Combining informative imaging methodologies with effective treatments to destroy tumors is of great importance for oncotherapy. Versatile nanotheranostic agents that inherently possess both diagnostic imaging and therapeutic capabilities are highly desirable to meet these requirements. Here, a simple but powerful nanoplatform based on polydopamine-coated gold nanostar (GNS@PDA), which can be easily diversified to achieve various function extensions, is designed to realize functional and anatomical imaging-guided photothermal oncotherapy. This nanoplatform intrinsically enables computed tomography/photoacoustic/two-photon luminescence/infrared thermal tetramodal imaging and can further incorporate fibroblast activation protein (FAP, a protease highly expressed in most of tumors) activatable near-infrared fluorescence imaging and Fe3+-based magnetic resonance imaging for comprehensive diagnosis. Moreover, GNS@PDA exhibits excellent photothermal performance and efficient tumor accumulation. Under the precise guidance of multimodal imaging, GNS@PDA conducts homogeneous photothermal ablation of bulky solid tumors (∼200 mm3) in a xenograft mouse model. These results suggest great promise of this extendable nanoplatform for cancer theranostics.

The limited penetration of imaging agents in solid tumor tissue and cells remains a great challenge to achieve ideal visibility in cancer diagnosis. Herein, a near infrared (NIR) light-driven Janus mesoporous silica nanomotor (JMS nanomotor) to promote magnetic resonance (MR) imaging in vivo is developed. The JMS nanomotors are prepared by depositing Au on the half-sphere surface of gadolinium-doped mesoporous silica nanoparticles. Under NIR irradiation, the nanomotors achieve efficient propulsion through thermophoresis in biological media. To prove the ability of nanomotors as active diagnostic imaging agents, both in vitro and in vivo experiments are carried out. In vitro studies confirm that NIR light can actively propel the JMS nanomotors to effectively seek, adhere to, and mechanically perforate the tumor cells to enhance the cellular uptake and MR imaging. To move a step further, in vivo investigations show that the JMS nanomotors also exhibit improved accumulation and deep penetration in the solid tumor model when exposed to NIR laser, where the MR imaging contrast is significantly enhanced in the majority of tumor tissue. Such JMS nanomotors provide new insight in precise cancer diagnosis and meanwhile MR imaging offers a new tool for real-time tracking of nanomotors in vivo.

Background Triple-negative breast cancer (TNBC) possesses special biological behavior and clinicopathological characteristics, which is highly invasive and propensity to metastasize to lymph nodes, leading to a worse prognosis than other types of breast cancer. Thus, the development of an effective therapeutic method is significant to improve the survival rate of TNBC patients. Results In this work, a liposome-based theranostic nanosystem (ILA@Lip) was successfully prepared by simultaneously encapsulating IR 780 as the photosensitizer and lenvatinib as an anti-angiogenic agent, together with banoxantrone (AQ4N) molecule as the hypoxia-activated prodrug. The ILA@Lip can be applied for the near-infrared (NIR) fluorescence diagnostic imaging of TNBC and its lymph node metastasis for multimodal therapy. Lenvatinib in ILA@Lip can inhibit angiogenesis by cutting oxygen supply, thereby leading to enhanced hypoxia levels. Meanwhile, large amounts of reactive oxygen species (ROS) were produced while IR 780 was irradiated by an 808 nm laser, which also rapidly exhausted oxygen in tumor cells to worsen tumor hypoxia. Through creating an extremely hypoxic in TNBC, the conversion of non-toxic AQ4N to toxic AQ4 was much more efficiency for hypoxia-activated chemotherapy. Cytotoxicity assay of ILA@Lip indicated excellent biocompatibility with normal cells and tissues, but showed high toxicity in hypoxic breast cancer cells. Also, the in vivo tumors treated by the ILA@Lip with laser irradiation were admirably suppressed in both subcutaneous tumor model and orthotopic tumor models. Conclusion Utilizing ILA@Lip is a profound strategy to create an extremely hypoxic tumor microenvironment for higher therapeutic efficacy of hypoxia-activated chemotherapy, which realized collective suppression of tumor growth and has promising potential for clinical translation.

