Nanomotors driven by endogenous enzymes are favored in biology and pharmacy due to their spontaneous driving and efficient biocatalytic activity, and have potential applications in the treatment of clinical diseases that are highly dependent on targeted effects. For diseases such as muscle atrophy, using energy molecules such as ATP to improve cellular metabolism is a relatively efficient treatment method. However, traditional adenosine triphosphate (ATP) therapies for muscle atrophy face limitations due to instability under physiological conditions and poor targeting efficiency. To address these challenges, we developed an endogenous proton-gradient-driven ATP transport motor (ATM), a nanomotor integrating chloroplast-derived FoF1-ATPase with a biocompatible flask-shaped organic shell (FOS). The ATM is synthesized by vacuum-injecting phospholipid-embedded FoF1-ATPase nanothylakoids into ribose-based FOS, enabling autonomous propulsion in acidic microenvironments through proton-driven negative chemotaxis (directional movement away from regions of higher proton concentration). This nanomotor converts proton gradients into ATP synthesis, directly replenishing cellular energy deficits in atrophic tissues. In vitro studies demonstrated high biocompatibility (>90% cell viability at 150 μg/mL) and pH-responsive motility, achieving speeds up to 4.32 μm/s under physiological gradients (ΔpH = 3). In vivo experiments using dexamethasone-induced muscle atrophy mice revealed that ATM treatment accelerated weight recovery and restored normal muscle morphology, with treated mice exhibiting cell sizes comparable to healthy controls (30–40 μm vs. 15–25 μm in untreated). These results highlight the ATM’s potential as a precision therapeutic platform for metabolic disorders, leveraging the natural enzyme functionality and synthetic material design to enhance efficacy while minimizing systemic toxicity.

Mitochondria, recognized as the cellular powerhouse, provide a continuous stream of energy essential for the rapid proliferation and elevated metabolic demands of tumor cells. Herein, a flexoelectric nanocatalyst, SrTiO₃/RGD/TPP (SRT), is engineered to disrupt mitochondrial proton gradients and ionic balance through ultrasound-induced flexoelectric catalysis. This interference impedes mitochondrial energy production, resulting in tumor cell apoptosis due to an inadequate energy supply. Upon ultrasound stimulation, the SRT experiences polarization due to the stress gradient, resulting in the separation of positive and negative charges, thereby generating a local electric field. The negative charges interact with protons (H⁺) in the mitochondrial intermembrane space to produce hydrogen (H₂), thereby reducing proton concentration and disrupting the mitochondrial proton gradient, subsequently inhibiting ATP synthesis. Concurrently, the local electric field modifies the mitochondrial membrane potential (MMP), opening the mitochondrial calcium uniporter (MCU) channels and facilitating the influx of calcium ions, resulting in calcium overload and mitochondrial dysfunction, ultimately severing the energy supply to tumor cells. This disruption of energy, when combined with immunotherapy, demonstrates significant tumor inhibition in both in vitro and in vivo models. Through the integration of flexoelectric catalysis and immunotherapy, this study demonstrates potent anti-tumor effects and proposes new research directions in nanomedicine.

Moisture-enabled electricity generation (MEG) is regarded as a highly promising technique for sustainable electric power. Carbon dots (CDs) are shown to have great promise in MEG devices due to their low-cost fabrication and tunable surface groups, but their potential in MEG devices has not been fully exploited. Herein, we demonstrate a simple and effective method to significantly enhance the output of a CD-based MEG device based on CD surface functionalization using a strong ionizing organic acid. Compared to a bare CD-based MEG device, a voltage output of a 1,5-naphthalenedisulfonic acid (1,5-NDSA) functionalized CD (S-CD)-based MEG device shows great performance, with a voltage that reaches up to 380 mV. To the best of our knowledge, this is the highest value reported for a CD-based MEG device to date. Our results show that the large number of protons produced by the S-CDs facilitates the creation of a strong proton gradient and improves ionic conductivity, thus leading to an enhancement in the electricity output. By simply connecting MEG devices in series, the overall assembled device can generate enough electricity to power an electronic calculator. Furthermore, the output of the as-prepared device can be greatly enhanced under the flow of moist air, which is expected to make it useful for windy and high humidity areas.

