Imaging techniques of upper tract urothelial carcinoma (UTUC) are presently limited. Upconversion particles (UCPs) could be used to target tumors for imaging. The present study aimed to assess the value of a nano‑UCP as a diagnostic probe for deep tumor tissue, including UTUC. Polymer‑coated water‑soluble UCPs were synthesized. The pH Low Insertion Peptide (pHLIP) polypeptide was synthesized using the solid phase method. The silane shell surface was modified to present amino or carboxyl groups. Succinimidyl 4‑(N‑maleimidomethyl)cyclohexane‑1‑carboxylate was used for the coupling of the polypeptide to the UCPs. An animal model of subcutaneous tumor was established in 4‑week old nude mice using UTUC cells. Urinary tract epithelial cancer T24 cells were injected into the diaphragm below the heart. PHLIP‑UCP solution (1 ml) was injected into the abdominal cavity of each animal. Optical detection was performed using a small animal living body multispectral imaging system. UCPs dispersed in chloroform emitted no light under natural light, while they emitted a green light when excited with a 980‑nm laser. The maximum emission wavelength of Ho3+‑doped UCPs was ~550 nm and the red emission region was ~650 nm. As the coated UCPs possessed a tendency to agglomerate and precipitate, the yield of the UCPs in the aqueous phase was reduced. Tumors could be successfully imaged in tumor‑bearing mice. NaYF4: Yb, Ho3+ UPCs could be used for the detection of UTUC, thus further studies are required to determine if it could be used in larger animals with deeper tumors.

Novel nanoparticulate contrast agents with low systemic toxicity and inexpensive character have exhibited more advantages over routinely used small molecular contrast agents for the diagnosis and prognosis of disease. Herein, we designed and synthesized PEGylated hybrid ytterbia nanoparticles as high-performance nanoprobes for X-ray computed tomography (CT) imaging and magnetic resonance (MR) imaging both in vitro and in vivo. These well-defined nanoparticles were facile to prepare and cost-effective, meeting the criteria as a biomedical material. Compared with routinely used Iobitridol in clinic, our PEG-Yb2O3:Gd nanoparticles could provide much significantly enhanced contrast upon various clinical voltages ranging from 80 kVp to 140 kVp owing to the high atomic number and well-positioned K-edge energy of ytterbium. By the doping of gadolinium, our nanoparticulate contrast agent could perform perfect MR imaging simultaneously, revealing similar organ enrichment and bio-distribution with the CT imaging results. The super improvement in imaging efficiency was mainly attributed to the high content of Yb and Gd in a single nanoparticle, thus making these nanoparticles suitable for dual-modal diagnostic imaging with a low single-injection dose. In addition, detailed toxicological study in vitro and in vivo indicated that uniformly sized PEG-Yb2O3:Gd nanoparticles possessed excellent biocompatibility and revealed overall safety.

Efficient delivery of photons to visceral organs is critical for the treatment of deep-seated tumors taking advantage of photo theranostics. Optical fiber can be regarded as a direct and facile photon pathway for targeting tumor lesion. However, current fiber theranostic strategies rely on the spatially separated optical fibers to realize diagnosis and therapy independently, resulting in low compactness, poor continuity of medical process, and incompatibility with current medical technologies. Herein, an integrated fiber-optic theranostic (iFOT) probe is developed that merges tumor microenvironment sensing and photothermal therapy by functionalizing the fiber with graphene/gold nanostar hybrid materials and hypoxic-responsive fluorophores. The iFOT probe can quickly detect the hypoxia of xenograft tumors of mice with high sensitivity. The tumors can be photothermally killed on-site through the same fiber probe tightly followed by detection, which presents a high cure rate. More importantly, the iFOT is highly adaptable to the conventional medical imaging and endoscopic techniques, which facilitates the imaging-assisted navigation and manipulation by use of the interventional trocar. The proposed strategy can be used as an effective endoscopic and interventional tool for tackling deep-situated tumor and may open a revolutionized pathway to bridge separate diagnosis and therapy process in the current stage.

Subcellular organelle targeted imaging and therapy are of enormous interest in cancer theranostics. However, the lack of tumor-selective organelle targeting has compromised their efficacy and safety. In this work, we found that the near-infrared (NIR) fluorophore hemicyanine (CyNH 2 ) can selectively target mitochondria with strong cytotoxicity through decreasing the mitochondrial membrane potential and increasing the intracellular reactive oxygen species (ROS) levels. A macrotheranostic probe (denoted as PLCy) based on conjugating CyNH 2 with an acetylated lysine group was developed with masked fluorescence and cytotoxicity, which could both be unmasked through sequential activation by cancer cells overexpressing histone deacetylases (HDACs) and cathepsin L (CTSL) enzymes for selective cancer cell mitochondria-targeted imaging and therapy. In vitro and in vivo studies confirmed that the specific fluorescence turn-on and toxicity were restored in cancer cells and efficiently inhibited tumor growth. This macrotheranostic probe with sequential enzyme activation and mitochondrial targeting is expected to have promising applications in imaging-guided cancer therapy with high specificity and efficiency. Statement of significance To improve the targeting efficiency and enhance the anti-cancer activities of macrotheranostic probe. We designed macrotheranostic probe PLCy that can be activated via sequential enzymes for selective tumor mitochondria targeting. More importantly, the activated CyNH 2 can decrease the mitochondrial membrane potential and elevate the reactive oxygen species level in cancer cells without light irradiation, which can further induce apoptosis of tumor cells for chemotherapy. Therefore, the use of sequential enzyme activation and mitochondria targeting strategies in the context of enzymatic activation may provide a general strategy for organelle-targeted imaging and therapy with high specificity and efficiency.

Non-alcoholic fatty liver disease (NAFLD) is a global health concern, but early diagnosis remains challenging due to the limitations of current methods, which are often invasive or lack sensitivity. To address this, we developed UIO-YB, a dual-responsive fluorescence probe combining water-soluble UIO carriers with a polarity-sensitive dye (YB) for precise NAFLD detection. UIO-YB exhibits dual fluorescence emission (red for YB, blue for UIO), enabling simultaneous detection of lipid accumulation (via YB’s polarity response) and oxidative stress markers (via UIO’s RNS reactivity). In vitro, UIO-YB detected lipid droplets in HepG2 cells within 2 h, surpassing Oil Red O in speed and simplicity. In vivo, it accurately visualized Hepatic steatosis in mice after 6 weeks of a high-fat diet, demonstrating high specificity and sensitivity. Unlike conventional dyes, UIO-YB operates without organic solvents, simplifying protocols while reducing background noise. UIO-YB’s water solubility and dual-response design overcome key limitations of existing probes, offering a rapid, scalable tool for early NAFLD detection. Its performance in cellular and preclinical models highlights its potential to advance NAFLD research and clinical diagnostics. This innovation paves the way for non-invasive, high-sensitivity imaging to improve disease monitoring and intervention. Graphical
