Rational construction of particle-in-tube structured NiO/CoO/polypyrrole as efficient nanozyme for biosensing

Wendong Zhu,Ya Cheng,Ce Wang,Xiaofeng Lu

Nanozymes are a new type of enzymes with ultrahigh environmental stability while facing the issues of low activity and poor specificity. Fabricating a confined structure has been verified to be an ideal strategy to optimize the catalytic performance of nanozymes. In this study, a particle-in-tube structured NiO/CoO/polypyrrole (PPy) is prepared via an electrospinning, calcination, vapor-phase deposition and thermal reduction route. Due to the novel confined structure and synergistic effect between different components, the boosted peroxidase (POD)-like properties are revealed. Furthermore, the prepared NiO/CoO/PPy nanotubes exhibit excellent long-term stability and multi-batch reproducibility for POD mimicking. In addition, a catalase (CAT)-like property is also achieved. Based on its desirable POD-like performance, a rapid colorimetric sensing platform to detect glutathione (GSH) is established with a low detection limit of 0.02 μM (S/N = 3) and favorable selectivity. We believe that this work presents a new strategy to construct high-efficiency nanozymes and excavates its potential in biosensing application.

A multifunctional carbon dots with near-infrared fluorescence for Au3+/Hg2+ and GSH detection and tumor diagnosis

Xiaojing Yang,Yingying Wang,Jingjing Xu,Mei-Xia Zhao

In recent years, carbon dots (CDs) have shown potential applications in fluorescence tracing and imaging due to their excellent optical and chemical properties. In this work, a new type of carbon dots (Cy7-CDs) with near-infrared absorption and emission was synthesized from cyanine dyes and polyethylene glycol by hydrothermal reaction. The prepared Cy7-CDs exhibited low toxicity and excellent biocompatibility, which were applied to detect the noble metal ions Au 3+ /Hg 2+ and glutathione via the ‘on–off-on’ mode. In addition, in vivo fluorescence images demonstrated that Cy7-CDs could efficiently accumulate in the tumor. Therefore, Cy7-CDs could act as a multi-functional probe for biological sensing and tumor diagnosis.

Fe3O4/Carbon Nanodot Hybrid Nanoparticles for the Indirect Colorimetric Detection of Glutathione

Na Luo,Zhengli Yang,Fenglin Tang,Dan Wang,Min Feng,Xiangjun Liao,Xiupei Yang

In recent years, carbon nanodots (CNDs)-based hybrid nanomaterials with enzyme-like properties have received considerable attention because of the simplicity of their preparation, storage, and separation, as well as cost-effectiveness compared with natural enzymes. Here, Fe3O4/CNDs hybrid nanoparticles (NPs) are prepared via a hydrothermal method, and they possess excellent peroxidase-like catalytic activity. During the synthesis, the formation and surface state of Fe3O4 NPs were extremely affected by solutions of CNDs, which served as solvents. Fe3O4/CNDs hybrid NPs with abundant hydroxyl and carboxyl groups exhibit better catalytic kinetics than natural horseradish peroxidase (HRP) to produce a rapid chromogenic reaction. Such characteristics from Fe3O4/CNDs hybrid NPs led us to develop a colorimetric method for rapid and selective determination of glutathione with the detection limit of 0.058 μM. Our approach offers a simple and novel method to prepare Fe3O4/CNDs hybrid NPs with modified CNDs, and their applications may be extended to biosensors, medical diagnostics, and environmental monitoring.

