Ratiometric fluorescent test strips based on CB-Ni2+@CDs probes for visual detection of histamine

Xiuying Liu,Si Kang,Wen Wang,Lijie Zhu,Wei Zhang,Pingping Wang,Zaixi Shu,Yiwei Tang

Histamine is a biogenic amine with various physiological functions. However, excessive consumption of histamine can lead to various symptoms, and pose a threat to human lives. A ratiometric fluorescent test strip for visual detection of histamine was developed based on CB-Ni 2+ @CDs probes. As the concentration of histamine increases, the test strips exhibit a transition in fluorescence signal from yellow-green to blue. The RGB values were extracted from the images, and used for quantitative analysis of histamine. The method had a linear range of 0–1.0 mM, with a detection limit of 0.086 mM. The test strips were employed for the detection of histamine, and the recovery rate was found to be in the range of 88.3% to 104.69%, indicating a high level of accuracy. The uniqueness of the test strips lies in their ability to be produced simply by mixing CB, Ni 2+ on a suitable polyvinyl alcohol/wood cellulose fiber substrate.

Gold nanoclusters immobilized paper for visual detection of zinc in whole blood and cells by coupling hydride generation with headspace solid phase extraction

Ke Huang,Rui Dai,Wenqing Deng,Shengjie Guo,Han Deng,Yao Wei,Fuling Zhou,Ying Long,Jun Li,Xin Yuan,Xiaoli Xiong

A novel visual detection method was developed for the determination of zinc (Zn) in whole blood and cells using gold nanoclusters (AuNCs) immobilized paper. The detection of Zn was based on a headspace-solid phase extraction (HS-SPE), visible fluorescence from the AuNCs coated paper was quenched by ZnH 2 generated from hydride generation (HG). The potential mechanism was investigated by using X-ray photoelectron spectroscopy (XPS) and Density Functional Theory (DFT). The selectivity was significantly increased because the zinc hydride was effectively separated from sample matrices by hydride generation. A limit of detection (LOD) of 3   μg   L −1 Zn 2+ and a relative standard deviation (RSD, n   =   7) of 2% at a concentration of 50   μg   L −1 of Zn 2+ were obtained when using a commercial fluorescence spectrophotometer as the detector. In visual detection, as low as 20   μg   L −1 of Zn 2+ in biological samples can be easily discriminated from the blank with the naked eye. The proposed method retains several unique advantages, including straightforward, rapid, affordable and equipment-free for visual detection.

Highly selective and sensitive determination of copper ion based on a visual fluorescence method

Linfeng Chen,Xike Tian,Chao Yang,Yong Li,Zhaoxin Zhou,Yanxin Wang,Fang Xiang

A novel ratiometric fluorescence sensor for rapid and on-site visual detection of Cu 2+ was designed and synthesized by integrating yellow-emission rhodamine fluorophore (RL) and red-emission CdTe@SiO 2 QDs. The as-prepared nanohybrid fluorescence sensor shows dual-emissions at 537   nm and 654   nm under a single excitation at 500   nm in the presence of Cu 2+ . Owing to the strong chelating ability of RL toward Cu 2+ , the yellow fluorescence of RL could be selectively enhanced while the red fluorescence of CdTe QDs is almost unchanged, leading to an obvious fluorescence color change from red to yellow, which could be used for visual and ratiometric detection of Cu 2+ . This nanohybrid sensor exhibits excellent selectivity, sensitivity and anti-interference to Cu 2+ detection and the detection limit is as low as 8.4   nM. Additionally, a simple device test strip for rapid and on-site detection of Cu 2+ has been designed by immobilizing the RL-CdTe@SiO 2 QDs on a common filter paper. This simple and effective paper-based sensor has a visual detection limit of 0.5   μM, showing its promising application for on-site and rapid sensing of Cu 2+ in pollution water.

