NIR-II fluorescence nanoprobe based on Erbium for fallopian tube diseases diagnosis

Guangxin Duan,Zhuxin Wei,Jingyu Zhang,Xuexiao Li,Shuwang Wu,Jianfeng Zeng,Ximing Wang,Hong Zhang,Ling Wen,Mingyuan Gao

Fallopian tube blockage-related diseases are the main cause of female infertility. Diagnosis of tubal patency and accurate determination of tubal obstruction lesions is of great value for infertility treatment. Conventionally, the common used hysterosalpingogram (HSG) has the disadvantages of radioactive hazard and high false positive rate. Thus, the development of a safe and accurate method which can replace traditional HSG for tubal patency diagnosis is urgently needed. Recently, fluorescence imaging in the second near-infrared region (NIR-II) was widely used in in-vivo imaging research due to its excellent tissue penetration and signal-to noise ratio. With the aid of fluorescence agents, NIR-II imaging has the potential to diagnose tubal patency, which, unfortunately, has not been explored. Herein, a kind of rare-earth Erbium-based nanoprobe (Er-RENPs) with outstanding NIR-II fluorescence property was synthesized. Based on the Er-RENPs, the potential of NIR-II fluorescence imaging was explored. Results show that the outline of fallopian tubes can be clearly visualized with the help of Er-RENPs. And the stenotic, occlusive fallopian tubes and hydrosalpinx can be diagnosed. Additionally, the Er-RENPs exhibit good biocompatibility. In this study, we reported that NIR-II rare-earth fluorescent nanoprobe can serve as a promising fluorescent agent, and exhibits considerable potential application for tubal diagnosis.

Soft Millirobot Capable of Switching Motion Modes on the Fly for Targeted Drug Delivery in the Oviduct

Yuan Liu,Jing Huang,Chu Liu,Zhongyi Song,Jiandong Wu,Qilong Zhao,Yingtian Li,Fuping Dong,Lei Wang,Haifeng Xu

Small-scale magnetic robots with fixed magnetizations have limited locomotion modes, restricting their applications in complex environments in vivo. Here we present a morphology-reconfigurable millirobot that can switch the locomotion modes locally by reprogramming its magnetizations during navigation, in response to distinct magnetic field patterns. By continuously switching its locomotion modes between the high-velocity rigid motion and high-adaptability soft actuation, the millirobot efficiently navigates in small lumens with intricate internal structures and complex surface topographies. As demonstrations, the millirobot performs multimodal locomotion including woodlouse-like rolling and flipping, sperm-like rotating, and snake-like gliding to negotiate different terrains, including the unrestricted channel and high platform, narrow channel, and solid–liquid interface, respectively. Finally, we demonstrate the drug delivery capability of the millirobot through the oviduct-mimicking phantom and ex vivo oviduct. The magnetization reprogramming strategy during navigation represents a promising approach for developing self-adaptive robots for performing complex tasks in vivo.

Oviduct-derived C-type natriuretic peptide improves bovine preimplantation embryo development in vitro by attenuating oxidative damage

Lin Wang,Zhiqi Cao,Zhaochen Wang,Zhenni Zhang,Juan Liu,Weiwei Wu,Juncheng Huang,Lei An,Jianhui Tian,Guangyin Xi

It is widely accepted that the poor quality of in vitro -produced (IVP) embryos becomes one of the most intractable issues currently limiting the outcomes of human assisted reproductive technology (ART). Although diverse molecules and pathways may be involved in this process, strategies to optimize culture conditions for the improvement of embryo development potential and the relevant mechanisms remain underexplored. Here, we demonstrate that the supplementation of C-type natriuretic peptide (CNP), a potential embryokine present in the oviduct, can improve the blastocyst formation rate and embryo quality, as evidenced by an increased total cell number and inner cell mass (ICM) cells, elevated embryonic cell proliferation, and reduced embryonic cell apoptosis. RNA-sequencing analysis suggests that CNP may regulate biological processes related to lipid metabolism and redox homeostasis in early embryos. Furthermore, our results reveal that CNP significantly reduces lipid droplet content, fatty acid storage, and the expression of lipogenesis-related genes. Notably, we show that CNP alleviates the elevated reactive oxygen species (ROS) levels and lipid peroxidation in in vitro embryos. These findings demonstrate that CNP supplementation is a feasible and effective approach to prevent deterioration in embryo quality and improve developmental potential by inhibiting excessive lipid droplet accumulation and elevated oxidative stress in embryos. Collectively, our study not only provides new insights into understanding the crosstalk between early mammalian embryos and the maternal reproductive tract but also presents a promising strategy for improving the success rate of human ART.