Ligands engineering of gold nanoclusters with enhanced photoluminescence for deceptive information encryption and glutathione detection

Mengyuan Chen,Zhenqiang Ning,Xue Ge,Erli Yang,Qian Sun,Fei Yin,Mingming Zhang,Yuanjian Zhang,Yanfei Shen

Gold nanoclusters (Au NCs) have appeared as an essential alternative to traditional quantum dots and fluorescent molecules for the development of intelligent stimuli-responsive photoluminescence (PL), but the low PL emission of Au NCs restricts their broad applications. Herein, we reported a simple yet effective strategy for preparing Au NCs with high PL by ligands engineering of 4-hydroxy-2-mercapto-6-methylpyrimidine (MTU) and L-Arginine (Arg). Owing to the rigidified shell and the ligand-to-metal charge transfer (LMCT) effects, it was found that the assembly of Arg ligand on MTU-protected Au NCs (Arg/MTU-Au NCs) led to a significantly enhanced PL in the alkaline solution up to 30 times. Moreover, utilizing the tunable LMCT, the Arg/MTU-Au NCs displayed rapid responses to multi-type ionic interaction in a reversible manner, such as H + /OH − and Cu 2+ /glutathione (GSH) pairs. Inspired by these intriguing ions-responsive LMCT and the associated switchable PL emission, the Arg/MTU-Au NCs were successfully used as excellent stimuli-responsive PL probes for intriguing deceptive information encryption and biosensing as well. This work would provide new insight into regulating the PL emission of Au NCs by ligands engineering and advance their potential applications in information encryption and bioassay.

Fabrication of Adjustable Au/Carbon Hybrid Nanozymes with Photothermally Enhanced Peroxidase Activity and Ultra-sensitivity for Glutathione Detection

Xinyan Chen,Yu Qi,Bin He,Yu Liang,Yu Lei,Jian Sun

Au nanozymes are extensively researched for their photothermal effect and catalytic performance, but overcoming the inherent defects of poor dispersibility and thermal stability through complementary materials will expand their prospects for biological applications. Herein, several novel CAu nanozymes were fabricated by in situ reduction of chloroauric acid on hollow carbon nanospheres (HCNs). Through regulating the number of reductions, sesame ball-shaped CAu (sCAu) with highly dispersed Au nanoparticles and diversity-shaped CAu (dCAu) were obtained. The number and morphology of loaded Au nanoparticles, absorption spectra, and hydrophilicity of CAu nanozymes were systematically characterized to demonstrate the flexibility of this novel method. The high-efficiency peroxidase-like sCAu0.3 nanozyme with hyperthermia-activated property was then screened for later bio-application. It is worth mentioning that its photothermal-promoted peroxidase-like activity could be achieved under near-infrared laser irradiation. Moreover, sCAu0.3 could specifically achieve glutathione detection in human blood samples. This method will provide a protocol for the regulation of CAu nanozymes to adapt to bio-detection applications.

Cascade-enhanced based-polyoxometalates nanozyme for glutathione detection and tumor cell disruption

Jinhang Guo,Hong Han,Hong Zhao,Deqi Jia,Ling Yin,Jingquan Sha

Nanozymes with biological enzyme activity show great promise in biochemical analysis and medicine, yet single-activity nanozymes were limited by low catalytic efficiency and strict catalytic environment requirements. Consequently, the development of nanozymes with multiple enzyme activities presents a significant challenge. In the work, a vanadium-capped polyoxometalates (POMs)-encapsulating metal-organic framework (MOF), [Cu 12 (Trz) 8 Cl][PMo 12 O 40 (VO) 2 ], was synthesized via hydrothermal synthesis, which shows multiple enzyme activities concluding oxidase (OXD), peroxidase (POD) and catalase (CAT) activities. In the catalytic procedure, a fraction of H 2 O 2 generated by the OXD is further subjected to the POD catalytic reaction and the remaining portion is transformed into O 2 through the CAT activity, in turn, supplies the driving force for the OXD-like catalytic process. This cascade reaction, in conjunction with the transformation between Cu 2+ and Cu + within the material, engenders a structure analogous to an interlocking "gear", which augments the catalytic efficacy as well as the adaptability to intricate environmental conditions of the [Cu 12 (Trz) 8 Cl][PMo 12 O 40 (VO) 2 ] enzyme. By capitalizing on this high catalytic efficiency, the rapid quantitative detection of glutathione (GSH) was established with the calculated limit of detection (LOD) 0.062 μM in the range of 5–60 μM. Surprisingly, the multi-enzymatic [Cu 12 (Trz) 8 Cl][PMo 12 O 40 (VO) 2 ] can concurrently enhance the generation of reactive oxygen species (ROS) and the depletion of GSH in the tumor microenvironment (TME), thereby inducing tumor cell apoptosis. This research holds promising application prospects in the fields of biotechnology, clinical diagnosis, and tumor therapy.

