Potential folate-producing strains and their applications in biofortification of fermented Moringa oleifera leaves powder

Huan Du,Yingyun Hu,Kaixiang Lu,Tao Li,Yang Tian,Yongjin Hu

Folate is an essential vitamin for the human body and must be obtained exogenously. It is recognized that some microorganisms are capable of producing folates during growth, revealing possibilities for innovations of natural folate biofortified foods. The aim of this study was to identify potential folate-producing strains to enhance folate content in fermented Moringa oleifera leaves powder (MOLP). Although most of the tested strains exhibited folate production, there were significant differences among them in terms of both quantity and distribution, whether intracellular or extracellular. 10 strains with potential for folate production were chosen for enhancing the folate content of MOLP. The two most promising strains, Lactiplantibacillus sp. LP1 and Saccharomyces cerevisiae Y-J-1, demonstrated high folate levels up to 1940 and 2115 μg/100 g dry matter, respectively. These levels were 1.6 and 1.8-fold the initial value in MOLP. Additionally, the fermentation of MOLP not only enhanced its nutritional quality but also preserved its significant bioactivity. This study provides the initial understanding of how selected microorganisms can naturally enrich folates in fermented MOLP, presenting an alternative solution to address the widespread issue of folate deficiency while promoting human health.

Effects of impregnate temperature on extraction of caffeoylquinic acid derivatives from Moringa oleifera leaves and evaluation of inhibitory activity on digestive enzyme, antioxidant, anti-proliferative and antibacterial activities of the extract

Yahya Saud Hamed,Mohamed Abdin,Guijie Chen,Hafiz Muhammad Saleem Akhtar,Xiaoxiong Zeng

Summary In an ultrasonic bath and for 30 min in the presence of water as extraction solvent in a ratio of 1:10 (g/mL), effect of extraction temperature on extraction of caffeoylquinic acids (CQAs) from Moringa oleifera leaves (MOL) was investigated. Gradually increasing in extraction temperature caused a growing in mono-CQA and declining in 3,5-diCQA. The extract obtained at 80 °C showed the highest antioxidant capacity, and IC 50 values for scavenging activities on DPPH radicals and ABTS radicals were 56.9 and 53.6 µg/mL, respectively. Compared with α-glucosidase and α-amylase, the extract showed the highest inhibition on pancreatic lipase (IC 50 of 0.073 mg/mL). Increasing the extraction temperature improved cytotoxicity of the extract on cancer cell lines of HeLa and HepG2. Compared with gram-negative Escherichia coli and Salmonella typhimurium , all the extracts showed higher inhibition on the growth of gram-positive Bacillus cereus and Staphylococcus aureus . Our results suggested that use of higher extraction temperature (80 °C) improved the biological activity of MOL extract.

Hierarchically porous carbon nanosheets derived from Moringa oleifera stems as electrode material for high-performance electric double-layer capacitors

Yijin Cai,Ying Luo,Hanwu Dong,Xiao Zhao,Yong Xiao,Yeru Liang,Hang Hu,Yingliang Liu,Mingtao Zheng

A facile one-step pyrolysis route for the synthesis of hierarchically porous carbon nanosheets (PCNSs) derived from Moringa oleifera stems (MOSs) is reported, in which no post-activation-process in needed. The as-prepared PCNSs possesses unique porous nanosheet morphology with high specific surface area of ca. 2250 m 2  g −1 , large pore volume of ca. 2.3 cm 3  g −1 , appropriate porosity as well as heteroatom doping (N and O), endowing outstanding electrochemical properties as electrode material for high-performance supercapacitors . The PCNS-based electrodes are investigated in various aqueous electrolytes including 1.0 M Na 2 SO 4 , 1.0 M H 2 SO 4 , and 6.0 M KOH. The PCNSs exhibit a maximum specific capacitance of ca. 283 F g −1 (0.5 A g −1 ), excellent rate capability (ca. 72% of capacitance retention even at an ultrahigh current density of 50 A g −1 ), and a tremendous long-term cycling stability in the three-electrode system. Moreover, the as-assembled PCNS-based symmetric supercapacitor shows a high energy density of ca. 25.8 Wh kg −1 (in 1.0 M Na 2 SO 4 electrolyte) and remarkable long-term cycling stability (almost no capacitance fade in aqueous electrolytes), indicating the promising of the as-prepared PCNSs for electrochemical energy storage and conversion .

