Quasi static pressure tolerant triboelectric textile sensor array for intelligent pulse diagnosis in traditional Chinese medicine

Xinyu Wang,Wenxuan Guo,Xiaodong Jiao,Zhengxiang Jin,Ce Chuai,Junjun Pan,Hao Sun,Qinglin Sun,Siyuan Wang,Peng Xu

Traditional Chinese Medicine (TCM) pulse diagnosis provides a non-invasive approach to assess physiological conditions through multidimensional pulse information and surface thermal cues reflecting body cold–heat states. However, current sensing platforms struggle to achieve high sensitivity, and a wide linear range under the quasi-static pressure required for pulse assessment, making faithful replication of the tactile diagnostic process challenging. Here, we present a polypyrrole(PPy)/MXene ( T i 3 C 2 T x ) based triboelectric smart textile sensor (TSTS) that captures key tactile features of TCM pulse diagnosis and simultaneously detects radial artery temperature via a dual-stage signal amplification design. The TSTS forms a bioinspired multiscale conductive interface in which nanoparticles and nanosheets assemble on textiles to enhance mechanical responsiveness and thermal sensitivity, while a silicone micro-protrusion and spacer layer optimize stress transfer and structural stability. The sensor exhibits high linearity (0.999), sensitivity (0.07 V/kPa), a low detection limit (0.02 kPa), rapid response time ( < 40 ms), and high thermal resolution (0.1 ∘ C), and reliably senses pulses under quasi-static pressures of 0–100 mmHg. Integrated with a pressure graded sensing protocol and a multiscale fusion cross attention neural network (MSFCN), the system with the TSTS array enables accurate TCM pulse classification and temperature inference for intelligent health monitoring.

Development of a p72 trimer–based colloidal gold strip for detection of antibodies against African swine fever virus

Geng Rui,Sun Yaning,Li Rui,Yang Jifei,Ma Hongfang,Qiao Zixuan,Lu Qingxia,Qiao Songlin,Zhang Gaiping

Abstract African swine fever virus (ASFV) causes a highly contagious and often lethal swine viral disease, and leads to tremendous economic losses to the swine industry. Unfortunately, there are no vaccines and effective antiviral agents available to prevent and control ASFV outbreaks. Therefore, it is necessary to develop simple and rapid strategies to monitor ASFV-infected pigs to restrain its spread. In the current study, ASFV capsid protein p72 was expressed along with its chaperone pB602L to form trimers in human embryonic kidney 293 (HEK293) cells. The p72 trimers were subsequently labeled with colloidal gold to develop a immunochromatographic strip. The strip showed high specificity to ASFV-positive serum and no cross-reactivity to other swine virus positive sera. Importantly, the strip showed a higher sensitivity of detecting ASFV antibodies in both positive standard serum and clinical serum samples than a commercial enzyme-linked immunosorbent assay (ELISA) kit. Taken together, these results demonstrate the strip as a reliable diagnostic tool against ASFV infection, which will be appropriate for application in prevention and control of ASFV. Key points • ASFV p72 trimers were successfully generated . • A colloidal gold strip was developed based on ASFV p72 trimers . • The strip is appropriate for detecting ASFV antibodies in the field .

A portable smartphone-based hemoglobin point-of-care testing platform for accurate anemia diagnostics

Qiangqiang Fu,Tao Qi,Ze Wu,Yongjian He,Shujuan Guan,Shihua Luo,Qi Zhang,Wenfeng Luo,Wei Xiao,Bo Situ,Lei Zheng

Anemia affects over 2 billion people worldwide, with the heaviest burden borne by women and children. At present, anemia is diagnosed by measuring hemoglobin (Hb) levels, which must be done in hospitals or commercial laboratories by skilled operators. In this work, we report a portable, affordable ($3), easy-to-operate (1 min) and accurate smartphone-based Hb analyzer (SHbA) that uses a drop of finger-pricked blood for anemia point-of-care test (POCT) applications. POCT of Hb was achieved using a smartphone ambient light sensor (ALS) to accurately measure the absorbance of colorimetric Hb biochemical analysis reagents in a microcuvette, as well as an Android-based application for results analysis. SHbA validation results agreed well with those reported by a hematology analyzer, and the SHbA has an anemia diagnosis sensitivity of 95.4% and specificity of 96.3% for venous blood (n = 360) and a sensitivity of 96.39% and specificity of 95.58% for fingertip blood (n = 475). In addition, SHbA exhibits excellent performance in the diagnosis and treatment guidance of anemia high-risk populations, including tumor chemotherapy patients (n = 424), pregnant women (n = 214) and thalassemia patients (n = 208). Importantly, volunteer self-testing results (n = 20) indicate that SHbA can be used for home-based anemia diagnosis and monitoring. SHbA has the advantages of high sensitivity and specificity while being cheap and easy to operate, making it widely applicable for the diagnosis and treatment of anemia, especially for high-risk patients in areas with poor medical resources.

