This paper developed a method for preparing ultrasound-responsive microgels based on reversible addition fragmentation chain transfer-hetero Diels–Alder (RAFT-HAD) dynamic covalent bonding. First, a styrene cross-linked network was successfully prepared by a Diels–Alder (DA) reaction between phosphoryl dithioester and furan using double-ended diethoxyphosphoryl dithiocarbonate (BDEPDF) for RAFT reagent-mediated styrene (St) polymerization, with a double-ended dienophile linker and copolymer of furfuryl methacrylate (FMA) and St as the dienophile. Subsequently, the microgel system was constructed by the HDA reaction between phosphoryl disulfide and furan groups using the copolymer of polyethylene glycol monomethyl ether acrylate (OEGMA) and FMA as the dienophore building block and hydrophilic segment and the polystyrene pro-dienophile linker as the cross-linker and hydrophobic segment. The number of furans in the dienophile chain and the length of the dienophile linker were regulated by RAFT polymerization to investigate the effects of the single-molecule chain functional group degree, furan/dithioester ratio, and hydrophobic cross-linker length on the microgel system. The prepared microgels can achieve the reversible transformation of materials under force responsiveness, and their preparation steps are simple and adaptive to various potential applications in biomedical materials and adaptive electrical materials.

Nanoparticles have been demonstrated to be effective in targeted drug delivery to tumor due to the enhanced permeability and retention (EPR) effect. However, the inhomogeneous distribution of the nanoparticles in the tumor and the slow release of the drug make the therapeutic effect unsatisfied. Here, we present reactive oxygen species (ROS)-responsive micelles comprising poly (ethylene glycol)-poly(propylene sulfide) (PEG-PPS) for targeted delivery and in situ release of drug. Upon the irradiation of ultrasound, the loaded sonosensitizer hypocrellin (HC) will generate ROS to trigger the disassembly of the micelles and meanwhile realize sonodynamic therapy (SDT) effect of cancer. The in vivo experiment indicates that the HC loaded PEG-PPS are biocompatible and much more efficacious than an equivalent amount of free HC in inhibiting the growth of cancer.

Implantable devices that integrate advanced nanomaterials with wireless energy systems represent a transformative approach to tackling brain tumors. Here, a millimeter-scale implantable device based on an MXene/polyvinyl alcohol (PVA) hydrogel is introduced, designed for precise and minimally invasive brain tumor therapy via tunable alternating electric fields. This device presents a novel approach by combining ultrasonic wireless energy transfer with triboelectric energy harvesting, eliminating the need for in vivo batteries and enabling continuous operation in a completely wireless manner. The system not only optimizes energy harvesting efficiency but also ensures the localized delivery of therapeutic electric fields directly to the tumor site. Its compact size and mechanical flexibility allow for precise implantation in the complex brain environment, offering a minimally invasive solution with enhanced safety and adaptability. Preclinical evaluations using U-87 MG orthotopic glioblastoma models demonstrate significant tumor growth inhibition and a marked improvement in survival rates, underscoring its therapeutic potential.

Self-assembly of asymmetric structures has been attractive for applications in the fabrication of hierarchical structures. However, the role of the electrostatic interaction in the self-assembly of charged asymmetric particles in organic solvents has not been fully understood. In this research, self-assembly of positively charged patchy micelles in organic solutions and the reversible ultrasound responsivity of the assemblies are studied. Surface micelles (s-micelles) with cross-linked/quaternized poly(2-(dimethylamino)ethyl methacrylate) (q-PDMAEMA) cores and poly(oligo(ethylene glycol) monomethyl ether methacrylate) (POEGMA) coronae were prepared on the surfaces of silica particles. Positively charged patchy micelles were synthesized upon cleavage of the s-micelles. In THF, the patchy micelles self-assemble into vesicles with positively charged q-PDMAEMA in the walls and POEGMA in the coronae. The average size of the vesicles is dependent on POEGMA chain length. The repelling electrostatic interaction between the positively charged q-PDMAEMA patches in the walls is unfavorable for the achievement of low free energy of the assemblies, but it is energetically favorable for the vesicles to overcome energy barriers in the dissociation of the structures under ultrasound irradiation. The vesicle structures present reversible ultrasound responsivity. Under sonication the vesicles formed by patchy micelles dissociate into transient nanostructures, including aggregates and isolated patchy micelles. When the ultrasound is switched off, the transient nanostructures self-assemble into vesicles. By contrast, crosslinked vesicles do not show ultrasound responsivity. This research provides a new approach to the fabrication of responsive assemblies based on asymmetric structures, and the assemblies could find potential applications in medicine and bioengineering.