Copper ions, implicated in processes such as oxidative stress and inflammation, are believed to play a crucial role in cardiovascular disease, a prevalent and deadly disease. Despite this, current diagnostic methods fail to detect early stage cardiovascular disease or track copper ion accumulation, limiting our understanding of the disease’s progression. Therefore, the development of noninvasive techniques to image copper ions in cardiovascular disease is urgently needed to enhance diagnostic precision and therapeutic strategies. In this study, we report the successful synthesis and application of a copper ion-activated photoacoustic probe, CS-Cu, which exhibits high sensitivity and selectivity toward copper ions both in vitro and in vivo. CS-Cu was able to noninvasively monitor the changes in copper ion levels and differentiate between different mice based on copper ions in urine. Furthermore, the probe demonstrated good photoacoustic stability and exhibited no significant toxicity in the mice. These findings suggest that CS-Cu could be a promising tool for early detection and monitoring of Cu2+ levels in vivo and urine, providing a new perspective on the role of copper ions in cardiovascular disease.

While targeted theranostics for cancer remains a pivotal research frontier, conventional ligand conjugation strategies exhibit persistent limitations in off-target accumulation and suboptimal tumor specificity, ultimately failing to achieve reliable detection of early-stage lesions or metastatic nodule while demonstrating insufficient therapeutic payload delivery. In this study, the manganese sulfide (MnS) nanoplatform was synthesized using transferrin (Tf) with tumor-targeting properties as carrier by a simple fabrication method. Notably, compared to clinically prevalent Gd-based contrast agents, Tf-MnS exhibited superior T1-weighted magnetic resonance imaging (MRI) contrast performance, with the longitudinal relaxation (r1) reaching 7.5253 mM-1 •s-1 which was significantly higher than 3.2915 mM-1 •s-1 of Gd-DTPA, and in the MRI of subcutaneous tumors and lung metastatic lesion in mice, the maximum relative signal-to-noise ratios reached 46.33% and 40.33% respectively. Remarkably, upon reaching the acidic tumor microenvironment, Tf-MnS disintegrated to release Mn2+ ions and hydrogen sulfide (H2S). The Mn2+ ions participated in Fenton-like reactions to produce cytotoxic hydroxyl radicals, while H2S concurrently inhibited catalase enzyme activity, thereby alleviating the insufficiency of hydrogen peroxide substrate and amplifying the therapeutic outcome. This synergistic mechanism endowed Tf-MnS with self-enhanced anti-tumor effect, inhibiting both lung metastatic lesion and subcutaneous tumors in mice of the Tf-MnS group, with a tumor inhibition rate of 54.26%. Collectively, this work proposes an innovative strategy for integrating accurate diagnosis and self-augmented therapy of tumors and lung metastatic lesion into a unified nanoplatform, offering a promising methodology for precision oncology.

To overcome the dual challenges of suboptimal therapeutic outcomes and delayed detection in hepatocellular carcinoma (HCC), a nucleus-targeting theranostic nanoplatform based on gadolinium-functionalized carbon dots (GdCDs) is presented. GdCDs were synthesized via a one-step hydrothermal method utilizing neutral red and citric acid as carbon precursors and gadolinium chloride hexahydrate as the paramagnetic source, showing good fluorescence (FL) and magnetic resonance imaging (MRI) capabilities. After modified with a nuclear localization sequence (NLS), GdCDs were loaded with doxorubicin (DOX) to yield the integrated theranostic platform: Nucleus-targeting gadolinium-functionalized carbon dot drug delivery system (NT-GdCDs-DDS) for FL/MRI dual-modality imaging and nucleus-targeting drug delivery. NT-GdCDs-DDS exhibited optimal excitation/emission wavelengths at 497/596 nm, a high longitudinal relaxivity (r 1 ) of 23.11 mM −1 s −1 , and a DOX loading efficiency of 61.9%. Results of antitumor activity assays in vitro demonstrated the viability of HepG2 cells incubated with 400 μg/mL NT-GdCDs-DDS for 24 h was 8.12%, demonstrating strong antitumor activity. Furthermore, a significant tumor growth suppression in mice was observed by treated with NT-GdCDs-DDS over a 15-day period, showing superiority to free DOX. These findings establish NT-GdCDs-DDS as a potent theranostic nanoplatform for simultaneous HCC imaging and therapy. Graphical abstract

