Identification and Functional Analysis of an Epsilon Class Glutathione S-Transferase Gene Associated with α-Pinene Adaptation in Monochamus alternatus

Mingyu Xue,Xiaohong Xia,Yadi Deng,Fei Teng,Shiyue Zhao,Hui Li,Dejun Hao,Wei-Yi Chen

Alpha-pinene is one of the main defensive components in conifers.Monochamus alternatus(Coleoptera: Cerambycidae), a wood borer feeding on Pinaceae plants, relies on its detoxifying enzymes to resist the defensive terpenoids. Here, we assayed the peroxide level and GST activity ofM. alternatuslarvae treated with different concentrations of α-pinene. Meanwhile, agstgene (MaGSTe3) was isolated and analyzed. We determined its expression level and verified its function. The results showed that α-pinene treatment led to membrane lipid peroxidation and thus increased the GST activity. Expression ofMaGSTe3was significantly upregulated in guts following exposure to α-pinene, which has a similar pattern with the malonaldehyde level. In vitro expression and disk diffusion assay showed that the MaGSTe3 protein had high antioxidant capacity. However, RNAi treatment of MaGSTe3 did not reduce the hydrogen peroxide and malonaldehyde levels, while GST activity was significantly reduced. These results suggested MaGSTe3 takes part in α-pinene adaptation, but it does not play a great role in the resistance ofM. alternatuslarvae to α-pinene.Keywords:glutathione S-transferase;detoxification enzymes;monoterpene

Effects of a Pathogenic Beauveria bassiana (Hypocreales: Cordycipitaceae) Strain on Detoxifying and Protective Enzyme Activities in Xylotrechus rusticus (Coleoptera: Cerambycidae) larvae

Jun-Nan Ding,Hui-Hui Zhang,De-Fu Chi

In recent years, biological methods have been used widely for controlling stem-boring pests in forests. In this study, the larvae of Xylotrechus rusticus (L.) (Coleoptera: Cerambycidae) were infected with 4 strains of 2 species of Beauveria (Hypocreales: Cordycipitaceae). Larval detoxifying and protective enzyme activities were measured at different times after infection. The results showed significant differences in the pathogenicity of the 4 strains. The most virulent strain, Beauveria bassiana (Bals.-Criv) Vuill. strain BbCC01, caused 93.3% mortality, with an estimated LT50 of 4.69 d. The detoxifying and protective enzyme activities in larvae infected with strain BbCC01 changed significantly. To resist the infection, the activities of the detoxifying enzymes (carboxylesterase, glutathione S-transferase, and acetylcholinesterase) first increased and then slowly decreased. The activities of the protective enzymes (catalase, superoxide dismutase, and peroxidase) showed a similar trend. The enzyme activities peaked at 72 to 96 h post-infection. In this study, the B. bassiana strain BbCC01 proved to exhibit high virulence towards X. rusticus larvae. This research provides important information regarding the biological control of X. rusticus and elucidates the patterns of detoxifying and protective enzyme activities in the susceptible larvae.

A novel detoxification strategy of Bombyx mori (Lepidoptera: Bombycidae) to dimethoate based on gut microbiota research

Tian Chao,Tang Jie,Zhu Qingyu,Guo Xiqian,Shu Qilong,Gu Zhiya,Li Fanchi,Li Bing

Bombyx mori (L.) (Lepidoptera: Bombycidae) is an important economic insect, and Exorista sorbillans (W.) (Diptera: Tachinidae) is an endoparasitic pest of larval B. mori. Dimethoate is less toxic to B. mori than E. sorbillans and is used in sericulture to controlling E. sorbillans. To investigate the effects of dimethoate treatment on the gut microorganisms and physiological functions of B. mori, 16S rRNA sequencing was used to analyzed the composition and structure of the gut microbiota. This study investigated their role in enhancing silkworm resistance by screening dominant populations after dimethoate treatment. The results indicated that dimethoate did not alter the composition of the dominant gut bacterial groups in silkworm; however, it significantly increased the abundance of the gut bacteria Methylobacterium and Aureimonas, and decreased the abundance of Enterobacterales, Bifidobacterium, Blautia, Collinsella, Faecalibacterium, and Prevotella. Eleven strains of dimethoate-resistant bacteria were selected through in vitro culture, all of which were unable to grow when dimethoate was used as a carbon source. Additionally, a germ-free silkworm model was established to assess detoxifying enzyme activity in the midgut. The results revealed that the gut symbiotic microbiota can enhance dimethoate resistance by increasing detoxification enzyme activity. This study identifies a novel pathway for silkworm resistance to dimethoate based on gut microbiota, providing new insights into the role of symbiotic gut bacteria in insecticide metabolism.