Osmotic energy is a kind of blue energy that has recently been identified as an additional source of clean energy. Using a membrane-based reverse electrodialysis (RED) process, this blue energy can be obtained from acidic industrial wastewater with different proton concentration gradients. However, this process demands high-performance membrane that can withstand harsh environments, possessing the advantages of wide pH tolerance, high-temperature resistance and chemical stability, developing such membranes remain a challenge. Herein, we report a two-dimensional (2D) lamellar Ti 3 C 2 T x MXene membrane-based RED device for osmotic energy capturing from proton gradients. Such a membrane exhibits a typical surface-charge-governed ion transport feature. Moreover, the MXene membrane-based energy harvesting device holds the merits of outstanding pH and temperature resistance. It exhibits an output power density of 6.5 W/m 2 and also demonstrates stability over 200 h at pH = 0, which is 30% higher than the commercialization benchmark (5 W/m 2 ). The osmotic power density can be further enhanced to 11.1 W/m 2 at 330 K, demonstrating excellent thermal and chemical stability. This work can help better understand protons' transport behaviors in MXene membranes and open new avenues for applications in sustainable power conversion and wastewater treatment.

Special proton-gradient-transfer acid complexes (PGTACs) in which the bonded protons are not equivalent and have gradients in transfer ability, acidity, and reactivity were reported. The acidity gradient of the protons gave the PGTACs excellent catalytic activity and selectivity in the esterification of terpenols. These PGTACs are “reaction-induced self-separation catalysts” and can be easily reused. The kinetics with PGTACs as catalyst in the esterification of geraniol were also studied for use in engineering design.

Molecule assembly and functionalization of protocells have achieved a great success. However, the yield efficiency of photophosphorylation in the present cell-like systems is limited. Herein, inspired by natural photobacteria, we construct a protocell membrane reconstituting motor protein for highly efficient light-mediated adenosine triphosphate (ATP) synthesis through a layer-by-layer technique. The assembled membrane, compartmentally integrating photoacid generator, proton conductor, and ATP synthase, possesses excellent transparency, fast proton production, and quick proton transportation. Remarkably, these favorable features permit the formation of a large proton gradient in a confined region to drive ATP synthase to produce ATP with high efficiency (873 ATP s–1). It is the highest among the existing artificial photophosphorylation systems. Such a biomimetic system provides a bioenergy-supplying scenario for early photosynthetic life and holds promise in remotely controlled ATP-consumed biosensors, biocatalysts, and biodevices.

Summary A Cu@MoS 2 microtube-based micromotor with a unique composite structure in which MoS 2 was “wrapped” on the surface of Cu microtubes confers excellent photoresponsive properties due to the unique hybridized structure. Micromotor swarms demonstrate spontaneous oscillatory motion in pure water with constant light input, without any external field changes or energetic toxic fuels. This macroscopic periodic oscillatory motion synchronized with the cycle of increasing and decreasing protons (H + ) produced by the micromotor. The H + gradient is the core of cellular ATP (adenosine triphosphate) synthesis and decomposition. Oscillating proton signals released by the micromotor can be coupled to cardiomyocytes to achieve synchronized oscillations of cell membrane potential/calcium ions and to initiate ATP hydrolysis in cardiomyocytes. The oscillatory behavior was considered to be caused by the different conversion levels of bistable reactions or by non-linear kinetics and mass transport. Specifically, MoS 2 photocatalytic hydrogen production, MoS 2 hydrolysis reactions, dynamic diffusive matter exchange from electroosmotic flow, and interfacial effects contribute to this phenomenon.
