Development of Global Chemical Profiling for Quality Assessment of Ganoderma Species by ChemPattern Software

Hui Zhang,Huijie Jiang,Xiaojing Zhang,Shengqiang Tong,Jizhong Yan

Triterpenoids are the major secondary metabolites and active substances in Ganoderma, considered as the “marker compounds” for the chemical evaluation or standardization of Ganoderma. A response surface methodology was used to optimize the ultrasonic-assisted extraction of triterpenoids. The extraction rate was 7.338 ± 0.150 mg/g under the optimum conditions: 87% ethanol, ratio of solid to liquid (w : v) 1 : 28, and ultrasound extraction time 36 min. Based on the high sensitivity and selectivity of HPLC-LTQ-Orbitrap-MSn, 24 components of triterpenoids were tentatively identified in the negative mode. Then, the global chemical profiling consisting of HPLC and TLC fingerprints generated by ChemPattern™ software was developed for evaluation of Ganoderma species. For fingerprint analysis, 11 peaks of triterpenoids were selected as the characteristic peaks to evaluate the similarities of different samples. The correlation coefficients of similarity were greater than 0.830. The cluster analysis showed a clear separation of three groups, and 11 peaks played key roles in differentiating these samples. The developed global chemical profiling method could be applied for rapid evaluation, quality control, and authenticity identification of Ganoderma and other herbal medicines.

Quality assessment of commercial dried ginger (Zingiber officinale Roscoe) based on targeted and non-targeted chemical profiles and anti-inflammatory activity

Liangliang He,Huifang Duan,Xintong Chen,Yuanshan Chen,Qingmei Mo,Junqing Huang,Huinan Zhao,Xinsheng Yao,Jiaxu Chen,Zhihong Yao

Dried ginger, a well-known medicine and food homologous production, has been widely circulated in China with high health benefits and economic value. Currently, there is still a lack of quality assessment on whether dried ginger in China exhibits chemically and biologically distinct properties, which creates a barrier to its quality control in commercial circulation. In this study, the chemical characteristics of 34 batches of common dried ginger samples in China were first explored using non-targeted chemometrics based on the UPLC-Q/TOF-MS analysis, leading to the identification of 35 chemicals that contributed to clustering into two categories, with sulfonated conjugates being the key chemically distinct components. By comparing the samples before and after sulfur-containing treatment and the further synthesis of a key differentiating component of [6]-gingesulfonic acid, it was then demonstrated that sulfur-containing treatment was the primary cause of the formation of sulfonated conjugates, as opposed to regional or environmental factors. Furthermore, the anti-inflammatory efficacy of dried ginger with high presence of sulfonated conjugates was significantly decreased. Consequently, for the first time, UPLC-QqQ-MS/MS was used to develop a targeted quantification method for 10 characteristic chemicals in dried ginger, allowing researchers to quickly determine whether dried ginger has been processed with sulfur and quantitatively evaluate the quality of dried ginger. These results provided an insight into the quality of commercial dried ginger in China and a suggested method for its quality supervision as well.

A facile method for determining the Fe(0) content and reactivity of zero valent iron

Shaolin Li,Yangyang Ding,Wei Wang,Hong Lei

Zero valent iron (ZVI) is increasingly used in large quantities for water treatment. In large-scale application of ZVI, a common question that engineers encounter is ZVI selection or its quality assurance and quality control (QA/QC). A facile method for assessing ZVI quality is proposed and studied in this work. The method is based on the Fe(III)–Fe(0) reaction that has a fast reaction rate, a known and simple stoichiometry, and an easy-to-assay product. The method is conducted by mixing the sample (e.g., testing ZVI) with concentrated Fe(III) solution (up to 3.0 g L−1) for 3–4 hours. The metallic iron (Fe(0)) content of the sample is obtained by assaying the final increase of the total dissolved iron, and its reactivity is quantified using the observed dissolution rate constants. The method is experimentally validated using 23 iron-based materials. Studies with reference iron powder and iron oxide demonstrate that the Fe(III) solution has satisfactory specificity in selecting Fe(0). Replicate experiments and t-test evaluation show the complete dissolution of the Fe(0) content in micron-scale ZVI and the high reproducibility of the method for measuring Fe(0). The accuracy and reliability of the method are studied using 15 commercial ZVIs of known Fe(0) purity, in a series of experiments using Fe(III), Cu(II) or trichloroethylene. The experiments show that the results from the proposed method are accurate and reliable, and that the method is a strong tool more effective than conventional techniques such as X-ray diffraction and BET-N2 adsorption. This work provides an easy and facile way for the quantitative assessment of the quality of metallic iron used for environmental clean-up, especially for the quick survey of a large number of iron samples on site. The method may also serve as a QA/QC procedure for ZVI in industrial-scale production.

