Attraction of adult Harmonia axyridis to volatiles of the insectary plant Cnidium monnieri

Zhiping Cai,Fang Ouyang,Jianwei Su,Xingrui Zhang,Chenglin Liu,Yunli Xiao,Jianping Zhang,Feng Ge

Plant volatiles mediate the interactions within plant-insect communities and influence the distribution and behavior of insects. Natural enemies often utilize the volatiles that released from plants to locate the profitable patches for foraging. The lady beetle Harmonia axyridis (Pallas) is a dominant generalist predator of various pests in agriculture fields, yet little is known about the chemical cues which the predator employs to locate its prey. In this study, we tested the attractiveness of volatiles emanating from healthy and aphid-infested insectary plant Cnidium monnieri (L.) Cuss. to H. axyridis adults in a Y-tube olfactometer. Next, using coupled gas chromatography – electroantennogram detection (GC-EAD) and gas chromatography – mass spectrometry (GC–MS) identified the volatile components which elicited electrophysiological activity in H. axyridis adults. Lastly, Y-tube olfactometer tests in laboratory and field traps were used to verify the effectiveness of the volatile components. In olfactometer trials, H. axyridis adults preferred healthy and aphid-infested plants versus control. In GC–MS and GC-EAD trials, two antenna-active components (1,2-diethylbenzene and p-diethylbenzene) were detected among the volatile blends emanating from healthy and aphid-infested plants; and in ensuing behavioral assays, H. axyridis adults were attracted to two synthetic volatile compounds. Under field conditions, the number of H. axyridis adults as recorded in traps for two synthetic volatile components at three different concentrations (100 mg/ml, 10 mg/ml, and 1 mg/ml) were significantly higher than those for mineral oil (control). These findings could provide an effective means for the cultivation of insectary plants to develop natural enemy attractants and support the biological control in the field.

Electrophysiological and behavioral responses of Tamarixia radiata (Hymenoptera: Eulophidae) to volatiles of nymphal Diaphorina citri (Hemiptera: Liviidae)

Liu Yan-Mei,Huang Yuan-Yuan,Wang Fei-Feng,Hu Yu-Wei,Zhang Zhi-Lin,Cuthbertson Andrew G S,Qiu Bao-Li,Sang Wen

Huanglongbing (HLB), a devastating citrus disease caused by Candidatus Liberibacter asiaticus, is efficiently vectored by the Asian citrus psyllid, Diaphorina citri Kuwayama (Hemiptera: Liviidae). Tamarixia radiata (Waterston) plays a crucial role as an ectoparasitoid, preying on D. citri nymphs. By collecting and identifying headspace volatiles from fifth instar nymphs of D. citri using a gas chromatograph–mass spectrometer (GC–MS), we obtained a collection of 9 volatile compounds. These compounds were subsequently chosen to investigate the electrophysiological and behavioral responses of female T. radiata. At a concentration of 10 μg/μl, 9 compounds were compared with cis-3-hexen-1-ol (control), resulting in trans-2-nonenal inducing the highest relative electroantennogram (EAG) value, followed by hexanal, heptanal, n-heptadecane, tetradecanal, n-tetradecane, n-pentadecane, 1-tetradecanol, and 1-dodecanol. The top 5 EAG responses of female T. radiata to these compounds were further investigated through EAG dose–response experiments. The results showed positive dose–responses as concentrations increased from 0.01 to 10 μg/μl. In Y-tube olfactometer bioassays, female T. radiata exhibited a preference for specific compounds. They were significantly attracted to tetradecanal at a concentration of 10 µg/µl and trans-2-nonenal at 0.01 µg/µl, while no significant attraction was observed toward hexanal, heptanal, or n-heptadecane. Our report is the first to demonstrate that volatiles produced by D. citri nymphs attract T. radiata, which suggests that this parasitoid may utilize nymph volatiles to locate its host.

