Preparation of immobilized pepsin for extraction of collagen from bovine hide

Youdan Duan,Haiming Cheng

In the extraction of collagens from mammalian tissues, the free pepsin used in the acid–pepsin extraction system is hard to recycle, and there is a risk of enzyme protein contamination in the extracted collagen products, which limits their applications. To solve this problem, an immobilized pepsin was successfully prepared via the covalent crosslinking of glutaraldehyde using a 3-aminopropyltriethoxysilane (APTES) surface modified silica clay as the support. The immobilized pepsin was applied for the extraction of collagen from bovine hide. The optimal immobilization process involves incubating pepsin with an initial concentration of 35 mg mL−1 and glutaraldehyde with 5% activated APTES modified silica clay at 25 °C for 60 min, by which the loading amount of pepsin was 220 mg g−1 and the activity of the immobilized pepsin was 4.2 U mg−1. The collagen extracted using acetic acid and the immobilized pepsin method retained its complete triple helix structure. This research thus details an effective separation method using pepsin for extraction of collagen via an acetic acid–enzyme method, where the extracted collagen may be a candidate for use in biomaterial applications.

Controlled delivery of 5-fluorouracil from monodisperse chitosan microspheres prepared by emulsion crosslinking

Tong Wan,Qianqian Zhang,Guocheng Jin,Shiai Xu

This work aims to determine the optimal conditions for emulsion cross-linking of chitosan (CHS) with various molecular weights using glutaraldehyde as a cross-linking agent to produce 5-fluorouracil-loaded CHS microspheres (5-FU/CHS). Their drug loading and encapsulation efficiencies are found to be in the range of 3.87–12.35% and 20.13–70.45%, respectively. The dynamic light scattering results show that 5-FU/CHS microspheres are micron-sized with a uniform size distribution, and the scanning electron microscopy results show that they are spherical. The results of thermogravimetric analysis, X-ray diffraction, and Fourier transform infrared spectroscopy demonstrate that 5-FU is successfully incorporated into the microspheres. The in vitro release tests show that 5-FU/CHS have a prolonged, pH-responsive release pattern of 5-FU, and the cumulative release rate under acidic condition is much larger than that under neutral conditions. The drug release kinetic analysis further demonstrates that the release of 5-FU can be well described by the Fickian diffusion model.

Selective Oxidation of Cyclopentene Over Amphiphilic Heteropolyacid Catalysts Loaded on Modified Activated Carbon

Xueying Liu,Jiahao Li,Bing Yuan,Fengli Yu

The synthesis of glutaraldehyde and 1,2-cyclopentanediol has great industrial significance. However, the highly selective synthesis of these two compounds, and particularly of glutaraldehyde, is challenging. To improve the reaction rate and product selectivity of cyclopentene oxidation using green hydrogen peroxide (H 2 O 2 , 30 wt%) as an oxidant, activated carbon (AC)-supported amphiphilic vanadium-doped heteropolyacid (HPA) catalysts (AC-COIMI-C n -HPA) were prepared by a series of modifications of AC, including acidification, imidazolylation, ammonium saltification, and HPAs loading. The structures and physicochemical properties of the catalysts were characterized by Fourier infrared spectroscopy, X-ray diffraction, thermogravimetric analysis, and nitrogen adsorption–desorption. The developed catalyst exhibits excellent catalytic properties and high selectivity toward glutaraldehyde. The AC carrier increases the number of effective active sites on the catalyst. The amphiphilicity of the catalyst increased the reaction rate. Vanadium doping of HPA enables fine-tuning of the oxidizability of the catalyst. The effects of the different active components of the catalyst, alkyl groups on the imidazole ring, solvent type and dosage, HPA loading on the catalyst, catalyst dosage, oxidant dosage, reaction temperature, and reaction time on the oxidation of cyclopentene were comprehensively examined. Finally, the optimized conditions were determined as follows: HPA doping by four V atoms (PW 8 V 4 ), octyl (C 8 ) linked to the imidazole, 1 mL of acetonitrile, HPA loading of 35 wt%, 500 mg of the catalyst (AC-COIMI-C 8 –35%PW 8 V 4 ), n(H 2 O 2 ):n (cyclopentene) = 2, reaction temperature and time of 40°C and 6 h, respectively. The conversion of cyclopentene was 98.8% and the total selectivity for glutaraldehyde and 1,2-cyclopentanediol was 91.6% (65.2% for glutaraldehyde and 26.4% for 1,2-cyclopentanediol). The catalyst was reused by centrifugation, and high cyclopentene conversion was maintained after three cycles.