Comprehensive assessment of matcha qualities and visualization of constituents using hyperspectral imaging technology

Yan Hu,Huahao Yu,Xinbei Song,Wei Chen,Lejia Ding,Jiayi Chen,Zhiyuan Liu,Yihang Guo,Dongyun Xu,Xuesong Zhu,Chuangchuang Zhou,Jingfei Zhang,Binhui Liao,Jihong Zhou,Xiaoli Li,Yuefei Wang,Yong He

Matcha, made from different tea leaves as raw material, exhibits diverse aromas and flavors. Therefore, there is an urgent need for a rapid, non-destructive method to assess the quality of matcha to ensure that these different characteristics are accurately assessed without compromising the integrity of the product. In this study, hyperspectral imaging technology (HSI) combined with machine learning methods enabled the first visual in situ assessment of matcha quality. The physicochemical contents of matcha were determined chemically. Qualitative and quantitative detection models for different types and grades were developed using HSI (containing Vis-NIR and NIR band). The results showed that hyperspectral data in the Vis-NIR were better than in the NIR band. The accuracy of XGBoost in modelling the classification of matcha grades reached 98.10 %. After feature selection using the random forest (RF) method, partial least squares regression (PLSR) was built to predicted the quality of matcha, which showed high prediction accuracy (test set R p 2  > 0.95). The model uses HSI to visually visualize spatial variations in constitutions (catechins, free amino acids, caffeine, soluble proteins, and soluble sugars) to show compositional differences between different types of matcha, providing a rapid non-destructive method for comprehensive assessment of matcha quality.

Integrated multidimensional spectra and multilevel data optimization for rapid origin tracing and quality assessment of Salvia miltiorrhiza

Rao Fu,Peng Chen,Wenhao Dong,Jia Qiao,Yu Li,Mingxuan Li,Li Zeng,Chunqin Mao,Tulin Lu,Chenghao Fei

The evaluation of the herbal medicine quality often involves time-consuming and labor-intensive high-precision instrument testing. This study used Salvia miltiorrhiza as a case study, with 120 samples collected from various regions. Hyperspectral data, Fourier Transform Near-Infrared data, heavy metals and harmful elements data, and active ingredient data were systematically gathered. Using multivariate statistical analysis, multimodal information integration, and deep learning algorithms, a geographical origin tracing model was developed, achieving an accuracy rate of 100%. Additionally, spectral-ingredient prediction models for five heavy metals and harmful elements and four active components were established, allowing the heavy metals and harmful elements and active components of Salvia miltiorrhiza to be rapidly and accurately assessed. This study compared the performance of the hyperspectral and FT-NIR techniques in herbal medicine quality control. The results indicated that hyperspectral demonstrated broader applicability and superior performance in predicting heavy metals and harmful elements contents, whereas FT-NIR was more effective in analyzing chemical constituents. This research provides a scientific basis for Salvia miltiorrhiza origin tracing and quality prediction while also confirming the potential of multidimensional spectra techniques in the quality evaluation.

A reliable detection method for nano-silver in functional textiles

Haifeng Yang,Junfang Li,Guangcheng Xi,Xingzhou Guo,Yahui Li,Hua Bai

Except for conventional functions, special textiles doped with nanoscale particles have shown quite attractive properties, such as antibacterial activity, deodorization, hydrophobicity, and ultraviolet/electromagnetic protection. However, facile and reliable methods for quality assessment of those functional nano-textiles are remarkably demanded for market surveillance and consumer protection. Herein, a novel detection method of silver in textiles was established for the comprehensive detection and a practical flow chart was also demonstrated as an experimental pathway including pretreatment, qualitative and quantitative analysis. The optimal ashing parameter (450°C for 1 h) in the pretreatment process was explored to improve detection sensitivity. The qualitative analysis was performed through X-ray diffraction for crystallinity, scanning electron microscopy for morphology, and energy-dispersive X-ray spectroscopy (EDS) for elements. The real existent status of the silver components could be decided via the flow chart. As shown with the detected sample, the content was calculated by the weight loss at 1050–1500°C from thermogravimetric analysis measurement and the accuracy of the quantitative process was also checked via inductively coupled plasma mass spectrometry. Finally, the detection limit of the method was investigated up to μg/g. The obvious advantage of the reported method was facile and reliable to achieve comprehensive identification from the dimension scale and crystal form to the component. Moreover, this analytical strategy could be also applied for the detection of other nanoscale metals or metal oxides in functional textiles.

