A carboxylesterase-activatable near-infrared phototheranostic probe for tumor fluorescence imaging and photodynamic therapy

Lihong Li,Qi Zhang,Jiaojiao Li,Yafei Tian,Jinyao Li,Wen Liu,Haipeng Diao

Phototheranostic probes have been proven to be a promising option for cancer diagnosis and treatment. However, near-infrared phototheranostic probes with specific tumor microenvironment responsiveness are still in demand. In this paper, a carboxylesterase (CES)-responsive near-infrared phototheranostic probe was developed by incorporating 6-acetamidohexanoic acid into a hemicyanine dye through an ester bond. The probe exhibits highly sensitive and selective fluorescence enhancement towards CES because CES-catalyzed cleavage of the ester bond leads to the release of the fluorophore. By virtue of its near-infrared analytical wavelengths and high sensitivity, the probe has been employed for endogenous CES activatable fluorescence imaging of tumor cells. Moreover, under 660 nm laser irradiation, the probe can generate toxic reactive oxygen species and efficiently kill tumor cells, with low cytotoxicity in dark. As far as we know, the probe was the first CES-responsive phototheranostic probe with both near-infrared analytical wavelengths and photosensitive capacity, which may be useful in the real-time and in situ imaging of CES as well as imaging-guided photodynamic therapy of tumors. Therefore, the proposed probe may have wide application prospect in cancer theranostics.

Dual-Triggered Near-Infrared Persistent Luminescence Nanoprobe for Autofluorescence-Free Imaging-Guided Precise Therapy of Rheumatoid Arthritis

Ruoping Wang,Junpeng Shi,Qian Zhang,Qiang Peng,Xia Sun,Liang Song,Yun Zhang

Rheumatoid arthritis (RA) is a common, chronic, and highly disabling autoimmune disease characterized by difficult treatment, long disease duration, and easy recurrence. The development and application of high-sensitivity theranostic probes for RA that will facilitate precise monitoring of disease progression and enable effective treatment are currently hotspots in the field of RA theranostics. In this study, mZMI@HA, a dual-triggered theranostics nanoprobe, is constructed based on near-infrared persistent luminescence nanoparticles (NIR-PLNPs) for precise RA treatment and therapeutic evaluation. This is the first reported use of high-sensitivity autofluorescence-free imaging based on NIR-PLNPs for precise RA treatment and therapeutic evaluation. Compared with the NIR fluorescence imaging probe-indocyanine green, the signal-to-background ratio of persistent luminescence (PersL) imaging is improved nearly 14-fold. Using PersL imaging to guide photothermal therapy and controllable drug release through NIR/pH-responsiveness, the progress of collagen-induced RA is relieved. Additionally, the therapeutic evaluation of RA by PersL imaging is consistent with clinical micro-computed tomography and histological analyses. This study demonstrates the potential of NIR-PLNPs for high-sensitivity imaging-guided RA treatment, providing a new strategy for RA precise theranostics.

Iridium oxide nanoparticles-based theranostic probe for in vivo tumor imaging and synergistic chem/photothermal treatments of cancer cells

Hui Zhang,LianXiao Zhang,Hua Zhong,Shuyan Niu,Caifeng Ding,Shaoping Lv

A novel iridium oxide nanoparticles-based probe was developed for in vivo fluorescence imaging of microRNA and synergistic chem/photothermal treatments of tumor. The theranostic nanoprobe was constructed by functionizing iridium oxide nanoparticles with split DNAzyme precursor and chemotherapeutic agent doxorubicin. The split DNAzyme precursor was used for specifically recognizing the target microRNA in cancer cells and then releasing the preloaded fluorescence for detection through DNAzyme cleavage reaction. Because the fluorescence was enhanced by cycle amplification process, the nanoprobe exhibited high sensitivity in the cell-free test. Both in vitro and in vivo fluorescence imaging studies proved the specificity and feasibility of the nanoprobe in tumor imaging. In order to enhance the effectiveness of cancer cell therapy, synergistic chem/photothermal treatments were designed on the nanoprobe. Based on the photothermally controlled drug release and chemotherapeutic treatment of doxorubicin, and the photothermal effect of iridium oxide nanoparticles, the nanoprobe exhibited excellent synergy effect on cancer cell treatment under near-infrared irradiation. In particular, in vivo therapeutic studies proved a great inhibition of the tumor growth. Therefore, this work provided a promising theranostic nanoprobe for both tumor imaging and synergistic treatment of cancer cells.