Development of an innovative capsule with three-dimension honeycomb architecture via one-step titration-gel method for the removal of methylene blue

Dichu Lin,Mei Shi,Xixi Wei,Bingxin Liu,Jiangfei Cao,Changsheng Peng

In this study, the capsules were prepared via the one-step titration-gel method by injecting spherical droplets of polyvinyl alcohol (PVA), sodium alginate (SA) and graphene oxide (GO) gelled mixture into the bath with calcium chloride (CaCl 2 ) and oversaturated boric acid (H 3 BO 3 ) solutions. The prepared capsules were then further modified with glutaraldehyde (GA). The formation mechanism of the prepared capsules was investigated. The morphology of the prepared capsules exhibited distinct micro-porous “3D honeycomb” pattern and hierarchical pore sizes distribution . It was also observed that GA not only acted as a co-cross-linked reagent in the fabricating process to increase the specific surface area of the capsules, but also offered exceptional tolerance property to work under a wide pH range (i.e. 2–12). The PVA-SA-GO capsules performed comparatively much better than PVA-SA capsules and they improved the adsorption capacity by up to 21.94%. Their adsorptive performance well followed the Langmuir isotherm. Moreover, the pH of the MB solution was evidently declined after adsorption, demonstrating the electrostatic attraction or ion-exchange might be the governing removal mechanism of methylene blue (MB) dye by the capsules. Importantly, the prepared capsules could be regenerated by simply washing with water and reused for at least 6 consecutive treatment cycles.

Glutaraldehyde cross-linked CDA/HBP-NH2 nanofiber membrane for adsorption of heavy metal ions in wastewater

Zang Chuanfeng,Han Xiangye,Dong Erying,Shen Feiyu,Yan Tingting,Wang Runyue,Zhang GuangyuYu

A new type of nanofiber membrane was prepared by mixed electrospinning of cellulose diacetate and amino-terminated hyperbranched polymer (HBP-NH2), and glutaraldehyde (GA) crosslinking was used to improve its water resistance and shape retention. pH, adsorption time, and initial solution concentration on Cu(II) and Cr(VI) ions were studied. Results showed that the prepared GA-CDA/HBP-NH2 nanofiber had good spinning uniformity, hydrophilicity, and adsorption effect on Cu(II) and Cr(VI) ions compared with the pure CDA nanofiber. The maximum adsorption capacity of GA-CDA/HBP-NH2 for Cu(II) and Cr(VI) was 50.2 and 176.6 mg g−1, respectively.

Casein nanogels as effective stabilizers for Pickering high internal phase emulsions

Shanshan Chen,Li-Ming Zhang

Pickering emulsion stabilized by colloidal particles is attracting increasing attention in recent years, and emulsions using food-grade or biocompatible particle stabilizers will be less restrained in biomedical applications. In this work, we demonstrated that the casein nanogels (CSNs) prepared by crosslinking casein, a natural protein with good biocompatibility, with glutaraldehyde could be used as effective stabilizers for Pickering high internal phase emulsions (HIPEs) without any other additives. The CSNs had an average diameter of 179 nm and a zeta potential of -16.4 mV at pH 7 with spherical morphologies. Emulsions were fabricated using CSNs as stabilizers under various conditions with three main variables including CSN concentration, pH and ionic strength , and characterized by fluorescence microscopy , photography and rheological measurement. It was found that CSNs could effectively adsorb on oil-water interface when their surface potential was close to neutrality under certain pH and ionic strength conditions and thereby assist in forming stable oil-in-water (O/W) Pickering HIPEs, even at a low CSN concentration of 0.5 wt%. To exploit the potential of these Pickering HIPEs in drug delivery, indomethacin (IDM) as a hydrophobic drug model was encapsulated into the oil phase. In vitro release experiment showed that the release behavior of IDM could be tuned by manipulating the CSN concentration. Overall, this work demonstrated that the casein nanogels could efficiently stabilize HIPEs and these HIPEs have a potential application in drug delivery system.

Micro foaming performance of scCO2-aid glutaraldehyde/hexametaphosphate/thermoplastic starch foams modified by alkali treatment and montmorillonite nano-platelets

Ke Wang,Ya-qiong Huang,Xiao-han Cheng,Jen-taut Yeh

The micro foaming performance, moisture resistance and dynamic viscosity of scCO2-aid glutaraldehyde/hexametaphosphate/thermoplastic tapioca starch (GA/SHMP/TOS) foams were considerably improved by proper NaOH treatment. The expansion ratio, resilience rate, dynamic viscosity values of these NaOH modified foams improved to a maximum, as the time for NaOH treatment approached a proper value. The dynamic viscosity, expansion ratio and resilience rate of the scCO2-aid GA/SHMP/TOS foams modified using 110 atm scCO2-pressure, the proper alkali treatment time, SHMP loading and varying montmorillonite (MMT) loadings improved further, as their MMT loadings approached a proper value of 2.5 part per hundred parts of tapioca starch (PHTOS). Relatively large dynamic viscosity (7.1x104 Pa·s), extremely large expansion ratio (∼75), cell density (1.1x109 cells/cm3) and/or resilience rate (∼80%) were acquired for the scCO2-aid GA/SHMP/TOS/MMT foam modified using the proper alkali treatment time and MMT loading. Thermal analyses results showed that crystallization onset temperatures and crystallization rates of scCO2-aid GA/SHMP/TOS/MMT foams modified using the proper alkali treatment time and varying MMT loadings improved to a highest value by adding 2.5 PHTOS of MMT nano-platelets. Possible reasons accounting for the considerably improved micro foaming performance of scCO2-aid GA/SHMP/TOS/MMT foams modified using the proper alkali treatment time and MMT loading are proposed in this study.
