Kiwifruit (Actinidia chinensis) is an important commercial crop in China, and the occurrence of diseases may cause significant economic loss in its production. In the present study, a new pathogen that causes brown leaf spot disease on kiwifruit was reported. The fungus was isolated from an infected sample and identified asFusarium graminearumbased on morphological and molecular evaluation. Koch’s postulates were confirmed when the pathogen was re-isolated from plants with artificially induced symptoms and identified asF. graminearum. Based on the biological characteristics of the pathogen, it was determined that: its optimal growth temperature was 25 °C; optimal pH was 7; most suitable carbon source was soluble starch; most suitable nitrogen source was yeast powder; and best photoperiod was 12 h light/12 h dark. Further investigations were conducted by determining 50% effective concentrations (EC50) of several active ingredients of biological fungicides againstF. graminearum. The results showed that among the studied fungicides, tetramycin and honokiol had the highest antifungal activity against this pathogen. Our findings provide a scientific basis for the prevention and treatment of brown leaf spot disease on kiwifruit. Keywords:kiwifruit;brown leaf spot disease;Fusarium graminearum;biological characteristics;active ingredients of biological fungicides

Background Protein extraction from plant leaves is a key step in plant proteomics analysis. However, traditional methods often face challenges, including low efficiency, incomplete protein coverage, and environmental concerns. These limitations highlight the need for developing more efficient and environmentally friendly extraction techniques. Ionic liquids (ILs) have shown promise in protein extraction due to their excellent solubility and enzyme compatibility, offering a potential solution to these issues. In this study, we explore the use of short and long alkyl chain ILs to improve protein extraction efficiency from plant leaves. Results Using tobacco K326 as a model plant, a stepwise extraction system was designed with 10% 1-ethyl-3-methylimidazolium chloride ([C 2 MIM]Cl) and 4% 1-dodecyl-3-methylimidazolium chloride ([C 12 MIM]Cl). Both [C 2 MIM]Cl and [C 12 MIM]Cl effectively disrupted the cellulose hydrogen-bond network, facilitating protein release from the plant cell wall, while [C 12 MIM]Cl also enhanced membrane protein solubility, significantly improving the overall protein extraction efficiency. The relative protein concentration achieved with this method was 2.28, which is notably higher than traditional methods using urea (1.21) and SDS (1.47). The method was applied to proteomic analysis of healthy and brown spot disease-affected tobacco leaves, identifying 6858 proteins in total. Among these, 758 proteins were differentially expressed, with 414 upregulated and 344 downregulated in diseased leaves, providing insight into disease-related protein expression. Significance This novel protein extraction method offers a highly efficient alternative to traditional approaches. It significantly enhances protein yield and solubility, providing valuable support for in-depth plant proteomics research. This method is particularly useful for studying plant disease responses, facilitating the identification of disease-related proteins and contributing to the understanding of plant-pathogen interactions.

Brown leaf spot disease caused by Nigrospora guilinensis on Phellodendron chinense occurs in a large area in Dayi County, Chengdu City, Sichuan Province, China each year. This outbreak has severely reduced the production of Chinese medicinal plants P. chinense and caused substantial economic losses. The bacterial isolate JKB05 was isolated from the healthy leaves of P. chinense , exhibited antagonistic effects against N. guilinensis and was identified as Bacillus megaterium . The following fermentation medium and conditions improved the inhibitory effect of B. megaterium JKB05 on N. guilinensis : 2% glucose, 0.1% soybean powder, 0.1% KCl, and 0.05% MgSO 4 ; initial concentration 6 × 10 6 cfu/ml, and a 42-h optimal fermentation time. A composite of 0.1% nano-SiO 2 JKB05 improved the thermal stability, acid-base stability and ultraviolet resistance by 16%, 12%, and 38.9%, respectively, and nano-SiO 2 was added to the fermentation process. The best formula for the wettable powder was 35% kaolin, 4% polyethylene glycol, 8% Tween, and 2% humic acid. The following quality test results for the wettable powder were obtained: wetting time 87.0 s, suspension rate 80.33%, frequency of microbial contamination 0.08%, pH 7.2, fineness 95.8%, drying loss 1.47%, and storage stability ≥83.5%. A pot experiment revealed that the ability of JKB05 to prevent fungal infections on P. chinense increased considerably and achieved levels of control as high as 94%. The use of nanomaterials significantly improved the ability of biocontrol bacteria to control this disease.