Emerging nanotechnologies in cancer therapy increasingly highlight the importance of integrating therapeutic and diagnostic functions into a single platform for enhanced efficacy and real-time monitoring. Here, we present a vortex‐shaped nanoring Fe 3 O 4 @MnO 2 @PDA (polydopamine) engineered for dual‐mode T 1 – T 2 magnetic resonance imaging (MRI)‐guided, dual‐thermal, and Fenton‐like chemodynamic therapy (CDT). The ring‐shaped nano Fe 3 O 4 core provides magnetothermal hyperthermia, while the MnO 2 layer, acting as a responsive nanozyme, catalyzes endogenous H 2 O 2 to generate oxygen and bolster CDT. Meanwhile, the PDA coating efficiently absorbs near‐infrared light, amplifying photothermal effects for tumor ablation. Under dual‐heating conditions, the temperature rise rate (2.05–2.92 times) and final equilibrium temperatures (1.57–2.27 times) exceed those of single‐mode heating, underscoring a robust synergistic effect. Additionally, due to the shell’s shielding effect weakening T 2 signals and the tumor microenvironment enhancing T 1 relaxation, the T 1 (5.75 Mm −1 s −1 ) to T 2 (91.56 mM −1 s −1 ) ratio is optimized specifically at tumor sites, thus achieving effective dual‐modal MRI contrast. Moreover, the smart MnO 2 design not only furnishes dynamic imaging enhancements but also enables environment‐triggered Mn 2+ release, offering further diagnostic and therapeutic benefits. In vitro and in vivo evaluations confirm significant tumor suppression with minimal systemic toxicity. These findings position nanoring Fe 3 O 4 @MnO 2 @PDA as a promising dual‐mode MRI‐guided, dual‐thermal, and chemodynamic platform for advanced cancer therapy.

Liver fibrosis, a pivotal pathological stage in the progression of chronic liver diseases to cirrhosis and hepatocellular carcinoma is characterized by liver sinusoidal endothelial cell (LSEC) capillarization, oxidative stress imbalance, and cell pyroptosis. Current clinical interventions show limited efficacy in reversing fibrosis, highlighting the urgent need for novel therapeutic strategies. In this study, we developed an L-arginine-loaded melanin-like nanozyme (L-Arg@MeNPs) that targets liver fibrosis through a triple-action mechanism: (1) sustained nitric oxiderelease from L-Arg restores LSEC fenestration, improving sinusoidal permeability; (2) the MeNPs exhibit catalase/superoxide dismutase-mimicking activity to scavenge reactive oxygen species, thereby blocking the NOD-like receptor pyrin domain-containing 3/caspase-1-mediated pyroptosis pathway; and (3) intrinsic photoacoustic/magnetic resonance dual-modal imaging enables real-time therapeutic monitoring. Overcoming the biosafety limitations of conventional metal-based nanozymes, this system demonstrates superior pH stability and metabolic degradability. In vivo studies confirm that L-Arg@MeNPs effectively ameliorate the fibrotic microenvironment and reduce excessive extracellular matrixdeposition, offering a targeted, safe, and theranostic solution for clinical liver fibrosis treatment.

In cancer biology, cysteine (Cys) is pivotal in maintaining the redox balance within tumor cells. To explore alterations in Cys concentration within osteosarcoma (OS) and hepatocellular carcinoma (HCC) cells, we devised a novel fluorescent (FL) probe named JYL-Cys , utilizing a thiol-sensitive dye, which specifically reacts with Cys to emit FL at 718 nm (λ ex = 660 nm). JYL-Cys exhibits a high response fold, a low limit of detection (0.15 μM), high selectivity, and effectively detects Cys under physiological conditions. A comprehensive suite of in vitro experiments, encompassing cell uptake studies and FL imaging analysis, was executed to assess the responsiveness of JYL-Cys across various cancer cell lines, specifically the OS cell line MG-63 and the HCC cell line HepG2. Furthermore, in vivo mouse model experiments were conducted to validate the probe's applicability and safety in live organisms. The probe demonstrated exceptional capability in visualizing the distribution of Cys within both OS and HCC cells through imaging, revealing markedly elevated Cys levels compared to normal cells. Notably, in vivo experiments confirmed the probe's safety for biological applications. Our experimental findings underscore that JYL-Cys is a valuable resource for delineating the biological significance of Cys in cancer progression and establish a foundation for future clinical applications, encompassing early diagnosis and prognosis assessment.

