High-Resolution Imaging of Biological Soft and Hard Tissues Using NIR Scintillators

Xinran Wang,Heng Dai,Yichun Liu,Linglu Kuang,Hao Song,Zhaowei Teng,Dawei Wang,Ting Wang,Xuhui Xu

Near-infrared (NIR) luminescence has emerged as a powerful tool in biological imaging due to its nondestructive nature, high spatiotemporal resolution, and deep tissue penetration. However, traditional NIR imaging often fails to provide detailed information about hard tissues such as bones. In this study, an approach for simultaneous imaging of soft and hard tissues using Lu 1.5 Mg 1.5 Al 3.5 Si 1.5 O 12 : Yb 3+ as an NIR scintillator is presented. Under X-ray irradiation, Lu 1.5 Mg 1.5 Al 3.5 Si 1.5 O 12 : Yb 3+ scintillator emits NIR emission with a peak at 970 nm with thermal quenching resistance, which is strongly absorbed by soft tissues, enabling soft tissue imaging. Additionally, the scintillator provides a high signal-to-noise ratio for X-ray imaging, allowing for high-resolution imaging of hard tissues, unaffected by ambient light, with a spatial resolution of up to 203 µm. By leveraging X-ray as the sole excitation source, this method provides a comprehensive view of internal biological structures, including vascular and skeletal tissues, with high resolution and minimal interference. This innovation holds great promise for advancing medical imaging technologies and enhancing diagnostic capabilities.

IGFBP3 enhances adipose-derived stem cell function in soft tissue injury repair via ITGB1 and ERK pathway activation

Tian Sirui,Yu Haiyang,Yang Ruoxuan,Wang Heshi,Zhao Baohong,Wang Danning

Soft tissue injury (STI) is a prevalent condition that requires effective therapeutic approaches. The focus of this investigation was to elucidate the molecular mechanisms linked to the IGFBP3 protein in adipose-derived stem cells (ADSCs) for STI repair, utilizing single-cell multiomics technology and a 3D bioprinting model. Establishment of a mouse-based STI model facilitated the comparison of cellular compositions and communication variances between wounded and normal tissues through single-cell RNA sequencing (scRNA-seq). High-throughput transcriptomics and bioinformatics analysis pinpointed IGFBP3 as a key target in ADSCs related to STI repair. In vitro experiments assessed IGFBP3's effects on ADSCs' epithelial cell differentiation, proliferation, and migration using various assays and lentivirus transfection to manipulate IGFBP3 expression. A 3D bioprinting technique was used to create an ADSCs-IGFBP3 peptide self-assembling hydrogel scaffold, characterized by Fourier-transform infrared spectroscopy, X-ray diffraction, SEM, and TEM. The scaffold's efficacy was validated in an animal model. Results showed nine cell subtypes in both normal and injured tissues, with increased ADSCs in STI tissues exhibiting enhanced connectivity and interactions. RNA-seq analysis confirmed IGFBP3 as crucial for ADSCs and STI. In vitro and 3D bioprinting experiments, along with animal model validation, confirmed IGFBP3's role in STI repair. Upregulation of IGFBP3 in ADSCs promoted epithelial cell differentiation by enhancing ITGB1 expression, activating the ERK pathway to boost cell proliferation and migration. This study highlights IGFBP3's significant role in ADSCs for STI repair, providing potential molecular targets for developing new treatments. The findings offer valuable insights into IGFBP3's mechanisms, aiding in advancing STI therapeutic strategies. Graphical 1. IGFBP3 enhances ADSC function in STI repair by activating ITGB1 and ERK pathways. 2. High IGFBP3 expression promotes ADSC differentiation into epithelial cells, improving tissue regeneration. 3. Single-cell multiomics and 3D bioprinting validate IGFBP3’s role in advancing STI treatment. 4. Findings present IGFBP3 as a promising molecular target for clinical STI therapy.

