Human hair keratin, a natural protein derived from human hair, has emerged prominently in the field of wound repair, showcasing its unique regenerative capabilities and extensive application potential. However, it is a challenge for the keratin to efficiently therapy the impaired wound healing, such as combined radiation-wound injury. Here, we report a keratin/chitosan (KRT/CS) film for skin repair of chronic wounds in in rats with combined radiation-wound injury. In brief, the KRT/CS film was characterized by scanning electron microscopy (SEM), mechanical property analysis, water absorption, and swelling analysis. A rat model of combined radiation-wound injury was employed to evaluate the therapeutic efficacy of the KRT/CS film. Finally, the systemic biotoxicity of KRT/CS film was assessed through histological analysis. The surface of KRT/CS film was uniform and smooth compared with the KRT film, and the mechanical property, swelling rate and water absorption rate of KRT/CS film were significantly improved, which can meet the application requirements of wound excipient dressing. Furthermore, the combined radiation-wound injury in rats was established that the wound closure rate was achieved 74.46% after 14 days of treatment with KRT/CS film, comparing to the single KRT membrane and commercially available Band-Aids. Histological analysis demonstrated that the amount of angiogenesis and collagen deposition in wounds treated with KRT/CS were significantly improved. These findings demonstrate the KRT/CS film as a promising therapeutic agent for combined radiation-wound injury. Graphical

The radiation damage of biomolecules, in particular with aliphatic compound, has been extensively studied. Morpholine is a typical six-membered aliphatic heterocyclic compound. In the present work, photoionization and dissociation of the morpholine monomer and subsequent fragmentations have been investigated by synchrotron vacuum ultraviolet photoionization mass spectrometry and theoretical calculations. The vertical ionization energy of morpholine monomer is 8.37±0.05 eV, which agrees reasonably well with a theo-retical value 8.41 eV of morpholine. Experimentally observed fragmentation of morpholine ( m/z =87 amu) gives rise to m/z =86 amu, m/z =57 amu, and m/z =29 amu. Based on experimental and theoretical results, it is found that the m/z =86 amu is produced by losing H atom, the m/z =57 amu is formed by the elimination of CH 2 O with a ring-opening process, the m/z =29 amu is generated by further dissociation of the fragment m/z =57 amu (C 3 H 7 N) + by the elimination of C 2 H 4 . This finding would provide valuable insight into the photo-damage of aliphatic compounds, which may be related to living cells and other biological system.

The long-term maintenance of hematopoietic stem cells (HSCs) relies on the regulation of endoplasmic reticulum (ER) stress at a low level, but the underlying mechanism remains poorly understood. Here, we demonstrate that suppression of ER stress improves the functions of HSCs and protects HSCs against ionizing radiation (IR)-induced injury. We identify epithelial membrane protein 1 (EMP1) as a key regulator that mitigates ER stress in HSCs. Emp1 deficiency leads to the accumulation of protein aggregates and elevated ER stress, ultimately resulting in impaired HSC maintenance and self-renewal. Mechanistically, EMP1 is located within the ER and interacts with ceramide synthase 2 (CERS2) to limit the production of a class of sphingolipids, dihydroceramides (dhCers). DhCers accumulate in Emp1 -deficient HSCs and induce protein aggregation. Furthermore, Emp1 deficiency renders HSCs more susceptible to IR, while overexpression of Emp1 or inhibition of CERS2 protects HSCs against IR-induced injury. These findings highlight the critical role played by the EMP1-CERS2-dhCers axis in constraining ER stress and preserving HSC potential.

Radioactive cesium (Cs) is a significant concern due to its role as a major byproduct of nuclear fission and its potential for radioactive contamination. Internal contamination with radioactive Cs is characterized by immoderate production of reactive oxygen species (ROS), resulting in severe radiation damage. Therefore, the development of therapeutic strategies should focus on enhancing the excretion of radioactive Cs and reducing radiation-induced oxidative damage. However, current therapeutic drugs like Prussian blue (PB) have limited efficacy in addressing these issues. In this study, we present Cu3[Fe(CN)6]2 (CuFe) nanoparticles, a Prussian blue analog (PBA), which can not only efficiently sequester Cs but also exhibit resistance against radiation damage. The results of the adsorption studies demonstrate that CuFe outperforms PB in terms of adsorption performance. Further mechanistic investigations indicate that the increased adsorption capacity of CuFe may be attributed to the presence of additional defects resulting from the [Fe(CN)6] missing linkers. Moreover, CuFe mimics the functions of catalase (CAT) and superoxide dismutase (SOD) by effectively eliminating O2˙− and H2O2 while scavenging ˙OH, thereby mitigating ROS induced by radiative Cs. Importantly, in vivo study confirms the efficient Cs decorporation capability of CuFe. The fecal cumulative excretion rate of CuFe reaches 69.5%, which is 1.45 times higher than that of PB (48.8%). These findings demonstrate that CuFe exhibits excellent Cs removal performance and ROS scavenging ability, making it an attractive candidate for the treatment of Cs contamination.