Background Biocompatible nanocarriers are widely employed as drug-delivery vehicles for treatment. Nevertheless, indiscriminate drug release, insufficient organ-specific targeting, and systemic toxicity hamper nanocarrier effectiveness. Stimuli-responsive nano-sized drug delivery systems (DDS) are an important strategy for enhancing drug delivery efficiency and reducing unexpected drug release. Methods This study introduces a temperature- and ultrasound-responsive nano-DDS in which the copolymer p-(MEO2MA-co-THPMA) is grafted onto mesoporous iron oxide nanoparticles (MIONs) to construct an MPL-p nano-DDS. The copolymer acts as a nanopore gatekeeper, assuming an open conformation at sub-physiological temperatures that allows drug encapsulation and a closed conformation at physiological temperatures that prevents unexpected drug release during circulation. Lactoferrin was conjugated to the nanoparticle surface via polyethylene glycol to gain organ-targeting ability. External ultrasonic irradiation of the nanoparticles in the targeted organs caused a conformational change of the copolymer and reopened the pores, facilitating controlled drug release. Results MPL-p exhibited excellent biocompatibility and rare drug release in circulation. When targeting delivery to the brain, ultrasound promoted the release of the loaded drugs in the brain without accumulation in other organs, avoiding the related adverse reactions, specifically those affecting the heart. Conclusion This study established a novel temperature- and ultrasound-responsive DDS that reduced systemic adverse reactions compared with traditional DDS, especially in the heart, and demonstrated excellent organ delivery efficiency.

Hydrogel dressings capable of infection monitoring and precise treatment administration show promise for advanced wound care. Existing methods involve embedd ingorganic dyes or flexible electronics into preformed hydrogels, which raise safety issues and adaptability challenges. In this study, an injectable hydrogel based smart wound dressing is developed by integrating food-derived anthocyanidin as a visual pH probe for infection monitoring and poly( L -lactic acid) microcapsules as ultrasound-responsive delivery systems for antibiotics into a poly(ethylene glycol) hydrogel. This straightforwardly prepared hydrogel dressing maintains its favorable properties for wound repair, including porous morphology and excellent biocompatibility. In vitro experiments demonstrated that the hydrogel enabled visual assessment of pH within the range of 5 ∼ 9.Meanwhile, the release of antibiotics could be triggered and controlled by ultrasound. In vivo evaluations using infected wounds and diabetic wounds revealed that the wound dressing effectively detected wound infection by monitoring pH levels and achieved antibacterial effects through ultrasound-triggered drug release. This led to significantly enhanced wound healing, as validated by histological analysis and the measurement of inflammatory cytokine levels. This injectable hydrogel-based smart wound dressing holds great potential for use in clinical settings to inform timely and precise clinical intervention and in community to improve wound care management.

Chronic diabetic wounds persist as a clinical healthcare challenge. In the hyperglycemic pathological milieu, excessive inflammation, peripheral neuropathy, and vascular disorders impede diabetic wound healing. Immunoregulation is substantially emphasized among these factors; however, research on neurovascular regeneration remains insufficient. Endogenous electric fields (EFs) can regulate cellular behaviors and significantly impact the wound healing process. Herein, a conductive composite hydrogel (CGGP) with ultrasound-responsive electrical stimulation based on interfacial ion/electron transfer for diabetic wound electrotherapy is proposed. CGGP is synthesized through physicochemical crosslinking of gelatin and polyvinyl alcohol (PVA) dual networks and incorporated with curcumin-loaded reduced graphene oxide (rGO). The conductivity of CGGP is comparable to that of skin. Notably, CGGP can generate electrical stimulation based on ion transfer at the fluid–solid interface (electrokinetic effects) and electron transfer at the curcumin-rGO interface in response to ultrasound. Moreover, CGGP exhibits an ultrasound-responsive release of curcumin for anti-inflammatory treatment. The ultrasound-responsive CGGP can promote the secretion of neurotrophic factors, vessel formation, and fibroblast migration. Furthermore, it facilitates local nerve regeneration, angiogenesis, and matrix remodeling in diabetic rat models, thereby accelerating diabetic wound healing. The sonoelectrical CGGP composite hydrogel holds great potential as a promising material for diabetic wound treatment.

