Experimental and theoretical study on the extraction of keratin from human hair using protic ionic liquids

Junlei Wang,Hongshuai Gao,Congwen Qin,Zhimin Zhao,Hanmeng Yuan,Jia Wei,Yi Nie

Keratin, the naturally derived biomaterials have been developed and widely applied in many different fields. Among the resources containing keratin, human hair is one of the most abundant natural fibers rich in keratin and one of the most abundant waste materials produced by humans. It is critical for both value-added human hair utilization and environmental protection if suitable solvents for the dissolution and regeneration of the keratin were developed. Ionic liquids (ILs) have been evidenced to be the green solvents to extract keratin from waste human hair. In this work, five novel Protic ILs were designed and synthesized in one step with inexpensive raw materials under mild conditions. The effect of different anions and cations, temperature, and water content on the dissolution ability of Protic ILs for human hair was investigated in depth. The best IL [MEA]HCOO with high solubilization capacity (9 h, 130 °C) for human hair was finally obtained by considering the time required for complete hair dissolution and the properties of regenerated keratin. The results of FTIR, XRD, and TGA showed that the α-helix structure of regenerated keratin was not destroyed. The recycling result indicates that the dissolution ability of [MEA]HCOO for human hair kept stable after 5 times recovery. Furthermore, the density functional theory (DFT) calculations and independent gradient model (IGM) analysis uncover the dissolution of human hair by ILs through synergistic interaction between the cations and anions of ILs.

Degradation and regeneration of feather keratin in NMMO solution

Ma Bomou,Sun Qisong,Yang Jing,Wizi Jakpa,Hou Xiuliang,Yang Yiqi

Chicken feather, a potential source of keratin, is often disposed as waste material. Although some methods, i.e., hydrolysis, reduction, and oxidation, have been developed to isolate keratin for composites, it has been limited due to the rising environmental concerns. In this work, a green solvent N -methylmorpholine N -oxide (NMMO) was used to extract keratin from chicken feather waste. Eighty-nine percent of keratin was extracted using 75% NMMO solution. However, the result from size exclusion HPLC showed that most of the keratin degraded into polypeptide with molecular weight of 2189 and only 25.3% regenerated keratin was obtained with molecular weight of 14,485. Analysis of amino acid composition showed a severe damage to the disulfide bonds in keratin during the extraction procedure. Oxidization had an important effect on the reconstitution of the disulfide bonds, which formed a stable three-dimensional net structure in the regenerated keratins. Besides, Raman spectra, NMR, FT-IR, XRD, and TGA were used to characterize the properties of regenerated keratin and raw chicken feather. In the end, a possible mechanism was proposed based on the results.

Hydration-responsive keratin fibers with ultra-high shape recovery via reversible protein conformational change and Au enhancement

Xiaoyun Xu,Zhuang Wang,Qi Zhang,Ke Zhang,Min Li,Jieqiong Yang,Yi Zhao,Qinfei Ke,Qingbao Guan,Jinlian Hu

Fabricating shape-memory materials from natural macromolecules has gained popularity to facilitate the development of bio-friendly smart materials and devices. However, current biobased shape-memory materials have shortcomings, including insufficient mechanical properties and low shape recovery rate in their real applications. Herein, this work aims to develop hierarchically structured keratin-based fibers with high strength and shape-memory properties in response to hydration. The gold nanoparticle is adopted for enhancement in the keratin system, and a well-aligned anisotropic structure with α-helix to the fiber axis is established in the regenerated keratin fibers by wet spinning. The strong crosslinking effect from Au S bonding endows the keratin fibers with outstanding shape-memory performance, exhibiting a shape fixity of ~92 % and a near-full shape recovery rate of ~96 %. In this system, the transition between α and β conformation under external force and water stimulation is assigned as the fundamental mechanism in response to hydration. In hydration and dehydration, the hydrogen bonds act as ‘switches’ that enable deformation, while the strong Au S bonds are ‘net points’ to maintain the original and temporary shapes. This strategy can promote metal nanomaterial anchoring as a methodology for fabricating shape-memory protein-based fibers with excellent mechanical properties and shape-memory performance.

