Proton nanomodulators for enhanced Mn2+-mediated chemodynamic therapy of tumors via HCO3− regulation

Yang Peng,Liu Shaojie,Chen Zhuang,Liu Weijing,Duan Deshang,Yang Zuo,Yan Haohao,Rao Zhiping,Zhang Xianghan,Zhang Ruili,Wang Zhongliang

Background Mn 2+ -mediated chemodynamic therapy (CDT) has been emerged as a promising cancer therapeutic modality that relies heavily on HCO 3 − level in the system. Although the physiological buffers (H 2 CO 3 /HCO 3 − ) provide certain amounts of HCO 3 − , the acidity of the tumor microenvironment (TME) would seriously affect the HCO 3 − ionic equilibrium (H 2 CO 3 ⇌ H + + HCO 3 − ). As a result, HCO 3 − level in the tumor region is actually insufficient to support effective Mn 2+ -mediated CDT. Results In this study, a robust nanomodulator MnFe 2 O 4 @ZIF-8 (PrSMZ) with the capability of in situ self-regulation HCO 3 − is presented to enhance therapeutic efficacy of Mn 2+ -mediated CDT. Under an acidic tumor microenvironment, PrSMZ could act as a proton sponge to shift the HCO 3 − ionic equilibrium to the positive direction, significantly boosting the generation of the HCO 3 − . Most importantly, such HCO 3 − supply capacity of PrSMZ could be finely modulated by its ZIF-8 shell thickness, resulting in a 1000-fold increase in reactive oxygen species (ROS) generation. Enhanced ROS-dependent CDT efficacy is further amplified by a glutathione (GSH)-depletion ability and the photothermal effect inherited from the inner core MnFe 2 O 4 of PrSMZ to exert the remarkable antitumor effect on mouse models. Conclusions This work addresses the challenge of insufficient HCO 3 − in the TME for Mn 2+ -mediated Fenton catalysts and could provide a promising strategy for designing high-performance Mn 2+ -mediated CDT agents to treat cancer effectively.

OH Regulator of Amorphous CrOx on Defect-Rich Ultrafine Pd Nanowires Boosts Electrocatalytic Ethanol Oxidation

Xiaotong Zhou,Songliang Liu,Huaifang Teng,Kun Ma,Weixin Miao,Xuejing Cui,Xin Zhou,Luhua Jiang

Reasonable construction of high activity and selectivity electrocatalysts is crucial to achieve efficient ethanol oxidation reaction (EOR). However, the oxidation of ethanol tends to produce CO species that poison the active centers of the EOR electrocatalysts. Herein, a unique amorphous CrO x -protected defect-rich ultrafine Pd nanowires (CrO x -Pd NWs) is developed. On the one hand, the CrO x layer can act as a protective layer to maintain the structure of the nanowire. On the other hand, it can play the role of OH regulator to optimize the adsorption energy barrier of intermediate species in Pd nanowire, thereby enhancing the ability of the catalyst to resist CO poisoning. The CrO x -Pd NWs exhibit excellent EOR performance with 3.64 times higher mass activity and 50 mV lower CO electro-oxidation potential than commercial Pd black. The results show that the CrO x layer promotes the dissociation of H 2 O into OH ads , while the CrO x transfers electrons to neighboring Pd atoms optimizing the electronic configuration of Pd, thus selectively oxidizing ethanol to acetate and preventing the formation of toxic * CO. This work provides an effective strategy for the synthesis of nanowire materials with oxide/metal interfaces and offers new ideas for the design of catalysts that can efficiently drive EOR.

