Cancer cell screening is very important for its treatment and prognosis. The screening techniques are required with high sensitivity and specific recognition. Here, an ultrasensitive tapered optical fiber (TOF) biosensor for cancer cell sensing is proposed. The unique property of the dispersion turning point (DTP) in the TOF is utilized to greatly improve the sensing sensitivity. Combined with numerical calculations, the TOF with the extremely high refractive index (RI) sensitivity (close to 18390 nm/RIU) near the DTP is achieved by precisely controlling the diameter of the TOF. In addition, biotin is used to functionalize the TOF for specific recognition of the biotin-overexpressing cancer cells. The sensitivity and selectivity of the TOF were demonstrated by the detection of the biotin receptor protein (streptavidin) and overexpressed cancer cells (lung epithelial adenocarcinoma cells, A549). The experiment results show that the TOF has a low limit of detection (LOD) for streptavidin and cancer cells, which are 28.73 pM and 49 cells/mL respectively. At the same time, the TOF showed the obvious response to another biotin-overexpressed cancer cell (breast cancer cells, 4T1), but is insensitive to normal cells (gastric mucosa cells, GES-1 and mouse epithelioid fibroblasts cells, L929). We believe that the proposed biosensor which exhibited extremely high sensitivity and good selectivity, will have potential application prospects in the field of chemical and biological detection.

Optical microfiber biosensors demonstrate exceptionally ultra-high sensitivity at the dispersion turning point (DTP). However, the DTP is highly susceptible to variations in dimensional and external environmental factors, and the spectral response is mismatched from preparation in air to application in a liquid environment, making the DTP difficult to control effectively. In this work, we propose a method that bridges the relationship between the interference spectra of air and aqueous environments. By counting the interference peaks in air, we can accurately predict the DTP position in liquids. Meanwhile, it provides a new balance between sensitivity and wide linear dynamic range, achieving wide dynamic range detection across various concentrations. The optical microfiber coupler (OMC) is fabricated using the hydrogen–oxygen flame melting tapering method. In addition, the concentration, temperature, and solvent used for the sensor’s biofunctional layer are optimized. Finally, in refractive index sensing, a maximum sensitivity of 1.17 × 105 ± 0.038 × 105 nm/RIU is achieved. For biosensing, a wide dynamic range detection of cardiac troponin I (cTnI) is realized at concentrations of 12–48 ng/mL, 120–480 pg/mL, and 120–480 fg/mL.

Objectives To forecast the development trend of current outbreak in Dalian, mainly to predict turning points of COVID-19 outbreak in Dalian, Liaoning province, China, the results can be used to provide a scientific reference for timely adjustment of prevention and control strategies. Methods During the outbreak, Bayesian framework was used to calculated the time-dependent reproduction number ( \({R_{t}}\) ), and then above acquired \({R_{t}}\) and exponential trend equation were used to establish the prediction model, through the model, predict the \({R_{t}}\) value of following data and know when \({R_{t}}\) smaller than 1. Results From July 22 to August 5, 2020, and from March 14 to April 2, 2022, 92 and 632 confirmed cases and asymptomatic infected cases of COVID-19 were reported (324 males and 400 females) in Dalian. The R square for exponential trend equation were 0.982 and 0.980, respectively which fit the \({R_{t}}\) with illness onset between July 19 to July 28, 2020 and between March 5 to March 17, 2022. According to the result of prediction, under the current strength of prevention and control, the \({R_{t}}\) of COVID-19 will drop below 1 till August 2, 2020 and March 26, 2022, respectively in Dalian, one day earlier or later than the actual date. That is, the turning point of the COVID-19 outbreak in Dalian, Liaoning province, China will occur on August 2, 2020 and March 26, 2022. Conclusions Using time-dependent reproduction number values to predict turning points of COVID-19 outbreak in Dalian, Liaoning province, China was effective and reliable on the whole, and the results can be used to establish a sensitive early warning mechanism to guide the timely adjustment of COVID-19 prevention and control strategies.