DOX-Conjugated keratin nanoparticles for pH-Sensitive drug delivery

Pengcheng Liu,Qiong Wu,Yanmei Li,Pengfei Li,Jiang Yuan,Xianwei Meng,Yinghong Xiao

Keratin is a good candidate for drug carrier due to its good biocompatibility, low immunogenicity, redox responsiveness, and abundant renewable sources. Herein, doxorubicin (DOX) was first conjugated with keratin through a pH-sensitive hydrazone linkage, and then prepared into particulate drug carrier via desolvation method. The size, morphology, and surface potential of keratin-DOX nanoparticles (KDNPs) were characterized by dynamic light scattering (DLS) and transmission electron microscopy (TEM). The drug release results showed that KDNPs performed an excellent pH-sensitive behavior under acidic tumor microenvironment. Cytotoxicity assay by MTT confirmed that KDNPs exhibited the enhanced cytotoxicity against A549 cells. Furthermore, KDNPs had higher therapeutic efficiency in vivo than free DOX . Hemolysis assay indicated that KDNPs was blood compatible. All the results identified that KDNPs are well suited as an ideal drug carrier.

The assembly of protein-templated gold nanoclusters for enhanced fluorescence emission and multifunctional applications

Ying Li,Yu Cao,Lai Wei,Jinjie Wang,Min Zhang,Xuexia Yang,Wenshuo Wang,Guang Yang

Protein-templated gold nanoclusters have attracted attention in fluorescence imaging due to their simple synthesis and good biocompatibility. However, limitations still exist such as poor colloid stability and undesirable fluorescence intensity. Here we describe the self-assembly of keratin-templated gold nanoclusters via a simple and mild preparation process, including keratin-templated synthesis of gold nanoclusters (AuNCs@Keratin), silver ions modification of AuNCs@Keratin (AuNCs-Ag@Keratin), and gadolinium ions-induced aggregation of AuNCs-Ag@Keratin (AuNCs-Ag@Keratin-Gd). It was demonstrated that the AuNCs-Ag@Keratin-Gd obtained an enhanced fluorescence intensity (6.5 times that of AuNCs@Keratin), high colloid stability for more than 4 months, and good biocompatibility. Moreover, the AuNCs-Ag@Keratin-Gd holds promise in multifunctional applications such as near-infrared (NIR) fluorescence imaging, magnetic resonance (MR) imaging, and redox-responsive drug delivery, extending the applicability of fluorescent gold nanoclusters, especially in biomedical fields. Statement of significance Assembly-induced fluorescence enhancement has been rarely reported on as it relates to the protein-templated gold nanoclusters (AuNCs). In this work, self-assembly of protein-templated AuNCs was developed for enhanced fluorescence intensity and multifunctional applications, including bioimaging and responsive drug delivery. A cysteine-rich protein, keratin , was utilized as the template to synthesize AuNCs, which underwent silver ion modification and gadolinium ion-induced aggregation. The silver modification of the keratin-templated AuNCs facilitated the formation of a dense aggregate after gadolinium ion-induced assembly, thus generating an enhanced fluorescence intensity. Such a mechanism was confirmed by fluorescence correlation spectroscopy analysis. We believe that this work will extend the applicability of the fluorescent gold nanoclusters, especially in biomedical fields, and provided an effective approach for the mechanism analysis of the assembly-induced fluorescence enhancement via fluorescence correlation spectroscopy.

A novel strategy for producing high-performance continuous regenerated fibers with wool-like structure

Guang Sheng Cao,Ze Ping Zhang,Min Zhi Rong,Ming Qiu Zhang

We proposed a novel approach to prepare high-performance continuous regenerated keratin fibers with wool-like structure by using the cortical cells and linear keratin from wool waste as reinforcement and adhesive, respectively. The spindle-shaped cortical cells were taken from wool waste based on the different responses of cortical cells and mesenchyme in wool to the treatments of H 2 O 2 oxidation and ultrasonication. The linear keratin was yielded through dissolving wool waste in the green solution consisting of starch derived dithiothreitol and protein denaturant sodium dodecyl sulfate. The recycled keratin fibers were produced by wet-spinning of the mixture solution comprising of cortical cells, linear keratin and toughener poly(ethylene glycol) diacrylate, and crosslinked by glutaraldehyde and 4,4′-methylenebis-(phenyl isocyanate). The cortical cells were aligned along the regenerated fibers axis and retained quite a few α-helical crystals of the intermediate filaments, benefitting improvement of mechanical properties. Consequently, the valuable chemical compositions and hierarchical microstructures of wool were largely inherited. Their mechanical properties, thermal stability, dyeing property, moisture absorption capability, and antistatic resistance resembled those of wool. The regenerated fibers contained 93.3 wt.% components of wool, and the amount of synthetic chemicals in the regenerated fibers was controlled to as low as 6.7 wt.%.