Preconcentration and Determination of Cadmium, Lead, and Cobalt in Moringa Oleifera (Moringaceae) Using Magnetic Solid-Phase Extraction and Flame Atomic Absorption Spectrometry

Shuangqiao Ju,Mousheng Liu,Yaling Yang

Magnetic solid-phase extraction was used for the preconcentration of cadmium, lead, and cobalt in Moringa Oleifera with determination by flame atomic absorption spectrometry. Fe3O4 nanoparticles were modified by oleic acid to form the sorbent for magnetic solid-phase extraction. The modified sorbent was characterized by transmission electron microscopy, infrared spectroscopy, and X-ray diffraction. The preconcentration was optimized for the mass of nanoparticles, pH, adsorption time, the presence of coexisting ions, solution volume, eluent, and reuse. The optimized limits of detection were 0.09, 0.26, and 0.54 µg/L for Cd(II), Pb(II), and Co(II) with relative standard deviations below 4.3%. The method was employed for the analysis of Moringa oleifera and recoveries between 90.0 and 102.0% were obtained.

Preparation of antioxidant peptides from Moringa oleifera leaves and their protection against oxidative damage in HepG2 cells

Tao Liang,Gu Fan,Liu Yan,Yang Min,Wu Xing-Zhong,Sheng Jun,Tian Yang

Moringa oleifera leaves are a kind of new food raw materials, rich in functional factors, M. oleifera leaves aqueous extract have antioxidant activity and M. oleifera leave protein is an important active ingredient in the aqueous extract. Numerous studies have shown that peptides have strong antioxidant activity. To reveal the antioxidant effects of M. oleifera (MO) leaves peptides, MO leave antioxidant peptides were isolated and prepared to clarify their antioxidant activity. MLPH1 (<1 kDa), MLPH3 (1~3 kDa), MLPH5 (3~5 kDa), and MLPH10 (5~10 kDa) fractions were obtained by the membrane ultrafiltration classification of MO leaves proteolytic hydrolysate (MLPH). MLPH1 was further separated by centrifugal filters, and the fraction separated by <1 kDa (MLPH1-1) was identified and analyzed by LC–MS/MS. The purpose of this study was to investigate the effect of MO leaves antioxidant peptide pretreatment on H2O2-treated HepG2 cells and to refine the antioxidant activity. The results showed that MLPH1 had the strongest antioxidant activity, and three MO leaves antioxidant peptides (LALPVYN, LHIAALVFQ, and FHEEDDAKLF) were obtained. The peptide with the sequence LALPVYN and a molecular weight of 788.44 Da had the strongest antioxidant activity. After 24 h of LALPVYN pretreatment, the cell viability and the CAT, GSH-Px, and SOD enzyme activity were significantly increased, and the MDA, ROS, and apoptosis rates were significantly decreased. These results provide a theoretical basis for further research on the antioxidant mechanism of MO leaves peptides.

Isolation, Purification of Phenolic Glycoside 1 from Moringa oleifera Seeds and Formulation of Its Liposome Delivery System

Shi Feng,Gong Mingjie,Adu-Frimpong Michael,Jiang Xia,Wang Xiaowen,Hua Qinyang,Li Tingyuan,Li Jiaying,Yu Jiangnan,Toreniyazov Elmurat,Cao Xia,Wang Qilong,Xu Ximing

In this study, N, N '-bis {4- [(α-L- rhamnosyloxy) benzyl]} thiourea (PG-1), a phenolic glycoside compound was purified from Moringa seed. The PG-1 has attracted extensive attention due to its anti-cancer, antioxidant, anti-inflammatory and hypoglycemic properties. However, some of its physicochemical properties such as oral bioavailability has not been studied. Herein, a highly purified PG-1 was extracted and incorporated in multiple layered liposomes (PG-1-L) to avoid its burst release and enhance oral bioavailability. After appropriate characterization, it was discovered that the obtained PG-1-L was stable, homogeneous and well dispersed with the average particle size being 89.26 ± 0.23 nm. Importantly, the in vitro release and in vivo oral bioavailability of PG-1-L were significantly improved compared with PG-1. In addition, MTT results showed that compared with the free PG-1, PG-1-L displayed obvious inhibitory effect on the HepG2 cells, while the inhibitory effect on healthy non-malignant 3T6 and LO-2 cells was not significant, indicating that PG-1-L had high safety. In conclusion, PG-1-L can be used as a promising delivery system and an ideal novel approach to improve the oral bioavailability and anticancer activity of PG-1. Graphical Abstract