A portable, one-tube CRISPR/Cas12a-based biosensor for sensitive and colorimetric detection of norovirus in foods without pre-amplification

Caihong Zhou,Jingjing Fan,Liangyuan Wang,Baoxue Gao,Chunhui Wei,Long Ma,Lijuan Yin

Norovirus (NoV) is a leading cause of nonbacterial acute gastroenteritis, primarily transmitted through contaminated food and water. Rapid, sensitive, and on-site detection is crucial for controlling NoV outbreaks and ensuring food safety. In this study, we developed a portable, one-tube CRISPR/Cas12a-driven colorimetric biosensor for the rapid and sensitive detection of NoV RNA without the need for pre-amplification. The biosensor integrates magnetic nanoprobes with gold nanoparticle (AuNP) bioprobes, connected via single-stranded DNA (ssDNA) linkers. Upon the specific recognition of NoV cDNA, the Cas12a protein’s trans -cleavage activity is activated, cleaving the ssDNA linkers and releasing AuNP bioprobes from the magnetic nanoprobes. This release results in a visually detectable red color change and enhanced UV–visible absorbance in the solution following magnetic separation. These changes can be easily detected using portable devices such as microplate readers or smartphones. This method achieves a detection limit of 10 copies/µL within 60 min, with no cross-reactivity from other viruses or bacteria. Validation using oyster samples showed perfect concordance with qPCR results. Designed for resource-limited settings, our biosensor can be adapted for detecting various pathogens by altering the target nucleic acids and crRNA sequences. This advancement offers significant potential for enhancing rapid diagnostics and outbreak management.

A capillary-based centrifugal indicator equipped with in situ pathogenic bacteria culture for fast antimicrobial susceptibility testing

Longyu Chen,Meijia Zhu,Zhiyong Wang,Hongliang Wang,Yongqiang Cheng,Ziwei Zhang,Xiaoxiao Qi,Yifan Shao,Xi Zhang,Hongwei Wang

Antimicrobial resistance has become a major global health threat due to the misuse and overuse of antibiotics. Rapid, affordable, and high-efficiency antimicrobial susceptibility testing (AST) is among the effective means to solve this problem. Herein, we developed a capillary-based centrifugal indicator (CBCI) equipped with an in situ culture of pathogenic bacteria for fast AST. The bacterial incubation and growth were performed by macro-incubation, which seamlessly integrated the capillary indicator. Through simple centrifugation, all the bacterial cells were confined at the nanoliter-level capillary column. The packed capillary column height could linearly reflect the bacterial count, and the minimum inhibitory concentration (MIC) was determined based on the difference in the column height between the drug-added groups and the control group. The AST results could easily be determined by the naked eye or smartphone imaging. Thus, the CBCI realized the combination of macro-bacterial incubation and early micro assessment, which accelerated the phenotypic AST without complex microscopic counting or fluorescent labelling. The whole operation process was simple and easy to use. AST results could be determined for E. coli ATCC strains within 3.5 h, and the output results for clinical samples were consistent with the hospital reports. We expect this AST platform to become a useful tool in limiting antimicrobial resistance, especially in remote/resource-limited areas or in underdeveloped countries.

A Multiplexed Three-Channel Detection System for Rapid Home-Based Diagnosis of Respiratory Viruses

Xuhui Huang,Yatian Zhang,Jingwei Chen,Zhijin Yang,Mengya Chen,Zhiwei Xue,Lizan Zhou,Zhaofeng Huang,Yule Zhang,Lulu Zheng,Dawei Zhang