Development of tumor marker detection and tumor treatment based on silver nanozymes

Yang Xuan,Yating Gao,Yinan Zhao,Wenliang Zhang,Xinyu Bian,Mingfan Zhang,Ruoyun Zhang,Shubiao Zhang

Nanomaterials with intrinsic enzyme activity (nanozymes) have been widely used as artificial enzymes in biomedicine, especially in tumor diagnosis and treatment. Herein, a kind of water-soluble silver nanozyme (Ag@GQD) was developed with a simple method, which showed two enzymatic activities (peroxidase and oxidase-like activity). Based on the excellent catalytic performance of oxidase-like activity, a simple and highly sensitive colorimetric detection strategy for glutathione (GSH) was established. What’s more, after absorbing DOX, Ag/DOX@GQD could realize the combined treatment of reactive oxygen species and chemotherapy. In conclusion, this study provided evidences that Ag@GQD is effective in tumor diagnosis and treatment, which provided new ideas for exploring nanozymes based on the integration of diagnosis and treatment.

Mn-Doped CeO2 Nanorods for the Colorimetric Sensing of Glutathione

Jie Yao,Chengyi Li,Jianxin Feng,Bei Wei,Rongrong Xing,Shurong Ban,Li Tang,Bin Liu

Nanozymes with high peroxide (POD)-like activity have great practical potential in the colorimetric sensing of biomolecules in the medical and biological fields. However, their applications are limited by some disadvantages such as their low catalytic activity and poor biocompatibility. While designing and synthesizing nanozymes, elemental doping is used as an effective method to improve their enzymatic activity. Herein, CeO2 nanorods doped with different concentrations of Mn were synthesized via the hydrothermal method. Mn doping effectively improved the POD-like activity of the CeO2 nanorods. Notably, MnCe0.1 exhibited superior POD-like activity to that of CeO2. The improvement in the catalytic activity of MnCe0.1 was attributed to the presence of more exposed active sites and more oxygen vacancies compared with those of CeO2 nanorods. Using its excellent POD-like activity, the developed system exhibited linear detection ranges of 10–100 μM and 2.5–20 μM and detection limits of 1.94 μM and 0.37 μM for H2O2 and glutathione detection, respectively. Additionally, the developed MnCeX nanomaterials exhibit remarkable temporal stability and reusability. Finally, considering the excellent POD-like activity of MnCe0.1 nanozymes, they have good prospects for cancer treatment.

A Co,N co-doped hierarchically porous carbon hybrid as a highly efficient oxidase mimetic for glutathione detection

Siqi Li,Liuting Wang,Xiaodan Zhang,Hongxiang Chai,Yuming Huang

The past few years have witnessed rapid development of nanozymes in various fields. However, analytical applications of oxidase nanozymes are limited. Here, a novel oxidase nanozyme was fabricated by one-pot pyrolysis of ZIF-67. Benefiting from self-template synthesis and a nitrogen-rich precursor, the resulting product is a highly N-doped hierarchically porous carbon with well-dispersed Co nanoparticles. It shows a typical oxidase mimicking property to efficiently catalyze oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB), 1,2-diaminobenzene, and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt by oxygen to produce a color reaction without the need for H 2 O 2 . Reactive oxygen species (ROS) measurements indicate that 1 O 2 and O 2 − radicals are the main ROSs in the TMB–Co,N co-doped hierarchically porous carbon (Co,N-HPC) system. The high surface area and hierarchical pores of Co,N-HPC facilitate rapid diffusion of dissolved oxygen to the catalytic active sites, resulting in the novel and efficient oxidase nanozyme characteristic of Co,N-HPC. On account of the inhibiting effect of glutathione on TMB oxidation, a rapid and highly sensitive colorimetric assay is proposed for its detection. This assay has a linear concentration range from 0.05 to 30 μM and a limit of detection of 36 nM (3 σ ). The proposed method was successfully applied to glutathione quantification in biological samples.