A MXene Modulator Enabled High-Loading Iodine Composite Cathode for Stable and High-Energy-Density Zn-I2 Battery

Dandan Li,Ying-Jie Zhu,Long Cheng,Sida Xie,Han-Ping Yu,Wei Zhang,Zhenming Xu,Ming-Guo Ma,Heng Li

Achieving both high iodine loading cathode and high Zn anode depth of discharge (DOD) is pivotal to unlocking the full potential of energy-dense Zn-I 2 batteries. However, this combination exacerbates the detrimental shuttle effect of polyiodide intermediates, significantly impairing the battery's reversibility and stability. Herein, this study reports an advanced high-loading iodine cathode (denoted as MX-AB@I) enabled by a multifunctional Ti 3 C 2 T x MXene modulator, which presents high stability and energy density in Zn-I 2 batteries. Through comprehensive experimental and theoretical analyses, the intrinsic regulating mechanisms are elucidated by which the MXene modulator effectively suppresses polyiodide shuttling, enhances iodine conversion kinetics, and dramatically improves Zn anode reversibility. With the aid of the MXene modulator, the MX-AB@I composite cathode achieves a high iodine mass loading of 23 mg cm −2 and realizes a practically high areal capacity of 4.0 mAh cm −2 . When paired with a thin Zn anode (10 µm), this configuration realizes a high Zn DOD of 78.7% and a high energy density of 171.3 Wh kg −1 , surpassing the majority of Zn-I 2 battery systems reported in the literature. This study presents an effective approach to designing high-loading iodine cathodes for Zn-I 2 batteries by leveraging MXene modulators to regulate critical electrochemical reaction processes.

Thermal management of Li-ion batteries with passive thermal regulators based on composite PCM materials

Kaixuan Li,Xiaole Yao,Zhicheng Li,Tongtong Gao,Weijun Zhang,Zhirong Liao,Xing Ju,Chao Xu

For battery thermal management , the phase change material (PCM) has demonstrated its advantages as a thermal buffer. In this paper, we proposed a novel thermal regulator that intelligently utilizes the volume changes to modulate heat transfer. The thermal regulator establishes a passive and negative feedback mechanism between the PCM and the cooling system, enabling a consistent and optimal operating battery temperature. This innovation negates the need for sensors or external logic devices, using a resilient valve passively adjusted by PCM volume changes. This in turn modulates coolant flow rate and battery temperature. Its thermal management performance is compared to other configurations like natural cooling, pure PCM, and composite PCM (cPCM). Extended dynamic tests are conducted at different ambient temperatures ( T amb ) for evaluation. The results indicate that batteries without thermal regulators fail to operate when T amb exceeds 30 °C. At T amb  = 35 °C, the thermal regulator with cPCM exhibits repeated switching and effectively keeps the battery temperature lower than 38.13 °C. At T amb  > 40 °C, both thermal regulators active to achieve faster heat dissipation. At 45 °C, the thermal regulator with cPCM successfully reduces the battery temperature to 35.02 °C. Integrating AlN in pure PCM lowers the battery module's peak temperature by 7.94 %. The thermal regulator also diminishes cyclic temperature variances, underlining its potential for enhancing battery thermal management .

Multifunctional Iron-Doped Hollow Mesoporous Silica Nanoregulator for Enhanced Tumor Chemodynamic Therapy

Jing Ling,Bei-Bei Wang,Xia-Nan Li,Rui Zhang,Ling-Feng Pan,Han Zhang,Ye-Ting Hu,Shi-Bo Wang,Xiang-Dong Kong

Chemodynamic therapy (CDT) is a promising method that uses endogenous hydrogen peroxide (H 2 O 2 ) to produce cytotoxic hydroxyl radicals (•OH) via Fenton reaction to kill tumor cells. However, the insufficient contents of H 2 O 2 and the presence of glutathione (GSH) can significantly reduce the therapeutic effect of CDT. Herein, a multifunctional nanoregulator (3-AT&MA@FHM) that combines Fe-doped hollow mesoporous silica nanoparticles (Fe-doped hMSN, or FHM) with 3-amino-1,2,4-triazole (3-AT) and maleimide (MA) are developed to overcome these challenges. After endocytosis by tumor cells, FHM part of the nanoregulator degrades in a mildly acidic intracellular environment and releases Fe 3+ for CDT. The subsequently released 3-AT serves as a catalase inhibitor to promote the accumulation of H 2 O 2, while MA acts as a GSH scavenger to decrease the GSH content in tumor cells. This multifunctional nanoplatform simultaneously regulates the contents of H 2 O 2 -the substrate for Fenton reaction and GSH-the main antioxidant, resulting in a significantly enhanced CDT effect. Moreover, organoids are used for safety and toxicity evaluation. The results of organoids experiments showed similar trends to those of cellular experiments, but MIO is more resistant to stress than cells. This study is expected to provide a novel idea for the design of highly efficient CDT nanosystems.

