Metabolomic analysis based on EESI-MS indicate blue LED light promotes aliphatic-glucosinolates biosynthesis in broccoli sprouts

Ahui Xue,Wenjing Liang,Shide Wen,Yuanyuan Gao,Xueyong Huang,Yuzhen Tong,Yingbin Hao,Liping Luo

Broccoli sprouts are rich in secondary metabolites, especially glucosinolates (GLs). The synthesis of GLs can be modified by light. In this study, extractive electrospray ionization-mass spectrometry (EESI-MS) was used to analyze the broccoli sprouts extracts (BSE) under different light quality treatments. Multivariable statistical methods were used to analyze the difference of different light quality treatments. Quantitative real-time polymerase chain reaction (qRT-PCR) was used to analyze the gene expression of key enzymes in the aliphatic-GLs biosynthesis pathway. Nineteen compounds, including 7 GLs, 3 phenolic acids, 6 amino acids and 3 organic acids were simultaneously detected in one minute by EESI-MS. Relative expression of MYB28 , BCAT4 , CYP79F1 , CYP83A1 and AOP3 were mostly upregulated in R5B5 (light ratios of red/blue 5:5) treatment. These findings suggest that blue light improving the accumulation of secondary metabolites in broccoli sprouts, and the R5B5 treatment was most beneficial. Furthermore, we established EESI-MS method to simultaneous and rapid detection of secondary metabolites in broccoli sprouts, especially GLs. This study could provide a reference for selecting proper LED lighting conditions to produce broccoli sprouts with high content of bioactive components, and provides light regulation technology and theoretical basis for the industrialized production of sprouts.

Effects of LED Light Quality on Broccoli Microgreens Plant Growth and Nutrient Accumulation

Luo Liping,Zhang Gaowei,Liang Wenjing,Wu Dong,Sun Qifang,Hao Yingbin

Broccoli microgreens represent a novel type of vegetable characterized by high levels of bioactive compounds. Here, we investigated the effects of light-emitting diode (LED) light quality on the development and nutritional uptake of broccoli microgreens. The plants were exposed to five distinct light treatments: white, single red, and red: blue at 5:1, 5:3, and 5:5 ratios. The soluble sugars, nutrients, and secondary metabolites of broccoli sprout extracts were examined using high-performance liquid chromatography and ultra-high-performance liquid chromatography quadrupole time-of-flight mass spectrometry under various light-quality conditions. In addition, the expression levels of genes encoding important enzymes in the glucosinolates (GLs) biosynthesis pathway were examined using quantitative real-time polymerase chain reaction. Overall, the findings demonstrated that mixed red-blue light, especially the 5:1 red: blue ratio, was beneficial for broccoli microgreens in terms of their hypocotyl length, fresh weight, edible rate, soluble proteins, vitamin C, total phenolics, total flavonoids, and GLs. Our findings provide a foundation and reference point for selecting the most appropriate LED light ratio in microgreens production.

Identification of volatile and flavor metabolites in three varieties of broccoli sprouts

Yu Xia,Ming-Yue Li,Syed Abdul Wadood,Han-Jun Hong,Yi Liu,Yu-Xuan Luo,Yi-Yan Wang,Hong-Yan Liu,Ren-You Gan

Broccoli sprouts are promising functional food sources and their taste and flavor play a pivotal role in the acceptance of consumers. In this study, the flavor profiles of three varieties of broccoli sprouts, namely Bi Lv, You Xiu, and Lv Hua, were comprehensively characterized using HS-SPME-GC/MS analysis. A total of 364 volatile and flavor components across 15 chemical classes were successfully identified. The results revealed a majority of volatile metabolites exhibiting upregulation during the germination process, leading to an enhancement in taste intensity after germination, particularly for umami and sweet tastes, which was associated with an increase in associated amino acids and sugar content. Although the total glucosinolate content in broccoli sprouts has decreased compared to seeds, it remains the primary contributor to the bitterness of broccoli sprouts. The present study elaborated on the flavor contribution of broccoli sprouts, supporting the cultivation and consumption of them as a nutritious food.

