An ultrasensitive biosensor based on three-dimensional nanoporous conducting polymer decorated with gold nanoparticles for microRNA detection

Yihui Ma,Nianzu Liu,Zhenying Xu,Jingjing Wang,Xiliang Luo

The rapid, selective and ultrasensitive assaying of biomarkers remains a challenge relevant to biosensor/diagnostic applications. Herein, a promising electrochemical biosensor based on three-dimensional (3D) nanoporous conducting polymer poly(3,4-ethylenedioxythiophene) (PEDOT) decorated with gold nanoparticles (AuNPs) was developed, in which methylene blue (MB) was employed as redox indicator for microRNA24 (miRNA24) detection. The unique porous construction was fabricated by a simple hard-template method and cyclic voltammetry (CV) electrochemical technique. AuNPs were electrodeposited onto the porous PEDOT surface to increase the interface conductivity and provide more active sites to load identification elements. The obtained porous biosensor based on conducting polymer PEDOT and AuNPs exhibited significantly enlarged specific surface area (about 2.4 times) and enhanced sensitivity (about 2.1 times), compared to the biosensor based on planar PEDOT. The developed porous biosensor achieved a miRNA24 quantification range from 1 fM to 10 nM with satisfying stability, high selectivity, and a low limit of detection (0.38 fM). The construction of biosensors based on porous conducting polymers decorated with AuNPs offers an effective strategy to increase the electroactive surface area of the sensing interfaces, so as to effectively enhance the sensitivity.

Dual-Parameter Modeling of Molecular Binding Lifetime Distribution for Ultrasensitive Biosensors

Wang Chen,Zhang Yiyang,Zeng Qiang,Yang Yuting,Yu Hui

Analyzing single-molecule binding kinetics offers an effective way to reduce the disturbance from nonspecific bindings in biosensors. Here we present a dual-parameter lifetime distribution modeling approach to detect specific binding signals in single-molecule sensors accurately. A proof of concept was demonstrated in the dynamic single-molecule sensing of microRNA using gold-nanoparticle labeled sandwiched assay with low-affinity probes. In this assay, a single molecule binding process was recorded with a large field-of-view plasmonic scattering microscope, and the lifetime distribution was quantified. A model involving two different exponential decaying constants was used to fit the lifetime distribution, providing accurate information about the number of nonspecific binding and specific binding events as well as their dissociation rates. We show both in simulations and experiments that by establishing the calibration curve with the number of specific binding events against different analyte concentrations, an ultra-low limit of detection at the femtomolar level could be achieved. The high sensitivity makes this approach a potential solution to detect low abundance biomarkers for disease diagnosis.

Constructing ultrasensitive biosensor for biomarker detection of early diseases through G-quadruplex probe coupled with aggregation-induced electrochemiluminescence platinum(II) complexes

Tingyi Huang,Li Tan,Wenyi Zhang,Ying Ma,Ziqi Lian,Benlin Hong,Jianqiang Hu

Highly sensitive and simple biomarker determination is crucial for diagnosis and treatment of early diseases. However, it remains a tremendous challenge for rapid, accurate and real time detection of biomarkers at the early stage of diseases due to its extremely low content and serious interference. Herein, we have successfully constructed an ultrasensitive electrochemiluminescence (ECL) biosensor based on 2,6-bis(2-benzimidazolyl)pyridine platinum(II) (BZIMPY-Pt) complexes with aggregation-induced electrochemiluminescence (AIECL) phenomenon, which was utilized to detect miRNA-155 at early diseases. The BZIMPY-Pt complexes with AIECL properties and had high affinity with G-quadruplex DNA compared to other square planar pyridyl Pt(II) complexes, which exhibited 13-fold enhancements in ECL intensity in the aggregated state. The constructed biosensor possessed high sensitivity, wide linear range (1 fM-10 nM), low limit of detection (∼0.22 fM), high selectivity and good stability and reproducibility for miRNA-155 detection. The ECL biosensor was successfully applied to determine miRNA-155 in the serum samples of early-stage diagnose. This AIECL-based strategy overcomes traditional limitations in low-abundance biomarker analysis through synergistic signal amplification and background suppression mechanisms, which is potential for clinical diagnosis of early diseases.

An ultrasensitive biosensor for colorimetric detection of Salmonella in large-volume sample using magnetic grid separation and platinum loaded zeolitic imidazolate Framework-8 nanocatalysts

Lei Wang,Xiaoting Huo,Lingyan Zheng,Gaozhe Cai,Yuhe Wang,Ning Liu,Maohua Wang,Jianhan Lin

Salmonella is the leading risk factor in food safety. Rapid, sensitive and accurate detection of Salmonella is a key to prevent and control the outbreaks of foodborne diseases caused by Salmonella . In this study, we reported a colorimetric biosensor for ultrasensitive detection of Salmonella Typhimurium using a magnetic grid separation column to efficiently separate target bacteria from large volume of sample and platinum loaded zeolitic imidazolate framework-8 ( [email protected] ) nanocatalysts to effectively amplify biological signal. The target Salmonella cells in large volume of sample were first separated and concentrated using the magnetic grid separation column with immune magnetic particle chains, then conjugated with the immune [email protected] nanocatalysts to mimic peroxidase for catalysis of hydrogen peroxide-3,3′,5,5′-tetramethylbenzidine, and finally determined by measuring the catalysate at characteristic wavelength of 450 nm. This proposed biosensor was able to separate ∼70% of target Salmonella cells from 50 mL of bacterial sample and quantitatively detect Salmonella from 10 1 to 10 4  CFU/mL in 2.5 h with the lower detection limit of 11 CFU/mL. The mean recovery for Salmonella in spiked chicken carcass was about 109.8%. This new magnetic grid separation method was first time reported for efficient separation of target bacteria from very large volume of sample to greatly improve the sensitivity of this biosensor and could be used with various biosensing assays for practical applications in routine detection of foodborne pathogens without any bacterial pre-enrichment.