Protective effects of resveratrol on honeybee health: Mitigating pesticide-induced oxidative stress and enhancing detoxification

Linlin Liu,Min Shi,Yanyan Wu,Jiali Hao,Junxiu Guo,Shanshan Li,Pingli Dai,Jing Gao

The widespread use of pesticides poses a significant threat to honeybee health by impacting their survival, behavior, immune function, and detoxification capacity. While phytochemicals such as resveratrol (RSV) have shown potential in mitigating oxidative stress and enhancing antioxidant defenses, their role in improving honeybee tolerance to pesticide exposure remains underexplored. In this study, we investigated the effects of RSV supplementation on honeybees exposed to three pesticides: dinotefuran (DIN), tebuconazole (TEB), and deltamethrin (DEL). The results showed that RSV supplementation significantly improved survival, feed intake, mobility, and gustatory sensitivity, indicating its protective effects against pesticide toxicity. Furthermore, RSV helped normalize impaired detoxification enzyme activities, including SOD, POD, catalase, and glutathione reductase, and reduced ROS levels and lipid peroxidation. Gene expression analysis revealed that RSV modulates Toll pathway-related genes like defensin and apidaecin , alleviating immune suppression caused by pesticides. Additionally, RSV influenced the insulin/insulin-like growth factor signaling (IIS) pathway by reducing ilp1 and inr1 expression, potentially mitigating metabolic stress. These findings demonstrate that protective effects of RSV may be linked to its ability to counter oxidative stress, restore mitochondrial function, and enhance energy metabolism. Furthermore, RSV is widely available, cost-effective, and easily incorporated into bee feed, making it feasible for large-scale application. This study highlights the protective role of RSV in pesticide detoxification in honeybees, offering new perspectives for honeybee health management and environmental toxicology research. By reducing the adverse effects of pesticides on honeybees, the application of RSV not only contributes to maintaining ecological balance but also supports sustainable agricultural practices. Future research should focus on optimizing its dosage, evaluating long-term effects, and investigating its impact on colony dynamics to facilitate its practical implementation in apiculture.

Down-Regulation of P450 Genes Enhances Susceptibility to Indoxacarb and Alters Physiology and Development of Fall Armyworm, Spodoptera frugipreda (Lepidoptera: Noctuidae)

Hafeez Muhammad,Li Xiaowei,Ullah Farman,Zhang Zhijun,Zhang Jinming,Huang Jun,Fernández-Grandon G. Mandela,Khan Muhammad Musa,Siddiqui Junaid Ali,Chen Limin,Ren Xiao Yun,Zhou Shuxing,Lou Yonggen,Lu Yaobin