Extraction of weak hydrophobic sulforaphane from broccoli by salting-out assisted hydrophobic deep eutectic solvent extraction

Wen-wen Deng,Xue-ping Mei,Zi-jun Cheng,Tian-xiang gan,Xuan Tian,Jiang-nan Hu,Chuan-ru Zang,Bo Sun,Jing Wu,Yin Deng,Reza. A. Ghiladi,George H. Lorimer,Gizem Keceli,Jun Wang

In order to extract sulforaphane (SFN) from broccoli via green and efficient ways, a novel method based on salting-out assisted deep eutectic solvent (DES) has been developed. Compared to known organic solvent- (such as dichloromethane, ethyl acetate, n -hexane, etc.) based liquid–liquid extraction, this new N 8881 Cl-based DES method exhibited excellent extraction efficiency for SFN, including a significant improvement due to the salting-out effect of KH 2 PO 4 . Under optimal conditions, 97.77 % of SFN was extracted by N 8881 Cl-EG DES and more than 82.5 % of SFN was recovered by activated carbon from DES. In addition, further studies with Kamlet-Taft parameters and density functional theory showed that the H-bond accepting capacity of hydrophobic DES, the existing vdW interaction, and the electrostatic interaction between N 8881 Cl-EG DES all contributed to efficient extraction of SFN. This is the first time that the underlying mechanism for SFN extraction by DES was revealed.

Antioxidant Capacity Changes and Untargeted Metabolite Profile of Broccoli during Lactic Acid Bacteria Fermentation

Feixiang Hou,Yanxue Cai,Jihui Wang

The purpose of this study was to reveal the changes in total phenolic content and antioxidant capacity of broccoli, and an untargeted metabolomics approach was developed to investigate the effect of lactic acid bacteria fermentation on the metabolome of broccoli florets. The results showed that the total phenolic content and antioxidant capacity significantly increased after fermentation. The untargeted metabolite profile showed that the main chemical components of fermented and unfermented broccoli are lipids and lipid-like molecules, organic acids and derivatives and organoheterocyclic compounds. Univariate and multivariate statistical analyses of the identified metabolites showed some metabolites such as sorbitol are upregulated after fermentation, and that other metabolites such as l-malic acid are downregulated after fermentation. Moreover, metabolite pathway analyses were used to study the identification of subtle but significant changes among groups of related metabolites that cannot be observed with conventional approaches. KEGG pathway analysis showed that metabolites are mainly enriched in the glucagon signaling pathway, pyruvate metabolism, glycolysis/gluconeogenesis and fructose and mannose metabolism after fermentation, compared with raw broccoli. The results of this study can help to further our understanding of the impact of LAB fermentation on bioactivity changes in and the metabolites profile of fermented broccoli, and the application of fermented broccoli in health foods and special dietary foods. Keywords:fermented broccoli;lactic acid bacteria;antioxidant capacity;untargeted metabolite profile

Insight into the Sulforaphane Content and Glucosinolate Profile of Broccoli Stems After Heat Treatment

Yu-Hong Zhang,Xue-Yi Liao,Zheng-Hong Li,Yu-Feng Guo,Ming-Xin Ma,Guo-Qiang Zhang

(1) Background: At the time of harvest, the stems of broccoli are frequently discarded. (2) Methods: In this study, the sulforaphane content and glucosinolate profile of broccoli stems were analyzed at different temperature treatments. (3) Results: Thermal treatment of broccoli stems for 1 h resulted in maximal sulforaphane content at 50 °C, with a subsequent progressive reduction in concentration correlating to elevated temperatures. Metabolomic analysis was conducted on broccoli stem samples subjected to 25 °C (CK), 50 °C, and 80 °C treatments. Among the 25 identified GSLs, the 50 °C-treated samples demonstrated significantly reduced GSL accumulation, whereas the 80 °C group exhibited maximal GSL retention. Indole derivatives predominated among the three GSL subclasses (aliphatic, aromatic, and indole), accounting for approximately 70% of total GSLs across all groups. The observed GSL depletion at 50 °C correlated with enhanced sulforaphane biosynthesis. Comparative analysis further indicated that 80 °C treatment induced a more pronounced elevation of indole GSLs compared to aliphatic and aromatic counterparts in broccoli stems. (4) Conclusions: The results demonstrated that indole GSLs in broccoli stems exhibit superior thermal stability. Moderate thermal treatments effectively enhance sulforaphane content, whereas exposure to 80 °C significantly increases total GSL content.