All-Solid-State Transparent Variable Infrared Emissivity Devices for Multi-Mode Smart Windows

Hulin Zhang,Xiang Zhang,Wenhai Sun,Mingjun Chen,Yingjun Xiao,Zhenmin Ding,Dukang Yan,Jianbo Deng,Zitong Li,Jiupeng Zhao,Yao Li

Smart windows that incorporate photothermal modulators (PMs) can independently regulate solar transmittance and infrared (IR) emissivity to improve building comfort and reduce energy consumption. Herein, a novel all-solid-state variable IR emissivity device (VED) is first designed and fabricated with a high visible irradiation transmittance ( T ′ vis = 0.79) and solar irradiation transmittance ( T ′ sol = 0.75) using an ITO/SiO 2 /ITO Fabry–Perot cavity structure. The VED exhibits different IR emissivity (ɛ) values at positive (ɛ P = 0.80) and negative (ɛ N = 0.38) bias, allowing for dynamic regulation of radiation by controlling the electrical conductivity of the indium tin oxide (ITO) layer. Furthermore, an all-solid-state PM with a structure of ITO/SiO 2 /ITO/Glass/ITO/NiO/ZrO 2 /Li/WO 3 /ITO, which is capable of independently regulating solar transmittance (Δ T' sol = 0.31) and IR emissivity (Δɛ 2.5–25 µm = 0.42), is fabricated. The multi-mode smart window incorporating PMs can achieve “bright,” “dark,” “warm,” and “cool” modes, making them suitable for deployment in diverse climate zones. The innovative smart window holds a massive potential for use in reducing building energy consumption.

Tumor-Specific Activated Polymeric Nanotuners Disrupt Positive Feedback Cycle of Hypoxia and Apoptosis Evasion for Potent Cancer Radiotherapy

Xiaoxue Hou,Chun Wang,Yu Zhao,Qian Wang,Dianyu Wang,Jingyu Zhao,Yang Liu,Fan Huang,Jianfeng Liu

Effective cancer radiotherapy is usually hindered by the self-perpetuating feedback cycle between tumor hypoxia and apoptosis evasion. Herein, a tumor-specific activated polymeric nanotuner is developed to boost radiotherapy outcomes by disrupting this vicious cycle. The designed nanotuner is composed of a proapoptotic peptide-engineered catalase core and a pH-detachable polymer shell. They can maintain the core-shell structure to against immune clearance and enzymatic degradation under the “turn-off” state. When reaching the tumor site, the nanotuners hold acid-responsive “turn-on” property by dissociating the polymeric shell, facilitating the tumor accumulation and cellular internalization of the exposed functional core. Subsequently, the internalized core of polymeric nanotuners efficiently decomposes endogenous hydrogen peroxide (H 2 O 2 ) into oxygen (O 2 ) for hypoxia alleviation, thus upregulating the expression of proapoptotic protein Smac. Furthermore, the apoptotic-inducing peptide modified on the core surface further boosts the Smac-induced apoptosis signal, intervening in tumor apoptosis evasion and ultimately realizing the efficient radiotherapeutic efficiency by blocking this vicious cycle. In vivo studies demonstrated that treatment with polymeric nanotuners remarkably enhances radiation-mediated tumor ablation without perceptible side effects. This study sheds light on the innovative attempt to specifically interfere with the feedback cycle in tumor radioresistance, pioneering the way for achieving safe and efficient cancer therapies.