Highly sensitive visual detection of catalase based on the accelerating decomposition of H2O2 using Au nanorods as a sensor

Simin Lu,Ling Chen,Ping Yang,Katarzyna Matras-Postolek

The detection of catalase associated with various diseases is very important due to the fact that it is frequently downregulated in human cancer tissues. Noble metal nanomaterial-assisted sensors have achieved rapid development for wide applications but few for detecting catalase. Thus, a novel colourimetric strategy has been developed for the selective and rapid visual detection of catalase by using the decelerating etching of gold nanorods (NRs). In the presence of hexadecyltrimethylammonium bromide and SCN−, which can reduce the redox potential of Au(I)/Au (0), Au NRs would be etched by H2O2 along the longitudinal direction. As a consequence, the colour of solutions changed from bluish green to pink accompanied by the localized surface plasmon resonance (LSPR) extinction peak being shifted to short wavelength. In contrast, the etching process would be prohibited in the presence of catalase, and the colour and LSPR extinction peak of Au NRs would remain unchanged. Furthermore, this Au NR based sensor is applied to test paper that could be used in the detection of catalase in real samples. The discussion of the mechanism indicates that such a sensing method has an ideal performance in terms of sensitivity, selectivity, and linearity.

An ink-jet printed dual-CD ratiometric fluorescent paper-based sensor for the visual detection of Cu2+

Ying Li,Fei Lu,Qing-zhi Li,Yi-hua Zhou,Jun Qian,Sheng Cao,Chen-yu Wang

Copper ion (Cu2+) plays an important role in the human body because it is beneficial for metabolism. However, an excessive or slight amount of Cu2+ can cause various symptoms. Therefore, it is necessary for human health to realize the trace and visual detection of Cu2+. Referring to traditional fluorescence test papers, the qualitative and semi-quantitative detection of Cu2+ could be realized by a dual-carbon dots (CDs) ratiometric fluorescent paper-based sensor with the advantages of environmental protection, portability and low cost. In this paper, the inkjet-printed test paper with the best mixing ratio of the two CDs has been researched to maximize the spectral energy transfer of ion detection (maximum color gamut expansion). Among them, the preparation method of b-CDs has been improved, increasing the photoluminescence quantum yield (PLQY) to 88.9%. The sensitivity detection limit of the double emission ratio sensor was 0.15 nM in solution, and the human eye can distinguish at least 3 μmol L−1 Cu2+ in the paper-based sensor. Compared with the traditional single-emission sensor, the human eye was more sensitive to the color change of the emission ratio sensor. The results indicate that we not only realized the micro detection of Cu2+ with convenient methods, but also provided a promising strategy for the visual detection of Cu2+.

Visual determination of Cu2+ through copper-catalysed in situ formation of Ag nanoparticles

Xun Yuan,Yi Chen

A new strategy was explored for the visual determination of Cu2+ using copper-catalysed in situ formation of Ag nanoparticles. In this method, only common reagents were used and the pre-synthesis and modification of nanoparticles are avoided. Ag+ can form a milk-white suspension (AgBr) with Br− in an aqueous solution composed of AgNO3, cetyltrimethylammonium bromide, ascorbic acid, bovine serum albumin, and NaNO3. The reaction will be stopped by addition of Cu2+, accompanied by a colour change from milk-white to orange or brilliant yellow. Cu+ (the reduction product of Cu2+) consumes the dissolved O2 and prevents the O2 from oxidizing the newly reduced Ag atoms (by ascorbic acid) back to Ag+, facilitating the further aggregation of Ag atoms to become Ag nanoparticles. The visible colour change was shown to be specific towards Cu2+ over most other metal ions. The limit of detection was 0.75 μM Cu2+ by the naked eye and 0.25 μM by spectrometer. Quantitation of Cu2+ was achieved in a linear range from 0.25 to 2.0 μM. This method was validated by measuring real water and serum samples, giving results agreeing well with the data reported and measured by inductively coupled plasma mass spectrometry. The recovery was 95.6–106% for untreated tap water and 96.0–100% for resin-pre-treated water and serum samples.

