed that 17 metabolites were closely related to blood deficiency. WAR-DBD and TAR-DBD significantly improved 13 and 8 metabolites among them, respectively. Further, fourteen genes were selected as promising targets for treating blood deficiency by searching GeneCards and CTD database. The results of molecular docking also showed that eight components of AR and three serum metabolites had strongest correlations with blood deficiency targets. The results further revealed that WAR-DBD was better than TAR-DBD against blood deficiency. The method provided a new way to compare the hematopoietic differences of AR of different cultivation methods. In addition, molecular docking plays an irreplaceable role in explaining the molecular mechanism of biological processes .

Protective effects of aqueous extract from Gei Herba on blood-deficiency mice: insights gained by a metabolomic approach

Ruru Zhao,Wenbi Mu,Xiaoning Wang,Sha Yang,Cancan Duan,Jianyong Zhang

With increasing tumor incidence, anemia (categorized as a blood deficiency in traditional Chinese medicine) caused by chemotherapy has become a major side effect worldwide. Gei Herba, a traditional Miao nation herb, has a prominent effect on the treatment of blood deficiency (BD). However, its application is limited owing to little fundamental research. Therefore, a GC-MS metabolomic approach was used to study the protective effect of aqueous extract from Gei Herba (AEG) on BD mice and its putative mechanism. In this study, 32 male mice were divided into four groups: a control group, a BD model group, and two groups subjected to AEG treatment at a daily dose of 0.15 or 0.30 g kg−1 for 8 d. After AEG treatment, the HGB and HCT levels in the blood of BD mice were significantly increased, the activity of superoxide dismutase was increased, and the histomorphology of the liver was improved. Furthermore, compared with those in the model group, the levels of eight significant metabolites [phosphoric acid, glycine, L-proline, ribitol, (Z,Z)-9,12-octadecadienoic acid, oleic acid, uridine and 4B2H-carbamic acid] in the liver were significantly changed by AEG. The findings of this study provide sound evidence regarding the protective effects of AEG in BD mice from both classical and metabolomic perspectives. The mechanisms of action of AEG could be related to regulation of linoleic acid metabolism and that of glycine, serine, and threonine metabolism.

Comprehensive Metabolomics and Network Pharmacology to Explore the Mechanism of 5-Hydroxymethyl Furfural in the Treatment of Blood Deficiency Syndrome

Zhang Wensen,Cui Na,Su Fazhi,Wang Yangyang,Yang Bingyou,Sun Yanping,Guan Wei,Kuang Haixue,Wang Qiuhong

Radix Rehmanniae (RR, from Radix Rehmanniae (Gaertn.) DC.) is a natural medicine used in traditional Chinese medicine (TCM) since ancient times for the treatment of blood disorders. RR is steamed to get Rehmanniae Radix Praeparata (RP), which has a tonic effect on blood; the content of 5-hydromethylfurfural (5-HMF) increases more than four times after steaming. Studies have shown that 5-HMF has positive pharmacological effects on cardiovascular and hematological disorders. This study aimed to explore and verify the impact of 5-HMF on rats with chemotherapy-induced blood deficiency syndrome (BDS). Rats were given cyclophosphamide (CP) and acetophenhydrazine (APH) to induce BDS, the coefficients of some organs (liver, spleen, and kidney) were measured, and a routine blood test examined the coefficients of several peripheral blood cells. Metabolomics and network pharmacology were combined to find important biomarkers, targets, and pathways. Western blot was used to detect the expression of CYP17A1 and HSD3B1 proteins in the spleen. All these findings suggested that the 5-HMF significantly increased the number of peripheral blood cells and reversed splenomegaly in rats. In addition, 5-HMF upregulated CYP17A1 and HSD3B1 protein expression in splenic tissues. Also, 5-HMF ameliorated chemotherapy-induced BDS in rats, and its therapeutic mechanism might depend on steroid hormone biosynthesis and other pathways. It acts on blood deficiency via multiple targets and pathways, which is unique to Chinese medicine.

Blood-Enriching Effects and Immune-Regulation Mechanism of Steam-Processed Polygonatum Sibiricum Polysaccharide in Blood Deficiency Syndrome Mice

Wang Juan,Wang Furong,Yuan Lixia,Ruan Hongsheng,Zhu Zhibiao,Fan Xiaoling,Zhu Lingyan,Peng Xin