Widely targeted metabolomic analysis and antioxidant properties of seeds and sprouts from different broccoli cultivars

Ming-Yue Li,Yi Liu,Kin Weng Kong,Fang Geng,Hong-Yan Liu,Ren-You Gan

Broccoli seeds and sprouts are popular food crops, but the nutritional values of different broccoli cultivars lack intensive research. In this study, three cultivars of broccoli seeds and their sprouts, including Lv Hua (LH), You Xiu (YX), and Bi Lv (BL), were selected to investigate the metabolites, antioxidant properties, total phenolic content (TPC), and total flavonoid content (TFC). Totally 10 categories and 1015 metabolites were identified. Comparing the sprouts of LH, YX, and BL with their respective seeds, 612, 621, and 602 differential metabolites were detected, of which 404 metabolites were shared among the three cultivars, and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis showed that the linoleic acid metabolism pathway was significant in three cultivars. Both the total and soluble fraction of LH had the highest values of antioxidant properties, TPC, and TFC. Overall, this study provided scientific support for the cultivar selection of broccoli germination and the development of their related products.

Hydrolysis before Stir-Frying Increases the Isothiocyanate Content of Broccoli

Yuanfeng Wu,Yuke Shen,Xuping Wu,Ye Zhu,Jothame Mupunga,Wenna Bao,Jun Huang,Jianwei Mao,Shiwang Liu,Yuru You

Broccoli is found to be a good source of glucosinolates, which can be hydrolyzed by endogenous myrosinase to obtain chemopreventive isothiocyanates (ITCs); among them, sulforaphane (SF) is the most important agent. Studies have shown that cooking greatly affects the levels of SF and total ITCs in broccoli. However, the stability of these compounds during cooking has been infrequently examined. In this study, we proved that the half-lives of SF and total ITCs during stir-frying were 7.7 and 5.9 min, respectively, while the myrosinase activity decreased by 80% after stir-frying for 3 min; SF and total ITCs were more stable than myrosinase. Thus, the contents of SF and total ITCs decreased during stir-frying largely because myrosinase was destroyed. Subsequently, it was confirmed that compared to direct stir-frying, hydrolysis of glucosinolates in broccoli for 90 min followed by stir-frying increased the SF and total ITC concentration by 2.8 and 2.6 times, respectively. This method provides large quantities of beneficial ITCs even after cooking.

Effect of Methyl Jasmonate on Phenolic Accumulation in Wounded Broccoli

Yuge Guan,Wenzhong Hu,Aili Jiang,Yongping Xu,Rengaowa Sa,Ke Feng,Manru Zhao,Jiaoxue Yu,Yaru Ji,Mengyang Hou,Xiaozhe Yang

In order to find an efficient way for broccoli to increase the phenolic content, this study intended primarily to elucidate the effect of methyl jasmonate (MeJA) treatment on the phenolic accumulation in broccoli. The optimum concentration of MeJA was studied first, and 10 μM MeJA was chosen as the most effective concentration to improve the phenolic content in wounded broccoli. Furthermore, in order to elucidate the effect of methyl jasmonate (MeJA) treatment on phenolic biosynthesis in broccoli, the key enzyme activities of phenylpropanoid metabolism, the total phenolic content (TPC), individual phenolic compounds (PC), antioxidant activity (AOX) and antioxidant metabolism-associated enzyme activities were investigated. Results show that MeJA treatment stimulated phenylalanine ammonia-lyase (PAL), cinnamate 4-hydroxylase (C4H), and 4-coumarin coenzyme A ligase (4CL) enzymes activities in phenylpropanoid metabolism, and inhibited the activity of polyphenol oxidase (PPO), and further accelerated the accumulation of the wound-induced rutin, caffeic acid, and cinnamic acid accumulation, which contributed to the result of the total phenolic content increasing by 34.8% and ferric reducing antioxidant power increasing by 154.9% in broccoli. These results demonstrate that MeJA in combination with wounding stress can induce phenylpropanoid metabolism for the wound-induced phenolic accumulation in broccoli. Keywords:wounded broccoli;methyl jasmonate;phenolic accumulation;phenylpropanoid metabolism;antioxidant capacity