The fall armyworm (FAW), Spodoptera frugiperda (J.E. Smith), is a pest of many important crops globally. Effective control is challenging, with the pest exhibiting resistance to different synthetic pesticides across various groups. However, the mechanisms employed by resistant insects for overexpression of relevant detoxification genes remain unclear. The activity of detoxification enzymes was investigated in this study. Additionally, using RNA interference (RNAi), a functional analysis was completed of two P450s genes in an indoxacarb resistant population of fall armyworms. Elevated resistance levels (resistance ratio = 31.37-fold) in indoxacarb-selected populations of FAW were observed after 14 generations. The qRT-PCR showed higher expression of two cytochrome P450 genes, CYP321A7 and CYP6AE43, in this selected population compared to the control population. RNAi was applied to knock down the P450 dsCYP321A7 and dsCYP6AE43 genes in the FAW larvae. Droplet feeding of the dsRNAs (CYP321A7 and CYP6AE43) via an artificial diet significantly increased mortality rates in the indoxacarb treated population. A shorter larval developmental time of FAW was detected in all dsRNAs-fed larvae. Correspondingly, larval mass was reduced by dsRNAs in indoxacarb resistant populations of fall armyworm. Larval feeding assays demonstrate that dsRNAs targeting, specifically of CYP321A7 and CYP6AE43 enzymes, could be a beneficial technique in the management of indoxacarb resistant populations. Further study on the potential use of dsRNA and its application should be conducted in efforts to counter the development of resistance in FAW against various insecticides in the field.

Multiple Insecticide Resistance and Associated Metabolic-Based Mechanisms in a Myzus Persicae (Sulzer) Population

Jinfeng Hu,Feng Chen,Jun Wang,Wenhua Rao,Lei Lin,Guocheng Fan

The green peach aphid,Myzus persicae(Sulzer) (Hemiptera: Aphididae), is an economically devastating crop pest worldwide. TheM. persicae(SEF-R) population of a cabbage field in China was tested for susceptibilities to 13 insecticides. Compared with the susceptible population (FFJ-S), extremely high and high resistance to beta-cypermethrin (324-fold) and imidacloprid (106.9-fold) was detected in SEF-R. More importantly, this is the first report of resistance in the fieldM. persicaepopulation to sulfoxaflor (32.4-fold), flupyradifurone (9.5-fold), pymetrozine (34.8-fold), spirotetramat (8.1-fold), flonicamid (5.8-fold), and broflanilide (15.8-fold) in China when compared with FFJ-S. The resistance factor decayed to a low level to sulfoxaflor and pymetrozine after 15 generations without any selection pressure. The resistance-related mutations (R81T and kdr) detected in SEF indicated target-site resistance to neonicotinoids and pyrethroids, respectively. Biochemical assays revealed the involvement of monooxygenase, carboxylesterase, superoxide dismutase, and peroxidase in a multi-insecticide resistance mechanism. The overexpression of P450s, esterases, and a UDP-glycosyltransferase might be responsible for the multi-insecticide resistance in SEF-R. The knockdown of CYP6CY3 in SEF-R increased its susceptibility to imidacloprid, thiacloprid, and thiamethoxam, which verified that P450s play vital roles in neonicotinoid metabolism. Our findings provide guidance for the rational use of insecticides to delay resistance development in GPA.Keywords:Myzus persicae;insecticides;resistance monitoring;detoxification enzymes;protective enzymes;metabolic mechanism

Exogenous 24-epibrassinolide activates detoxification enzymes to promote degradation of boscalid in cherry tomatoes

Yajie Yang,Linchun Mao,Weiliang Guan,Xiaobo Wei,Yelin Shao,Zisheng Luo,Xingyu Lin,Li Li

BACKGROUND Boscalid is often used to extend the storage time of postharvest cherry tomato. Pesticide residue has become an issue of food safety. This study sought to investigate the spatial distribution of boscalid residue in cherry tomato fruits and to determine the effect of 24-epibrassinolide (EBR) in promoting boscalid degradation. RESULTS Boscalid could quickly penetrate into cherry tomatoes, but mainly remained in the peel. The migration of boscalid from the peel into the core was a time-consuming and complex process during storage. After 72 h, boscalid residues in the pulp and the core began to accumulate gradually. The exogenous application of EBR activated peroxidase, glutathione reductase and glutathione S -transferase, and effectively promoted the degradation of boscalid by a maximum decrease of 44.8% in peel, 54.0% in pulp and 71.2% in core. CONCLUSION As one of the common pesticides, boscalid had a strong ability to enter the cherry tomato and thus become a potential risk for public consumption. Therefore, rational use of pesticides is recommended. The results of this study indicate that the possible risk of boscalid residue could be alleviated by EBR pretreatment through activating detoxification enzymes. © 2020 Society of Chemical Industry