A two-dimensional thin Co-MOF nanosheet as a nanozyme with high oxidase-like activity for GSH detection

Miaomiao Han,Mengzhen Ren,Zhaohui Li,Lingbo Qu,Lanlan Yu

Nanozymes with the advantages of excellent enzyme-mimicking activity, low cost and high stability have displayed great application potential in diverse fields as replacements for natural enzymes. In this study, a new nanozyme based on an ultrathin two-dimensional metal–organic framework nanomaterial D-ZIF-67 was prepared and characterized. D-ZIF-67 exhibited remarkable oxidase-like activity, which was attributed to the large specific surface area of the two-dimensional sheet structure, as well as its numerous exposed active sites compared with crystal MOFs. In the catalytic process of D-ZIF-67, the hydroxyl radical and superoxide anion played an essential role and facilitated the oxidation of the substrate 3,3′,5,5′-tetramethylbenzidine (TMB) by oxygen. The reducing agent glutathione (GSH) inhibited the catalytic activity of D-ZIF-67 besides reducing the oxidized oxTMB, based on which a simple, sensitive and accurate colorimetric method was developed to determine GSH, with the detection limit of 229.2 nM. This work mainly proposes a new two-dimensional metal–organic framework with high oxidase-like activity and provides a simple and practical platform for visual GSH detection by naked eyes.

Controlled synthesis of titanium dioxide/molybdenum disulfide core-shell hybrid nanofibers with enhanced peroxidase-like activity for colorimetric detection of glutathione

Wendong Zhu,Maoqiang Chi,Mu Gao,Ce Wang,Xiaofeng Lu

Synergistic effects play a crucial role in improving the catalytic activity of enzyme-like reactions. The preparation of hybrid nanomaterials for enzyme mimicking that display synergistic enhanced catalytic activity remains a formidable challenge. Titanium dioxide (TiO 2 )/molybdenum disulfide (MoS 2 ) core-shell hybrid nanofibers were synthesized as efficient peroxidase mimics via a three-step approach involving electrospinning, calcination, and hydrothermal treatment. The resulting TiO 2 /MoS 2 hybrid nanofibers exhibited synergistically enhanced peroxidase-like catalytic activity relative to the TiO 2 nanofibers or MoS 2 nanosheets alone. Based on the high peroxidase-like activity of the TiO 2 /MoS 2 hybrid nanofibers, a simple colorimetric approach for the detection of l -glutathione (GSH) was developed, with a detection limit as low as 0.05 μM. This study provides a simple and sensitive sensing platform for the detection of GSH, with prospective applications in colorimetric sensing, environmental monitoring, and medical diagnosis.

Bimetallic Cu@Co-MOFs Mimic Peroxidase for Colorimetric Detection of Glutathione

Yaohua Gu,Jiping Han,Ni Zhang,Weisong Yan,Yue Guo,Huiyi Tan,Changyi Yang,Fuke Wang,Huiqin Yao

Nanozyme possess the advantages of high enzyme simulation activity, low cost, and excellent stability. As a substitute for natural enzymes, nanozyme have demonstrated significant potential in various fields. In this study, a bimetallic organometallic framework Cu@Co-MOFs was synthesized via the hydrothermal method using terephthalic acid (H2BDC) as a ligand and characterized. Cu@Co-MOFs exhibits outstanding peroxidase activity (POD) by catalyzing the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) and other color-developing substrates through •OH formation. A simple, rapid, and reliable colorimetric method for quantitatively detecting glutathione (GSH) was developed based on the Pod-like activity of Cu@Co-MOFs. The linear range of this method was 1–1200 μM with a detection limit of 0.72 μM. This work not only proposes a bimetallic organic framework with high POD as a straightforward and practical platform for visually detecting GSH but also provides a strategy to enhance the performance of nanomaterials.