Green synthesis of broccoli-derived carbon quantum dots as effective photosensitizers for the PDT effect testified in the model of mutant Caenorhabditis elegans

Shereen M. Elsherbiny,Changsheng Shao,Adolf Acheampong,Mahmoud A. Khalifa,Chao Liu,Qing Huang

The clinical application of photodynamic therapy (PDT) is still limited because of the drawbacks of the traditional photosensitizers, such as low singlet oxygen (1O2) quantum yield and the problem of photobleaching. Herein, carbon quantum dots (CQDs) derived from broccoli natural biomass as a carbon source were fabricated via a simple hydrothermal method and showed outstanding PDT ability as an effective photodynamic agent tested in Caenorhabditis elegans (C. elegans) models. The as-prepared broccoli-derived CQDs (BCQDs) showed excellent water solubility and optical properties and could generate singlet oxygen (1O2) effectively under irradiated light with a wavelength of 660 nm. The in vivo experiment revealed that the PDT efficiency of the BCQDs was dependent on the induction of germline apoptosis through the cep-1/p53 pathway. Further investigation confirmed the DNA damage of the worm by the BCQDs after sufficient light irradiation, which was tested by measuring the egl-1-fold induction in hus-1(op244), and cep-1(w40) mutants that have a loss of function in the genes involved in DNA damage response such as hus-1 (DNA checkpoint gene) and cep-1/p53 (tumor suppressor). The lack of germline apoptosis in the loss of function mutants egl-1(n487), hus-1(op244), and cep-1(w40) exposed to light irradiation compared with the control proved the necessity of these genes in DNA damage-induced germline apoptosis. Therefore, this work has not only provided a new photodynamic agent but also introduced C. elegans as an easy and high-throughput model for the rapid evaluation of the efficiency of PDT.

Broccoli ingestion increases the glucosinolate hydrolysis activity of microbiota in the mouse gut

Yuanfeng Wu,Yuke Shen,Ye Zhu,Jothame Mupunga,Ligen Zou,Chao Liu,Shiwang Liu,Jianwei Mao

After consuming broccoli, isothiocyanates can be produced by the hydrolytic action of myrosinase from plant and/or microbiota. Using male C57BL/6 mice, the present study investigated the effects of broccoli ingestion on the myrosinase-like activity, NAD(P)H:quinone oxidoreductase 1 (NQO1) activity, diversity and composition of the gut microbiota. Compared with the control group, continuously feeding raw or hydrolysed broccoli increased the myrosinase-like activities of the colon and caecum contents, and also improved the NQO1 activity of the colon mucosa. Significant difference between the broccoli and control feeding groups were found. 16S rRNA gene sequencing indicated that broccoli ingestion profoundly affected the composition of the gut bacteria community. The correlation between the gut bacterial community composition and microbiota myrosinase-like activity was also studied. However, one type of glucosinolate, sinigrin, had no effect on these activities, indicating that broccoli component(s), other than glucosinolate, had increased the NQO1 and myrosinase-like activities.

Structural characterization of a broccoli polysaccharide and evaluation of anti-cancer cell proliferation effects

Lishan Xu,Jingjing Cao,Wenrong Chen

Broccoli is a widely consumed vegetable with abundant amount of nutrients, which bring numerous beneficial effects on human health. The structural information of water-soluble polysaccharides in broccoli was eludicated for the first time in this work. A purified polysaccharide fraction (BPCa) was obtained by column chromatography. It comprised of arabinose (Ara), galactose (Gal), rhamnose (Rha) with a molar ratio of 5.3:0.8:1.0. Nuclear magnetic resonnance spectra data revealed that α- l -1,5-Ara f and α- l -1,3,5-Ara f are present in the backbone, while α- l -Ara f terminal was attended in side chain. α- l -1,2-Rha p was found to be linked to α- l -1,5-Ara f in heteronuclear multiple bond correlation spectra. The presences of β- d -1,4-Gal p and α- d -1,4-Gal p A were also detected. Furthermore, BPCa showed significant anti-cancer cell proliferation activities against HepG2, Siha and MDA-MB-231 carcinoma cell lines. The results indicated that BPCa had a good potential to be applied as functional food additives.