Recent common clinical treatments for implant bacterial infections involve replacing inert implants and using antibiotics. However, these methods remain limited in their effectiveness for pathogen clearance, immune regulation, and osteogenesis. In this study, we developed a Zr-doped heterointerface of SrTiO 3 and Hap (SrTiZrO 3 /Hap) heterojunction coating with single-atom Zr doping and heterogeneous interfaces designed for ultrasound-responsive antimicrobial activity and bone formation. Under ultrasound, the mechanical force exerted by SrTiZrO 3 /Hap enhances its physical puncture and sonodynamic activity, synergizing with the metalloimmunotherapy effect of Zr 4+ for efficient antimicrobial activity. The primary mechanism enhancing sonodynamic activity involves local interfacial polarization from single-atom Zr doping, achieving piezoelectric catalysis in conjunction with electronic polarization from the built-in electric field. SrTiZrO 3 /Hap achieved a 99.3% antibacterial rate against S. aureus and 99.7% against E. coli under ultrasound. Additionally, SrTiZrO 3 /Hap promoted osteogenic differentiation after ultrasound irradiation by activating the PI3K/Akt pathway via its piezoelectric, needle-like topological surface and the release of functional ions, thus accelerating bone repair.

An in-depth investigation of the ultrasonic response of mechanophores containing weak covalent bonds within nanosized aggregates is vital for ultrasound imaging, drug delivery, and sensors. In this work, well-defined amphiphilic block SP-[poly(methyl methacrylate- b -N-isopropylacrylamide)] 2 SP-[p(MMA- b -NIPAM)] 2 ( P1 ) and amphiphilic random, SP-[poly(methyl methacrylate- r -N-isopropylacrylamide)] 2 SP-[p(MMA- r -NIPAM)] 2 ( P2 ) copolymers with SP mechanophore embedded at midpoint polymer chain are prepared by RAFT (reversible addition-fragmentation chain transfer) polymerization using a pre-synthesized novel functional bi (trithiocarbonate) and spiropyran (SP)-based RAFT reagent. Both P1 and P2 copolymers self-assemble in spherical micelles in AcCN/water mixed solvents, and several assemblies with different water contents are fabricated. The structural differentiation between block P1 and random P2 dominates the inverse influence of water content on the aggregate sizes of micelles. Under ultrasound irradiation , the SP mechanophore is activated within both micelles, and the amount of activated SP mechanophore increases progressively with prolonged sonication time. For P1 , the mechanochemical reaction (mechanochromism) kinetics in an aggregated state and a single chain state are different, while that of P2 micelles and liner are similar, inferred to be associated with the structure difference between both micelles. The mechanoactivation of SP mechanophore within micelles in AcCN/water mixed solvents is higher than in dissolved state due to the micellization of P1 and P2 , entangling the hydrophobic PMMA segments and partially swelling in the micellar core. Moreover, an increase in the medium's dielectric constant around the core's SP is also observed, promoting the SP mechanoactivation. This approach may provide a new strategy for preparing novel mechanophore-centered amphiphilic copolymers with colorful structures and high hydrophilicity . These can self-aggregate into nanoparticles in aqueous or mixed solutions, exhibiting effective effects of ultrasonic responsiveness. We expect this work to push forward the in-depth studies on the ultrasonic responsive mechanism of assembly and help open new research avenues into the mechanochemical reactions within nanosized aggregates.