Behavioral responses of Anagrus nilaparvatae to common terpenoids, aromatic compounds, and fatty acid derivatives from rice plants

Guofeng Mao,Jiaxin Tian,Ting Li,Hatem Fouad,Hassan Ga'al,Jianchu Mo

The attraction of parasitoids to their hosts is a key issue in efforts to enhance biological control efficiency. This study was conducted to identify which rice plant volatiles are attractive to females of the rice planthopper egg parasitoid Anagrus nilaparvatae Pang et Wang (Hymenoptera: Mymaridae) by using Y-tube olfactometer behavior assays. Our results demonstrated that 10 of 17 selected volatiles – that is, 3-carene, linalool oxide, linalool, benzyl alcohol, benzaldehyde, methyl salicylate, ( E )-2-hexenal, ( E )-2-hexen-1-ol, ( E )-3-hexen-1-ol, and hexanal – from three major groups of rice plant volatiles – terpenoids, aromatic compounds, fatty acid derivatives – attracted female A. nilaparvatae at various doses. Two full-component mixtures of 10 behaviorally active compounds at different ratios were more attractive to wasps compared with individual volatiles. Blends composed of the 10 behaviorally active compounds in their optimally attractive doses (i.e., each in the dose that elicited the strongest response in the wasps) or at equal mass ratio may be considered as attractants for A. nilaparvatae and thus help to control rice planthoppers.

Volatiles of the trap crop sunflowers: Effects on the behaviour of adult female Conogethes punctiferalis moths

Jia Zhou,Li-Lin Chen,Yu-Tong Zhu,Evan Siemann,Nian-Feng Wan

Sunflowers ( Helianthus annuus ) are potential trap crops to attract the yellow peach moth ( Conogethes punctiferalis ) in orchards but the volatile compounds released from sunflowers and their effects on C. punctiferalis behaviour are not clear. Here we used solid phase micro-extraction to collect volatiles released from flower disks of sunflowers, identified them by gas chromatography–mass spectrometry, and examined the attractiveness of some of the compounds to mated female adult C. punctiferalis moths in Y-tube olfactometer choice tests. We detected nine volatile compounds (α-Pinene, β-Phellandrene, Camphene, β-Pinene, 3-Carene, D-Limonene, Eucalyptol, γ-Terpinen, and Valencene) with consistently large peak areas for α-Pinene and β-Phellandrene. Among tested volatiles, both α-Pinene and β-Phellandrene were attractive to C. punctiferalis in olfactometer tests but this was only significant for higher concentrations of β-Phellandrene. This suggests that β-Phellandrene may promote C. punctiferalis oviposition on sunflower disks. This study provides insights into volatile chemical mediated responses of insect herbivores to trap crops.

Electroantennogram and behavioural responses of Plutella xylostella to volatiles from the non-host plant Mentha spicata

Xi-Zhong Yan,Zhi-Yu Wang,Li Ma,Cheng-Fei Song,Jin-Yu Zhao,Chen-Zhu Wang,Chi Hao

Plutella xylostella Linnaeus (Lepidoptera: Plutellidae) is one of the most important pests of cruciferous vegetables. Mechanically damaged Mentha spicata Linnaeus (Lamiales: Lamiaceae) strongly repels P. xylostella adults; however, the repellent mechanism remains unclear. In this study, the repellent mechanism was explored based on the electroantennogram (EAG) and behavioural responses of P. xylostella to volatiles from the damaged M. spicata . First, the active volatile compounds in the headspace volatiles of damaged M. spicata were identified by gas chromatography-electroantennographic detection (GC-EAD) and gas chromatography–mass spectrometry (GC–MS). Furthermore, the EAG and behavioural responses of P. xylostella to these compounds were investigated. The GC-EAD results indicated that four compounds, 2,4-dimethylheptane, 4-methyloctane, 2-methylnonane, and myrcene induced EAG responses in female antennae. 2-methylnonane and myrcene elicited strong dose-dependent EAG responses starting at a dose of 1 μg/μL. Choice tests in a four-arm olfactometer showed significant repellent effects of the four compounds on P. xylostella adults at 100 μg/μL, and the repellent indices of 2,4-dimethylheptane, 4-methyloctane, 2-methylnonane, and myrcene were 70.0%, 76.7%, 66.7%, and 73.3% to mated females, respectively. Finally, oviposition repellence assays showed that female adults were significantly repelled by these compounds, and the oviposition repellent index for myrcene was 23.9%. 2,4-dimethylheptane, 4-methyloctane, 2-methylnonane, and myrcene mediate the repellence of wounded M. spicata to P. xylostella adults, suggesting that the four compounds contribute towards developing behavioural regulators of this important pest.