Microwave-assisted extraction of polysaccharides from Moringa oleifera Lam. leaves: Characterization and hypoglycemic activity

Chun Chen,Bin Zhang,Qiang Huang,Xiong Fu,Rui Hai Liu

Microwave-assisted extraction of Moringa oleifera Lam. leaf polysaccharides was investigated using a response surface methodology. The following conditions were found to generate an experimental yield of 2.96   ±   0.11%, which closely agreed with the predicted value of 2.89%: time 70   min, microwave power 700   W, temperature 70   °C and a liquid/solid ratio of 35   mL/g. After the MLPs were treated with graded precipitation of 40%, 60% and 80% (v/v) ethanol solution, the respective average molecular weight polysaccharide fractions, namely MLP-1 (9.68   ×   10 3   kDa), MLP-2 (6.29   ×   10 3   kDa) and MLP-3 (4.86   ×   10 3   kDa), were obtained. These three fractions were composed of xylose, mannose, glucose, galactose and arabinose but with differing molar ratios. They had similar Fourier transform infrared spectroscopy but their surface morphology differed as determined by scanning electron microscopy. The Congo red assay indicated that the three fractions were not helical polysaccharides. The three polysaccharide fractions exhibited different rheological properties. MLP-1 exhibited pseudoplastic flow behaviors, while MLP-3 displayed near-Newtonian flow behaviors over the entire accessible frequency range. In addition, MLP-3 had stronger inhibitory effects on α-amylase and α-glucosidase than MLP-1 and MLP-2. The results suggest that MLP-3 has potential for the treatment of diabetes.

Comparison of the performance of Moringa oleifera seed protein and polyaluminum chloride in removal of humic acid from water

Jingxi Tie,Zhihong Zheng,Xiaomei Zheng,Yaru Hao

The water-soluble protein was extracted from Moringa oleifera seed press cake and tested its performance as a natural coagulant for removing humic acid (HA) from water solution when comparing to polyaluminum chloride (PAC), a commercial coagulant. Influencing factors including pH, coagulant dosage, temperature and suspended solid were investigated in this study. The results indicated that the coagulating efficiencies of HA using the Moringa oleifera seed protein (MOSP) were better than those by PAC under all the experimental conditions at the same dosage. MOSP could increase the zeta potential more efficiently than PAC. The Fourier transform infrared spectra revealed that no new compounds were generated during the coagulation process. Hence, adsorption and charge neutralization could be the main mechanism contributing to the HA removal using MOSP as coagulant. MOSP is an ideal natural coagulant for HA removal.

Improved protein extraction from Moringa oleifera seeds using deep eutectic solvents: Mechanistic insights and protein characterization

Lang Wang,Tao Fei,Xiang Li,Qin Sun,Xiaoze Liu,Lu Wang

This study employed novel green solvents, deep eutectic solvents (DES), for protein extraction from Moringa oleifera seeds. Two DESs systems including choline chloride-malic acid (ChCl-MA 2 ) and choline chloride-urea (ChCl-U) exhibited the highest protein extraction efficiency, reaching 31.415 % and 31.945 %, respectively. Molecular dynamics (MD) simulation results revealed that the DESs systems showed smaller Root Mean Square Deviation (RMSD), Solvent Accessible Surface Area (SASA), and Radius of Gyration (Rg) values, along with a significant number of hydrogen bonds. The enhanced extraction efficiency of DESs was attributed to the formation of intermolecular hydrogen bonds. Both hydrogen bonding and hydrophobic interactions promoted protein aggregation, thereby stabilizing the protein structure and preserving its integrity. Spectroscopic analyses further indicated that proteins extracted with DESs displayed a more stable and ordered structure. However, DES-extracted proteins exhibited lower surface hydrophobicity (2000–4000) and reduced solubility (40 %–50 %), which significantly compromised their functional properties. Although DES-treated proteins displayed a superior foaming capacity (57.333 %), their emulsifying properties (3.464 m 2 /g and 1.584 m 2 /g, respectively) were comparatively unsatisfactory. In conclusion, DES represent a promising green and efficient extraction method for protein extraction. This work provides valuable insights into DES-mediated protein extraction and offers guidance for the efficient extraction of high-quality proteins from Moringa oleifera seeds.