“All-in-one” hydrolyzed keratin protein-modified polyacrylamide composite hydrogel transducer

Yang Gao,Song Gu,Fei Jia,Qian Wang,Guanghui Gao

Conductive hydrogels as wearable devices meet the basic demands of mechanical flexibility and smart sensing. However, achieving a combination of adequate skin adhesion, long-term stability and visible human-sensor interface are still challengeable. Here, a simple and effective strategy was proposed to fabricate “all-in-one” hydrolyzed keratin protein-modified polyacrylamide composite hydrogel transducer. Hydrolyzed keratin endowed hydrogel with lower modulus and ultra-stretchability to minimize wearing-related tension and discomfort. Specially, robust skin adhesion was obtained to achieve precise sensing without delamination. Impressively, the hydrogel had high transparency and exhibited wide-range temperature tolerance (−20 °C to 50 °C). Besides, the hydrogel with stable conductivity was extremely sensitive to pressure and strain at a wide range. As a result, the obtained transducer could not only detect human motion, respiratory mode and vocal-cord vibration, but also identify handwriting and facial expression. This work shed a new light on the design of next-generation skin-like electronic devices with applications in human-machine interface, ionic skin, artificial intelligence and healthcare monitoring.

Highly cadmium and lead adsorption performances of magnesium-rich nitrogen-self-doped keratin-based biochar

Deyun Li,Langjia Feng,Huayi Chen,Kerun Ren,Yonglin Liu,Xueming Lin,Yongtao Li,Yulong Zhang

Cadmium (Cd) and lead (Pb) pollution are significant environmental issues. Metal- and heteroatom-co-doped biochar demonstrated enhanced characteristics and adsorption performance compared to the original biochar. Therefore, magnesium-rich nitrogen-self-doped keratin-based biochar (MNBCs) was developed for the adsorption of Cd and Pb in this study. The incorporation of magnesium increased pH levels, specific surface area, and pore volume. The experimental results showed that MNBC3 exhibited maximum adsorption capacities of 295.63 mg·g −1 for Cd and 640.48 mg·g −1 for Pb. The adsorption processes of Cd and Pb on MNBCs followed the pseudo-second-order kinetic model and the Langmuir isotherm model. The formation of Mg 3 (OH) 5 Cl·4H 2 O and MgO in MNBCs enhanced ion exchange and precipitation of Cd(OH) 2 and Pb 3 (CO 3 ) 2 (OH) 2 . FTIR and XPS analyses confirmed the participation of oxygen- and nitrogen-containing functional groups in surface complexation reactions with Cd and Pb. In summary, the adsorption mechanisms of cadmium and lead by MNBCs mainly include pore filling, ion exchange, precipitation, and functional group complexation. The results of this experiment provide references for the resource utilization of waste chicken feathers and the remediation of heavy metal-polluted water.

Catalytic Generation of Nitric Oxide from Poly(ε-caprolactone)/Phosphobetainized Keratin Mats for a Vascular Tissue Engineering Scaffold

Pengfei Li,Yanfang Wang,Xingxing Jin,Jie Dou,Xiao Han,Xiuzhen Wan,Jiang Yuan,Jian Shen

Tissue-engineered vascular graft (TEVG) is a promising alternative to meet the clinical demand of organ shortages. Herein, human hair keratin was extracted by the reduction method, followed by modification with zwitterionic 2-methacryloyloxyethyl phosphorylcholine (MPC) through thiol–Michael addition to improve blood clotting nature. Then, phosphobetainized keratin (PK) was coelectrospun with poly(ε-caprolactone) (PCL) to afford PCL/PK mats with a ratio of 7:3. The surface morphology, chemical structure, and wettability of these mats were characterized. The biocomposite mats selectively enhanced adhesion, migration, and growth of endothelial cells (ECs) while suppressed proliferation of smooth muscle cells (SMCs) in the presence of glutathione (GSH) and GSNO due to the catalytic generation of NO. In addition, these mats exhibited good blood anticoagulant activity by reducing platelet adhesion, prolonging blood clotting time, and inhibiting hemolysis. Taken together, these NO-generating PCL/PK mats have potential applications as a scaffold for vascular tissue engineering with rapid endothelialization and reduced SMC proliferation.