Preparation and Heat-Setting of Warp-Knitted Silk Patches for Soft Tissue Repairs

Linya Yan,Jinfeng Wang,Ying Mao,Hongshi Zhao,Wenxing Chen,Wangyang Lu

Currently, synthetic materials for soft tissue repair are non-biodegradable and rigid, while biodegradable natural polymer materials exhibit low mechanical strength. This study focuses on creating biodegradable silk patches with superior mechanical properties using warp knitting. Four kinds of silk patches were made, degummed, and their structural parameters and mechanical properties were evaluated. An orthogonal experiment was conducted to investigate the effects of heat-setting temperature and time on the patches' mechanical properties. The results show that the #D patch, with its moderate thickness and density, large and concentrated pores, high strength, and low elongation, closely resembles natural soft tissue. After heat setting at 120°C for 3 min, the mechanical properties significantly enhanced (up to 17.1%). The patch exhibited a longitudinal tensile strength of 276.81 N, transverse tensile strength of 330.73 N, burst strength of 1566.5 N, longitudinal tear strength of 307.33 N, transverse tear strength of 124.66 N, longitudinal suture pullout strength of 53.28 N, and transverse suture pullout strength of 73.89 N. This method of using warp knitting and digital weaving technology allows for the preparation of silk patches suitable for soft tissue repair, with high strength, stability, and porosity, making them ideal for tendon and rotator cuff repairs.

Lipoic acid-based stable underwater adhesive with repeatable adhesion and recyclability

Zhenchun Li,Zhanhe Zhang,Xu Ji,Wenyu Wang,MingLu Xu,Huijuan Cui,Chuang Du,Lei Wang

The development of fast, recyclable, and stable adhesion is crucial across multiple fields. Adhesives based on lipoic acid (LA) as a raw material have garnered significant attention; however, their susceptibility to closed-loop depolymerization after polymerization greatly limits their applications. We have designed a stable underwater adhesive through a straightforward preparation process that exhibits high adhesive strength, excellent mechanical properties, and resistance to swelling, making it suitable for soft tissue repair under high burst pressure. This adhesive effectively seals isolated organ breakage and achieves instant adhesion to a variety of substrates. Notably, repeatable adhesion exceeding one hundred cycles has been accomplished. A simple and effective polymerization reaction is employed to prevent depolymerization, allowing for the long-term storage of the adhesive. The adhesive is degradable, biocompatible, and sustainable, enabling reprocessing multiple times to create a robust adhesive interface. The adhesive can be continuously produced using closed-loop recyclable monomers. Additionally, it is utilized in human electromyogram (EMG) and electrocardiogram (ECG) applications, where it effectively collects stable and realistic physiological signals. This study presents a novel, straightforward, and environmentally friendly strategy for designing a stable LA-based underwater adhesive. Furthermore, it offers new insights for the design and application of conductive underwater adhesives.

An injectable hyaluronic acid/lithium calcium silicate soft tissue filler with vascularization and collagen regeneration

Jinzhou Huang,Jianmin Xue,Jimin Huang,Xinxin Zhang,Hongjian Zhang,Lin Du,Dong Zhai,Zhiguang Huan,Yufang Zhu,Chengtie Wu

The significance of collagen and vascular in skin augmentation have been recognized in recent years. However, current skin tissue fillers, e.g. hyaluronic acid (HA) or HA-based hydrogel, fail to meet the perfect augmentation requirements due to their inadequate long-term support effect and the lack of tissue-inducing activity. Herein, an injectable skin filler containing hyaluronic acid (HA) hydrogel and lithium calcium silicate (LCS, Li 2 Ca 4 Si 4 O 13 ) bioceramic microspheres was developed for skin tissue fillers, owing to the excellent biological function of silicate bioceramics. The HA-LCS fillers could be easily injected through a tiny standard medical needle (27 G) with force of less than 36 N, and showed good biocompatibility both in vitro and in vivo . Furthermore, the bioactive ions released from HA-LCS fillers significantly enhanced the expression of vascularization-related genes and collagen-related genes. Importantly, the HA-LCS fillers not only stimulated the regeneration of mature blood vessels, but also promoted collagen secretion in dermal skin and filling area. This study not only presented an injectable filler with enhanced regeneration of blood vessels and collagen, but also provided a new strategy for developing tissue-induced fillers based on bioactive components of silicate bioceramics.