Electrophysiological and behavioural responses of Plutella xylostella L. (Lepidoptera: Plutellidae) to thirteen non-host plant essential oils

Chengfei Song,Xinyao Ma,Jiao Liu,Li Ma,Xingtao Qie,Xizhong Yan,Chi Hao

The diamondback moth (DBM), Plutella xylostella (L.), is a cosmopolitan obligate pest of cruciferous crops. Currently, DBMs management is largely dependent on broad-spectrum synthetic pesticides, which has led to the adverse effects on the environment, human health, and nontargeted organisms. Thus, alternative control methods are needed. The essential oils (EOs) derived from bioactive species may be developed as effective alternatives for the management of DBMs that is safe and eco-sustainable than conventional pesticides. In this study, we investigated electrophysiological (EAG) and behavioral responses of DBM adults to the thirteen non-host EOs of Acorus calamus , Ageratum conyzoides , Artemisia argyi , Chenopodium ambrosioides , Cymbopogon citratus , Foeniculum vulgare , Ocimum basilicum , Pelargonium hortorum , Perilla frutescens , Pogostemon cablin , Polygonum hydropiper , Rosmarinus officinalis , and Ruta graveolens in order to contribute to the development of low environmental impact management alternatives. EAG recordings revealed all the tested EOs significantly elicited EAG responses of DBM females, and the males were sensitive to the eleven EOs except A. conyzoides and O. basilicum . Meanwhile, EAG recordings of DBMs exhibited dose-dependent responses to these tested EOs in most cases. In Y-tube olfactometer bioassays, the EOs of A. calamus , C. citratus , P. hortorum , P. hydropiper , and R. graveolen demonstrated prominent repellent effects on DBM females at concentrations of 10 μg/μL or higher. The results of oviposition bioassays found the number of eggs on the two plants with the five tested EOs were less than that on the two plants with paraffin oil. And the oviposition indexes of the plants that fixed with the EOs of P. hortorum , R. graveolen , A. calamus , P. hydropiper , C. citratus , and paraffin oil were –71.81%, –59.43%, –57.46%, –51.11%, –35.41%, and 0.09%, respectively. These findings indicate that the five EOs, especially P. hydropiper EO, provide active and safe tools for natural repellents to prevent and control DBMs infestations of cruciferous vegetables.

Electrophysiological and laboratory behavioral responses of a leaf beetle pest of elm, Ambrostoma quadriimpressum, to selected plant volatiles and essential oils

Yan-Chen Wang,Peng Li,De-Fu Chi

The leaf beetle Ambrostoma quadriimpressum Motschulsky (Coleoptera: Chrysomelidae) is distributed in China, Siberia, and elsewhere in northeastern Asia. This pest feeds only on buds and leaves of elm trees (Ulmaceae) and inhibits elm trees sprouting, increasing the risk of damage and destruction by other pests. We investigated selected plant compounds that could be used for the development of semiochemical-based push-pull methods for the control of this elm pest. Electroantennogram (EAG), Y-tube olfactometer, and wind tunnel bioassays were conducted to test the electrophysiological and behavioral response of A. quadriimpressum to nine individual volatiles and five essential oils. Individual volatiles and essential oils were selected based on their activity against insects in previous studies. The individual volatiles and essential oils were dissolved in dichloromethane and adsorbed by filter paper. Using a pipette, odor stimuli were delivered as 0.5-s puffs of air into a continuously humidified air stream in the odor delivery tube at 400 ml per min. Ambrostoma quadriimpressum adults display strong EAG responses to l mol l −1 isoeugenol, (1 R )-(+)-α-pinene, 1% pepper oil (derived from the pericarp of Piper chinense Miq.), and peppermint oil [derived from the aerial parts of Mentha haplocalyx (Briq.)] compared to the control. In behavioral assays conducted using a Y-tube olfactometer and a wind tunnel, adult beetles were attracted by (1 R )-(+)-α-pinene but repelled by 0.1% pepper oil. The results provide a basis for the development of a ‘push-pull’ strategy to manage A. quadriimpressum .

Identification of carbon dioxide receptors in the tropical bed bug, Cimex hemipterus (Hemiptera: Cimicidae)

Delong Kong,Zitong Wang,Hui Guo,Tao Lin,Dingxin Jiang,Hualong Qiu,Tao Ma,Chunsheng Jia,Lufei Xie,Yongyue Lu,Feng Liu,Changlu Wang,Desen Wang