Polygonatum sibiricum Red. has been used as a medicinal herb and nutritional food in traditional Chinese medicine for a long time. It must be processed prior to clinical use for safe and effective applications. However, the present studies mainly focused on crude Polygonatum sibiricum (PS). This study aimed to investigate the chemical properties, blood-enriching effects and mechanism of polysaccharide from the steam-processed Polygonatum sibiricum (SPS), which is a common form of PS in clinical applications. Instrumentation analyses and chemistry analyses revealed the structure of SPS polysaccharide (SPSP). A mice model of blood deficiency syndrome (BDS) was induced by acetylphenylhydrazine (APH) and cyclophosphamide (CTX). Blood routine test, spleen histopathological changes, serum cytokines, etc. were measured. The spleen transcriptome changes of BDS mice were detected by RNA sequencing (RNA-seq). The results showed that SPSP consists predominantly of Gal and GalA together with fewer amounts of Man, Glc, Ara, Rha and GlcN. It could significantly increase peripheral blood cells, restore the splenic trabecular structure, and reverse hematopoietic cytokines to normal levels. RNA-seq analysis showed that 122 differentially expressed genes (DEGs) were obtained after SPSP treatment. GO and KEGG analysis revealed that SPSP-regulated DEGs were mainly involved in hematopoiesis, immune regulation signaling pathways. The reliability of transcriptome profiling was validated by quantitative real-time PCR and Western blot, and the results indicated that the potential molecular mechanisms of the blood-enriching effects of SPSP might be associated with the regulating of JAK1-STAT1 pathway, and elevated the hematopoietic cytokines (EPO, G-CSF, TNF-α and IL-6). This work provides important information on the potential mechanisms of SPSP against BDS.

Gut microbial analysis combined with metabolomics reveal the mechanism of stachyose on blood deficiency syndrome in rats

Wensen Zhang,Na Cui,Fazhi Su,Yanping Sun,Biao Li,Meng Liu,Yuanning Zeng,Bingyou Yang,Qiuhong Wang,Haixue Kuang

Stachyose (ST), a naturally occurring compound extracted from cucumbers and legumes, holds great promise as a natural therapeutic agent. In this study, the impact of ST on blood deficiency syndrome (BDS) induced by cyclophosphamide (CP) and acetylphenylhydrazine (APH) was investigated in a rat model. Subsequently, the levels of erythropoietin (EPO), granulocyte colony-stimulating factor (G-CSF), tumour necrosis factor-alpha (TNF-α), and interleukin-6 (IL-6) were quantified in rat blood and peripheral blood cells. Furthermore, haematoxylin and eosin (HE) staining was employed to elucidate morphological alterations in the spleen tissue. To gain comprehensive insights into the therapeutic potential and underlying mechanisms of ST against BDS, we used 16S rRNA gene sequencing and serum and spleen metabolomic analyses. Our findings revealed a significant ameliorative effect of ST on BDS. Notably, at the phylum level, Firmicutes and Bacteroidota exhibited marked variations in abundance, while at the genus level, discernible changes were observed in the abundances of Lactobacillus and Bifidobacterium , among others. The therapeutic effects of ST on BDS were proposed to be due to its modulation of phenylalanine metabolism and glycine, serine and threonine metabolism and its impact on the restructuring of the gut microbiota.

Metabolomic strategies and biochemical analysis of the effect of processed Rehmanniae radix extract on a blood-deficient rat model

Wang Yang-yang,Zhou Ning,Shan Zeng-fu,Ke Ying-ying,Liu Zhen,Liu Zhen-hui,Feng Wei-sheng,Zheng Xiao-ke

Background Rehmanniae Radix (RR), an herb with numerous pharmacological effects, is widely used in traditional Chinese medicine for the treatment of blood deficiency syndrome, either alone or in combination with other herbs. However, the mechanism by which processed Rehmanniae Radix (PRR) improves blood enrichment efficacy has not been clearly defined. Methods Ultra-performance liquid chromatography coupled to quadrupole time-of-flight mass (UPLC-Q-TOF/MS) and biochemical methods were combined to explore the hematopoietic functional mechanisms of PRR on blood deficiency in a rat model, as well as the potential active ingredient for blood enrichment efficacy. The pharmacological effects of PRR were evaluated on a rat blood deficiency model induced by cyclophosphamide in combination with 1-acetyl-2-phenylhydrazine. The blood routine index, including white blood cell (WBC), red blood cell (RBC), and platelet (PLT) counts, as well as hemoglobin (HGB) level, and the changing metabolite profile based on urine and serum were assessed. Nontargeted metabolomic studies, combined with biochemical analyses, were employed to clarify pharmacological mechanisms. Results PRR significantly increased the blood routine index levels and reversed the levels of SOD, GSH, and ATP. The PRR group was similar to the control group, as determined from the metabolic profile. All of the 60 biomarkers, representing the typical metabolic characteristics of the blood-deficient rat model, mainly involved energy metabolism dysfunction, the peripheral circulation system, and oxidative damage in the body. This improvement may be attributed to changes in polysaccharide and sixteen non-polysaccharide compounds in PRR, which were caused by processing RR with rice wine. Conclusions The strategies of integrated metabolomic and biochemical analyses were combined, revealing the biological function and effective mechanism of PRR.