Double functional polymer brush-grafted cotton fiber for the fast visual detection and efficient adsorption of cadmium ions

Jizhen Jia,Changkun Liu,Lin Wang,Xiaoyan Liang,Xiaoyan Chai

In this study, a multi-functional cotton fiber was prepared and investigated for the visual detection and enhanced adsorption of cadmium (Cd) ions in aqueous solutions. SI-ATRP technology was used to graft poly (glycidyl methacrylate) (PGMA) and poly (3-sulfopropyl methacrylate potassium salt) (PSMP) successively on the surface of the cotton fiber. Then, diethylenetriamine (DETA) and 5,10,15,20-tetrakis(1-methy-4-pyridinio)porphyrin tetra(p-toluenesulfonate) (TMPyP) were immobilized onto the PGMA and PSMP polymer brush, respectively, to form the Cotton-PGMA(DETA)-PSMP(TMPyP) (abbreviated as C-PD-PT) material. C-PD-PT showed enhanced Cd ion adsorption with the maximum adsorption capacity of 1.6215 mmol/g derived from the Langmuir model fitting. In addition, The C-PD-PT material exhibited simple visual detection of Cd ions with a remarkable color change from brownish to green. A linear relationship between the light absorbance of C-PD-PT-Cd (C-PD-PT adsorbed with Cd ions) and the initial concentrations of Cd ions was established, which would be used for the fast determination of Cd ion concentrations in aqueous solutions.

A lateral flow assay for copper(II) utilizing catalytic and stem-loop based signal amplification

Wang Yulong,Wang Limin,Zhang Cunzheng,Liu Fengquan

A DNAzyme-based catalytic and stem-loop based amplification scheme is used in a Cu(II)-specific lateral flow assay (LFA). Three test lines with given cut-off value on the test strip are set as the signal indicating zone for semiquantitative analysis by the number of red color bands that appear after lateral flow. The colored bands are generated by accumulation of gold nanoparticles. Four detection ranges can be visualied: (a) 0–2 ng·mL −1 (= negative); 2–50 ng·mL −1 ; 50–200 ng·mL −1 and > 200 ng·mL −1 of Cu(II) (= positive). The visual detection limit is thus considered as being 2 ng·mL −1 which is much lower than the U.S. EPA limit in drinking water (1.25 μg·mL −1 ). The highly specific DNAzyme, the strong multiple-turnover catalytic target recycling property and highly efficient amplification strategy warrant the high specificity, sensitivity and rapidity of this LFA. Conceivbly, this detecton scheme can be extended to other metal ions by proper choice of the ion-specific DNAzyme. Schematic presentation of a semiquantitative lateral flow test strip for Cu 2+ analysis by three visual cut-off value test lines using catalytic and stem-loop based signal amplification strategy.

A visual assay and spectrophotometric determination of LLM-105 explosive using detection of gold nanoparticle aggregation at two pH values

He Yi,Cheng Yang

We report a simple, rapid, and sensitive assay for visual and spectrophotometric detection of the 2,6-diamino-3,5-dinitropyrazine-1-oxide (LLM-105) explosive. The assay is based on different interactions between LLM-105 and gold nanoparticle (AuNP) dispersions at two pH values, leading to the formation of dispersed or aggregated AuNPs. Two AuNP dispersions at two pH values were applied to recognize and detect LLM-105 instead of traditional AuNP dispersion under an aptotic pH to improve the anti-interference ability. The developed assay showed excellent sensitivity with a detection limit of 3 ng/mL, and the presence of as low as 0.2 μg/mL LLM-105 can be directly detected with the bare eye. This sensitivity is about six orders of magnitude higher than that of the reported traditional assays. Additionally, the assay exhibited good selectivity toward LLM-105 over other explosives, sulfur-containing compounds, and amines. A simple, sensitive, and selective assay for LLM-105 was developed based on the pH-dependent interaction between the LLM-105 explosive and gold nanoparticle dispersion

Superwettable microchip for visual detection of date-rape drug in beverages

Zhenping Li,Hao Xu,Yukun Du,Zhongyu Huang,Jing Li,Lian Liu,Ying Dong,Xiao-Yu Yang