Data Fusion of ion Mobility Spectrometry Combined with Hierarchical Clustering Analysis for the Quality Assessment of Apple Essence

Sha Min,Zhang Zhengyong,Gui Dongdong,Wang Yanbo,Fu Linglin,Wang Haiyan

A fingerprinting approach was developed by means of high-performance ion mobility spectrometry with direct electrospray ionization (ESI-HPIMS) for the quality consistency and authentication of apple essence which contain many analytes. Thirty-four apple essence samples of the same brand and commercialized as a same product but brewed in four different manufacturers were used to establish the fingerprints. Hierarchical clustering analysis (HCA) was performed to evaluate the similarity and variation of these samples. A combined data matrix was constructed with the use of individual data matrix of ion mobility spectrum in positive and negative ion modes. For comparison, ion mobility spectrum fingerprints in positive and negative ion modes respectively were also applied to the quality assessment of the same samples. Finally, our study demonstrated that the fusion of fingerprints did indeed provide more information and could be used to comprehensively conduct the quality consistency evaluation and discrimination of apple essences from similar products. It is suggested that this fingerprint approach is suitable for analysis of other complex, multianalyte substances.

E-eye and FT-NIR combined with multivariate algorithms to rapidly evaluate the dynamic changes in the quality of Gastrodia elata during steaming process

Jiu-ba Zhang,Bin Wang,Yun-fei Zhang,Yi Wu,Ming-xuan Li,Ting Gao,Tu-lin Lu,Zhen-hua Bian,Lian-lin Su

Gastrodia elata (GE) is traditionally subjected to steaming, and steaming duration plays a crucially important role in determining GE quality. This study examined the variations in bioactive components during the steaming process and proposed the utilization of electronic eye and Fourier Transform near-infrared (FT-NIR) spectroscopy for quality assessment. The findings revealed that the levels of parishin E parishin B, parishin A, and gastrodin initially rose and subsequently declined, while 4-Hydroxybenzyl alcohol exhibited a rapid decrease followed by stabilization. With prolonged steaming, the brightness of GE decreased, while the red and yellow tones became more pronounced and the color saturation increased. FT-NIR divided the steaming process into three stages: 0 min (raw GE), 0–9 min (partially steamed GE), and 9–30 min (fully steamed GE), and the partial least squares regression models effectively predicted the levels of five components. Overall, this study provided valuable insights into quality control in food processing.

Rapid quality assessment of Gentianae Macrophyllae Radix based on near-infrared spectroscopy and capillary electrophoresis

Zhiyong Zhang,Nan Hao,Long Wang,Shunan Li,Xinhao Yan,Xuting Guo,Yunlan Lian,Wenlong Li

The aim of this study was to establish a rapid quality assessment method for Gentianae Macrophyllae Radix (RGM) using near-infrared (NIR) spectra combined with chemometric analysis. The NIR spectra were acquired using an integrating sphere diffuse reflectance module, using air as the reference. Capillary electrophoresis (CE) analyses were performed on a model P/ACE MDQ Plus system. Partial least squares-discriminant analysis qualitative model was developed to distinguish different species of RGM samples, and the prediction accuracy for all samples was 91%. The CE response values at each retention time were predicted by building a partial least squares regression (PLSR) calibration model with the CE data set as the Y matrix and the NIR spectra data set as the X matrix. The converted CE fingerprints basically match the real ones, and the six main peaks can be accurately predicted. Transforming NIR spectra fingerprints into the form of CE fingerprints increases its interpretability and more intuitively demonstrates the components that cause diversity among samples of different species and origins. Loganic acid, gentiopicroside, and roburic acid were considered quality indicators of RGM and calibration models were built using PLSR algorithm. The developed models gave root mean square error of prediction of 0.2592% for loganic acid, 0.5341% for gentiopicroside, and 0.0846% for roburic acid. The overall results demonstrate that the rapid quality assessment system can be used for quality control of RGM.