Cytochrome P450s from the Chinese white pine beetle, Dendroctonus armandi (Curculionidae: Scolytinae): Expression profiles of different stages and responses to host allelochemicals

Lulu Dai,Mingyuan Ma,Chunyan Wang,Qi Shi,Ranran Zhang,Hui Chen

Bark beetles oxidize the defensive allelochemicals from their host trees to both detoxify and convert these materials into components of their pheromone system. The ability of this insect to adapt to its chemical environment might be explained by the action of major detoxification enzymes such as cytochrome P450s (or CYPs). Sixty-four sequences coding for P450s were identified, and most of the transcripts were found to be expressed in the larvae, pupae and adults of Dendroctonus armandi . To gain information on how these genes help D. armandi overcome the host defense, differential transcript levels of the CYP genes were observed between sexes and within the sexes. Significant differences were observed among developmental stages, in feeding on the phloem of Pinus armandi and in exposure to stimuli ((±)-α-pinene, (S)-(−)-α-pinene, (S)-(−)-β-pinene, (+)-3-carene, (±)-limonene and turpentine oil) for 8 h. We investigated the effect of sex and generations on the survivorship of individual D. armandi that were exposed to host volatiles at concentrations comparable to constitutive and induced levels of defense using fumigant exposure to understand the ability of the beetles to tolerate host defensive chemicals. The differential transcript accumulation patterns of CYP genes of these bark beetle provided insight into the ecological interactions of D. armandi with its host pine.

Stability, Inheritance, Cross-Resistance, and Fitness Cost of Resistance to λ-Cyhalothrin in Cydia pomonella

Chao Hu,Cong Zhang,Yi-Fan Tang,Yu-Xi Liu,Zi-Nan Xia,Yan Wang,Wei-Tu Li,Ping Gao,Yu-Ting Li,Yun-Tong Lv,Xue-Qing Yang

Insecticides are commonly utilized in agriculture and forestry for pest control, but their dispersal can pose hazards to humans and environment. Understanding resistance, inheritance patterns, and fitness costs can help manage resistance. A λ-cyhalothrin-resistant population (LCR) of Cydia pomonella, a global pest of pome fruits and walnuts, was obtained through selective insecticide breeding for 15 generations, showing stable moderate resistance (23.85-fold). This population was cross-resistant to deltamethrin (4.26-fold) but not to β-cypermethrin, chlorantraniliprole, chlorpyrifos, and avermectin. Genetic analysis revealed the resistance was autosomal, incompletely dominant, and controlled by multiple genes. Increased activity of glutathione S-transferases and cytochrome P450 monooxygenases (P450s) played a primary role in resistance, with specific genes up-regulated in LCR, and exhibited significant expression in midgut. LCR also exhibited fitness costs, including delays in development, reduced fecundity, and slower population growth. These findings contribute to understanding λ-cyhalothrin resistance in C. pomonella and can guide resistance management strategies.

Effects of poplar secondary metabolites on performance and detoxification enzyme activity of Lymantria dispar

Zhenyue Wang,Faidah Arina Nur,Jingyi Ma,Jianguo Wang,Chuanwang Cao

To identify the effects of poplar secondary metabolites on Lymantria dispar , six poplar secondary metabolites (i.e., caffeic acid , salicin , rutin , quercetin , flavone , and catechol) and three mixtures containing characteristic secondary metabolites in poplar were selected. Mixture 1 contained flavone and salicin, mixture 2 contained salicin, caffeic acid, and catechol , and mixture 3 contained flavone, catechol, and caffeic acid. Mixtures were added to artificial diets used to feed 2nd instar L. dispar larvae. The effects of different secondary metabolites on larval growth and development , antifeedant activity, nutrient utilization, and detoxifying enzymatic activity were investigated. Results revealed that there were different influences on L. dispar larvae. The maximum antifeedant rate of flavone was 87.58%. Larvae treated with mixture 2 had a significantly longer development time of 5.61 d with a survival rate of 38.75% for 15 d, which is lower than a single secondary metabolite. No L. dispar larvae survived on feeding diets containing flavone for 7 d. An increase in GST and P450 activities in larvae was significantly induced during the 72 h feeding on artificial diets containing experimental secondary metabolites. After treatment containing salicin and flavone for 24–72 h, P450 activity increased at first then decreased. These results provide a foundation for further investigation on the host selection and underlying adaptation mechanisms in L. dispar .