Functionalized Rhodium Nanoparticles as Antimicrobial Agents for Treatment of Drug-Resistant Skin and Soft Tissue Infections

Qingqin Tan,Zhenxing Chen,Lei Liu,Xi Huang

Skin and soft tissue infections (SSTIs) are among the most common bacterial infections reported in outpatients. Drug-resistant bacteria are the major cause of treatment failure and increased mortality rate in patients with SSTIs, posing significant challenges to human health. In this study, new-generation rhodium nanoplates (RhNPs) and glycol chitosan- and polydopamine-functionalized RhNPs (Rh@GCS) are developed for the treatment of drug-resistant SSTIs. RhNPs exhibited favorable antibacterial activity against methicillin-resistant Staphylococcus aureus (MRSA) and Ag-resistant MRSA. The modified Rh@GCS exhibited enhanced antibacterial activity and can directly kill various drug-resistant bacteria by increasing the permeability of cell membranes, including gram-positive MRSA and gram-negative multidrug-resistant Escherichia coli ( E . coli ) and Pseudomonas aeruginosa (PA). Moreover, Rh@GCS effectively inhibited bacterial growth and promoted the healing of skin lesions in MRSA-induced SSTI mouse models. These results suggest that Rh@GCS is a promising nonantibiotic antimicrobial agent for the treatment of drug-resistant SSTIs.

Sensitive detection of cell-derived force and collagen matrix tension in microtissues undergoing large-scale densification

Xiang Wang,Qiang Gao,Xiaoning Han,Bing Bu,Longfei Wang,Aoqi Li,Linhong Deng

Mechanical forces generated by cells and the tension of the extracellular matrix (ECM) play a decisive role in establishment, homeostasis maintenance, and repair of tissue morphology. However, the dynamic change of cell-derived force during large-scale remodeling of soft tissue is still unknown, mainly because the current techniques of force detection usually produce a nonnegligible and interfering feedback force on the cells during measurement. Here, we developed a method to fabricate highly stretchable polymer-based microstrings on which a microtissue of fibroblasts in collagen was cultured and allowed to contract to mimic the densification of soft tissue. Taking advantage of the low-spring constant and large deflection range of the microstrings, we detected a strain-induced contraction force as low as 5.2 µN without disturbing the irreversible densification. Meanwhile, the microtissues displayed extreme sensitivity to the mechanical boundary within a narrow range of tensile stress. More importantly, results indicated that the cell-derived force did not solely increase with increased ECM stiffness as previous studies suggested. Indeed, the cell-derived force and collagen tension exchanged dramatically in dominating the microtissue strain during the densification, and the proportion of cell-derived force decreased linearly as the microtissue densified, with stiffness increasing to ∼500 Pa. Thus, this study provides insights into the biomechanical cross-talk between the cells and ECM of extremely soft tissue during large-extent densification, which may be important to guide the construction of life-like tissue by applying appropriate mechanical boundary conditions.

An Artificial Piezoelectric-Conductive Integrated Peri-Implant Gingiva Enables Efficient Bacterial Inhibition and Soft-Tissue Integration

Han Wen,Liu Zhiqing,Yu Hao,Zhang Yaqi,Mei Enhua,Wang Wei,Chen Feng,Cao Wentao,Qi Shengcai

Peri-implantitis is the main reason for dental implant failure. Optimizing electroactivity at the interface between dental implants and tissue is essential for enhancing integration and preventing bacterial invasion. Here, a bioinspired piezoelectric-conductive integrated peri-implant gingiva (PiG) with simultaneously enhanced antibacterial efficacy and soft-tissue integration, which is based on a flexible piezoelectric film and conductive polymer network, is presented. The piezoelectricity of PiG is achieved through the electrospinning of polyvinylidene fluoride/BaTiO 3 /MXene on a polydopamine-modified plasma-activated Ti surface, whereas the conductive property of PiG is achieved by the in situ polymerization of 3,4-ethylenedioxythiophene monomers. Under ultrasonic irradiation, PiG can promote the formation of neutrophil extracellular traps and reactive oxygen species, thus achieving synergistic and efficient piezodynamic killing of Staphylococcus aureus ( S. aureus ) and Escherichia coli (E. coli) . Additionally, piezoelectricity-enabled electrical stimulation endows PiG with enhanced fibroblasts adhesion, proliferation, and collagen secretion. As a demonstration, ultrasound irradiation of PiG-grafted Ti implanted in a subcutaneous implantation rat model efficiently eliminates the S. aureus infection and rescues the implant with increased soft-tissue integration. The concept of an artificial PiG is anticipated to open new avenues for the development of high-performance implant materials, potentially extending their lifespans. Graphical abstract

Gelatin-crosslinked pectin nanofiber mats allowing cell infiltration

Xiaoqi Shi,Sisi Cui,Xiaoyu Song,Alex P. Rickel,Hanna J. Sanyour,Jia Zheng,Junli Hu,Zhongkui Hong,Yifa Zhou,Yichun Liu

Pectin nanofiber mats are promising tissue engineering scaffolds but suffer from poor cell infiltration. In this study, gelatin, a collagen derived cell adhesive protein, was used to crosslink the electrospun nanofibers of periodate oxidized pectin. Cell culture experiment results demonstrated that cells were able to grow into the gelatin-crosslinked pectin nanofiber mats rather than only spread on mat surface. The nanofiber mats showed moderate mechanical strength, with a maximum tensile strength of up to 2.3 MPa, an ultimate tensile strain of up to 15%, and were capable of degrading gradually over 4 weeks or even longer periods in simulated body fluids. Thus, gelatin-crosslinked pectin nanofiber mats hold a great potential for soft tissue regeneration.