Acute respiratory syndrome coronavirus (SARS-CoV-2), influenza A virus (Flu A), and influenza B virus (Flu B) are highly contagious pathogens that can cause similar clinical manifestations and often co-infect, posing significant challenges for differential diagnosis and disease management. Early and accurate detection of these viruses is crucial for curbing their respiratory transmission. Therefore, there is an urgent need to develop a reliable, rapid, sensitive, and user-friendly tool for detecting multiple respiratory pathogens. Here, we present a triple-channel immunochromatographic system (TCIS) based on quantum dot fluorescent microspheres (QDFM), which includes immunochromatographic test strips and a detection platform integrated with optical and electronic modules. The TCIS utilizes a smartphone for fluorescence image extraction and filtering algorithms, enabling automatic image denoising and quantitative data analysis. Combined with high quantum yield luminescent materials, the system significantly enhances detection sensitivity. The limits of detection (LODs) for Flu B nucleoprotein, Flu A nucleoprotein, and SARS-CoV-2 N protein were as low as 0.301 ng/mL, 0.227 ng/mL, and 0.348 ng/mL, respectively. To evaluate its clinical performance, we tested saliva samples from 15 Flu A patients and 20 healthy controls, demonstrating a sensitivity of 100% and a specificity of 95.2%. This work highlights the potential of TCIS as an efficient, accessible, and accurate home-based diagnostic tool, offering a promising solution for early diagnosis.

CONVERT: Dynamic crRNA Reconfiguration for Universal One-Pot CRISPR/Cas12a-Based Nucleic Acid Detection

Songkuan Zhuang,Xiaojun Huang,Muyan Diao,Ruiling Dong,Yueyu Dong,Jun Liu,Yizhen Liu

Current one-pot CRISPR diagnostics necessitate meticulous control of nucleic acid hybridization parameters or extensive modification of CRISPR components to achieve complete enzymatic silencing, a fundamental bottleneck limiting their robustness and generalizability. Here, we resolve this challenge through dynamic crRNA reconfiguration, a paradigm-shifting strategy that exploits the intrinsic structural duality of CRISPR RNA. We present CONVERT (CRISPR One-Pot Nucleic acid detection Via Engineered crRNA Reconfiguration Technology), a universal platform where nontarget intact crRNA acts as a universal suppressor, achieving complete Cas12a inhibition during RPA (recombinase polymerase amplification) by irreversible enzyme sequestration. Target detection is initiated through programmable conversion to split crRNA activators, bypassing sequence-specific design constraints entirely. This crRNA conformational switching mechanism is implemented via photocleavable linker cleavage and subsequent assembly with presupplied truncated crRNA (tcrRNA), activating trans-cleavage signal amplification. Three transformative advances emerge: First, the endogenous crRNA engineering eliminates dependency on exogenous nucleic acid blockers or chemical modifications, reducing optimization costs and time. Second, near-total enzymatic suppression ensures zero cross-talk between amplification and detection phases. Third, the unified workflow enables contamination-resistant operation through spatiotemporal control, requiring no physical partitioning. Validated for Enterocytozoon bieneusi detection, the system demonstrates attomolar sensitivity of 1 aM, 100% diagnostic concordance with qPCR, and absolute specificity against related pathogens. By effectively decoupling CRISPR regulation from sequence-context constraints through rational crRNA structural plasticity, this work establishes a generalized framework for next-generation molecular diagnostics.

An energy-embodied paralleled liquid manipulation for equipment-free, quantitative multiplexed liver function monitoring

Chao Liang,Jingyang Zhao,Pingdong Wei,Kang Wei,Hanqing Jiang

Comprehensive liver function monitoring is vital for liver damage diagnoses. However, multiplexed testing with clinical accuracy often requires paralleled liquid manipulation to avoid reagent interference and nonspecific adsorption, making it restricted to centralized facilities and specialists. Here, we report an equipment-free, quantitative, and multiplexed point-of-care testing (POCT), the energy-embodied POCT (EE-POCT). The driving energy (pressured gas) and liquid reagents are embodied in separate rigid-flexible chambers through a mechanical pressure sensing mechanism to achieve a paralleled, interference-free, and multiple-reagent operation with no need for external equipment. The EE-POCT achieves quantitative results for nine liver-related indicators from 180 microliters of whole blood within 4 minutes. Validated with 103 clinical specimens, it shows high concordance with a clinical gold standard analyzer (Pearson coefficients >0.9). The EE-POCT platform not only advances liver health diagnostics but also sets a precedent for developing versatile, accessible medical technologies that can significantly affect global health outcomes.