Electron radiation damage hinders the high-resolution cryogenic transmission electron microscopy (cryo-TEM) imaging of macromolecules. In this work, sodium ascorbate (SA) was used to eliminate reactive oxygen species generated during electron radiation, thereby enhancing the electron radiation resistance of macromolecular samples in cryo-TEM experiments. The effectiveness of SA on protecting sample morphology and structure was investigated for representative types of macromolecular samples, including collagen, cellulose, liposome, and apoferritin, and compared with other radical scavengers. Our findings revealed that SA provided the most effective protection for collagen, increasing its dose limit by more than 40%. Similar effects were observed for cellulose and apoferritin. Importantly, the addition of SA does not change the three-dimensional structure of apoferritin, as confirmed by a 2.2 Å resolution single particle analysis, further indicating the practicality of the method.

This study proposes a novel method for the detection of inert gas radon using a label-free, specific, fluorescence-sensing aptamer in the context of PW17-OG system. This method utilizes the cyanine dye OliGreen (OG) as a signal reactor and the aptamer PW17 as a fluorescent identification probe. When OG integrates into the free curling PW17, a strong fluorescence signal is generated. After radon decays, the long lived naturally occurring radon progeny Pb being disposed and introduced to the system. Lead ions induce PW17 to form a stable G-quadruplex, thereby inhibiting the interaction between OG and PW17 and resulting in a reduction of the fluorescence intensity . The fluorescence intensity show a good linear relationship with lead ion and the radon concentration ( D ), thereinto, We fitted linear regression of radon concentration in the range of 0.92–4.22 (×10 4 Bqhm −3 ) to receive a good relationship between Δ F and the concentration of radon with the detection limit of 1963 Bqhm −3 . This method has been successfully applied for detecting standard cumulative concentration of radon and the detection limit reached the national standard of China. This sensitive method can exclude radiation damage in field testing, furthermore, it explores a new field in biological analysis using an aptamer to detected inorganic, gaseous, and radioactive materials.

Radiotherapy is one of the most important clinical cancer treatments, which works mainly by delivering a prescribed radiation dose to the tumor tissues. However, high doses of radiation may also lead many irreversible damages to the surrounding normal tissues. Thereby, how to effectively reduce these side-effects has been a significant factor in influencing cancer therapeutic effect. In this work, we synthesized the hollow PtPd nanocubes with high-index facets, and investigated the radiation protection capability in vitro and in vivo . Our results showed the PtPd nanocrystals can decrease the ROS level and improve the survival rate of radiated cells. Meanwhile, survival rate of radiated mice can significantly increase from 0 to 30% after PtPd treatment. Consequently, the enzyme and ROS level in radiated mice can be recovered.

A label-free, highly sensitive, and selective biosensor for detection of interleukin-6 (IL-6), which is an important inflammatory and cancer progression cytokine, has been achieved by combining gold nanoparticles (NPs) array surface-enhanced Raman spectroscopy (SERS) substrate and DNA aptamer. The SERS substrate consisted of Au NPs array is functionalized with the aptamer which is composed of the IL-6 recognition sequence and the output signal reporter. Upon recognition of target IL-6 in serum, the aptamer changes its conformation, resulting in the corresponding change of the output Raman intensity ratio (I 660 /I 736 ), with which the quantitative evaluation of IL-6 in 10 -12 -10 -7 M range can be realized. This SERS approach was employed to detect IL-6 in mice serum for inspecting the effect of drug and bacterial infection, and particularly, the radiation injury in the treated animals. This work therefore exemplifies that the aptamer-based SERS biosensor can serve as a promising tool for quick and convenient medical diagnosis applications.

Mesenchymal stromal cells (MSCs) are believed to migrate to injury sites, release chemical attractants, and either recruit local stem cells or modulate the immune system positively. Although MSCs are highly desired for their potential to reduce inflammation and promote tissue regeneration, their limited lifespan restricts their applications. This study presents a simple approach for protecting MSCs with epigallocatechin-3-gallate (EGCG) and magnesium (Mg) based metal-organic framework coatings (E-Mg@MSC). The layer strengthens MSCs resistant to harmful stresses and creates a favorable microenvironment for repair by providing Mg to facilitate MSCs' osteogenic differentiation and using EGCG to neutralize excessive reactive oxygen species (ROS). E-Mg@MSC serves as a treatment for hematopoietic injury induced by ionizing radiation (IR). Coated MSCs exhibit sustained secretion of hematopoietic growth factors and precise homing to radiation-sensitive tissues. In vivo studies show substantial enhancement in hematopoietic system recovery and multi-organ protection. Mechanistic investigations suggest that E-Mg@MSC mitigates IR-induced ROS, cell apoptosis, and ferroptosis, contributing to reduced radiation damage. The system represents a versatile and compelling strategy for cell-surface engineering with functional materials to advance MSCs therapy.