Melt-based embedded printing for freeform fabrication of overhanging and flexible polycaprolactone scaffolds

Xiangbin Zeng,Zijie Meng,Zhennan Qiu,Jiankang He,Jinhai Fan,Dichen Li

We developed a novel melt-based embedded printing strategy to fabricate polycaprolactone (PCL) structures with complex overhanging geometries within a thermally stable supporting matrix. By optimising the concentration of the supporting matrix, process parameters, and inter-filament thickness, continuous PCL filaments can be stably printed with the smallest width of 50.6 ± 4.7 μm. The one-step in situ post-solidification process provides a unique approach to regulate the surface morphology of the printed filaments, and reduce structural anisotropy. Various 3D structures with overhanging geometries, such as truss structures and flexible scaffolds were successfully fabricated by melt-based embedded printing. The effective modulus of the printed flexible PCL scaffolds can be widely tuned in the range of 867.4 ± 21.6 to 9.1 ± 1.2 kPa by adjusting the design parameters. We envision that the presented strategy might provide an innovative tool to fabricate flexible polymeric scaffolds with complex structural organisations for soft tissue engineering.

Constructing an Injectable Multifunctional Antibacterial Hydrogel Adhesive to Seal Complex Interfaces Post–Dental Implantation to Improve Soft Tissue Integration

Yubing Hong,Siwei Wang,Abudusaimi Maimaiti,Jiarun Li,Dongying Li,Qinmei Wang,Wei Teng

Soft tissue integration (STI) around dental implants determines their long-term success, and the key is to immediately construct a temporary soft tissue-like barrier to prevent bacterial invasion after implantation and then, promote STI. In response to this need, an injectable multi-crosslinked hydrogel (MCH) with abilities of self-healing, anti-swelling, degradability, and dry/wet adhesion to soft tissue/titanium is developed using gallic acid-graft-chitosan, oxidized sodium alginate, gelatin, and Cu 2+ with water and borax solution as solvents, whose properties can be controlled by adjusting its composition and ratio. MCH can not only immediately build a sealing barrier to block the bacterial invasion in the oral simulation environment but also deliver outstanding antibacterial efficacy through the synergism of trapping bacteria and releasing bactericidal agents such as chitosan, gallic acid, aldehyde, and Cu 2+ . Moreover, MCH has an adjustable ROS-scavenging ability imparted by gallic acid, chitosan, and gelatin to reduce inflammation and can control the release of Cu 2+ . Based on these, it is believed that by injecting MCH around implants (percutaneous/transmucosal) after surgery, a universal non-aggressive strategy to promote STI can be developed for long-term implant success.

Chitosan ducts fabricated by extrusion-based 3D printing for soft-tissue engineering

C.Q. Zhao,W.G. Liu,Z.Y. Xu,J.G. Li,T.T. Huang,Y.J. Lu,H.G. Huang,J.X. Lin

Three kinds of methods based on extrusion and 3D printing and different acidic solutions (formic acid (FA), acetic acid (AA), glycolic acid (GA) and lactic acid (LA)) were applied for manufacturing the CS ducts. The tensile properties and preliminary cytotoxicity were measured for selecting the optimal ratio of CS slurry. The 3D printability of CS slurry was also studied. The tensile strength, Young’s modulus, and fracture strain were tested for evaluating the degree of mechanical matching to soft-tissue. The optimal solvent to CS was 30 wt.% GA solution. The CS slurry possessing shear-thinning properties was suitable for 3D printing. The tensile strength, Young’s modulus, and fracture strain of the CS rods were 10.98 ± 0.61 MPa, 12.38 ± 1.19 MPa, and 146.03 ± 15.05 %, correspondingly. The CS ducts manufactured by 3D printing had an excellent mechanical matching to soft-tissue, outstanding biocompatibility and have great potential for soft-tissue restorations.
