Physicochemical and thermal characteristics of Moringa oleifera seed oil were studied. The refractive index of Moringa oleifera seed oil at ambient temperature was 1.46. The acid, peroxide, saponification, and iodine values content of Moringa oleifera seed oil were 2.35 g KOH/kg oil, 7.2 meq O 2 /kg oil, 189.84 g KOH/kg oil, and 66.78 g I 2 /100 g oil, respectively. Moringa oleifera seed oil contains ten fatty acids. Among 10 kinds of fatty acids in Moringa oleifera seed oil, oleic acid is the most abundant fatty acid comprising 70.85% of total oil followed by palmitic acid (8.98%). The unsaturated fatty acids represented by oleic acid are mainly distributed in the Sn-2 position, which is as high as 95.59%. It contains a high level of unsaturated fatty acids. Moringa oleifera seed oil contains 233.29 mg/kg α-tocopherol, 9.04 mg/kg β-tocopherol, and 10.13 mg δ-tocopherol, respectively. Determination of 10 elements in Moringa oleifera seed oil by ICP-MS. Among them, calcium has the highest content. Moringa oleifera seed oil remained stable up to 305 °C with a negligible degradation. Moringa oleifera seed oil has high nutritional value and broad development prospect. This research provides a prospect for the comprehensive development and utilization of Moringa oleifera seed oil. The water-soluble protein was extracted from Moringa oleifera seed press cake and tested its performance as a natural coagulant for removing humic acid (HA) from water solution when comparing to polyaluminum chloride (PAC), a commercial coagulant. Influencing factors including pH, coagulant dosage, temperature and suspended solid were investigated in this study. The results indicated that the coagulating efficiencies of HA using the Moringa oleifera seed protein (MOSP) were better than those by PAC under all the experimental conditions at the same dosage. MOSP could increase the zeta potential more efficiently than PAC. The Fourier transform infrared spectra revealed that no new compounds were generated during the coagulation process. Hence, adsorption and charge neutralization could be the main mechanism contributing to the HA removal using MOSP as coagulant. MOSP is an ideal natural coagulant for HA removal.

This study employed novel green solvents, deep eutectic solvents (DES), for protein extraction from Moringa oleifera seeds. Two DESs systems including choline chloride-malic acid (ChCl-MA 2 ) and choline chloride-urea (ChCl-U) exhibited the highest protein extraction efficiency, reaching 31.415 % and 31.945 %, respectively. Molecular dynamics (MD) simulation results revealed that the DESs systems showed smaller Root Mean Square Deviation (RMSD), Solvent Accessible Surface Area (SASA), and Radius of Gyration (Rg) values, along with a significant number of hydrogen bonds. The enhanced extraction efficiency of DESs was attributed to the formation of intermolecular hydrogen bonds. Both hydrogen bonding and hydrophobic interactions promoted protein aggregation, thereby stabilizing the protein structure and preserving its integrity. Spectroscopic analyses further indicated that proteins extracted with DESs displayed a more stable and ordered structure. However, DES-extracted proteins exhibited lower surface hydrophobicity (2000–4000) and reduced solubility (40 %–50 %), which significantly compromised their functional properties. Although DES-treated proteins displayed a superior foaming capacity (57.333 %), their emulsifying properties (3.464 m 2 /g and 1.584 m 2 /g, respectively) were comparatively unsatisfactory. In conclusion, DES represent a promising green and efficient extraction method for protein extraction. This work provides valuable insights into DES-mediated protein extraction and offers guidance for the efficient extraction of high-quality proteins from Moringa oleifera seeds.

Gastrodigenin rhamnopyranoside (GR) is a hepatoprotective compound that exists in Moringa oleifera seeds. However, the UPLC-MS/MS method for the determination of GR ( in-vitro / in-vivo ) is lacking clarification. Herein, this study established the UPLC-MS/MS technique, which was effective and sensitive for the investigation of the pharmacokinetics and biodistribution of GR in rats and mice. The separation was achieved with a Shim-pack XR-ODS III C 18 column (2.0 × 75 mm, 1.6 μm) at 40 °C, while the mobile phase (Acetonitrile/0.1% Formic acid =12:82, v /v) was at an eluting rate of 0.2 mL/min. The Multiple Reaction Monitoring (MRM) was selected for quantification, i.e. , m / z [M + HCOO] − 314.9 → 269 for GR and m/z [M + HCOO] - 182.85 → 137 for Tyrosol as the internal standard. The calibration curves were linearly ranged from 10 to 2500 ng/mL ( r ≥ 0.999) with a lower-limit-of-quantification (LLOQ) of 10 ng/mL in the various biological samples (plasma, liver, heart, lung, spleen, brain, kidney). The intra- and inter-day precision was within 5%, while accuracy ranged from −11.4% - 8.33%. Recovery and matrix effect were with 80.32 to 101.31% and 90.36 to 103.76%, respectively, in a reasonable range. After oral and intravenous administration, GR was detected within 3 h but decreased rapidly in plasma, indicating fast elimination. Also, GR was quickly distributed in the various tissues, particularly in the kidney and spleen. The results demonstrated that the established UPLC-MS/MS method was highly linear, precise and accurate with the potential to be used for the quantitative analysis of GR in-vivo .

The hypoglycemic peptide MoHpP-2 from Moringa oleifera seeds exhibits excellent α-glucosidase inhibitory activity, but its amelioration in insulin resistant (IR) cells and type 2 diabetes mellitus (T2DM) mice is still unclear. Therefore, the present study aimed to investigate the hypoglycemic effect of MoHpP-2 on C2C12/IR cells, and gut microbiota and metabolite changes of T2DM mice using 16S rRNA high-throughput sequencing and metabolomics. MoHpP-2 within the concentration range could effectively enhance the glucose consumption levels and antioxidant capacity in C2C12/IR cells. MoHpP-2 intervention improves the blood sugar levels, liver function, and organ indices, reverses oxidative damage, and enhances the antioxidant capacity of T2DM mice. Furthermore, it increases the diversity of cecum microorganisms, decreases the ratio of Firmicutes to Bacteroides and some pathogenic bacteria , and reshapes the gut microbial composition of T2DM mice. Moreover, the metabolomic analysis results showed that MoHpP-2 intervention might promote unsaturated fatty acid biosynthesis, inhibit bile acid formation, and regulate linoleic acid metabolism and amino acid metabolism related to oxidative stress, thereby improving insulin resistance and sensitivity in T2DM mice. Overall, the experimental results provide insights into the hypoglycemic effect of MoHpP-2 in vitro and in vivo and its application in hypoglycemic functional food products.