Fast separation of triterpenoid saponins using supercritical fluid chromatography coupled with single quadrupole mass spectrometry

Yang Huang,Tingting Zhang,Haibo Zhou,Ying Feng,Chunlin Fan,Weijia Chen,Jacques Crommen,Zhengjin Jiang

Triterpenoid saponins (TSs) are the most important components of some traditional Chinese medicines (TCMs) and have exhibited valuable pharmacological properties. In this study, a rapid and efficient method was developed for the separation of kudinosides, stauntosides and ginsenosides using supercritical fluid chromatography coupled with single quadrupole mass spectrometry (SFC-MS). The separation conditions for the selected TSs were carefully optimized after the initial screening of eight stationary phases. The best compromise for all compounds in terms of chromatographic performance and MS sensitivity was obtained when water (5–10%) and formic acid (0.05%) were added to the supercritical carbon dioxide/MeOH mobile phase. Beside the composition of the mobile phase, the nature of the make-up solvent for interfacing SFC with MS was also evaluated. Compared to reversed phase liquid chromatography, the SFC approach showed higher resolution and shorter running time. The developed SFC-MS methods were successfully applied to the separation and identification of TSs present in Ilex latifolia Thunb. , Panax quinquefolius L. and Panax ginseng C.A. Meyer . These results suggest that this SFC-MS approach could be employed as a useful tool for the quality assessment of natural products containing TSs as active components.

Quality Assessment of Different Species and Differently Prepared Slices of Zedoray Rhizome by High-Performance Liquid Chromatography and Colorimeter with the Aid of Chemometrics

Fei Sun,Xiaolu Yang,Fei Liu,Ying Zhang,Shumei Wang,Hui Cao,Jiang Meng

In this study, high-performance liquid chromatography (HPLC) and colorimeter were applied to evaluate the quality of different species and differently prepared slices of Zedoray Rhizome samples with the aid of chemometric tools. Fifty batches of Zedoray Rhizome samples from different species and forty-two batches of Zedoray Rhizome samples from differently prepared slices were collected. The quantitative method was developed using HPLC to simultaneously determine the contents of twelve chemical ingredients in Zedoray Rhizome. The colour parameters L, a, and b were measured by a colorimeter. Then, the collected data were analyzed by the principal component analysis and Pearson correlation analysis. The results showed that the proposed method was capable of accurately determining the contents of the twelve chemical ingredients and the colour parameters for the collected samples. There was a dramatic difference in the contents of the chemical ingredients and in the colour parameters among different species and differently prepared slices of Zedoray Rhizome samples. This study reveals that combining HPLC, colorimeter, and chemometric tools can provide a new approach to comprehensively evaluate the quality of Zedoray Rhizome samples.

Integrated sediment quality assessment through biomarker responses and bioavailability measurements: Application in Tai Lake, China

Hong-xue Qi,Hui-zhen Li,Ping Ma,Jing You

A weight of evidence (WoE) framework has been applied to assess sediment quality of a typical freshwater lake, Tai Lake in China, where the sediments were contaminated by various chemicals but showed no acute lethality to the benthic invertebrate, Chironomus dilutus . A quantitative scoring method was employed to integrate three lines of evidence (LoE), including adverse effects in life cycle bioassays, biomarker responses, and bioavailability-based chemical analysis. Six biomarkers were determined in C. dilutus after the exposure to the sediments from Tai Lake and provided sensitive indication of sublethal effects at the molecular level. The biomarkers included cytochrome P450, glutathione S -transferase, carboxylesterase, acetylcholinesterase, catalase , and lipid peroxidation. The changes of the biomarkers were summarized for individual sampling sites by computing the integrated biomarker response (IBR) indices. Complementary information was also confirmed by the interrelationship of the LoEs. The IBR indices gained before pupation correlated well with the impairments of emergence of the midges, and altered acetylcholinesterase was corroborated by the detection of chlorpyrifos , an organophosphate pesticide . The relationship between bioavailable toxic units estimated by Tenax extractable concentrations of chemicals in sediment and the observed toxicity in the midges helped to identify the putative toxicity contributors to C. dilutus . Overall, the WoE method clearly distinguished the contaminated sites and ranked them by the level of contamination. Sediment-associated pesticides, particularly γ -hexachlorocyclohexane and chlorpyrifos, were the possible contributors to chronic toxicity to the midges.