Siddique Farzana,Ahmad Salman,Hussain Ashiq,Firdous Nida,Nisar Rizwan,Bilal Muhammad,Najam Ayesha,Salik Abdullah,Zia Muhammad,Elkhedir Abdeen Elsiddig

Yoghurt is a one of the commonly consumed nutritious dairy products, which lacks certain bioactive compounds that are present in abundance in many plants. While, a significant range of phytonutrients are present in the leaves of Moringa oleifera . Thus, the purpose of this work is to develop a functional yoghurt, which could promote the health of the consumers to its bioactive components. In the present work, the buffalo milk yoghurt supplemented with Moringa oleifera leaves powder [at 0% (T 0 ), 0.2% (T 1 ), 0.4% (T 2 ), 0.6% (T 3 ) and 0.8% (T 4 )] was prepared to check the variations in the physicochemical, bioactive, and antioxidant properties, during 21 days of refrigerated storage. The results showed that the fat content was decreased from T 0 (4.42%), to T 4 (3.37%), and decreased from 4.40 to 4.38% during storage period of 21 days. The highest protein content was observed in T 4 (4.15%), while the lowest in T 0 (3.61%), and this protein content also deceased during storage. The moisture content was high in T 0 and lowest in T 4 , while increased in all samples during the storage. The pH and total plate count decreased significantly from T 0 to T 4 , whereas with the storage the pH decreased and total plate count was increased. A significant increase in the TPC, TFC and antioxidant activity of yoghurt samples was observed, with increasing the level of incorporation of Moringa oleifera leaves powder, as T 4 presented the highest values for these parameters. However, as storage days passed from 0 to 21, the TPC, TFC and antioxidant activity of all the samples was significantly decreased. Minerals analysis also revealed a significant increase in the Ca, K, Na, Fe and Zn content of the yoghurt, as a result of increasing the level of Moringa oleifera leaves powder, as T 4 was found to be having the highest amounts of these minerals. Current study concludes that Moringa oleifera leaves powder could be an excellent fortification ingredient for the development of functional yoghurt, which due to the presence of bioactive compounds could provide pharmacological benefits.

Effects of Heat Treatment on Physicochemical Properties of Moringa oleifera Lam. Leaf Protein

Chuyu Xi,Wenjie Li,Zhiguo Xu,Jing Xie,Xiaoyu Gao,Dan Feng,Yang Tian,Shuang Song

M. oleiferaleaves represent a novel and nutritious food. Prior research has demonstrated thatM. oleiferaleaves can elicit allergic responses in BALB/c mice. Based on these findings, further studies were conducted to investigate the effects of heat treatment on the allergenicity, particle size, zeta potential, total sulfhydryl (TSH) content, hydrophilicity and hydrophobicity, ultraviolet spectrum, and intrinsic fluorescence spectrum ofM. oleiferaleaf protein. Additionally, in vitro digestion experiments were carried out to gain further insights into the protein’s behavior under these conditions. The experiment simulated the alterations inM. oleiferaleaf protein during the processes of cooking and digestion. The findings of this experiment can provide certain guidance for the processing ofM. oleiferaleaf products. The hydrophilicity, hydrophobicity, transmembrane region, antigen index, calcium binding site, spatial structure, and homology ofM. oleiferaleaf fructose 1,6 bisphosphate aldolase (FBA) were simulated and calculated based on the amino acid sequence of the 36 kDa allergen. These parameters collectively serve to indicate the allergenic activity of the peptide. The findings of the analysis align with the outcomes of the sensitization experiments, suggesting that the FBA ofM. oleiferaleaves is indeed consistent. In conjunction with the heat treatment experiments, this research can inform the preparation ofM. oleiferaleaf foods and provide a foundation for further investigation intoM. oleiferaleaf allergens.