Carbon dioxide (CO 2 ) plays a crucial role in the host search/localization process of bed bugs, Cimex lectularius L., and Cimex hemipterus (F.). Current research on the CO 2 perception by bed bugs mainly focuses on their behavioral responses, and the molecular mechanisms are still unclear. In addition, existing research has mainly been conducted on C. lectularius with very little research on C. hemipterus . In this study, we investigated the behavioral response of C. hemipterus to CO 2 and analyzed the role of antennae in the CO 2 sensing process. Then, potential CO 2 receptor genes were screened through antennal RNA sequencing and tissue-specific expression profiling analysis. Finally, the function of CO 2 receptors was further validated using RNA interference. Results indicated that increasing the CO 2 concentration in the air not only stimulated the activity of C. hemipterus , but also presented a directional attraction effect on them. CO 2 , with a concentration of 0.06%–1.8%, had a significant attraction effect on C. hemipterus . Removal of the antennae led to the loss of bed bugs’ preference for CO 2 . Four candidate CO 2 receptor genes ( ChGR1 , ChGR2 , ChGR3 , and ChGR4 ) were identified through transcriptomic analysis of the bed bug antennae, and 3 of them ( ChGR2 , ChGR3 , and ChGR4 ) were highly expressed in the antennae. Silencing ChGR2 or ChGR4 individually led to a significant reduction or even loss of CO 2 sensing ability in C. hemipterus . In conclusion, CO 2 can induce the host searching behavior of C. hemipterus ; moreover, ChGR2 and ChGR4 are crucial for detecting CO 2 in C. hemipterus .

Behavioural response of female Culex pipiens pallens to common host plant volatiles and synthetic blends

Yu Bao-Ting,Ding Yan-Mei,Mo Jian-Chu

Background Most mosquito species need to obtain sugar from host plants. Little is known about the chemical cues that Culex pipiens pallens use during their orientation to nectar host plants. In this study, we investigated the behavioural responses of female Cx. pipiens pallens to common floral scent compounds and their blends. Methods Behavioural responses of female Cx. pipiens pallens to 18 individual compounds at different concentrations were determined in the olfactometer bioassays. A synthetic blend composed of behaviourally active compounds was formulated, and its attractiveness to mosquitoes was tested. Several most attractive compounds constituted a reduced blend, and its attractiveness was tested against the solvent and the full blend, respectively. Mosquito response in the olfactometer was analyzed by comparing the percentages of mosquitoes caught in the two arms by χ2 test (observed versus expected). Results Fifteen of the 18 compounds were attractive to female Cx. pipiens pallens in the dose-dependent bioassays, with the exception of β-pinene, acetophenone and nonanal. (68.00 ± 2.49) % mosquitoes responded to the full blend composed of these 15 compounds on their optimal doses when tested against the solvent, with the preference index at 46.11 ± 3.57. Six individual compounds whose preference indices were over 40 constituted the reduced blend, and it attracted (68.00 ± 1.33) % mosquitoes when tested against the solvent while its preference index was 42.00 ± 3.54. When tested against the full blend simultaneously in the olfactometer, the reduced blend could attract (45.00 ± 2.69) % of released mosquitoes, which was as attractive as the full blend. Conclusion sOur results demonstrate that female Cx. pipiens pallens is differentially attracted by a variety of compounds at different concentrations. Alteration of the concentration strongly affects the attractiveness of the synthetic blend. Several floral scent volatiles might be the universal olfactory cues for various mosquito species to locate their nectar host plants, which could be potentially used in trapping devices for surveillance and control of them.

Antennal and behavioral responses of female Maruca vitrata to the floral volatiles of Vigna unguiculata and Lablab purpureus

Pan Wang,Na Zhang,Li-Lin Zhou,Sheng-Yun Si,Chao-Liang Lei,Hui Ai,Xiao-Ping Wang

In many herbivorous insects, the attraction to suitable host plants is in part mediated by similar olfactory cues from different host plants. This study investigated the electrophysiological and behavioral responses of female Maruca vitrata Fabricius (Lepidoptera: Crambidae) to floral volatiles from two host plants, Vigna unguiculata (L.) Walpers and Lablab purpureus (L.) Sweet (both Fabaceae). Qualitative and quantitative differences were found between the odor profiles of two host plants by gas chromatography-mass spectrometry (GC-MS). Nine electroantennography-active compounds were detected in the headspace collections of the two host plants by means of coupled GC-electroantennographic detection. In a wind tunnel, female behaviors (upwind flight and source approach) were not affected by stimuli or their interaction with host plants, but the behaviors were influenced by host plants. A mixture of the nine bioactive compounds, including limonene, 1,3-diethylbenzene, benzaldehyde, acetophenone, 4-ethylbenzaldehyde, 1-(4-ethylphenyl)-ethanone, 2-methyl-3-phenylpropanal, 1H-indol-4-ol, and 1,1′-(1,4-phenylene)bis-ethanone mimicking L. purpureus (in a ratio of 0.4:9.7:2.4:5.7:78.1:4.8:100:0.6:30.3) attracted significantly more females approaching the sources compared with the mixture of compounds mimicking V. unguiculata (in a ratio of 0.3:1.4:1.6:3.7:50.8:3.2:100:4.2:16.7). Further bioassays revealed that a subtractive 4-component blend [limonene, 1,3-diethylbenzene, 4-ethylbenzaldehyde, and 1-(4-ethylphenyl)-ethanone in a ratio of 0.4:9.7:78.1:4.8] was essential for maximal attraction. Our study suggested that female M. vitrata might use similar odors from V. unguiculata and L. purpureus to locate suitable hosts and that an artificial lure, containing the major floral volatiles released by the two host plants, might be useful in exploring efficiency monitoring and/or control strategies of this moth in the field.