Metal–organic frameworks (MOFs) demonstrate immense potential in the development of drug carriers for cancer treatment and imaging guidance due to their adjustable particle size, excellent biodegradability, and substantial porosity. In this study, we constructed a nanodrug delivery system, P@F-FD, based on a metal–organic framework. This system is coloaded with the anticancer drug Adriamycin, further modified using 5-FAM, and encapsulated with polydopamine. It exhibits high drug-carrying capacity, satisfactory biocompatibility, pH-responsiveness, and efficient drug-release capability. P@F-FD achieves highly efficient pancreatic cancer therapy through a cascaded mechanism: Initially, the system leverages nanoparticle size-dependent effects for targeted tumor accumulation; subsequently, under the acidic tumor microenvironment, the polydopamine coating undergoes protonation-induced structural transformation to intelligently trigger controlled release of the antitumor drug doxorubicin; simultaneously, the activated theranostic module facilitates synchronous magnetic resonance anatomical imaging via iron components while enabling real-time navigation through covalently conjugated fluorescent probes, collectively guiding precise therapeutic intervention at the tumor site. This innovative application of a nanodelivery organometallic framework not only regulates drug release efficiently but also enhances the platform for pancreatic cancer therapy, providing a strategy for the development of “diagnostic-integrated” nanomaterials for combined cancer therapy.

Rapid and specific detection of influenza A viruses is critical for pandemic response. However, traditional lateral flow immunoassays (LFIAs) face challenges in sensitivity, signal reliability, and cross-reactivity. Herein, a dual-aptamer-functionalized lateral flow platform is designed, integrating upconversion nanoparticles (UCNPs) and gold nanoparticles (AuNPs) to mitigate these bottlenecks through a synergistic mechanism: near-infrared-excited luminescence resonance energy transfer (LRET) for fluorescence modulation, dual-aptamer domain recognition for specific binding, and fluorescence-colorimetric dual readouts for orthogonal validation. Specifically, LRET serves as the core signal transduction mechanism, with aptamer1-conjugated UCNPs as donors and aptamer2-conjugated AuNPs as acceptors, regulated by target-induced proximity. Dual-aptamers recognizing distinct hemagglutinin (HA) epitopes enhance specificity, minimizing cross-reactivity with non-target subtypes (H5N1, H6N1, etc.) while ensuring high-affinity H1N1 HA binding. Such synergy enables significant signal amplification, achieving a fluorescent limit of detection (LOD) of 2.814 ng mL −1 (2.84–5.33-fold better than traditional AuNP-LFIAs) with a linear range of 4–10 ng mL −1 . Importantly, the dual-mode design integrates upconversion fluorescence and AuNPs colorimetry, mitigating variability, ensuring precision, and enabling 20-minute qualitative detection (up to 100 ng mL −1 ) for point-of-care testing (POCT). Thus, this work presents a scalable POCT paradigm via apt-LRET integration, leveraging dual-aptamer recognition and LRET-mediated amplification for on-site viral diagnostics with pandemic-response timeliness.

Most of the existing single-component nanostructures cannot provide comprehensive diagnostic information, and their treatment strategies always have to combine other therapeutics as a complementary for effective biomedical application. Here, we adopted a facile approach to design a theranostic nanoflower (NF) with robust efficacy for comprehensive tumor diagnosis and quadruple synergistic cancer therapy. The NF is equipped with a metallic hybrid of several functional elements and flower-like superstructures and thus shows excellent in vitro and in vivo theranostic performance. It shows high X-ray attenuation coefficiency for the Bi element, strong near-infrared (NIR) plasmon absorbance and singlet oxygen (1O2) generation ability for the Mo element, and great photothermal conversion efficiency (54.7%) because of enhanced photoabsorption of the petal structure. Moreover, the NF realizes a very high doxorubicin-loading efficiency (90.0%) and bimodal pH/NIR-responsive drug release, posing a promise as a controlled drug carrier. The NF also shows excellent performance at trimodal magnetic resonance/X-ray computed tomography/photoacoustic imaging for comprehensive tumor diagnosis. To our best knowledge, it is the first time that integrating at least seven functions into one biomedical nanomaterial for well-rounded tumor theranostics has been reported. This “all-in-one” NF opens a new perspective in developing novel and efficient multifunctional nanotheranostics.