All-Season Dual-Mode Thermolator Based on Radiative Cooling and Solar Heating Technology

Yunqi Wang,Xinyu Tan,Xiongbo Yang,Guiguang Qi,Chao Shi,Shengyu Chen

Passive radiative cooling and solar heating have received a great deal of scholarly attention as green and sustainable ways to maintain thermal homeostasis. However, most of the current passive radiative cooling and solar heating materials are optically static and cannot adapt to seasonal dynamic weather changes. Herein, we developed an HPC/CaCl2/C dual-mode device (HCC) that autonomously senses changes in ambient temperature to switch cooling/heating modes. When the ambient temperature reaches the low critical solution temperature (LCST ≈ 37 °C) of the hydrogel, the cooling side of HCC gradually turns white to reflect sunlight and radiative cooling happens by means of a high infrared emissivity (0.97) in the atmospheric window band (8–13 μm). The heat-absorbing effect of the heated side accelerates the phase transition of the hydrogel and makes it change to the cooling mode more quickly. When the ambient temperature is higher than LCST, at an irradiance of 850 W/m2, the HCC-20 coating leads to a cavity cooling temperature of approximately 12.3 °C and the theoretical cooling power is 109.3 W/m2. When the ambient temperature is lower than the LCST, the upper hydrogel layer is transparent and HCC-20 transforms into a heating function. Under weather conditions with the irradiance of 500 W/m2 and a maximum ambient temperature of 6 °C, the average heating capacity is up to around 8 °C. The HCC dual-mode device can automatically switch between cooling and heating modes and has great application potential in fields such as buildings and automobiles, providing a favorable way to mitigate energy crises and reduce greenhouse gas emissions.

Tunable-emission microwire heterojunctions for optical waveguides

Hongyan Xia,Yibin Zhu,Xueliu Chen,Tingkuo Chen,Dao Zhang,Xiang Li,Fangcheng Shen,Haiming Jiang,Kang Xie

Multi-component hybrid optical heterojunctions assembled from chromophore-doped polymer microwires are one of the first choices of building blocks for the construction of complex/integrated functional photonic devices, because they not only have the advantages of the individual components (optical confinement from one-dimensional microstructures, high emission efficiency from chromophore and flexibility from polymer matrix), but also bring novel performances to the system by the synergetic interactions between multiple components which cannot be obtained from single components. Herein, we propose a strategy to construct different waveguide heterojunctions through arrangement and combination of two kinds of polymer doped optical microwires via micromanipulation. One of them (MW1) contains carboxyl-fluorescein and photochromic spiropyran which will undergo photoisomerization upon external light stimuli, producing tunable fluorescence (FL) emission for MW1 due to the adjustable fluorescence resonance energy transfer. The other (MW2) is embedded with the dye rhodamine 6G whose FL always remains almost unchanged. Waveguide logic gate, switch and modulator within these heterojunctions can be well realized by changing the external light stimuli and excitation position. Our study indicates that these tunable-emission microwire heterojunctions composed of responsive dye molecules would provide a novel tactic in developing multi-functional advanced waveguides for integrated micro/nanophotonic devices and circuits.

Roles of Response Regulators in the Two-Component System in the Formation of Stress Tolerance, Motility and Biofilm in Salmonella Enteritidis

Mengjun Hu,Zhuoan Zhou,Chenqi Liu,Zeqiang Zhan,Yan Cui,Shoukui He,Xianming Shi

Two-component systems (TCS) of Salmonella enterica serovar Enteritidis are composed of a histidine kinase and a response regulator (RR) and represent a critical mechanism by which bacteria develop resistance to environmental stress. Here, we characterized the functions of RRs in TCS in the formation of stress tolerance, motility and biofilm using twenty-six S . Enteritidis RR-encoding gene deletion mutants. The viability results unraveled their essential roles in resistance to elevated temperature (GlrR), pH alterations (GlrR, TctD, YedW, ArcA and YehT), high salt (PhoB, BaeR, CpxR, PhoP, UvrY and TctD), oxidative stress (PhoB, YedW, BaeR, ArcA, PhoP, UvrY, PgtA and QseB) and motility (ArcA, GlnG, PgtA, PhoB, UhpA, OmpR, UvrY and QseB) of S . Enteritidis. The results of the crystal violet staining, microscopy observation and Congo red binding assays demonstrated that the absence of ArcA, GlnG, PhoP, OmpR, ZraR or SsrB in S . Enteritidis led to a reduction in biofilms and an impairment in red/dry/rough macrocolony formation, whereas the absence of UvrY exhibited an increase in biofilms and formed a brown/smooth/sticky macrocolony. The results indicated the regulatory effects of these RRs on the production of biofilm matrix, curli fimbriae and cellulose. Our findings yielded insights into the role of TCSs, making them a promising target for combating S . Enteritidis.