Gamma-hydroxybutyric acid (GHB), which is used in drug-facilitated sexual assault, is typically delivered in the form of gamma-butyrolactone (GBL) in beverage carriers. In response to the growing need of methods for rapid and on-site sensing of the drugs, we carried out an investigation aimed at developing a superwettable microchip for visual detection of the date-rape drug GBL in adulterated beverages. The superwettable microchip is comprised of Fe 3 O 4 nanoparticles which aggregate to form a reactive spot on a superhydrophobic fabric. The spot captures and confines microdroplets containing GBL on the superhydrophobic substrate. Fe(III), generated by HCl promoted reaction of the Fe 3 O 4 spot, then participates in a colorimetric reaction with GBL to form a naked eye observable colored complex. Quantitative determination of GBL in beverages is achieved by analyzing the red proportion of a smartphone camera derived image. The detection range of this assay protocol is ca. 0.1–10 mg mL −1 , which meets the requirements to detect potentially dangerous concentration of GBL in drinks. The detection stability of the superwettable microchip for GBL in water and common beverages is maintained over at least 30 cycles.

Gelatin-based photonic hydrogels for visual detection of pathogenic Pseudomonas aeruginosa

Yujie Gao,Yu Chen,Miaomiao Li,Lizhen Jia,Lianbin Zhang,Jintao Zhu

Identification or detection of pathogenic microbes is one of the most important aspects to prevent and/or treat bacterial infection. In this study, we report a facile yet versatile strategy for preparation of gelatin-based photonic hydrogel sensors for visual detection of Pseudomonas aeruginosa ( P. aeruginosa ). The photonic hydrogels are generated by in-situ photopolymerization of acrylamide (AM), N , N ’-methylenebis-(acrylamide) (BIS), and gelatin methacrylate (GelMA) with the self-assembled one-dimensional (1D) chain-like structures of carbon-encapsulated Fe 3 O 4 nanoparticles (Fe 3 O 4 @C NPs). In the presence of P. aeruginosa , the hydrolysis of gelatin triggered by gelatinase reduces the crosslinking density of photonic hydrogel, expands the hydrogel in volume, increases the lattice spacing of the colloidal particles, and red-shifts the reflection spectra of the photonic hydrogel. The optical appearance variation of the photonic hydrogel, resulting from the reflection peak shift, can be observed by naked eyes. Besides, we show that the photonic hydrogels exhibit negligible appearance changes with the presence of Escherichia coli ( E. coli ), indicating their potential for distinguishing various bacteria by a simple structural color variation. This finding provides a proof-of-concept for a colorimetric sensor for visual detection of P. aeruginosa and demonstrates the great potential of the photonic hydrogels in visual detection of pathogens.

Visual chiral recognition of aromatic amino acids with (S)-mandelic acid-based ionic liquids via complexation

Xiaofei Ma,Yingxiang Du,Xinqi Zhu,Jiangxia Yang

Recently, chiral ionic liquids have attracted increasing attention in analytical chemistry . However, only a few papers focus on the application of them in visual chiral recognition. Herein, two functionalized chiral ionic liquids derived from (S)-mandelic acid (1-butyl-3-methylimidazolium mandelate , CIL1 and N-butyl-N-methylpyrrolidinium mandelate, CIL2) were prepared for visual chiral recognition of aromatic amino acids for the first time. In the presence of Cu(II) and appropriate solvents, visual enantiomeric responses of phenylalanine , tryptophane, tyrosine and phenylglycine were observed. Relying on solubility or color differences, all chiral recognition could be finished within 5 min. The potential mechanism was investigated by means of infrared spectroscopy, ultraviolet spectroscopy, thermal gravity analysis, elemental analysis and scanning electron microscope. Results revealed that CuSO 4 interacted with CIL1 and D-tryptophane in the ratio of 1:1.96:0.43 in relevant precipitate, and the different stability of complex was responsible for the chiral recognition. In addition, resolution of racemic tryptophane was performed, which offered excellent enantiomeric excess values (94.2% for CIL1 and 95.1% for CIL2 in solid phase). The proposed ionic liquids had strong enantioselectivity for aromatic amino acids and great potential in visual chiral recognition.