A pH- and ultrasound dual-responsive drug release pattern was successfully achieved using mesoporous silica nanoparticles (MSNs) coated with polydopamine (PDA). In this paper, the PDA shell on the MSN surface was obtained through oxidative self-polymerization under the alkaline condition. The morphology and structure of this composite nanoparticle were fully characterized by a series of analyses, such as infrared (IR), transmission electron microscopy, and thermogravimetric analysis. Doxorubicin hydrochloride (DOX)-loaded composite nanoparticles were used to study the performances of responsive drug storage/release behavior, and this kind of hybrid material displayed an apparent pH response in DOX releasing under the acidic condition. Beyond that, upon high-intensity focused ultrasound exposure, loaded DOX in composite nanoparticles was successfully triggered to release from pores because of the ultrasonic cavitation effect, and the DOX-releasing pattern could be optimized into a unique pulsatile fashion by switching the on/off status. From the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay, it was observed that our blank nanoparticles showed no toxicity to HeLa cells, but DOX-loaded nanoparticles could inhibit the growth of tumor cells. Furthermore, these composite nanoparticles displayed an effective near-IR photothermal conversion capability with a relatively high conversion efficiency (∼37%). These as-desired drug delivery carriers might have a great potential for future cancer treatment that combine the chemotherapy and photothermal therapy.

Given that platinum-based drugs are widely used clinically as chemotherapeutic agents, their severe toxic side effects have attracted significant attention. Consequently, the development of novel nanoprodrugs based on low-toxicity tetravalent platinum (Pt(IV)) complexes holds substantial research value. Herein, we discovered that coumarin derivatives exhibit inherent antitumor efficacy and significantly enhance superoxide anion radicals (•O 2 − ) generation in aqueous solutions under ultrasound (US) irradiation. Given that •O 2 − is known to mediate the reduction of Pt(IV) to divalent platinum (Pt(II)), we engineered an US-responsive dual-drug nanoprodrug (P-cisPt(IV)@5-MOP). This nanoprodrug was prepared by covalently conjugating Pt(IV) and methoxy polyethylene glycol hydroxyl (mPEG-OH) to a poly( l -glutamic acid) (PLG) carrier, followed by encapsulating coumarin derivatives. Under low-intensity US irradiation (1.5 W/cm 2 , 1 MHz, 10 min), P-cisPt(IV)@5-MOP achieved a Pt(IV) reduction rate of 91.4%. Furthermore, upon US exposure, its half-maximal inhibitory concentration (IC 50 ) against 4T1 breast cancer cells decreased dramatically from 25.7 µmol/L to 0.1 µmol/L. Remarkably, this system combined with US therapy yielded a tumor inhibition rate of 90.9%, with 40% of tumor-bearing mice achieving complete eradication of tumors, while exhibiting low systemic toxicity. Collectively, this work not only identifies a novel sonosensitizer capable of generating •O 2 − but also develops a new class of ultrasound-activatable Pt(IV) nanoprodrug.

Stimuli-responsive microcarriers have received considerable attention in a variety of fields, including disease diagnosis, drug delivery, sensing, and imaging. Here, we report the generation of multiple-responsive perfluorocarbon-loaded magnetic hydrogel microcapsules (PMHMs) with uniform size for magnetic controlled ultrasound (US) and laser activation. The PMHMs are fabricated by a novel coaxial interface shearing (CIS) method based on the mechanism of liquid bridge formation and fracture. Perfluorocarbon and iron oxide magnetic nanoparticles are used as US-responsive and photothermal absorption medium, respectively, and magnetic nanoparticles are also used for magnetic-controlled targeting. Moreover, the size, structure, and function of the prepared biocompatible PMHMs can be precisely controlled by adjusting the process parameters of CIS. It is indicated that the PMHMs have different US- and light-responsive characteristics, mainly because of the difference of their activation mechanisms. It is demonstrated that laser has better activation resolution and can achieve site-specific activation and drug release of PMHMs. The multiple-responsive features imply that the PMHMs fabricated by CIS may provide an effective drug release platform for biomedical and pharmaceutical applications.