Effects of azadirachtin on detoxification-related gene expression in the fat bodies of the fall armyworm, Spodoptera frugiperda

Yu Haikuo,Yang Xianmei,Dai Jinghua,Li Yuning,Veeran Sethuraman,Lin Jintian,Shu Benshui

The fall armyworm, Spodoptera frugiperda, has become a worldwide pest and threatens world food production. A previous study indicated that azadirachtin, the most effective botanical insecticide for S. frugiperda , inhibits larval growth of the insect. The effect of azadirachtin on the tissues of the larvae, however, remains to be determined. In this study, the effects of azadirachtin on the structure of fat bodies were analyzed. Comparative transcriptomic analysis was conducted between controls and samples treated with 0.1 μg/g azadirachtin for 7 days to explore potential relevant mechanisms. The expression of 5356 genes was significantly affected after azadirachtin treatment, with 3020 up-regulated and 2336 down-regulated. Among them, 137 encode detoxification enzymes, including 53 P450s , 20 GSTs , 27 CarEs , 16 UGTs , and 12 ABC transporters . Our results indicated that azadirachtin could destroy fat body structure and change the mRNA levels of detoxification-related genes. The up-regulated genes encoding detoxification enzymes might be related to detoxifying azadirachtin. Our results elucidate a preliminary mechanism of azadirachtin detoxification in the fat bodies of S. frugiperda larvae.

Exploration of Synergistic Pesticidal Activities, Control Effects and Toxicology Study of a Monoterpene Essential Oil with Two Natural Alkaloids

Jianwei Xu,Min Lv,Shanshan Fang,Yanyan Wang,Houpeng Wen,Shaoyong Zhang,Hui Xu

With the increasing development of pest resistances, it is not easy to achieve satisfactory control effects by using only one agrochemical. Additionally, although the alkaloid matrine (MT) isolated fromSophora flavescensis now utilized as a botanical pesticide in China, in fact, its pesticidal activities are much lower in magnitude than those of commercially agrochemicals. To improve its pesticidal activities, here, the joint pesticidal effects ofMTwith another alkaloid oxymatrine (OMT) (isolated fromS. flavescens) and the monoterpene essential oil 1,8-cineole (CN) (isolated from the eucalyptus leaves) were investigated in the laboratory and greenhouse conditions. Moreover, their toxicological properties were also studied. AgainstPlutella xylostella, when the mass ratio ofMTandOMTwas 8/2, good larvicidal activity was obtained; againstTetranychus urticae, when the mass ratio ofMTandOMTwas 3/7, good acaricidal activity was obtained. Especially whenMTandOMTwere combined withCN, the significant synergistic effects were observed: againstP. xylostella, the co-toxicity coefficient (CTC) ofMT/OMT(8/2)/CNwas 213; againstT. urticae, the CTC ofMT/OMT(3/7)/CNwas 252. Moreover, the activity changes over time of two detoxification enzymes, carboxylesterase (CarE) and glutathioneS-transferase (GST) ofP. xylostellatreated withMT/OMT(8/2)/CN, were observed. In addition, by scanning electron microscope (SEM), the toxicological study suggested that the acaricidal activity ofMT/OMT(3/7)/CNmay be related to the damage of the cuticle layer crest ofT. urticae.Keywords:matrine;1,8-cineole;ternary complex;pesticidal activity;detoxification enzymes;SEMKey Contribution:Joint pesticidal activities ofMT–OMT–CNwere related to the mass ratio of ternary complex and controlling pests. These results will lay the foundation for the future study of different combinations ofMT–OMT–CNand the use ofCNas a synergist with other bioactive plant natural products as pesticidal agents in crop protection.