Photosensitizer-Free Nanophosphor-Based theranostic probe for controllable photodynamic and photothermal visualization therapy

Mengjie Sun,Zhouyu Zhang,Chang Yin,Zi-Jin Wei,Zichao Yan,Kai Long,Yuan Tian,Yifei Wang,Wei Wang,Zhi Yuan

Photodynamic therapy (PDT) has emerged as a powerful candidate for cancer intervention as for the merits of least-invasiveness, multidrug resistance-free and spatiotemporal selectivity. Whereas, clinical photosensitizers (PSs) suffer from microsecond-level reactive oxygen species (ROS) lifetime requiring 20–30 min continuous exterior light stimulation, deficient photostable nature and off-target phototoxicity in nonmalignant tissues. Persistent luminescence nanoparticles (PLNPs), are proven to exert long-lasting ROS production after the excitation cessation (hours to days), and serve as photostable imaging agent for diagnose. Herein, a traceable theranostic agent, ZP@PCM PLNPs is prepared by modifying polyaniline (PANI) onto Zn 1.2 Ga 1.6 Ge 0.2 O 4 : Cr 3+ (ZGGC) to obtain ZP PLNPs and wrapping phase-change material (PCM) as outer protective shell. Solid-phase PCM isolates inner ZP PLNPs from surrounding oxygen during circulation to circumvent off-target photosensitization. Once ZP@PCM PLNPs accumulated within tumors, external laser triggered PANI assisted photothermal therapy (PTT) initiates, and thermal melting of PCM shell occurs leading to the exposure of ZP PLNPs to switch on PDT. PANI also facilitates PDT efficacy optimization, enabling ZP PLNPs of 2-fold ROS yield compared to ZGGC PLNPs, sustained up to 48 h upon 10 min of illumination. ZP@PCM PLNPs with long-term, moderate therapy modality presents ameliorated anti-tumor effect, achieving tumor inhibition rate of 92.2 % over that of ∼ 46 % with individual ZGGC PLNPs, and shows satisfactory biosafety without tissues damage or inflammatory response. Meanwhile, the autofluorescence-free visualization potency helps to self-report therapeutic outcomes and determine treatment duration. Overall, ZP@PCM PLNPs provide a promising PSs-free alternative candidate for finely controlled PDT/PTT and visualization tumor therapy.

Mesoporous silica nanoparticles loaded with fluorescent coumarin-5-fluorouracil conjugates as mitochondrial-targeting theranostic probes for tumor cells

Xiaojing Yang,Di-Feng Chen,Lin-Song Li,Xue-Jie Zhao,Mei-Xia Zhao

In this study, a nanodrug carrier (mesoporous silica nanoparticle (MSN)-SS-cysteamine hydrochloride (CS)-hyaluronic acid (HA)) for targeted drug delivery was prepared using MSNs, in which HA was used as a targeting ligand and blocking agent to control drug release. Coumarin is a fluorescent molecule that targets mitochondria. Two conjugates (XDS-DJ and 5-FUA-4C-XDS) were synthesized by chemically coupling nitrogen mustard and 5-fluorouracil with coumarin, which was further loaded into MSN-SS-CS-HA nanocarriers. MTT analysis demonstrated that the nanocomposite MSN-SS-CS@5-FUA-4C-XDS/HA displayed stronger cytotoxicity toward HCT-116 cells than HeLa or QSG-7701 cells. Furthermore, MSN-SS-CS@5-FUA-4C-XDS/HA was able to target the mitochondria of HCT-116 cells, causing decreased mitochondrial membrane potential and excessive production of reactive oxygen species. These results indicate that MSN-SS-CS@5-FUA-4C-XDS/HA has the potential to be a nanodrug delivery system for the treatment of colon cancer.