Volatiles from Cotton Plants Infested by Agrotis segetum (Lep.: Noctuidae) Attract the Larval Parasitoid Microplitis mediator (Hym.: Braconidae)

Mengyu Li,Shike Xia,Tao Zhang,Livy Williams III,Haijun Xiao,Yanhui Lu

Herbivore-induced plant volatiles (HIPVs), chemicals produced by plants infested by herbivorous insects, can act as kairomones that recruit natural enemies of the pest herbivore.Agrotis segetum(Denis and Schiffermüller) is a common, important pest of seedling cotton in Xinjiang Province, China, and the braconidMicroplitis mediator(Haliday) is an important mortality factor of this pest’s larvae. In olfactometer tests, which included healthy foliage, infested foliage, or infested roots,M. mediatorpreferredA. segetum-infested cotton plants to healthy cotton plants. In GC-MS analyses of plant-emitted volatiles, we found that compounds emitted increased 14.9- and 13.3- fold after leaf infestation and root infestation, respectively, compared to healthy control plants. The volatiles were mainly p-xylene, nonanal, tetradecane, decanal, benzaldehyde, β-caryophyllene, and humulene, while linalool was only present in the leaf-infestation treatment. In addition, principal component analysis indicated that all 18 compounds were associated with the infested plants, especially β-caryophyllene, p-xylene, and decanal. Based on the above studies and previous functional evaluations of the volatile compounds, it can be demonstrated that these compounds play a crucial role in modulating the interactions betweenA. segetumandM. mediatorand regulating parasitoid behavior. It may be possible to enhance the biological control ofA. segetumbyM. mediatorthrough the application of HIPVs.Keywords:turnip moth;parasitoid wasp;herbivore-induced plant volatiles;behavioral response;biological control

Orexin-A attenuated motion sickness through modulating neural activity in hypothalamus nuclei

Leilei Pan,Shuifeng Xiao,Zichao Xu,Wenping Li,Long Zhao,Ling Zhang,Ruirui Qi,Junqin Wang,Yiling Cai

Background and Purpose We evaluated the hypothesis that central orexin application could counteract motion sickness responses through regulating neural activity in target brain areas. Experimental Approach Thec effects of intracerebroventricular (i.c.v.) injection of orexin-A and SB-334867 (OX 1 antagonist) on motion sickness-induced anorexia, nausea-like behaviour (conditioned gaping), hypoactivity and hypothermia were investigated in rats subjected to Ferris wheel-like rotation. Orexin-A responsive brain areas were identified using Fos immunolabelling and were verified via motion sickness responses after intranucleus injection of orexin-A, SB-334867 and TCS-OX2-29 (OX 2 antagonist). The efficacy of intranasal application of orexin-A versus scopolamine on motion sickness symptoms in cats was also investigated. Key Results Orexin-A (i.c.v.) dose-dependently attenuated motion sickness-related behavioural responses and hypothermia. Fos expression was inhibited in the ventral part of the dorsomedial hypothalamus (DMV) and the paraventricular nucleus (PVN), but was enhanced in the ventral part of the premammillary nucleus ventral part (PMV) by orexin-A (20 μg) in rotated animals. Motion sickness responses were differentially inhibited by orexin-A injection into the DMV (anorexia and hypoactivity), the PVN (conditioned gaping) and the PMV (hypothermia). SB-334867 and TCS-OX2-29 (i.c.v. and intranucleus injection) inhibited behavioural and thermal effects of orexin-A. Orexin-A (60 μg·kg −1 ) and scopolamine inhibited rotation-induced emesis and non-retching/vomiting symptoms, while orexin-A also attenuated anorexia with mild salivation in motion sickness cats. Conclusion and Implications Orexin-A might relieve motion sickness through acting on OX 1 and OX 2 receptors in various hypothalamus nuclei. Intranasal orexin-A could be a potential strategy against motion sickness.