Preparation of keratin/chlorhexidine complex nanoparticles for long-term and dual stimuli-responsive release

Xuelian Zhi,Yanfang Wang,Pengfei Li,Jiang Yuan,Jian Shen

Nanoscale polyion complex formation via the electrostatic complexation of a polyelectrolyte and a charged drug is the most convenient method for building a drug delivery system that simultaneously realizes the carrier preparation and drug embedding. Herein, we prepared keratin/chlorhexidine complex nanoparticles (KCNPs) based on a drug-induced ionic gelation technique without using a crosslinker, organic solvent or surfactant. These KCNPs exhibited good stability even after having been long-standing for 14 days. The KCNPs were characterized using FTIR, DLS, SEM and TEM. It was found that these nanoparticles had a spherical morphology with a diameter of about 180 nm, and a negatively charged surface with a zeta potential of about −39.1 mV. The cell toxicity of the KCNPs at different dosage levels was evaluated using the MTT assay method, indicating their slight cytotoxicity at lower dosages. The antibacterial activity against E. coli and S. aureus was determined using the zone of inhibition method. It seemed that the KCNPs had better antibacterial activity against S. aureus than against E. coli. Drug delivery profiles showed that the chlorhexidine (CHX) loaded nanoparticles exhibited both pH- and glutathione-responsive character. These keratin-based complex nanoparticles can be regarded as a valuable stimuli-responsive strategy for the delivery of anticancer agents.

Characterization of the keratin/polyamide 6 composite fiber's structure and performance prepared by the optimized spinning process based on the rheological analysis

Bo Li,Yanli Sun,Jinbo Yao,Yanqin Shen,Hailiang Wu,Jiaying Li,Meihui Yang

The complex chemical structure of polypeptide and the imperfection of processing technology cause the mechanical properties of regenerated keratin to be hard and brittle. This defect seriously affects the application prospects of keratin materials. To solve the above problems, α-lipoic acid modified keratin (KER) was blended with Polyamide 6 (PA6) and prepared into composite fibers via the wet-spinning method in this work. The spinnability and spinning conditions of the KER/PA6 blend solution were analyzed by rheological theory. The results illustrated that keratin solution will easily form a gel state under certain temperatures and concentrations, which was not conducive to the preparation of regenerated fiber. When the temperature was 45 °C and the mass fraction was 10 %, the viscosity and rheology of the solution were appropriate. The rheological properties of the blend solution showed that too much keratin would make the solution easy to gel, which was not conducive to the preparation of regenerated fibers and may affect the fiber properties. On this basis, the prepared composite fibers were characterized to explore the macromolecular aggregation state of keratin and PA6 in fibers. FT-IR and XRD results proved that there was no chemical reaction between keratin and PA6 in the composite fibers, which belonged to physical blending. At the same time, the two polymers had good compatibility and can be blended at the molecular level. SEM, DSC , and tensile strength test results indicated that when the proportion of keratin was too high, the structure and properties of the composite fibers will have obvious defects, which was consistent with the rheological analysis. Therefore, the blend ratio of keratin/PA6 was determined to be 3:7. Under this condition, the fibers exhibited a homogeneous structure and good thermal properties, especially its mechanical properties were close to wool fibers. The KER/PA6 composite fibers show important research value and can also provide technical reference for the development of regenerated biomass materials.

A high-performance S-TENG based on the synergistic effect of keratin and calcium chloride for finger activity tracking

Shuai Zhang,Shuo Meng,Ke Zhang,Zhuang Wang,Xiaoyun Xu,Chuanwei Zhi,Shuo Shi,Jinlian Hu

Triboelectric nanogenerator (TENG) is one of the key research directions for future human-computer interaction (HCI), biomedicine, and environmental protection. Bio-based materials are an essential branch of many degradable materials. Keratin has attracted much attention due to its advantages of easy access, biodegradability, and good biocompatibility . A highly sensitive single-electrode TENG (S-TENG) based on CaCl 2 /PVA/keratin and Ecoflex with micro-domes is designed. The excellent stability (18,000 cycles) and stretchability (200%) of the sensor are confirmed by research. Moreover, the presence of CaCl 2 and keratin can significantly increase the S-TENG's output voltage, and the reason is also explored by the density functional theory (DFT) method. Through simulation, it is found that the decrease of the HOMO (highest occupied molecular orbital)-LUMO (lowest unoccupied molecular orbital) gap and the increase of the electrostatic potential are the root causes of the voltage increase. Relying on the excellent characteristics of the S-TENG, finger curvatures, gestures, and object shapes are recognized. Among them, the accuracy of object shape recognition by machine learning algorithm reached 98.1%. This study provides a new method for improving the output efficiency and prolonging the service life of S-TENG and confirms the feasibility of the electron cloud trap model to explain biomass triboelectric materials by keratin and CaCl 2 .