Dual-signal assay integrating colorimetric and pH biosensing pathways for simultaneous detection of multiple pathogenic bacteria

Jin Wen,Zhaoyang Fang,Shuyao Chen,Wanyi Xu,Yongjun Liu,Hongwen Wu,Hongliang Tan

The increasing prevalence of multi-pathogen contamination highlights the critical need for advanced diagnostic technologies capable of accurately detecting multiple pathogens simultaneously. In this study, we developed a dual-signal assay that synergistically integrates colorimetric and pH biosensing pathways to detect multiple pathogenic bacteria simultaneously. This assay employs the unique properties of rhodamine B (RhB) and urease to create two distinct and independent signaling transduction pathways: RhB provides a characteristic colorimetric absorbance, while urease catalyzes the hydrolysis of urea, generating a pH-modulating signal. The spatiotemporal separation between RhB absorbance and the pH signal can effectively eliminate crosstalk interference, ensuring exceptional accuracy and reliability. Implemented through a dual-recognition strategy, the assay uses Fe 3 O 4 -functionalized phenylboronic acid nanoparticles to capture a wide range of bacteria. Pathogen-specific probes, featuring immunoglobulin G- and aptamer-conjugated ZIF-90 composites encapsulating either RhB or urease, enable precise bacterial identification. The assay exhibits remarkable sensitivity, with detection limits as low as 6 CFU/mL for Staphylococcus aureus and 8 CFU/mL for Escherichia coli . Moreover, it demonstrates excellent accuracy in actual samples, with recovery rates exceeding 97.75 % and a relative standard deviation below 4.99 %. We believe that this study can offer a new insight into the development of multiplex assays for simultaneous multi-target detection.

Huaqing Sun,Jian Li,Liang Li,Chunyu Wang,Xiujuan Liu,Ming Shi,Zhiwei Liu,Kun Li

Lung cancer is the most common malignancy worldwide. Early diagnosis helps to reduce mortality and improve survival. Aptamers are widely used in cancer screening because of their high specificity, good stability and low cost. In this study, using the specific aptamer of lung cancer serum, the sandwich method colloidal gold test strip was prepared by isothermal amplification technique and the principle of nucleic acid hybridisation for the early diagnosis of lung cancer. The results showed that the test strip was positive in 8 patients with lung cancer, which was consistent with the actual cases. The test strip can accurately identify lung cancer patients. The concentration range of nucleic acid detection is 1 × 10 −4 - 7 × 10 −4 mol/L, and the detection limit is 0.67 mM. The test strip detection method has low cost and simple operation, and provides a reference for the development of home portable tumor early detection.

Development of plate-type and tubular chemiluminescence immunoassay against African swine fever virus p72

Miao Chun,Shao Junjun,Yang Sicheng,Wen Shenghui,Ma Yunyun,Gao Shandian,Chang Huiyun,Liu Wei

Abstract African swine fever (ASF) is a highly contagious and fatal viral disease that has caused huge economic losses to the pig and related industries worldwide. At present, rapid, accurate, and sensitive laboratory detection technologies are important means of preventing and controlling ASF. However, because attenuated strains of African swine fever virus (ASFV) are constantly emerging, an ASFV antibody could be used more effectively to investigate the virus and control the disease on pig farms. The isolation of ASFV-specific antibodies is also essential for the diagnosis of ASF. Therefore, in this study, we developed two chemiluminescence immunoassays (CLIAs) to detect antibodies directed against ASFV p72: a traditional plate-type blocking CLIA (p72-CLIA) and an automatic tubular competitive CLIA based on magnetic particles (p72-MPCLIA). We compared the diagnostic performance of these two methods to provide a feasible new method for the effective prevention and control of ASF and the purification of ASFV. The cut-off value, diagnostic sensitivity (Dsn), and diagnostic specificity (Dsp) of p72-CLIA were 40%, 100%, and 99.6%, respectively, in known background serum, whereas those of p72-MPCLIA were 36%, 100%, and 99.6%, respectively. Thus, both methods show good Dsn, Dsp, and repeatability. However, when analytical sensitivity was evaluated, p72-MPCLIA was more sensitive than p72-CLIA or a commercial enzyme-linked immunosorbent assay. More importantly, p72-MPCLIA reduced the detection time to 15 min and allowed fully automated detection. In summary, p72-MPCLIA showed superior diagnostic performance and offered a new tool for detecting ASFV infections in the future. Key points • Two chemiluminescence immunoassay (plate-type CLIA and tubular CLIA) methods based on p72 monoclonal antibody (mAb) were developed to detect ASFV antibody. • Both methods show good diagnostic performance (Dsn (100%), Dsp (99.6%), and good repeatability), and p72-MPCLIA detects antibodies against ASFV p72 with high efficiency in just 15 min.