Biodegradable calcium sulfide-based nanomodulators for H2S-boosted Ca2+-involved synergistic cascade cancer therapy

Chuchu Lin,Chenyi Huang,Zhaoqing Shi,Meitong Ou,Shengjie Sun,Mian Yu,Ting Chen,Yunfei Yi,Xiaoyuan Ji,Feng Lv,Meiying Wu,Lin Mei

Hydrogen sulfide (H 2 S) is the most recently discovered gasotransmitter molecule that activates multiple intracellular signaling pathways and exerts concentration-dependent antitumor effect by interfering with mitochondrial respiration and inhibiting cellular ATP generation. Inspired by the fact that H 2 S can also serve as a promoter for intracellular Ca 2+ influx, tumor-specific nanomodulators ( [email protected] ) have been constructed by encapsulating calcium sulfide (CaS) and indocyanine green (ICG) into methoxy poly (ethylene glycol) -b- poly (lactide -co- glycolide) (PLGA-PEG). [email protected] can achieve tumor-specific biodegradability with high biocompatibility and pH-responsive H 2 S release. The released H 2 S can effectively suppress the catalase (CAT) activity and synergize with released Ca 2+ to facilitate abnormal Ca 2+ retention in cells, thus leading to mitochondria destruction and amplification of oxidative stress. Mitochondrial dysfunction further contributes to blocking ATP synthesis and downregulating heat shock proteins (HSPs) expression, which is beneficial to overcome the heat endurance of tumor cells and strengthen ICG-induced photothermal performance. Such a H 2 S-boosted Ca 2+ -involved tumor-specific therapy exhibits highly effective tumor inhibition effect with almost complete elimination within 14-day treatment, indicating the great prospect of CaS-based nanomodulators as antitumor therapeutics.

Sialic Acid-Functionalized Pyroptosis Nanotuner for Epigenetic Regulation and Enhanced Cancer Immunotherapy

Jiahao Hu,Yuchan You,Luwen Zhu,Jucong Zhang,Yanling Song,Jingyi Lu,Xinyi Xu,Xiaochuan Wu,Xiajie Huang,Xiaoling Xu,Yongzhong Du

The efficacy of immune checkpoint blockade (ICB) in promoting an immune response against tumors still encounters challenges such as low response rates and off-target effects. Pyroptosis, an immunogenic cell death (ICD) mechanism, holds the potential to overcome the limitations of ICB by activating and recruiting immune cells. However, the expression of the pyroptosis-related protein Gasdermin-E(GSDME) in some tumors is limited due to mRNA methylation. To overcome this obstacle, sialic acid-functionalized liposomes coloaded with decitabine, a demethylation drug, and triclabendazole, a pyroptosis-inducing drug are developed. This nanosystem primarily accumulates at tumor sites via sialic acid and the Siglec receptor, elevating liposome accumulation in tumors up to 3.84-fold at 24 h and leading to the upregulation of pyroptosis-related proteins and caspase-3/GSDME-dependent pyroptosis. Consequently, it facilitates the infiltration of CD8 + T cells into the tumor microenvironment and enhances the efficacy of ICB therapy. The tumor inhibition rate of the treatment group is 89.1% at 21 days. This study highlights the potential of sialic acid-functionalized pyroptosis nanotuners as a promising approach for improving the efficacy of ICB therapy in tumors with low GSDME expression through epigenetic alteration and ICD.
